Description
본 발명은 암 치료의 분야에 관한 것이다.
최근, 진행암(advanced cancer)을 대상으로 한 증식형 바이러스(replicative virus)에 의한 바이러스 요법(virotherapy)의 임상연구가 진행되고 있다. 바이러스 요법이란, HSV-1이나 아데노바이러스(adenovirus) 등의 증식형 바이러스를 종양세포에 감염시켜, 바이러스 증식에 수반되는 바이러스의 살세포 효과(cell-killing effect)에 의해 종양의 치유를 도모하는 치료법이다. HSV-1이나 아데노바이러스의 경우, 종양 치료용 증식형 바이러스는, 바이러스 게놈(viral genome)에 유전자 조작을 가한 변이 바이러스로, 종양 내에서의 복제능을 유지하면서, 인간 정상조직에서의 병원성이 최소한으로 억제되어 있다. 종양세포에 감염된 치료용 증식형 바이러스는, 세포 내에서 복제되고, 그 과정에서 감염세포는 사멸한다. 증식된 바이러스는 주위의 종양세포에 재감염되어, 항종양작용을 확산시킨다(Alemany R. et al., Replicative adenoviruses for cancer therapy. Nat Med 1:938-943, 1995). 암에 대한 바이러스 요법은, 수술·방사선요법·화학요법이라고 하는 종래 치료법과의 병용이 가능하고, 고형암 일반에 적용이 가능한 것, 반복투여가 가능한 것, 사이토카인 등의 치료 유전자를 바이러스 게놈에 직접 삽입하여 항종양 효과를 증강시킬 수 있는 등, 그의 응용범위가 넓어, 실용면에서 우수하다. 보다 효과적인 바이러스 요법을 개발할 수 있다면, 암 치료에 크게 공헌할 수 있을 것으로 기대된다.

발명의 개시
발명이 해결하고자 하는 과제
본 발명은, RNA 바이러스가 도입된 수상세포(dendritic cell)를 포함하는 항암제를 제공한다. 또한 본 발명은, RNA 바이러스가 도입된 수상세포를 제조하는 공정을 포함하는, 항암제의 제조방법을 제공한다. 또한 본 발명은, RNA 바이러스가 도입된 수상세포를 사용한 암의 치료방법을 제공한다.
과제를 해결하기 위한 수단
본 발명자 등은, 게놈 복제능을 가지는 RNA 바이러스를 수상세포에 도입하면, 수상세포가 활성화되어, 우수한 제암효과(anticancer effect)를 발휘하는 것을 발견하였다. 수상세포를 매개로 하여 RNA 바이러스를 암에 주입한 경우의 암 증식에 대한 억제효과는, RNA 바이러스를 직접 암에 주사하는 것보다도 유의하게(significantly) 높았다. 수상세포에 RNA 바이러스를 도입하는 공정은 체외에서 행할 수 있기 때문에, 종래의 바이러스 요법에 비하여 바이러스 도입조건을 엄밀하게 제어하는 것이 가능하고, 또한, 수상세포에 감염되지 않은 바이러스를 제거함으로써 안전성을 높이는 것도 가능하다. 또한, RNA 바이러스를 도입한 수상세포에 의한 제암효과는, 감염성 바이러스 입자를 방출하지 않는 결손형 바이러스(defective virus)를 사용한 경우에도 동일하게 발견되었다. 즉, RNA 바이러스를 도입한 수상세포에 의한 제암효과는, RNA 바이러스가 수상세포 내에서 게놈 RNA를 복제하는 것이 중요하고, 감염성 바이러스 입자를 방출하여 주위의 세포에 감염을 반드시 확산시킬 필요는 없다. 따라서, 예를 들면 바이러스 엔벨로프 단백질(viral envelope protein) 등의, 감염 바이러스 입자의 형성에 필수의 단백질을 코드하는 바이러스 유전자를 결손시킴으로써 감염성 바이러스 입자의 형성능을 없앤 안전성이 높은 RNA 바이러스를 사용하여, 바이러스 요법을 행하는 것도 가능해진다.
즉 본 발명은, RNA 바이러스가 도입된 수상세포를 포함하는 항암제, 상기 항암제의 제조방법, 및 RNA 바이러스가 도입된 수상세포를 사용한 암의 억제방법에 관한 것이고, 보다 구체적으로는 청구항의 각 항에 기재된 발명에 관한 것이다. 또한 동일한 청구항을 인용하는 청구항에 기재된 발명의 하나 또는 복수의 조합으로 되는 발명은, 그들의 청구항에 기재된 발명에 이미 의도되어 있다. 즉 본 발명은,
[1] 게놈 복제능을 갖는 RNA 바이러스가 도입된 수상세포를 포함하는 항암제,
[2] [1]에 있어서, RNA 바이러스가 외래 단백질을 코드하지 않는 항암제,

도면의 간단한 설명
도 1은, 말초혈 단구 농축세포(monocyte-enriched peripheral blood cell)의 단핵 세포(mononuclear cell)에 유래하는 수상세포의 표현형을 나타내는 도면이다. PI로 판별할 수 있었던 생세포(viable cell)에 게이트(gate)를 걸고, 항CD11c-PE 결합항체 및 항HLA-class Ⅱ(DR, DP, DQ) FITC 결합항체를 사용하여, CD11c 및 HLA-class Ⅱ(DR, DP, DQ)의 발현을 관찰하였다(왼쪽 매트릭스(matrix)). 추가로 CD11c 및 HLA-class Ⅱ(DR, DP, DQ)가 모두 양성인 게이트를 선택하여, 1) 항CD14-APC 결합항체, 2) 항CD1a-APC 결합항체, 3) 항CD80-biotin 결합항체(2차적으로 스트렙트아비딘(streptavidin)-APC로 염색)를 사용하여, 각각의 발현 레벨을 CD11c와의 도트 플롯(dot plot)으로 나타내었다(오른쪽 3개의 매트릭스). 또한, 실시예 중의 "Class Ⅱ"는 HLA-DR, DQ, DP를 모두 인식하는 항체를 사용한 결과를, "HLA-DR"은 HLA-DR을 특이적으로 인식하는 항체를 사용한 결과를 나타낸다.

발명을 실시하기
위한 최선의 형태
본 발명은, 게놈 복제능을 가지는 RNA 바이러스가 도입된 수상세포를 포함하는 항암제를 제공한다. 본 발명에 있어서 RNA 바이러스란, RNA 게놈을 갖는 바이러스를 말한다. 본 발명에 있어서 RNA 바이러스는, 바람직하게는 바이러스의 라이프 사이클(life cycle)에 있어서 RNA를 주형(template)에 RNA 합성을 행하는 바이러스이다. RNA 바이러스로서는, 수상세포에 있어서 게놈 RNA를 복제하는 목적의 RNA 바이러스여도 되고, 야생형 바이러스여도, 약독형 바이러스(attenuated virus)나 온도 감수성 바이러스(temperature-sensitive virus) 등의 변이형 바이러스여도 된다. 또한, 천연 바이러스(자연발생한 바이러스)여도, 재조합 바이러스여도 된다. RNA 바이러스에는, 단일 가닥 RNA 바이러스(single-stranded RNA virus)(플러스 가닥 RNA 바이러스(plus strand RNA virus) 및 마이너스 가닥 RNA 바이러스(minus strnad RNA virus)를 포함한다), 및 이중 가닥 RNA 바이러스(double-stranded RNA virus)를 포함한다. 또한 엔벨로프를 갖는 바이러스(엔벨로프 바이러스; enveloped viruses) 및 엔벨로프를 갖지 않는 바이러스(비엔벨로프 바이러스; non-enveloped viruses)를 포함하지만, 바람직하게는 엔벨로프 바이러스가 사용된다. 본 발명에 있어서 RNA 바이러스에는, 구체적으로는 이하의 과(family)에 속하는 바이러스가 포함된다.

본 발명에 있어서, 게놈 복제능을 가지는 RNA 바이러스가 도입된 수상세포란, 게놈 복제능을 가지는 RNA 바이러스의 게놈 RNA를 포함하는 수상세포로서, 상기 RNA가 코드하는 바이러스 단백질에 의해, 상기 RNA가 수상세포 내에서 복제되는 세포를 말한다. RNA 바이러스의 게놈 RNA와, 상기 RNA에 결합하는 바이러스 단백질은, 세포 내에서 리보뉴클레오프로테인(ribonucleoprotein)(RNP) 복합체를 형성하여, 세포 내에서 상기 게놈 RNA를 복제한다. 이 RNP는 뉴클레오캡시드(nucleocapside)라고도 불리운다. 즉, 본 발명에 있어서 게놈 복제능을 가지는 RNA 바이러스가 도입된 수상세포는, 게놈 복제능을 가지는 RNA 바이러스의 리보뉴클레오프로테인(뉴클레오캡시드)을 갖는 수상세포를 의미한다.
RNA 바이러스가 도입된 수상세포를 얻기 위해서는, RNA 바이러스를 수상세포 또는 그의 전구세포(precursor cell)에, 감염성 RNA 바이러스 입자를 접촉시켜 감염시키면 된다. 또는, 감염성 바이러스 입자를 사용하지 않아도, 게놈 복제능을 가지는 RNA 바이러스에 포함되는 RNP를 세포에 도입하거나, 또는 감염성이 없는 바이러스 입자로서, 상기 RNP를 포함하는 바이러스 입자(비감염성 바이러스 입자, 또는 바이러스 유사 입자(virus-like particle)(VLP)라고 불리운다)를 세포에 도입해도 된다. 바이러스 입자로부터 엔벨로프 또는 피막(coat)을 제거한 RNP(바이러스 코어(viral core))일지라도, 수상세포에 도입되면, 세포 내에 있어서 바이러스 게놈 RNA를 복제할 수 있다(WO97/16538; WO00/700550). 또한, 바이러스 게놈 RNA 및 상기 게놈 RNA의 복제에 필요한 바이러스 단백질(마이너스 가닥 RNA 바이러스인 경우 N, P 및 L 단백질)을 코드하는 발현 벡터를 수상세포에 도입하여, 수상세포 내에서 RNP를 형성시켜도 된다. Biol. Chem.(1997) 272, 16578-16584). 따라서, 엔벨로프가 제거된 RNP를 수상세포에 도입하는 것에는 이점이 있다(WO 00/70055).
RNA 바이러스가 도입되면, 수상세포 내에서 바이러스 게놈의 복제가 일어나, 수상세포의 활성화가 유도되어 성숙 수상세포로 분화된다. 얻어진 성숙 수상세포는, T세포를 활성화하는 능력을 보유하고 있다. RNA 바이러스를 도입한 수상세포는 면역계를 활성화하는 높은 능력을 가지고 있어, 종양에 투여함으로써 제암작용(anticancer effect)을 발휘할 수 있다. RNA 바이러스의 수상세포로의 감염은, 예를 들면 배양액 또는 생리식염수 등의 목적의 생리적 수용액 중에서 in vitro(또는 ex vivo)로 행할 수 있다. 본 발명은, 체내로부터 꺼낸 수상세포 또는 그의 전구세포를 체외에서 RNA 바이러스와 접촉시켜, 바이러스를 도입 후에 체내에 되돌리는 ex vivo의 항종양 치료에 있어서 유용하다. RNA 바이러스를 체외에서 감염시키는 경우는, RNA 바이러스를 미성숙 수상세포에 접촉, 또는 미성숙 수상세포를 포함하는 세포 분획(cell fraction)과 혼합하는 것이 바람직하다. 수상세포는 RNA 바이러스를 도입함으로써 활성화할 수 있지만, 그것과는 별도로, 세균 또는 리포폴리사카라이드(lipopolysaccharide)(LPS) 등과의 접촉에 의해서도 활성화할 수 있다. 수상세포를 이와 같은 방법으로 별도 활성화시키는 경우는, 활성화한 후 RNA 바이러스를 도입해도 되지만, 바이러스의 도입효율이 저하되지 않도록 하기 위해, 활성화의 조작을, 바이러스의 도입 전이 아니라, 바이러스를 도입한 후(또는 바이러스를 수상세포에 접촉시키는 것과 동시)에 행하는 것이 바람직하다.
바이러스와 수상세포 또는 그의 전구세포와의 접촉에 있어서는, MOI(다중감염도(multiplicity); 세포 한 개당 감염 바이러스수)는 1~500 사이로 하는 것이 바람직하고, 더욱 바람직하게는 2~300, 더욱 바람직하게는 3~200, 더욱 바람직하게는 5~100, 보다 바람직하게는 7~70이다. RNA 바이러스와 수상세포와의 접촉은 짧은 시간으로도 충분하여, 예를 들면 1분 이상, 바람직하게는 3분 이상, 5분 이상, 10분 이상, 또는 20분 이상 접촉시키면 되고, 예를 들면 1~60분 정도, 보다 특정하면 5분~30분 정도이면 된다. 물론, 그 이상의 시간 접촉시켜도 되고, 예를 들면 수일간 또는 그 이상 접촉시켜도 된다.
수상세포(Dendritic cell; DC)란, 성숙 상태에 있어서 수지상 형태(dendritic morphology)를 취하고, 항원을 제시하여 T세포를 활성화시키는 능력을 가지는 세포이다. 수상세포에는, 생체내 각종 조직기관에 분포하는 골수세포 유래의 수지상 형태를 취하는 세포군, 및 골수 또는 혈액 유래의 줄기세포(stem cell)로부터, in vitro에서 사이토카인 등을 이용하여 분화 유도를 가한 생체 내 조직기관에 분포하는 수지상 형태를 취하는 세포와 동등한 세포군이 포함된다. 구체적으로는, 수상세포에는, 예를 들면 림프구계 수상세포(Th2로의 유도 또는 면역 관용(immune tolerance)을 유도하는 것이어도 된다), 골수구계 수상세포(일반적으로 사용되는 수상세포. 미숙 수상세포 및 성숙 수상세포를 포함한다), 랑게르한스 세포(Langerhans cell)(피부의 항원제시세포에서 중요한 수상세포), 상호 연결세포(림프절, 비장의 T세포 영역에 있고, T세포로의 항원제시에 작용하고 있는 것으로 생각되고 있는 세포), 여포 수상세포(follicular dendritic cell)(B세포로의 항원제시세포로서 중요, 항원과 항체 복합체, 항원과 보체 복합체(complement complex)를 항체 리셉터(antibody receptor), 보체 리셉터에 의해, 수상세포상에 제시함으로써, B세포에 항원제시하고 있다.) 등을 포함하는 것으로, 바람직하게는, MHC 클래스 Ⅰ 및 클래스 Ⅱ를 고발현(高發現)하고 있고, 더욱 바람직하게는 CD11c를 발현하고 있는 세포이다.
또한, 수상세포는, 수지상 형태를 가지고, CD11c, HLA-class Ⅱ(HLA-DR, -DP, 또는 -DQ), CD40, 및 CD1a로 이루어진 군으로부터 선택되는 표면 마커의 두개 이상이 양성 세포여도 된다. 본 발명에 있어서 수상세포는, 보다 바람직하게는, HLA-class Ⅱ+ 및 CD11c+의 세포, 보다 바람직하게는 CD1a+, HLA-class Ⅱ+, 및 CD11c+의 세포이고, 또한 T세포 마커(CD3), B세포 마커(CD19, CD20), NK세포 마커(CD56), 호중구 마커(neutrophil marker)(CD15), 단구 마커(CD14)를 발현하고 있지 않은 세포이다. RNA 바이러스의 도입에 사용되는 수상세포 집단의 CD14+의 비율은, 예를 들면 10% 이하, 바람직하게는 5% 이하, 더욱 바람직하게는 1% 이하가 좋다.
또한, 본 발명에 있어서 수상세포에는 성숙 수상세포 및 미성숙 수상세포가 포함된다. 미성숙 수상세포란, T세포 활성화능력이 낮은 수상세포를 말한다. 구체적으로는, 미성숙 수상세포는, LPS(1 micro-g/㎖)를 첨가하여 2일간 배양하고 성숙을 유도한 수상세포에 비하여, 항원제시능이 1/2 미만, 바람직하게는 1/4 미만인 것이면 된다. 항원제시능은, 예를 들면, 알로 T세포 활성화능(혼합 림프구 시험; 알로 T세포와 수상세포의 혼합 배양으로, T세포 대 수상세포의 비율은 1:10으로 배양, 바람직하게는 그 비율을 변화시켜서 배양한 것으로, 배양종료 8시간 전에 3H-thymidine을 첨가하여, 그 T세포의 DNA 내의 흡수(incorporation)량에 의해, T세포 증식능력을 정량한 것: 도 21 및 도 22 참조; 문헌 Gene Therapy 2000; 7; 249-254), 또는 펩티드를 사용한 특이적 세포상해성 T세포(CTL)의 유도능시험(수상세포에, 어느 항원의 어느 Class Ⅰ 구속성의 기지의 펩티드를 첨가하여, 수상세포를 채취한 것과 동일한 건강한 정상인 말초혈의 T세포를 공배양(3일째 이후는 IL-2를 25 U/㎖, 바람직하게는 100 U/㎖)(바람직하게는 21일간 3회의 수상세포에 의한 자극, 더욱 바람직하게는 14일간 2회의 수상세포의 자극)하여, 얻어진 이펙터 세포(effector cell) 를 51Cr로 라벨된 타겟 세포(펩티드의 구속성 Class Ⅰ 양성의 종양세포)와 20:1, 10:1, 5:1, 및 2.5:1, 바람직하게는 100:1, 50:1, 25:1, 및 12.5:1로 4시간 혼합 배양하고 타겟 세포의 51Cr 유출량(遊出量)으로 정량한 것; 도 23 참조; 문헌 Arch Dermatol Res 2000; 292; 325-332)에 의해 정량할 수 있다. 또한 미성숙 수상세포는, 바람직하게는 항원 식세포능을 가지고, 더욱 바람직하게는 T세포 활성화를 위한 보조자극을 유도하는 수용체의 발현이 저발현(예를 들면 상기와 같이 LPS 유도한 성숙 DC에 비하여 유의하게) 또는 음성이다. 한편으로, 성숙 수상세포란 T세포 활성화 등을 위한 항원제시능력이 높은 수상세포를 말한다. 구체적으로는, 성숙 수상세포는, LPS(1 micro-g/㎖)를 첨가하고 2일간 배양하여 성숙을 유도한 수상세포의 항원제시능의 1/2 이상, 바람직하게는 동등 이상의 항원제시능을 가지는 세포여도 된다. 또한 성숙 수상세포는, 바람직하게는 항원 식세포능이 낮거나, 또는 갖지 않고, 더욱 바람직하게는 T세포 활성화를 위한 보조자극을 유도하는 수용체의 발현이 높은 것을 말한다. 또한, 수상세포의 활성화란, 미성숙 수상세포로부터 성숙 수상세포로의 이행을 말하고, 활성화 수상세포에는 성숙 수상세포 및 활성화 자극에 의해 보조자극을 유도하는 CD80, CD86의 발현을 상승시키고 있는 도중 경과의 수상세포가, 그의 범주에 포함된다. CD11c 양성의 수상세포에 있어서는, CD83 양성이 성숙 수상세포의 지표가 된다.
예를 들면, 성숙 수상세포는, 바람직하게는 CD40, CD80, CD86 및 HLA-class Ⅱ의 발현이 강하게 양성인(strongly positive) 세포이면 된다. 더욱 바람직하게는, 성숙 수상세포는 CD83를 발현한다. 예를 들면, CD80, CD83 , 및 CD86로 이루어진 군으로부터 선택되는 마커를 토대로, 미성숙 수상세포와 성숙 수상세포를 분별할 수 있다. 미성숙 수상세포에서는 이들의 마커는 약하거나, 바람직하게는 음성이지만, 성숙 수상세포에서는 양성이다.
상기와 같이, 미성숙 수상세포는, 통상, 높은 식세포능을 보유하고 있다. 수상세포에 LPS(1 micro-g/㎖)를 첨가하여 2일간 배양하면, 수상세포는 활성화되어 식세포능은 저하된다. 식세포능은, 수상세포 내로의 소분자의 흡수량 또는 흡수 세포(uptaking cell)의 비율을 측정하여 알 수 있다. 바람직하게는, 식세포능은, 수상세포 내로의 소분자의 흡수량으로 결정된다. 예를 들면, 1 micro-m 정도의 착색 비즈(colored beads)를 사용하여, 수상세포 내로의 비즈의 흡수를 측정할 수 있다. 4℃에서 양성이 되는 백그라운드(background)를 빼고 정량한다. 높은 식세포능이란, 수상세포 내로의 소분자의 흡수량이, 수상세포를 상기와 같이 LPS(1 micro-g/㎖)로 2일간 자극한 수상세포의 4배 이상, 보다 바람직하게는 5배 이상, 보다 바람직하게는 6배 이상의 식세포능을 말한다. 또는, 소분자의 흡수 세포의 비율이 2배 이상, 보다 바람직하게는 3배 이상이다. 낮은 식세포능이란, 수상세포 내로의 소분자의 흡수량이, LPS(1 micro-g/㎖)로 2일간 자극한 수상세포의 4배 미만, 보다 바람직하게는 2배 미만, 보다 바람직하게는 1.5배 미만이다. 또한, 소분자의 흡수 세포의 비율로 측정한 경우에, 2배 미만, 보다 바람직하게는 1.5배 미만이다.
성숙 수상세포의 판별은 당업자가 통상 행하고 있고, 상기의 각 마커 및 그의 발현의 측정방법도 당업자에게 주지이다. 예를 들면, CD11c는 약 150 kD의 접착 당단백(p150, 인테그린 알파 사슬(integrin alpha chain))이다. Med., 1998, 187: 1019-1028; Blood, 1996, 87: 4520-4530)가 제거되지 않고 남아, 접착성 CD14 세포로부터 분화된 DC 뿐만 아니라, 그들의 전구체(precursor)로부터 분화된 DC를 함께 회수하는 것이 가능하다고 생각되어진다. 이에 따라, 세포상해성을 경감하는 것을 기대할 수 있다.
예를 들면, T세포, NK세포, B세포 등에 특이적인 항체를 사용하여, 이들의 세포를 제거함으로써, 수상세포를 농축하는 것이 가능하다. 구체적으로는, 예를 들면, CD2, CD3, CD8, CD19, CD56, CD66b로부터 선택되는 표면 마커 또는 그 임의의 조합의 발현이 낮거나 또는 음성의 세포(low or negative cell)를 얻는 것이 바람직하다. 보다 바람직하게는, CD2, CD3, CD8, CD19, CD56, 및 CD66b의 전부가 낮거나 또는 음성의 세포이다. 그 때문에, 이들의 마커에 대한 항체를 사용하여, 이들의 마커를 발현하는 세포를 제거하면 된다(Hsu 등(1996) Nature Med. 2: 52). 또한, 음성 선택에 있어서는, 본 실시예에 나타내어지고 있는 것과 같은 다가항체(polyvalent antibody)를 사용하여 행할 수 있고, 또는 자기 세포 분리(magnetic cell seperation)(MACS)를 위해 비즈 등을 사용해도, 동일한 셀렉션을 실시하는 것이 가능하다. 혈구 분리 등을 사용하여 단핵구를 채취하는 등, 세포를 대량으로 조정하는 경우는 비즈를 사용하는 것이 바람직하다. 예를 들면, 생체로부터 얻은 세포용액으로부터 단구를 농축(enrich)하고, 이에 대하여 음성 선택을 행하여 조제한 수상세포를 본 발명에 있어서 적합하게 사용할 수 있다.
또한, RNA 바이러스를 도입하기 전에 접착세포에 의해 얻어진 말초혈 단구를 수상세포로 분화시킨 것을 선택하면, 바이러스의 도입효율이 저하되는 경우가 있다. 본 발명에서 사용되는 수상세포는 이것에 한정되는 것은 아니지만, 미성숙 수상세포의 비율을 저하시키지 않도록, RNA 바이러스를 접촉시키기 전의 세포배양은, 고체 지지체(solid support)(예를 들면, 배양 접시 또는 병(bottle) 등의 배양용기)에 접착된 세포를 선택하는 공정을 포함하지 않는 것이 바람직하다. 즉, 본 발명은, RNA 바이러스와 수상세포의 접촉 전 24시간 이내에, 고체 지지체에 부착된 세포를 선택하는 공정을 포함하지 않는 방법을 제공한다. 보다 바람직하게는, RNA 바이러스와 수상세포와의 접촉 전 2일, 3일, 5일, 또는 7일 이내에, 고체 지지체에 부착된 세포를 선택하는 공정을 포함하지 않도록 하면 된다.
또한, 이것에 한정되는 것은 아니지만, RNA 바이러스를 접촉시키기 전에, CD14+ 세포를 선택하는 공정을 포함하지 않는 것이 바람직하다. 즉, 본 발명은, RNA 바이러스와 수상세포의 접촉 전 24시간 이내에, CD14+ 세포를 선택하는 공정을 포함하지 않는 방법을 제공한다. 보다 바람직하게는, RNA 바이러스와 수상세포와의 접촉 전 2일, 3일, 5일, 또는 7일 이내에, CD14+ 세포를 선택하는 공정을 포함하지 않도록 하면 된다.

본 발명에 있어서는, 수상세포 또는 그의 전구세포(예를 들면, CD11c+ 세포 또는 CD34+ 세포)를 고농도로 포함하는 세포 분획에 RNA 바이러스를 혼합하는 것이 바람직하다. 전구세포란, 적당한 사이토카인(즉 G-CSF, GM-CSF, TNF-alpha, IL-4, IL-13, SCF, Flt-3 리간드, c-kit 리간드, 또는 그들의 조합)의 존재하에서 수상세포로 분화시킬 수 있는 세포를 말하고, 바람직하게는 4주일 이내, 보다 바람직하게는 20일 이내, 보다 바람직하게는 18일 이내, 보다 바람직하게는 16일 이내에 수상세포로 분화시킬 수 있는 세포이다. 이와 같은 세포에는, CD34+ 줄기세포를 들 수 있다. 수상세포로의 분화는, 예를 들면, SCF(50 ng/㎖), GM-CSF(500 U/㎖), TNF-alpha(50 ng/㎖)의 존재하에서 3일 정도 배양 후, SCF(50 ng/㎖), GM-CSF(500 U/㎖), IL-4(250 U/㎖), TNF-alpha(50 ng/㎖)의 존재하에서 배양함으로써 실시하면 된다. 또한 세포 분획이란, 세포의 분리(또는 분획)에 의해 얻어진 세포 집단이다. 세포 분획은, 세포 및 약학적으로 허용되는 담체를 포함하는 조성물이어도 된다. 담체로서는, 생리식염수, 인산 완충 생리식염수(PBS), 배양액, 혈청 등, 생세포를 현탁할 수 있는 목적의 용액을 들 수 있다. 본 발명의 방법에 있어서 RNA 바이러스의 접촉에 사용되는 세포 분획은, 전체 생세포 중의 수상세포 및/또는 그의 전구체의 비율이, 예를 들면 30% 이상, 바람직하게는 40% 이상, 바람직하게는 50% 이상, 바람직하게는 60% 이상, 바람직하게는 70% 이상이다.
또한, RNA 바이러스에 접촉시키는 수상세포 중에는, 미성숙 수상세포가 포함되는 것이 바람직하다. RNA 바이러스를 혼합하는 수상세포를 포함하는 세포 분획은, 전체 생세포 중의 미성숙 수상세포의 비율이, 예를 들면, 10% 이상, 바람직하게는 20% 이상, 보다 바람직하게는 30% 이상, 보다 바람직하게는 40% 이상, 보다 바람직하게는 50% 이상, 보다 바람직하게는 60% 이상, 보다 바람직하게는 70% 이상이다.
RNA 바이러스와 수상세포를 조합시킨 항암제는 우수한 성질을 가지고 있다. 예를 들면 RNA 바이러스를 사용하면, 바이러스의 감염만으로 활성화 수상세포가 얻어져, 나중의 성숙 수상세포를 얻기 위한 공정을 생략할 수 있다. 수상세포를 면역 부활에 사용하기 위해서는 활성화가 필요하기 때문에, 바이러스의 감염만으로 활성화를 일으키는 것에는 이점이 있다. 또한, 이 성질을 이용하여, in vitro에서 T세포 이입요법에 필요한 활성화 T세포, 특히 세포상해성 T세포 등을 효율적으로 단기간에 유도할 수 있다. 바이러스를 도입하지 않은 수상세포로는 CTL을 유도할 수 없고, 다른 바이러스 벡터에서 지금까지 보고되어 있는 성질로부터, 다른 바이러스 벡터의 유전자 도입만으로는 in vitro에서의 CTL 유도는 곤란하다. 따라서, RNA 바이러스는, 바이러스의 도입만으로 T세포를 활성화(CTL 유도)가 가능하다고 하는 이점을 가지고 있다(도 21~23 참조).
본 발명의 항암제의 제조에 있어서는, RNA 바이러스를 줄기세포에 유전자 도입한 후에, 수상세포를 분화시켜도 된다. 예를 들면 센다이 바이러스를 줄기세포에 유전자 도입한 후에, 수상세포로의 분화를 유도한 경우, 유전자 도입효율은 70% 가까이에 달한다. 이것은, 레트로바이러스 및 렌티바이러스 벡터(retroviral vector and lentivirus vector)의 개변형에 필적한다. 아데노바이러스 벡터는, 도입 후의 에피솜(episome)의 희석에 의해 발현이 저하되기 때문에, 줄기세포부터 유전자 도입하는 것은 곤란하다. 게놈 복제형 RNA 바이러스가 도입된 수상세포의 조제는, 줄기세포에 도입 후에 수상세포 분화를 행하는 방법, 및 말초혈 단핵구로부터 분화시킨 수상세포에 유전자를 도입하는 방법 중 어느 방법에 의해서도 가능하다.
또한, RNA 바이러스를 감염시킬 때에 MOI를 높게(예를 들면, 10 이상, 바람직하게는 20 이상, 보다 바람직하게는 30 이상, 예를 들면 40 이상, 더 나아가서는 50 이상)하면, 세포상해성에 현저한 영향 없이, 안정되게 거의 100%의 도입효율로 세포에 도입하는 것을 기대할 수 있다. 더욱이, 숙주 염색체에 게놈을 삽입하지 않는 RNA 바이러스를 사용하면, 숙주 게놈 개변에 기인하는 종양 발생 등의 리스크가 낮다고 하는 이점도 가지고 있다. 이 점에서, 레트로바이러스 이외의 RNA 바이러스가 적합하게 사용된다.
RNA 바이러스로서는, 재조합 바이러스가 아니어도, 천연의 바이러스로도 사용하는 것이 가능하고, 각 RNA 바이러스의 정제방법, 증식방법, 단리주의 취득방법에 대해서는, 바이러스학 실험학 각론, 개정 2판(국립예방위생연구소 학우회편, 마루젠, 1982)을 참조할 수 있다. 예를 들면, 파라믹소바이러스과(Paramyxoviridae)의 센다이 바이러스 등 파라인플루엔자 바이러스는, 각 형 모두 원숭이 신장(MK2), 인간 태아 폐(human fetal lung), 신장, 양막(amnion)의 초대 배양세포, 및 트립신 첨가 Vero 세포에서 잘 증식하여(상기와 동일, p334; Itoh H et al., Jap. J. Med. Sci. Biol. 23, 227 (1970)) 회수할 수 있다. 정제는, 수크로오스 밀도 구배원심법(sucrose density gradient ultra centrifugation), 평형원심법(equilibrium centrifugation) 등으로 정제할 수 있다(p336). 홍역 바이러스는, 원숭이 유래의 각종 세포에서 잘 증식 가능하여(Matsumoto M, Bact. Rev. 30, 152(1966)) Vero 세포가 가장 범용되고 있지만, CV1, FL, KB, HeLa HEp2 등을 사용하여 증식 가능하다(p351). 광견병 바이러스 등의 라브도바이러스과(Rhabdoviridae)에서는 조직배양법을 사용하여, BHK, CE 및 Vero 세포 등이 사용되고 있다. 정제법으로서는, 감염 3~4일의 배양액을 pH 7.4 이상으로 보정하고, 저속 원심으로 세포편(cell debris)을 제거하고 농축한다(p376). 라싸 바이러스(lassa virus) 등의 아레나바이러스과(Arenaviridae)는, in vitro로 계대되어 있는 거의 모든 배양세포에서 잘 증식하지만, HK-21/13S 세포에 감염시킨 후, 한천(agar) 중에 현탁한 형태로 배양함으로써 증식이 가능하다(Sedwik W. D., J. Virol. 1, 1224 (1967))(p240). 풍진 바이러스 등의 토가바이러스과(Togaviridae)는, 초대 아프리카 녹색 원숭이(primary African green monkey kidney)(GMK)세포, Vero, BHK21, RK13, 초대 메추라기(primary quail) 또는 닭 배세포, R66, 및 SIRC 세포 등 광범위에 걸친 각종 배양세포 중에서 증식한다. 비교적 수량(yield)이 높은 바이러스를 얻기 위해서는 BHK21 또는 Vero 세포가 자주 사용된다(p227). 인플루엔자 바이러스 등의 오르토믹소바이러스과(Orthomyxoviridae)는, 발육 계란이나 MDCK 세포에 있어서 증식할 수 있다(p295). 정제는 원심분획법, 적혈구로의 흡착·유출(release)에 의한 정제(Laver W. G., Fundamental Techniques in Virology, 82(1969)) 등으로 정제할 수 있다(p317).
RNA 바이러스는, 천연으로부터 단리된 바이러스여도, 유전자 재조합에 의해 인위적으로 생성된(generate) 바이러스여도 된다. 또한, 감염세포에 있어서 게놈 RNA를 복제하는 능력을 보유하는 한, 야생형 바이러스가 갖는 어느 하나의 바이러스 유전자에 변이 및/또는 결손이 있는 것이어도 된다. 예를 들면, 바이러스의 엔벨로프 단백질 또는 피막 단백질(coat protein)을 코드하는 적어도 하나의 유전자에 변이 또는 결손을 갖는 바이러스를 적합하게 사용할 수 있다. 이와 같은 바이러스는, 감염세포에 있어서는 RNA 게놈을 복제할 수는 있지만, 감염성 바이러스 입자를 형성할 수 없다. 따라서, 주위에 감염을 확대할 우려가 없기 때문에 안전성이 높다. 예를 들면, 마이너스 가닥 RNA 바이러스에 있어서는, F, H, HN, 또는 G 등의 엔벨로프 단백질 또는 스파이크 단백질(spike protein)을 코드하는 적어도 하나의 유전자, 또는 그들의 조합이 포함되어 있지 않은 바이러스를 사용할 수 있다(WO00/70055 및 WO00/70070; Li, H.-O. et al., J. Virol. 74(14) 6564-6569(2000)). 게놈 복제에 필요한 단백질(예를 들면, N, P 및 L 단백질)을 게놈 RNA에 코드하고 있는 경우, 감염세포에 있어서 게놈을 증폭시킬 수 있다. 즉, 적어도 N, L 및 P 단백질을 코드하는 게놈 RNA와 그들의 단백질을 포함하는 RNP, 및 상기 RNP를 포함하는 바이러스 입자는, 본 발명의 항종양제의 제조에 있어서 수상세포에 도입되는 것으로서 적당하다. 결손형 바이러스를 제조하기 위해서는, 예를 들면, 결손되어 있는 유전자 산물 또는 그것을 상보(相補)할 수 있는 단백질을 바이러스 생산세포에 있어서 외래적으로 공급한다(WO00/70055 및 WO00/70070; Li, H.-O. et al., J. Virol. 74(14) 6564-6569(2000)). 단, 예를 들면 마이너스 가닥 RNA 바이러스에 있어서는, M 단백질 유전자를 갖는 바이러스인 경우, F 단백질이나 HN 단백질 등의 스파이크 단백질을 코드하는 유전자를 갖지 않아도 비감염성 바이러스 입자(VLP)가 방출되기 때문에, 바이러스 단백질을 상보하지 않고 VLP를 제조하는 것이 가능하다(WO00/70070). 또한, 엔벨로프 단백질 유전자를 아무것도 갖지 않아도, N, L, P 단백질, 및 게놈 RNA로 되는 RNP는 세포 내에 있어서 증폭할 수 있기 때문에, 세포 용해물(cell lysate)로부터 원심분리 등에 의해 RNP를 회수할 수 있다. 얻어진 VLP나 RNP는, 목적의 트랜스펙션 시약과 혼합하여 수상세포에 도입함으로써, 항종양제를 제조할 수 있다.
또한, 변이형 RNA 바이러스를 사용하여 본 발명의 항종양제를 제조하는 것도 가능하다. 예를 들면, 엔벨로프 단백질이나 피막 단백질에 있어서 다수의 온도 감수성 변이가 알려져 있다. 이들의 온도 감수성 변이 단백질 유전자를 갖는 RNA 바이러스를 본 발명에 있어서 적합하게 사용할 수 있다. 온도 감수성 변이란, 저온(예를 들면 30℃ 내지 32℃)에 비하여, 바이러스 숙주의 통상의 온도(예를 들면 37℃ 내지 38℃)에 있어서 유의하게 활성이 저하되는 변이를 말한다. 이와 같은, 온도 감수성 변이를 갖는 단백질은, 허용 온도(저온)하에서 바이러스를 제작할 수 있기 때문에 유용하다.
예를 들면, 마이너스 가닥 RNA 바이러스의 M 유전자의 온도 감수성 변이로서는, 센다이 바이러스의 M 단백질에 있어서의 G69, T116, 및 A183로 이루어진 군으로부터 임의로 선택되는 부위의 아미노산 치환을 들 수 있다(Inoue, M. et al., J. G.(1992) Proc. Natl. Acad. Sci. USA 89: 10915-10919)의 값이 3 이하, 바람직하게는 2 이하, 보다 바람직하게는 1 이하, 더욱 바람직하게는 0 이하의 아미노산으로 치환한다. 구체적으로는, 센다이 바이러스 M 단백질의 G69, T116, 및 A183 또는 다른 바이러스 M 단백질의 상동부위를, 각각 Glu(E), Ala(A), 및 Ser(S)로 치환할 수 있다. 또한, 홍역 바이러스 온도 감수성주(temperature-sensitive strain) P253-505(Morikawa, Y. et al., Kitasato Arch. 또한, HN 유전자의 온도 감수성 변이로서는, 예를 들면 센다이 바이러스의 HN 단백질의 A262, G264, 및 K461로 이루어진 군으로부터 임의로 선택되는 부위의 아미노산 치환을 들 수 있다(Inoue, M. et al., J. Virol. 2003, 77: 3238-3246). 바람직한 일례를 들면, 센다이 바이러스 HN 단백질의 A262, G264, 및 K461 또는 다른 바이러스 HN 단백질의 상동부위를, 각각 Thr(T), Arg(R), 및 Gly(G)로 치환한다. 또한, 예를 들면, 유행성 이하선염 바이러스(mumps virus)의 온도 감수성 백신주 Urabe AM9를 참고로, HN 단백질의 464 및 468번째 아미노산으로 변이 도입하는 것도 가능하다(Wright, K. E. et al., Virus Res. 2000: 67; 49-57).
또한 마이너스 가닥 RNA 바이러스는, P 유전자 또는 L 유전자에 변이를 갖고 있어도 된다. 이와 같은 변이로서는, 구체적으로는, SeV P 단백질의 86번째 Glu(E86)의 변이, SeV P 단백질의 511번째 Leu(L511)의 다른 아미노산으로의 치환, 또는 다른 마이너스 가닥 RNA 바이러스 P 단백질의 상동부위의 치환을 들 수 있다. 구체적으로는, 86번째 아미노산의 Lys로의 치환, 511번째 아미노산의 Phe로의 치환 등을 예시할 수 있다. 또한 L 단백질에 있어서는, SeV L 단백질의 1197번째 Asn(N1197) 및/또는 1795번째 Lys(K1795)의 다른 아미노산으로의 치환, 또는 다른 마이너스 가닥 RNA 바이러스 L 단백질의 상동부위의 치환을 들 수 있고, 구체적으로는 1197번째 아미노산의 Ser로의 치환, 1795번째 아미노산의 Glu로의 치환 등을 예시할 수 있다. P 유전자 및 L 유전자의 변이는, 지속 감염성(persistent infectivity), 2차 입자 방출의 억제, 또는 세포상해성 억제의 효과를 현저하게 높일 수 있다. 또한, 엔벨로프 단백질 유전자의 변이 및/또는 결손을 조합시킴으로써, 이들의 효과를 극적으로 상승시킬 수 있다.
또한 엔벨로프 바이러스를 사용하는 경우는, 바이러스가 본래 갖는 엔벨로프 단백질과는 상이한 단백질을 엔벨로프에 포함하는 바이러스를 수상세포에 감염시켜도 된다. 예를 들면, 바이러스 제조시에, 목적의 외래성 엔벨로프 단백질을 바이러스 생산세포로 발현시킴으로써, 이것을 포함하는 바이러스를 제조할 수 있다. 이와 같은 단백질에 특별히 제한은 없고, 포유동물 세포로의 감염능을 부여하는 목적의 단백질이 사용된다. 구체적으로는, 예를 들면 수포성 구내염 바이러스(Vesicular stomatitis virus; VSV)의 G 단백질(VSV-G)을 들 수 있다. VSV-G 단백질은, 임의의 VSV주에 유래하는 것이어도 되고, 예를 들면 Indiana 혈청형 주(J. Virology 39: 519-528(1981)) 유래의 VSV-G 단백을 사용할 수 있지만, 이것에 한정되지 않는다. 본 발명에 있어서 사용되는 RNA 바이러스는, 다른 바이러스 유래의 엔벨로프 단백질을 임의로 조합시켜 포함할 수 있다.
RNA 바이러스는, 게놈 RNA 중에 외래 유전자를 코드해도 되고, 하지 않아도 된다. 수상세포에 도입됨으로써, 외래 단백질을 코드하지 않는 RNA 바이러스일지라도 항암작용을 발휘하기 때문에, 외래 유전자는 반드시 필요하지는 않다. 따라서 본 발명에는, 야생형 RNA 바이러스나 천연으로부터 단리된 바이러스(변이주를 포함한다) 등의 목적의 RNA 바이러스를 이용할 수 있는 이점이 있다. 예를 들면 본 발명에 있어서 사용하는 RNA 바이러스로서는, 암 치료효과를 가지는 단백질을 코드하지 않는 RNA 바이러스를 사용할 수 있다. 이와 같은 바이러스로서는, 암 치료효과를 가지지 않는 목적의 외래 단백질을 코드하는 RNA 바이러스가 포함되고, 예를 들면 RNA 바이러스의 도입을 검출하기 위해, 녹색 형광단백질(GFP), 루시페라아제(luciferase), 각종 펩티드 태그(peptide tag) 등의 마커 단백질(marker protein)을 코드하는 RNA 바이러스 등을 사용할 수 있다. 또는, RNA 바이러스에 제암작용을 돕는 외래 유전자를 추가로 삽입함으로써, 제암작용을 보다 높이는 것도 가능하다.
외래 유전자를 갖는 재조합 RNA 바이러스의 재구성은 공지의 방법을 이용하여 행하면 된다. 구체적인 순서는, 전형적으로는, (a) RNA 바이러스 게놈 RNA를 코드하는 cDNA를, 바이러스 입자형성에 필요한 바이러스 단백질을 발현하는 세포로 전사시키는 공정, (b) 생성된 바이러스를 포함하는 배양상청(culture supernatant)을 회수하는 공정에 의해 제조할 수 있다. 바이러스 단백질은, 전사시킨 바이러스 게놈 RNA로부터 발현되어도 되고, 게놈 RNA 이외로부터 트랜스(trans)에 공급되어도 된다. 게놈 RNA에 있어서 입자형성에 필요한 바이러스 유전자가 결손되어 있는 경우는, 그 바이러스 유전자를 바이러스 생산세포에서 별도 발현시켜, 입자형성을 상보한다. 바이러스 단백질이나 RNA 게놈을 세포 내에서 발현시키기 위해서는, 상기 단백질이나 게놈 RNA를 코드하는 DNA를 숙주세포에서 기능하는 적당한 프로모터(promotor)의 하류에 연결한 벡터를 숙주세포에 도입한다. 전사된 게놈 RNA는, 바이러스 단백질의 존재하에서 복제되어, 감염성 바이러스 입자가 형성된다. 엔벨로프 단백질 등의 유전자를 결손하는 결손형 바이러스를 제조하는 경우는, 결손하는 단백질 또는 그의 기능을 상보할 수 있는 다른 바이러스 단백질 등을 바이러스 생산세포에 있어서 발현시킨다.

게놈 복제능을 가지는 마이너스 가닥 RNA 바이러스가 도입된 수상세포의 제조방법을 보다 구체적으로 나타내면, 게놈 복제에 필요한 바이러스 단백질(N, P 및 L)을 발현하는 세포에, 바이러스 게놈 RNA(마이너스 가닥) 또는 그의 상보가닥(플러스 가닥)을 도입 또는 전사시킨다. 또한, 표적으로 하는 암이 발현되는 암 항원 펩티드를 수상세포에 제시시키는 것도 가능하다. 수상세포에 제시시키는 항원은, RNA 바이러스에 코드시키거나, 또는 RNA 바이러스를 도입한 수상세포에 첨가(즉 펄스(pulse))하거나, 또는 다른 목적의 벡터로 발현시킬 수 있다. 종양 항원은 종양세포에 특이적인 것(즉, 종양세포에 존재하지만, 비종양세포에는 존재하지 않는 것)이 바람직하지만, 동일한 타입의 비종양세포보다도 종양세포에 높은 레벨로 존재하는 것이어도 된다. 또한, 암 항원 단백질 전장이어도, 그의 부분 펩티드여도 된다. 수상세포에 있어서 제시되는 펩티드를 동정함으로써, 그의 펩티드를 합성하여 사용할 수 있다. 항원 펩티드는, 수상세포 표면의 MHC 분자에 결합하여 세포 표면에 제시되어, 면역응답이 유도된다.
CTL이 주된 이펙터(effector)로서 작용하는 경우는, 항원으로서는 세포 내외에 발현하는 목적의 종양 항원을 사용할 수 있다. 수상세포를 사용하여, CD4 T세포의 활성화로부터 계속되는 B세포의 활성화에 의한 항체 생산을 야기하여, 항체를 이펙터로서 작용시키는 경우에는, 항원으로서는 세포 표면에 표출하는 것이 바람직하고, 예를 들면, 세포 표면 수용체 또는 세포 접착 단백질을 사용할 수 있다. 종양 항원의 예로서는, 난소암 등으로 하는 Muc-1 또는 Muc-1 유사 뮤신 탠덤 리피트 펩티드(mucin tandem repeat peptide)(미국특허 제5,744,144호), 자궁경부암(cervical cancer)을 일으키는 인간 유두종 바이러스 단백질 E6 및 E7, 흑색종 항원 MART-1, MAGE-1, -2, -3, gp 100 및 티로시나아제(tyrosinase), 전립선암 항원 PSA, 그 밖에도, CEA(Kim, C. et al., Cancer Immunol. 또한, 이들 사이토카인의 개변체를 발현하도록 유전자 개변해도 된다. 예를 들면, 전구체(precursor form) 및 성숙체(nature form)의 2개의 형태를 갖는 사이토카인(예를 들면, 시그널 펩티드의 절단에 의해 활성 프래그먼트(active fragment)를 생성하는 것, 또는 단백질의 한정분해에 의해 활성 프래그먼트를 생성하는 것 등)에 대해서, 전구체 또는 성숙체 중 어느 하나를 발현하도록 유전자 개변해도 된다. 그 밖의 개변체(예를 들면, 사이토카인의 활성 프래그먼트와 이종 서열(heterologous sequence)(예를 들면, 이종 시그널 펩티드) 사이의 융합 단백질)를 사용해도 된다.
RNA 바이러스가 도입된 수상세포는, 필요에 따라서 약리학적으로 허용되는 목적의 담체 또는 매체(예를 들면 생리식염수, 링거액(Ringer’s solution), 배양액, 혈청 등)와 조합시킬 수 있다. 또한, 필요에 따라서 원심 등에 의해 농축되어, 배양액 또는 생리식염수 등의 생리적 용액 중에 재현탁되어도 된다. 본 발명에 의해서 조제되는 수상세포는, 암에 대한 유효한 면역요법에 있어서 유용하고, 종양 항원을 코드하는 유전자가 도입된 수상세포 또는 그의 수상세포로 자극된 T세포에 의한 면역 감작(Immune sensitization)은, 환자에 있어서 항종양 작용을 유도하는 유효한 방법이 된다. 본 발명은, 본 발명의 방법에 의해 얻어진 수상세포의 항암치료에 있어서의 사용에 관한 것이다. 또한 본 발명은, 본 발명의 방법에 의해 얻어진 수상세포의, 항암제(또는 제암제, 암 증식억제제 등)의 제조에 있어서의 사용에 관한 것이다. 또한 본 발명은, RNA 바이러스 및 수상세포의, 항암제(또는 제암제, 암 증식억제제 등)의 제조에 있어서의 사용에 관한 것이다.
얻어진 수상세포는, DC 백신(DC vaccine)으로서 유용하다. 면역원성을 높이기 위해, RNA 바이러스가 도입된 수상세포에는, 사이토카인, 콜레라 독소, 살모넬라 독소 등의 면역 촉진제를 첨가하는 것도 가능하다. 또한 백반, 불완전 프로인트 애주번트(incomplete Freund's adjuvant), MF59(오일 에멀젼), MTP-PE(마이코 박테리아(mycobacteria) 세포벽 유래의 muramyl tripeptide), 및 QS-21(soapbark tree Quilaja saponaria 유래) 등의 애주번트를 조합시키는 것도 가능하다.
또한 본 발명은, RNA 바이러스 및 수상세포를 포함하는 패키지로서, 상기 수상세포를 암 억제에 사용하는 것의 기재를 포함하는 패키지에 관한 것이다. RNA 바이러스 및 수상세포는, 각각의 용기 중에 포함되어 있어도 되고, 동일한 용기에 포함되어 있어도 된다. 또한 본 발명은, RNA 바이러스가 도입된 수상세포를 포함하는 패키지로서, 상기 수상세포를 암 억제에 사용하는 것의 기재를 포함하는 패키지에 관한 것이기도 하다. RNA 바이러스 및 수상세포는, 배양액 또는 생리식염수 등의 용액 중에 현탁되어 있으면 된다. 암 억제에 사용하는 것이란, 예를 들면, RNA 바이러스가 도입된 수상세포, 또는 그것을 포함하는 조성물이, 종양 증식억제, 암의 축퇴(縮退), 암 치료, 암환자의 처치·치료, 암환자의 연명, 또는 항암제로서 사용되는 것이다. 상기 기재는, 패키지에 직접 기재되어 있어도 되고, 상기 기재를 포함하는 종이나 스티커를 패키지에 포함하고 있어도 된다. 패키지는, RNA 바이러스 및/또는 수상세포를 포함하는 용기여도 되고, 그 경우, 용기는, 예를 들면 병(bottle), 튜브(tube), 비닐 봉지(plastic bag), 바이알(vial), 시린지(syringe) 등이어도 된다. 또한 본 발명의 패키지는, 상기 용기를 수납하는 백 또는 바깥상자(outer box) 등을 포함해도 된다. 패키지에는, 수상세포의 투여방법이 기재된 설명서를 포함해도 되고, 수상세포 투여를 위한 시린지, 카테터(catheter), 및/또는 주사바늘 등을 추가로 포함해도 된다.
본 발명에 의해 제조되는 제암제는 체내에 접종되기 때문에, 그곳에 포함되는 수상세포는 세포 증식성을 없애 놓으면 보다 안전하다. 예를 들면, 제대혈 유래의 단구는 분화 유도함으로써 증식능이 극도로 저하되는 것이 알려져 있지만, 세포 백신으로서 보다 안전하게 이용하기 위해, 가열처리, 방사선처리, 또는 마이토마이신 C(mitomycin C) 처리 등으로 처리하여, 백신으로서의 기능을 남긴 채, 증식성을 없앨 수 있다. 예를 들면, X선 조사를 이용하는 경우, 총방사선량 1000~3300 Rad로 조사할 수 있다. 마이토마이신 C 처리법은, 예를 들면, 수상세포에 25~50 micro-g/㎖의 마이토마이신 C를 첨가하여, 37℃, 30~60분간 보온처리할 수 있다. 열에 의한 세포처리방법은, 예를 들면, 50~65℃에서 20분간 가열처리를 행할 수 있다.
수상세포의 투여시에는, 애주번트 효과를 높이는 사이토카인류를 조합시키는 것도 유효하다. 이와 같은 유전자로서는, 예를 들면, ⅰ) IL-2와 단일 사슬 IL-12와의 조합(Proc. Natl. Acad. Sci. USA 96(15): 8591-8596, 1999), ⅱ) IL-2와 인터페론-γ(미국특허 제5,798,100호), ⅲ) 단독으로 사용되는 과립구 콜로니 자극인자(GM-CSF), ⅳ) GM-CSF와 IL-4의 조합(J. Neurosurgery 90(6), 1115-1124(1999)) 등을 들 수 있다.
RNA 바이러스가 도입된 수상세포는, 환자 자신의 T세포를 in vivo에서 자극하는 데에 유용하고, 또는 이 수상세포는 T세포를 in vitro에서 자극하는 것에도 유용하다. 감작한 T세포를 환자에게 투여하고, 엑스 비보 면역요법을 매개로 하여 환자의 종양면역을 자극하는 것도 가능하다.
본 발명은, 수상세포에 의해 자극된 T세포를 포함하는 항암제의 제조방법으로서, (a) RNA 바이러스를 수상세포 또는 그의 전구세포에 도입하는 공정, (b) 상기 세포를 성숙 수상세포로 분화시키는 공정, (c) 상기 성숙 수상세포에 암 항원을 제시시키는 공정, (d) 상기 성숙 수상세포를 T세포에 접촉시키는 공정을 포함하는 방법에 관한 것이다. RNA 바이러스를 도입한 수상세포는 T세포를 활성화하여, CTL을 유도한다. 수상세포에서 제시시키는 항원은, RNA 바이러스로부터 발현시킨 암 항원(또는 그의 프로세스된 산물)이어도 되고, 바깥으로부터 수상세포에 펄스해도 된다. 얻어진 T세포는 암 치료를 위해 사용할 수 있다. 인 비트로로 T세포와 수상세포를 접촉시키는 경우, 환자로부터 T세포를 채취하여, 수상세포와 접촉시킨 후, T세포를 엑스 비보 투여하는 것이 바람직하다.
또한 본 발명은, 본 발명의 방법에 의해 제조한 수상세포를 사용하여, 암을 억제하는 방법에 관한 것이다. 예를 들면, 암환자에 있어서 항종양면역을 자극하는 치료를 행할 수 있다. 이 방법은, 수상세포를 투여하는 공정을 포함하는 방법이다. 구체적으로는, 게놈 복제능을 가지는 RNA 바이러스의 RNP 복합체를 갖는 수상세포의 치료상 유효량을, 환자에게 투여하는 공정을 포함하는 방법이다. 본 방법에 의해, 본 발명의 수상세포를 투여하지 않는 경우에 비하여, 암의 증식을 억제하는 것을 기대할 수 있다. RNA 바이러스는, 외래 유전자를 갖지 않는 것이어도 되고, 또는 암 항원, 면역자극성 사이토카인, 혈관 신생을 저해하는 단백질 등의 하나 또는 복수를 코드하는 유전자를 갖는 것이어도 된다. RNA 바이러스가 수상세포에 도입됨으로써, 수상세포는 활성화되기 때문에, 외래 유전자를 갖지 않는 RNA 바이러스를 도입한 수상세포를 암에 투여하는 것에 의해서도, 암에 대한 환자의 면역계를 활성화시킬 수 있다. 이 수상세포에, 암 항원 펩티드를 펄스하여, 목적의 항원을 제시시킴으로써, 암 억제효과를 보다 높인 수상세포를 얻을 수 있다.
본 발명은, 목적의 고형암(solid cancer)에 적용할 수 있고, 예를 들면 설암(tongue cancer), 치은암(gingival cancer), 악성 림프종, 악성 흑색종(멜라노마), 상악암(maxillary cancer), 비암(nose cancer), 비강암(nasal cavity cancer), 후두암(laryngeal cancer), 인두암(pharyngeal cancer), 신경교종(glioma), 수막종(meningioma), 신경교종, 폐암, 유방암, 췌장암, 소화기암(식도암, 위암, 십이지장암, 대장암), 편평상피암(squamous cell carcinoma), 선암(adenocarcinoma), 폐포상피암(alveolar cell carcinoma), 고환 종양(testicular tumor), 전립선암, 갑상선암, 간암, 난소암, 횡문근육종(rhabdomyosarcoma), 섬유육종(fibrosarcoma), 골육종(osteosarcoma), 연골육종(chondrosarcoma) 등을 들 수 있다. 대상이 되는 암은, 바람직하게는 상피암이고, 보다 바람직하게는 피부 편평상피암, 피부 기저세포암(basal cell carcinomas of the skin), 피부 보웬병(Bowen’s disease of the skin), 피부 파제트병(Paget's disease of the skin), 피부 악성 흑색종 등을 포함하는 피부암이다.
RNA 바이러스를 도입한 수상세포를 투여하는 경우는, 일반적으로는 105~109 세포, 바람직하게는 106~108 세포, 보다 바람직하게는 약 107 세포를 환자의 암 병소에 투여한다. 암 병소란, 암조직 또는 그의 주위(예를 들면 암으로부터 5 ㎜ 이내, 바람직하게는 3 ㎜ 이내)의 영역을 말한다. 또한 투여량은, 암의 타입이나 스테이지, 도입 유전자의 유무 등에 의해 적절히 조절되어도 된다. RNA 바이러스는 외래 유전자를 탑재하고 있지 않아도 항종양 효과를 기대할 수 있지만, IFN-beta 유전자 또는 가용성 FGF 수용체 유전자 등을 RNA 바이러스에 탑재시킴으로써 보다 높은 효과를 얻을 수 있다. 또한, 수상세포를 종양에 투여하기 전에, 수상세포에 종양항원을 접촉시키면 보다 높은 효과를 얻을 수 있다. 수상세포로의 종양항원의 접촉은, 수상세포와 종양세포의 세포 용해물(cell lysate)을 혼합하는 방법, 종양항원 펩티드를 수상세포에 펄스하는 방법, 또는 수상세포에 종양항원 유전자를 도입하여 발현시키는 방법 등을 사용할 수 있다. 또한, IFN-beta, 가용성 FGF 수용체, 또는 그들을 코드하는 유전자를 탑재하는 목적의 벡터를, 본 발명의 수상세포와 함께 암 병소에 직접 주입함으로써도 항종양 효과가 얻어진다. 즉, RNA 바이러스를 도입한 수상세포의 투여와, IFN-beta 또는 가용성 FGF 수용체에 의한 항종양처치와의 조합은, 본 발명에 있어서 적합하다.
수상세포에 의해 활성화된 T세포를 투여하는 경우는, 예를 들면, T세포는 1 ㎡의 체표면적당 약 105~109 세포, 바람직하게는 106~109 세포, 보다 바람직하게는 108~109 세포의 용량으로, 정맥내 주입에 의해 투여될 수 있다(Ridell 등, 1992, Science 257: 238-241을 참조). 주입은, 목적의 간격(예를 들면, 매월)으로 반복될 수 있다. 투여 후의 레시피언트(recipient)는, 필요에 따라 임의의 부작용에 대해서, T세포 주입의 사이 또는 주입 후에 모니터되어도 된다. 이 때, T세포는 수상세포를 얻은 환자와 동일한 환자로부터 얻는 것이 바람직하다. 또는, T세포를 환자로부터 채취하고, T세포를 자극하기 위해 사용하는 수상세포는, HLA 적합성의 건강한 정상의 도너(donor)에 유래해도 된다. 또는 반대로, 수상세포를 환자로부터 채취하고, T세포는 HLA 적합성의 건강한 정상의 도너에 유래해도 된다.
수상세포 또는 T세포의 투여 횟수는, 1회 또는 임상상 용인 가능한 부작용의 범위에서 복수회 가능하며, 1일의 투여 횟수에 대해서도 동일하다. 투여대상으로서는 특별히 제한은 없지만, 예를 들면, 닭, 메추라기, 마우스, 랫트, 개, 돼지, 고양이, 소, 토끼, 양, 염소, 원숭이, 및 인간 등을 포함하는 조류, 포유동물(인간 및 비인간 포유동물)을 들 수 있다. 인간 이외의 동물에 투여하는 경우는, 예를 들면, 목적의 동물과 인간과의 체중비로 상기의 투여량을 환산한 양을 투여할 수 있다.
이하, 실시예에 의해 본 발명을 더욱 상세하게 설명하지만, 본 발명은 이들 실시예에 제한되는 것은 아니다. 또한, 본 명세서 중에 인용된 문헌은, 모두 본 명세서의 일부로서 포함된다.
A. 도입효율의 검토
(실험 1)
건강한 정상인으로부터 단구를 음성 선택으로 농축(enrichment)하였다. 단구의 농축을 위한 음성 선택은, RosetteSepTM-human monocyte enrichment cocktail(Stem Cell Technology Inc.)을 사용하였다. 즉, tetrameric antibody(항체 2분자가 결합되어 있는 항체로, 한쪽은 적혈구를 인식하는 항글리코포린 A 항체(anti-glycophorin A antibody), 다른 쪽이 단핵구의 표면 항원을 인식하는 항체로 되어 있다)를 사용하여, 제거하려는 세포를 적혈구에 결합시키고, Ficoll PaqueTM Plus(Pharmacia Biotech Inc.)로 제거함으로써 실시하였다. 이 음성 선택에 의해, CD2, CD3, CD8, CD19, CD56, CD66b를 발현하고 있는 세포가 제거되고, 남은 세포를 단구 농축세포로서, 다음의 DC의 분화 유도에 사용하였다. 이 때의 CD14+ 세포는 65-80%였다. 단구 농축세포에 GM-CSF(500 U/㎖)와 IL-4(250 U/㎖)를 첨가하여, endotoxin free RPMI+10% FCS로 배양 후, DC의 작성을 행하였다. 3-4일째에 절반의 배양상청을 동일한 조성의 새로운 배양액으로 배양액 교환을 행하였다. 보조자극분자(Costimulatory molecule), 및 CD11c, HLA-class Ⅱ(DR, DP, DQ), 및 CD1a의 발현이 양성인 것을 확인하고, 그 밖의 리니지 마커(lineage marker)(CD3, CD56, CD19, CD15 및 CD14)를 표출하고 있지 않은 것도 확인하였다(도 1 및 비제시 데이터). 이 세포를 사용하여, 바이러스의 도입효율을 검토하였다. 이 시점에서, 생세포의 90~98%가 DC 마커(CD11c, HLA-class Ⅱ(DR, DP, DQ))를 발현하고 있었다.
또한, 본 실시예에 있어서의 셀렉션은 상기 키트(kit)를 사용하였지만, 항체로 코트된 자기 비즈(magnetic beads)를 사용해도, 동일한 셀렉션을 실시하는 것이 가능하다. 혈구분리 등을 사용하여 단핵구를 채취하는 등, 세포를 대량으로 조제하는 경우는, 비즈를 사용하는 것이 바람직하다.
(실험 2)
실험 1로부터 얻어진 DC(분화 유도 후 7일째)에, 녹색 형광 단백질(GFP)을 발현하는 센다이 바이러스(SeV-GFP)(전파형, WO00/70070)를 MOI를 다양하게 감염시켜, 경시적으로 세포수의 변화, GFP의 발현, 보조자극분자(costimulatory molecules)의 발현 정도에 대해서 검토하였다. 그 결과, MOI에 대해서는, MOI 20 이상에서 %GFP는 최대가 되었다(도 2~5). GFP의 평균 형광강도(mean fluorescence intensity; MFI)에 대해서는, MOI 100까지 높임으로써 더욱 상승시키는 것이 가능하다(비제시 데이터). 또한, 8일째까지 GFP의 MFI는 상승하였다. 보조자극분자(CD 80 및 CD 86)에 대해서는, 전체적으로 MOI 20 이상에서 최대가 되었다. 세포수의 감소에 대해서는, MOI에서 1~20의 사이는 그다지 변화가 없지만, MOI 50에서는 약간 감소하는 경향이 보였지만 유의차는 없었다(도 6).
(실험 3)
MOI 20에서 DC에 SeV-GFP를 감염시켜, 경시적으로 FACS를 사용하여, GFP의 발현을 검토하였다. 그 결과, 2주 이후는 발현이 저하되지만(세포수도 감소), 2개 월까지는 발현세포를 확인할 수 있었다(도 7). 이하의 실시예에서 나타내는 바와 같이, RNA 바이러스의 감염으로 DC는 활성화된다. 따라서, RNA 바이러스를 사용한 DC로의 유전자 도입은 임상응용으로서, 백신으로의 응용이 가능하다. 투여는 인 비보로도 엑스 비보로도 가능하지만, 예를 들면, 엑스 비보 투여에 의해 RNA 바이러스를 감염시킨 DC를 빈번히 투여함으로써, 장기에 걸쳐 체내에 있어서의 유전자 발현을 지속시키는 것이 가능하다.
(실험 4)
활성화와 감염효율을 검토하였다. 활성화의 유무로 바이러스의 감염효율이 변화하는지 여부의 검토를 행하였다. 7일간 배양 후의 DC에 2일간 LPS(1 micro-g/㎖)로 자극한 후, SeV-GFP를 MOI 30에서 감염, 2일 후에 FACS로 GFP를 해석하였다. 반대로 SeV-GFP 감염 2일 후에 LPS 자극(2일간)을 동일한 조건에서 시행하였다.(도 8 및 9)
결과; 인간에서는, LPS로 활성화시킨 후에, %GFP에 있어서 60% 가까이의 양성을 인정하였다. 이것에 대하여 마우스 DC에서는 양성률은 매우 낮았다(비제시 데이터). 그러나 인간에게 있어서도 MFI는 매우 낮고, 활성화시킨 후의 DC에는 유전자 도입효율의 극단적인 저하를 인정하였다. 이것에 대하여, SeV를 도입 후에 LPS로 자극해도, 유전자 도입효율은 변화하지 않았다. 이 결과는, RNA 바이러스를 도입한 DC를 얻기 위해서는, 미성숙 DC, 즉 활성화를 받고 있지 않은 DC를 사용하는 것이 바람직한 것을 나타내고 있다.
(실험 5)
감염에 필요한 접촉시간의 검토를 행하였다(도 10). 그 결과, 약 30분 이하에서 유전자 도입이 가능한 것이 판명되었다.
(실험 6)
다른 바이러스 벡터에서의 보고에서 CD34 세포에 유전자 도입하여, DC 분화 유도로 유전자 도입 DC 제작에 성공한 보고가 있다(J. Immunol. Meth. 2002; 153-165). SeV-GFP로도 동일한 방법을 시도하였다. 인간 제대혈로부터 CD34 마이크로비즈를 사용하여, CD34 양성 줄기세포를 분리(CD34>90%)하고, MOI 0, 10, 100에서 감염시킨 후, 잘 세정하였다. 그 세포를 RPMI+10% FCS에 SCF(50 ng/㎖), GM-CSF(500 U/㎖), TNF-alpha(50 ng/㎖)를 첨가하여, 3일간 배양 후, SCF(50 ng/㎖), GM-CSF(500 U/㎖), IL-4(250 U/㎖), TNF-alpha(50 ng/㎖)를 첨가한 배지에서 계대(절반량의 배지를 3~4일에 교환)한 것을 바이러스 감염으로부터 13일째에 GFP의 발현을 검토하였다. 그 결과, 유전자 도입효율은 65~70%에 달하고, 다른 벡터 이상으로 GFP의 발현효율이 좋은 DC가 제작되었다. 감염 후의 DC는 보조자극분자의 발현의 해석으로부터, 감염시키지 않은 것과 비교하여 활성화를 받고 있는 것이 회수되었다(도 11 및 12).
이상의 실시예로부터, RNA 바이러스는 렌티바이러스, 레트로바이러스에 비교하여, 도입효율은 현격히 우수하고, 아데노바이러스에 뒤지지 않는 도입효율을 매우 간편하고 신속하게 얻을 수 있는 것이 실증되었다. 또한, 다른 벡터에서는 활성화 마커는 변동하지 않지만, RNA 바이러스의 감염에 의해 DC의 활성화를 유도할 수 있는 것이 판명되었다.
B. 도입 후의 DC 기능의 평가
(실험 1)
DC에 SeV-GFP를 MOI 30-50에서 감염시켜, 1일 후에 LPS로 자극(2일간)하고, 그 다음, 보조자극분자의 발현을 검토하였다. 대조(control)로서, LPS 자극만, SeV-GFP 감염만, 및 LPS 자극도 SeV-GFP 감염도 없는 조건에 대해서 비교 검토하였다.
결과; 얻어진 결과로부터, SeV 감염만으로도 DC의 활성화가 일어나는 것이 나타내어졌다.
LPS와 필적하는 것; CD80(+) HLA-DR(-) CD83(-)
LPS보다 강한 것; CD86(+) CCR7(-)
LPS보다 약한 것; CD40(-)
(+)는 LPS+SeV로 상승 효과가 있는 것을 나타낸다. (도 13-15)
(실험 2)
MOI 30에서 DC에 SeV-GFP를 감염(감염 후 3일째, 감염 후 1일째에, 어떤 그룹은 LPS로 자극)시켜, 실험 1과 동일한 군에 대해서, 식세포능력(phagocytic activity)을 검토하였다(1 micro-m PCV-RED latex-microspheres를 사용. 막대 그래프는 4℃에서 양성이 되는 백그라운드를 뺀 것).
결과; 활성화 마커에서도 본 바와 같이, SeV로 감염시킨 것은, 활성화 때문에, 식세포능의 저하가 보였다. 특히, GFP의 발현이 높은 것일수록 식세포능이 낮았다. 따라서, 예를 들면, DC로 종양항원을 제시시키기 위해 종양의 용해 물(lysate)을 사용하는 경우는, RNA 바이러스를 DC에 도입하기 전에 용해물(lysate)과 DC를 공배양하는 것이 바람직하다(도 16-17).
(실험 3)
RNA 바이러스에 의한 수상세포의 활성화에 수반되는 수상세포 사이토카인 생산능을 검토하기 위해, 7일간의 배양으로 얻어진 단구 유래 수상세포(MoDC)를 12웰의 플레이트에서 48시간(8×105/2 ㎖/well: 배지는 X-vivo15TM+2% 자가혈청+GM-CSF(500 U/㎖)+IL-4(250 U/㎖), 이하의 군의 조건에서 배양한 상청 중의 TNF-alpha, IL-1beta, IL-6, IL-8의 값을 LuminexTM system으로 측정하였다. SeV의 감염은 MOI 30에서 2일간 배양하였다.
자극받지 않은 군(Unstimulated group): 배지만의 군
요막강액군(Allantoic fluid group): SeV의 부유액인 계란 요막강액(SeV는 포함하지 않는다) 60 micro-L 첨가한 군
UV-SeV-GFP군: SeV-GFP 용액을 자외선 조사하여, 복제(replication) 능력을 제거한 용액 60 micro-L 첨가한 군
SeV-GFP: SeV-GFP 용액을 60 micro-L 첨가한 군(replication-competent SeV)
결과: UV 조사를 하지 않는 복제 가능한 SeV(replication-competent SeV)로 GFP를 유전자 도입한 수상세포만 TNF-alpha, IL-1beta, IL-6을 생산, IL-8의 생산을 증대하였다(도 18). 이 때의 수상세포상의 CD40, CD80, CD83, CD86, HLA-DR의 발현 상승은 복제 가능한 SeV(replication-competent SeV)만으로 유도되었다(도 19 및 20). 이것은, SeV를 수상세포에 유전자 도입하는 것 만으로, 면역응답시에 중요한 염증성 사이토카인(proinflammatory cytokine)의 수상세포에 의한 생산을 야기할 수 있는 것을 의미하고 있다. 또한, UV 처리한 SeV에서는, 사이토카인 생산을 유도할 수 없었기 때문에, 수상세포 유전자 도입시에 있어서의 수상세포의 막상의 리셉터와 SeV의 접촉에 의한 수상세포의 활성화라기보다도, SeV의 감염 후에 발생하는 바이러스 게놈 RNA의 증폭 과정이, 수상세포의 활성화에 중요하다는 것도 시사하고 있다.
(실험 4)
RNA 바이러스에 의한 수상세포의 활성화에 수반되는 수상세포 항원 제시능을 검토하기 위해, 상기와 동일한 실험군에 대해서, 그들의 DC를 3000 rad의 방사선을 조사한 후, T세포의 활성화능력에 대해서 검토하였다. (dose를 다양한 DC와 순화(CD3+>95%)의 알로 또는 신제닉 T세포(allo or syngenic T cell)와 3일간 공배양하였다). SeV-GFP에 대한 반응의 지표로서, 신제닉 T세포를 사용하였다.
결과; DC비와 T세포의 양이 적어 차가 비교적 현저하지 않지만, SeV 감염 단독으로 LPS에 필적하는 알로 T세포 자극성을 가지고 있는 것이 나타내어졌다(도 21). 또한, DC는 방사선 조사를 하지 않고 사용하는 것도 가능하다.
(실험 5)
RNA 바이러스에 의한 수상세포의 활성화에 수반되는 수상세포 항원 제시능을, 현재 가장 수상세포 성숙화 능력이 강력하다고 여겨지고 있는 사이토카인 칵테 일 자극에 의한 항원 제시능과 비교하였다. 7일간의 배양에서 얻어진 인간 단구 유래 수상세포(MoDC)를 12웰의 플레이트에서 48시간 배양하였다{1×106/2 ㎖/well: 배지는 X-vivo15TM+2% 자가혈청+GM-CSF(500 U/㎖)+IL-4(250 U/㎖), 이하의 군의 조건에서 배양}. 또한 RNA 바이러스로서, F 유전자를 결실하고, 온도 감수성의 변이 M 및 HN 단백질(M 유전자: G69E, T116A, A183S, HN 유전자: A262T, G264R, K461G) 유전자를 탑재하며, 또한, 지속 감염형 변이를 갖는 변이 P 및 L 단백질(P 유전자: L511F, L 유전자: N1197S, K1795E) 유전자를 탑재하는 센다이 바이러스(SeV-dFMtsHNtsPLmut-GFP(SeV/TSdF라고도 약칭한다))도 비교로서 사용하였다(WO2003/025570; Inoue M, et al. J Virol 2003; 77:3238-3246; Inoue M, et al. Mol. Ther. 2003; 7(5):S37). 이 바이러스는, 감염된 세포에 있어서 감염성 바이러스 입자를 형성하는 능력을 잃고 있다.

48시간 후에 얻어진 MoDC에 30 Gy 조사한 후, 그의 MoDC(4×104/웰로부터 6.25×102/웰)와 알로의 말초혈 유래 T세포(1×105/웰){알로의 말초혈로부터, RosetteSepTM-human T cell enrichment kit(Stem Cell Technologies, Vancouver, Canada)를 사용하여 얻어진 96% 이상의 순도의 T세포}를 4일간 배양한 후, 각 웰에 1 μCi의 [3H]-thymidine을 첨가하고, 8시간 후의 [3H]-thymidine의 흡수량을 Beta Plate System(Pharmacia LKB Biotechnology, Uppsala, Sweden)으로 카운트하였다. 배지는 X-vivo15TM+2% 자가혈청을 사용하였다. 나타내어진 도면의 X축은 웰당 배양 MoDC수/웰당의 T세포수(=1×105), Y축은 [3H]-thymidine 흡수량(cpm)을 나타낸다(도 22).
결과: 자극하지 않은 수상세포와 비교하여, SeV-GFP를 감염시킨 수상세포는 알로의 T세포를 유의하게 증식시키고, 그 능력은, 양성 대조(positive control)에서, 현재 가장 수상세포 성숙화 능력이 강력하다고 여겨지고 있는 사이토카인 칵테일 자극과 동등 이상이었다. 이 항원 제시능력의 정도는 F 결손형 HN 온도 감수성 SeV(SeV-dFMtsHNtsPLmut-GFP)를 감염시킨 수상세포에서도 거의 동등하였다(도 22).
C. 암 항원 특이적 CTL의 유도
A에 기재된 방법으로 인간 말초혈(HLA-A 0201의 건강한 정상의 도너)로부터, CD14+ 세포를 농축하여, x-vivo 15TM(Cambrex사제)+2% 자가혈청(autologous serum)을 배지로서, GM-CSF(500 U/㎖), IL-4(250 U/㎖)를 첨가(3~4일에 한번, 반량의 배지를 교환), 미숙 수상세포를 제작하였다. 제작한 미숙 수상세포를 다음의 3군으로 나누고, 추가로 48시간 GM-CSF(500 U/㎖), IL-4(250 U/㎖)의 존재하에 배양하였다.
1군 아무것도 첨가하지 않는다
2군 SeV-GFP의 감염(MOI 30)
3군 사이토카인의 칵테일(IL-1β 50 ng/㎖, IL-6 500 ng/㎖, IFN-α 2500 U/㎖, TNF-α 100 ng/㎖, PGE2 20 micro-M)에 의한 자극
그 다음, 수상세포를 회수하여, MART-1 펩티드(EAAGIGILTV(서열번호: 1))를 펄스(50 micro-g/㎖; 3시간)하고, 수상세포를 채취한 것과 동일한 건강한 정상인 말초혈의 T세포를 음성 선택으로 농축(CD3+>97%)하여, 펩티드 펄스한 상기 3군의 수상세포와 7일간 배양하였다(X-vivo 15TM+2% 자가혈청)(3~4일마다, 또는 배지의 황변시에 절반량의 배지를 교환하였다. 최초의 자극시는 IL-2 없이 T세포와 수상세포를 혼합 배양하고, 3일째부터 IL-2를 100 U/㎖ 첨가 개시하였다.). 이것을 2회 반복하여, 각각의 혼합 배양으로부터 세포를 회수하고, CTL 어세이의 이펙터 세포로서 사용하였다.
타겟 세포(target cell)는, T2세포(HLA-A2+의 인간으로부터 얻어진 T세포-B세포 하이브리도마(hybridoma)로 TAP 결손 세포주)를 사용하였다. 이 세포는, TAP(class I으로의 트랜스포터(transporter)가 없기 때문에, 세포질 내의 단백의 분해에 의해 생산된 펩티드를 Class I으로 유도할 수 없는 것으로부터, 펩티드를 외래로부터 첨가하면 그 펩티드가 Class I으로 로드(load)되어, Class I의 발현이 일어난다. 이 타겟을 변이 MART-1 펩티드(ELAGIGILTV(서열번호: 2))(상기의 자극에서 사용한 펩티드에 대하여 T세포 리셉터 인식부는 변화없이, HLA-A2의 결합을 강하게 한 것)를 펄스한 것, 인플루엔자의 펩티드(Flu; third party로서의 펩티드; GILGFVFTL(서열번호: 3))를 펄스한 것을 제작하여, Cr로 세포를 라벨하고, 상기 3군의 이펙터 T세포와 이 2종류의 타겟을 20:1, 10:1, 5:1, 2.5:1로 4시간 혼합 배양하여 CTL 활성을 조사하였다.
이하에 실험의 조합을 정리하였다.
결과; 상기 3군의 DC로 활성화를 시키지 않은 DC(MART1 펩티드+)로 T세포를 자극하여도, MART-1 특이적 CTL은 유도할 수 없지만, 양성 대조(positive control)로서 사이토카인으로 활성화(현재 종양 면역의 수상세포 요법에서 가장 강하게 활성화할 수 있는 방법)시킨 수상세포로, T세포를 자극하면 MART-1 특이적 CTL을 유도할 수 있었다(타겟에 펄스하는 변이 MART-1 펩티드로 바꿔, 자극에 사용한 MART-1 펩티드를 사용해도 동일한 결과가 얻어졌다). SeV를 도입한 수상세포를 사용한 경우, 양성 대조와 동일한 정도의 CTL 활성이 얻어졌다(도 22). 즉, CTL 어세이로 판정한 경우, SeV를 감염시키는 것만으로 수상세포는 활성화되어, 사이토카인으로 활성화시킨 수상세포와 동일한 레벨로 in vitro로 CTL을 유도할 수 있는 것이 나타내어졌다. T세포 부활화로서 SeV를 사용하면, 표적 유전자의 도입과 함께 활성화를 일어킬 수 있기 때문에, 사이토카인 등의 활성화 인자를 첨가할 필요가 없어져, 비용 절감, 시간 절감, 세포의 생존성의 유지에 기여한다.
D. RNA 바이러스 도입 DC에 의한 암 증식억제
본 실시예는, RNA 바이러스의 인 비보 및 엑스 비보 투여에 의한 종양의 치료방법의 일례를 나타낸다.
(실험 1)
종양 모델로서, MHC Class Ⅰ을 매우 낮은 레벨로밖에 발현하지 못하고 면역원성이 부족한 B16 흑생종의 이식 모델을 채용하였다. 종양 모델 마우스에는, C57BL/6 마우스(6~8주령, 암컷)(일본 찰스 리버)를 사용하고, 수상세포는 C57BL/6 마우스(8주령, 암컷)(일본 찰스 리버)로부터 채취하였다. 수상세포는, C57BL/6 마우스의 대퇴골로부터 골수를 채취하여, SpinSepTM, murine hematopoetic progenitor enrichmentcocktail(항CD5 항체, 항CD45R 항체, 항CD11b 항체, 항Gr-1 항체, 항TER119 항체, 항7/4 항체, Stem Cell technology)을 사용하고, T세포를 제거한 후, IL-4 및 GM-CSF를 첨가하여 1주간 배양하여 얻었다. 1×105/100 micro-L의 B16 흑색종 세포를 day 0에 마우스의 복부 피하(s. c.) 접종하였다. Day 10, 17, 및 24에 활성화 자극을 가하지 않는 수상세포, LPS로 활성화시킨 수상세포(LPS DC), 또는 SeV-GFP 또는 마우스 인터페론(mouse interferon) β를 발현하는 SeV-IFN β를 도입하여 활성화시킨 수상세포(각각 SeV GFP DC 또는 SeV IFN β DC)를 종양 주위에 투여하였다. 이 때, 수상세포에 종양항원(B16의 냉동과 해동(freeze and thaw)에 의한 종양 용해물)을 펄스한 후 투여하는 실험도 행하였다. 이들과는 별도로, 종양 접종 후 10일째(day 10)에 SeV-IFN β를 직접, 종양내 주입(intratumoral injection)하여 항종양 효과를 조사하는 실험도 행하였다.
수상세포로의 SeV의 도입에서는, 상기와 같이 1주간 배양한 수상세포에 SeV-IFN β를 MOI 40에서 감염시켜 8시간 배양하였다. 종양항원을 펄스하는 경우는, 상기와 같이 1주간 배양한 수상세포를 회수하여, 종양항원이 되는 종양 용해물(tumor lysate)을 펄스(DC: tumor lysate=1:3)하고, 18시간 배양한 후, SeV-IFN β를 MOI 40에서 감염시켜 8시간 배양하였다. 그 다음, 이들의 수상세포를 회수하여, 5×105~10×105 세포수를 마우스의 종양 주위에 투여하였다.
도 24에 나타내는 바와 같이, SeV-IFN β의 직접 종양내 주입 및 수상세포를 매개로 한 엑스 비보 투여는, 모두 종양 증식을 억제하였다. 특히 DC/SeV-IFN β로 처리한 마우스에서는, 매우 강한 종양 억제효과가 관찰되었다.
상기의 각 치료군에 있어서의 항종양 효과를 보다 상세하게 검토하였다. 내츄럴 킬러(natural killer)(NK) 세포 활성을 어세이하기 위해, 상기의 각 치료군에 대해서, 3회의 DC 요법 종료 7일 후의 마우스로부터 비장을 적출하여, 이펙터 세 포(effector cell)를 작성하였다. 타겟으로서 Yac-1을 사용하여, 51Cr 방출 어세이(release assay)를 행하였다. 또한, T 림프구의 세포상해성을 어세이하기 위해, 상기의 NK세포 활성 어세이에 사용한 비장세포의 나머지를 사용하고, B16의 종양항원인 TRP-2 펩티드와 함께, 5일간 배양한 세포를 이펙터 세포로서 사용해서, mTRP-2 펩티드를 펄스한 EL-4 타겟 세포와 공배양하여, 51Cr 방출 어세이를 행하였다. 특이적 51Cr 방출의 비율은 이하와 같이 계산하였다.
[(시료의 cpm-자발 방출의 cpm)/(최대 방출의 cpm-자발 방출의 cpm)]×100
여기에서, 최대 방출은 1% triton X와 함께 인큐베이트한 타겟 세포, 자발 방출은 배양액만으로 인큐베이트한 타겟 세포를 사용하였다.
내츄럴 킬러(NK) 세포의 활성화는, 직접 SeV를 주입한 마우스에만 보이고, 수상세포 투여군에서는 검출되지 않았다(도 25). 이것에 대하여, 세포상해성 T 림프구(cytotoxic T-lymphocytes, CTL)의 활성화는, DC/LPS 처리군 및 DC/SeV-IFN β로 처리한 마우스에서 가장 강하고, DC/SeV-GFP 처리군에서는 그것보다도 약간 약하며, SeV-IFN β의 직접 주입군에서는 검출되지 않았다(도 26). 종양 용해물의 펄스는, 종양 증식에도 CTL 응답에도 유의하게 영항을 끼치지 않았다. 이와 같이, SeV에 의해 면역자극성 사이토카인 유전자를 도입한 수상세포를 사용한 종양 면역치료는, 강한 항종양 치료효과를 발휘할 수 있는 것이 나타내어졌다. DC/LPS 처리군과 DC/SeV-IFN β 처리군 사이에서 CTL 활성에 약간의 상이함이 있는 것에도 불구하고, 동일한 정도의 항종양 효과가 발견되었다. 이와 같이, SeV-IFN β의 직접 투여는 NK세포를 강하게 활성화시키는 한편으로, 수상세포를 매개로 한 간접투여는 CTL 활성을 유도하는 것이 판명되었다. 따라서, 이들을 조합시킨 치료는 보다 효과를 발휘할 수 있는 것이 기대된다.
(실험 2)
흑색종 세포주 B16F1(ATCC CRL-6323) 1×105개를, C57BL/6 마우스(6~8주령, 암컷)의 복측 피하로 접종하였다(n=4). 접종 후 5일(day 5) 및 12일(day 12)에, 특별한 치료 유전자를 갖지 않는 SeV(GFP 발현 SeV; SeV-GFP), 인간 가용형 FGF 수용체를 발현하는 센다이 바이러스(SeV-sFGFR), 또는 인간 PDGFRα 가용형을 발현하는 SeV-hsPDGFRα, 각 1×108 PFU를 종양내 주사하고, 이후, 종양 사이즈를 경시적으로 측정하였다. 그 결과, 어느 SeV 투여군에 있어서도, SeV 비투여군에 비하여 종양 사이즈가 유의하게 축소되었다(도 27). 이와 같이, SeV의 인 비보 투여는, 치료 유전자를 갖지 않는 것일지라도 항종양 효과를 발휘할 수 있었다. 가용형 FGF 수용체를 발현하는 SeV를 투여한 경우는, SeV-GFP 투여군 보다도 강한 종양 증식 억제효과가 확인되고, 가용형 PDGFRα를 발현하는 SeV를 투여한 경우의 항종양 효과는 가장 현저하여, 종양 사이즈는 거의 증가하지 않았다.
(실험 3)
흑색종 세포주 B16F1(ATCC CRL-6323) 1×105개를, C57BL/6 마우스(6주령, 암컷)의 복측 피하로 접종하였다(n=4). 그것과는 별도로, C57BL/6 마우스(6~8주령, 암컷)의 골수세포를 채취하여, SpinSepTM(STEM CELL TECHNOLOGIES, INC)에 의한 CD45R, CD5, CD11b, TER119, Gr-1, 7-4의 음성 선택으로 추출한 세포를 GM-CSF 250 IU/㎖, IL-4 250 IU/㎖ 존재하에 7일간 배양하여, 마우스 골수세포 유래의 수상세포를 얻었다. 그 수상세포에, 치료 유전자를 갖지 않는 센다이 바이러스(GFP 발현 SeV; SeV-GFP)를 MOI 60, 또는 인간 가용형 PDGFα 수용체를 발현하는 센다이 바이러스(SeV-hsPDGFRα), 종양 항원 TRP2를 발현하는 센다이 바이러스(SeV-TRP2), 또는 종양 항원 gp 100을 발현하는 센다이 바이러스(SeV-gp 100)를 각각 MOI 20씩 감염시켜, 유전자 도입을 행하였다.
B16F1의 접종 후 10일(day 10) 및 17일(day 17)에, SeV-GFP 또는 SeV-hsPDGFRα를 도입한 수상세포 1×106개를 종양내 주사하고, 이후, 종양 사이즈를 경시적으로 측정한 결과를 도 28에 나타내었다. 상기 바이러스의 인 비보 투여와 동일하게, SeV-GFP를 도입한 수상세포를 투여한 마우스에서도, SeV 미도입 수상세포를 투여한 마우스에 비하여 종양 사이즈가 유의하게 축소되었다. SeV에 의한 항종양 효과는, 수상세포를 사용한 엑스 비보 투여쪽이, SeV의 인 비보 투여 보다도 현저하였다(도 28). 가용형 PDGFRα를 발현하는 SeV를 투여한 수상세포의 엑스 비보 투여에서는 항종양 효과는 더욱 상승하고, 종양 사이즈는 거의 증가하지 않았다.
본 발명에 의해, RNA 바이러스가 도입된 수상세포를 유효성분으로 하는 항암제가 제공되었다. RNA 바이러스의 도입은 수상세포의 활성화를 야기하기 때문에, 도입 후의 사이토카인 등에서의 활성화처리의 공정이 생략 가능하고, 세포의 생존성(viability)의 유지나 비용 삭감, 더 나아가서는 ex vivo에서의 조작시간의 삭감에 기여할 것으로 생각된다. 본 발명은, RNA 바이러스와 수상세포를 조합시킨 새로운 바이러스 요법을 가능하게 한다.

Claims (10)
- 게놈 복제능을 가지는 센다이 바이러스가 도입된 수상세포를 포함하는 항암제로서, 상기 수상세포는, (i) 미성숙 수상세포에 상기 센다이 바이러스를 접촉시키는 공정, 또는 (ii) 수상세포의 전구세포에 상기 센다이 바이러스를 접촉시켜, 상기 전구세포를 미성숙 수상세포로 분화시키는 공정을 포함하고, 상기 미성숙 수상세포에 있어서 상기 센다이 바이러스의 게놈이 복제되는 단계, 상기 수상세포가 성숙화하는 단계를 포함하는 방법에 의해 생성되는 성숙 수상세포인 항암제.
- 제1항에 있어서, 전구세포가 CD34+ 세포인 항암제.
- 제1항에 있어서, 센다이 바이러스가 상기 바이러스에 대한 외래 단백질을 코드하지 않는 항암제.
- 제1항에 있어서, 센다이 바이러스가 감염성 바이러스 입자를 형성하지 않는 바이러스인 항암제.
- 제1항에 있어서, 센다이 바이러스가 가용성 FGF 수용체 또는 IFN-β를 코드하는 항암제.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 센다이 바이러스가 감염성 또는 비감염성 바이러스 입자인 항암제.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 센다이 바이러스가 게놈 RNA-단백 복합체인 항암제.
- 게놈 복제능을 가지는 센다이 바이러스를 (i) 미성숙 수상세포에 도입하는 공정, 또는 (ii) 수상세포의 전구세포에 상기 센다이 바이러스를 도입하여, 상기 전구세포를 미성숙 수상세포로 분화시키는 공정을 포함하고, 상기 미성숙 수상세포에 있어서 상기 센다이 바이러스의 게놈이 복제되며, 상기 수상세포가 성숙화하는 단계를 포함하는 항암제의 제조방법.
