Description
본 발명은 항균 조성물과 이를 사용하는 방법에 관한 것이다. 구체적으로, 본 발명은 4급 아민류, 산화제 또는 할로겐 화합물로 구성된 실버 디하이드로겐 시트레이트의 조성물 및 이 조성물을 사용하여 다양한 기질에 대해 항균 효과를 부여하는 방법에 관한 것이다.
무수한 항균 화합물이 알려져 있다. 미생물의 번식을 막거나 살균하는 화합물이 다양한 목적으로 사용되어 왔고, 다수의 제품에 사용되어 제품의 유효 기간을 연장시켜 왔고, 최종 사용자들, 이들의 소유물 및 환경에 항균 혜택을 부여하는 데에 사용되어 왔다. 상술한 바와 같이 일부 항균 화합물은 4급 암모늄 화합물, 산화제 및 할로겐 화합물을 포함한다.

과산화수소와 같은 산화제는 살균제로 사용되어 왔다. 과산화수소는 피부 및 상처의 소독 및 급수 장치를 소독하는 살균제로 알려져 있다. 과망간산칼륨(KMnO4)은 냉각탑, 증발 응축기, 공기 정화 장치, 냉각 액체 저장 용기 및 인간 음료수에 대해 살균, 살진균, 살조제(殺藻劑)로 사용되는 산화제이다. 또한, 과망간산칼륨은 수족관 및 관상용 연못에서 어류의 외부 감염을 제어하는데 사용된다. 포타슘 모노퍼설페이트와 같은 모노퍼설페이트는 수영장이나 온천에서 부차적인 소독을 위한 살균제 및 쇼킹 에이전트로 사용된다. 상기 쇼킹 에이전트(shocking agent)로서 사용될 때, 모노퍼슐페이트는 클로라민을 산화 분해시키며, 이에 의해 유리 염소의 유효 농도 혹은 염소 처리수 중의 유효 농도를 증가시킨다. 언급할 수 있는 다른 산화제로는 과산화 벤조일과 과붕산 나트륨이 있는데, 이들 물질도 역시 항균 화합물로 사용된다. 산화제가 매우 효과적인 살균제라고 해도, 자연환경에서 빨리 분해되는 경향이 있어 장기간에 걸쳐 항균효과를 제공하는 데에 적합하지 않다.

본 발명은 실버 디하이드로겐 시트레이트, 시트르산 및 산화제로 구성된 항균 조성물을 사용하는, 경질 표면을 처리하는 다른 방법을 제공한다. 항균 조성물 은 소정 농도의 실버 디하이드로겐 시트레이트 및 산화 화합물을 포함한다. 조성물의 처리를 위해 적합한 실버 디하이드로겐 시트레이트의 농도는 약 0.1 ppm 및 100 ppm 범위이고, 조성물 처리에서 적절한 산화제의 농도는 약 0.2 ppm 내지 20 ppm이다. 상기 항균 조성물은 스테인레스 스틸 혹은 조리대와 같은 경질 표면에 적용된다. 이러한 방법은 미생물 침투로부터 광범위한 잔여물 보호를 제공한다. 일단 산화제가 소산되고 실버 디하이드로겐 시트레이트 잔여물이 존속하면, 확대된 항균력이 경질 표면에 제공되어 어떤 물질도 상기 표면과 접촉하게 될 수 있다.
본 발명은 나아가 실버 디하이드로겐 시트레이트, 시트르산 및 염소 또는 브롬으로 구성되는 항균 조성물을 가지고 식품 아티클이나 비-식품 아티클과 같은 기질을 처리하는 방법을 제공한다. 항균 조성물을 위한 적절한 실버 디하이드로겐 시트레이트의 농도는 적어도 약 0.1, 특히 적어도 약 0.5 ppm이다. 상기 조성물의 처리에서 적절한 염소 또는 브롬의 농도는 적어도 약 0.2 ppm, 특히 약 0.2 내지 20 ppm 범위이다. 조성물의 처리는 경질 표면, 식품 아티클, 또는 비식품 아티클인 기질에 적용된다. 이 방법은 상승작용 있는 항균 효능을 제공한다.

1: 실버 디하이드로겐
시트레이트
저장 용액의 생성
물을 역삼투 유닛에 유입시켜 반투막을 통과시킴으로써 불순물을 제거하고, 탈이온수를 만든다. 순도 99%인 무수 시트르산 물과 섞어서 200 갤런 중 20% 중량 용액(1갤런의 물에 796g의 시트르산)을 만든다. 시트르산 20%인 200 갤런 용액을 양극 및 음극을 가지는 이온 챔버에 넣고, 각 전극은 999 정제 은 200 트로이 온스로 이루어진다. 양극 및 음극은 적어도 2.0 mm 이격되어, 두 전극 사이로 시트르산 용액이 흐르도록 하였다. 양 및 음의 전도체를 포함하는 이온 발생 컨트롤러(IGC) 전력 공급 장치를 양극 및 음극에 부착하였다. 상기 IGC는 1분 간격의 극성 변화와 함께 17 볼트에서 5 암페어 전류를 인가하였고, 매 9초마다 진동시켰다. 공정 전반에 걸쳐, 전극의 간격이 조절되어 5 암페어 17 볼트 출력을 유지하였다. 전류의 흐름으로 인해 양극과 음극 사이에서 이온 흐름을 발생시켰고, 희석된 시트르산 용액 안에 유리 은 이온을 생성하였다. 은 이온은 시트르산 용액 내에서 시트르산과 작용하여 실버 디하이드로겐 시트레이트 용액을 생성하였다. 20% 시트르산 용액을 원하는 은 이온 농도가 얻어질 때까지 분당 50 갤런으로 144 시간 동안 이온 챔버를 통해 재순환시켰다. 다음에, 실버 디하이드로겐 시트레이트 용액을 침전 과정 동안 고체가 형성되도록 하기 위해 가라앉혔다. 결과물은 은 이온의 농도가 2400 ppm인 실버 디하이드로겐 시트레이트 용액이다. 이후, 이 용액은 저장 용액이라 한다. 저장 용액은 하기의 실험예 마다 즉각적으로 사용되거나 나중에 사용을 위해 저장될 수 있다.

실험예 1로부터 얻어진 0.003% 실버 디하이드로겐 시트레이트 저장 용액을 첨가하기 전에 상기 (a)-(b)로부터 얻어진 중간 조성물을 약 15분간 섞었다. 다음, 모든 구성성분을 1 시간의 남은 시간 동안 섞었다. 그 결과 얻어진 조성물(실험 용액 (a)-(d))은 실험예 3에서와 같이 사용된다.

실험예
3으로부터 얻은 조성물의 항균 효과
대장균 실험 균주(E. coli PIPSA, German H.)를 35℃에서 24 시간 동안 배양했다. 세포를 10분간 원심 분리해서 수확하고 버터필드 인산 버퍼 (BPB pH. 7.2)로 두 번 세척했다. 상기 세포를 BPB에서 재현탁하여 대략 108 CFU/ml의 세포현탁액을 얻었다. (목표 접종 레벨은 최종 실험 용액 내에서 대략 106 CFU/㎖였다).
세 세트의 구멍 없는 유리 슬라이드를 준비했다. 각각의 실험 슬라이드 세트에 대해, 하나는 실험예 3의 실험 용액으로 처리하였고, 다른 하나는 실험예 3의 중간 실험용액으로 처리하였다. 각각의 슬라이드에 대장균을 접종하였다.
접종 후, 실험 슬라이드를 1시간 및 24시간 동안 저장하였다. 그 다음, 지시된 시간에 슬라이드를 빼내어 24시간 동안 배양하였다. 통과(pass)라는 기록은 박테리아 성장이 없음을 나타내고, 실패(fail)이라는 기록은 박테리아가 성장하였음을 나타낸다. 하기 표 5의 결과는 실버 디하이드로겐 시트레이트 및 4급 암모늄 화합물의 결합의 잔여 활성을 보여주며, 이 특성은 4급 암모늄 화합물만으로 구성된 조성물에 의해 구비되는 것이 아님을 보여준다.

Claims (17)
- 실버 디하이드로겐 시트레이트, 시트르산 및 4급 암모늄 화합물, 산화제 및 할로겐 화합물로 구성된 군으로부터 선택된 2차 항균제로 구성된 것인 조성물.
- 제 1 항에 있어서, 상기 2차 항균제는 4급 암모니아인 것을 특징으로 하는 조성물.
- 제 1 항에 있어서, 상기 2차 항균제는 산화제인 것을 특징으로 하는 조성물.
- 제 3 항에 있어서, 상기 산화제는 과산화수소, 포타슘 모노퍼설페이트 및 과망간산칼륨으로부터 선택된 것을 특징으로 하는 조성물.
- 제 1 항에 있어서, 상기 2차 항균제는 할로겐 화합물인 것을 특징으로 하는 조성물.
- 제 5 항에 있어서, 상기 할로겐은 염소, 차아염소산, 브롬 혹은 요오드인 것을 특징으로 하는 조성물.
- 제 1 항에 있어서, 분산제를 더 포함하는 것인 조성물.
- 제 7 항에 있어서, 상기 분산제는 계면활성제인 것을 특징으로 하는 조성물.
