Claims (27)
- Ig 융합 단백질을 제조하는 데 있어서, 면역반응에 관여하는 일정 단백질의 유효작용부위를 연쇄체화시킴으로써 연쇄체 형태의 Ig 융합 단백질을 제조하는 방법.
- 제 1항에 있어서, Ig을 코딩하는 유전자와 면역반응에 관여하는 일정 단백질의 유효작용부위를 코딩하는 유전자로부터, 단순 이량체 형태의 Ig 융합 단백질을 코딩하는 유전자를 제조하는 단계;상기 제조된 단순 이량체 형태의 Ig 융합 단백질을 코딩하는 유전자의 서열과 상기 면역반응에 관여하는 일정 단백질을 코딩하는 유전자의 유효작용부위의 서열에 동일한 제한효소 인식서열을 중합효소 연쇄반응을 이용하여 삽입하는 단계;상기 제한효소 인식서열과 일치하는 제한효소를 사용하여 상기 단순 이량체 형태의 Ig 융합 유전자의 서열과 상기 일정 단백질을 코딩하는 유전자의 유효작용부위의 제한효소 인식서열 부위를 절단하는 단계; 및상기 절단된 부분을 라이게이즈로 연결하여 연쇄체 형태의 Ig 융합 유전자를 제조하는 단계를 포함하는 연쇄체 형태의 Ig 융합 단백질을 제조하는 방법.
- 제 2항에 있어서, 상기 연쇄체 형태의 Ig 융합 유전자를 벡터에 삽입하는 단계;상기 연쇄체 형태의 Ig 융합 유전자를 포함하는 벡터를 난소 세포에 도입하여 형질전환체를 제조하는 단계;상기 형질전환체를 배양하는 단계; 및 상기 형질전환체로부터 상기 형질전환체가 생성한 연쇄체 형태의 Ig 융합 단백질을 정제하는 단계를 더 포함하는 연쇄체 형태의 Ig 융합 단백질을 제조하는 방법.
- 제 2항 또는 제 3항에 있어서, 상기 Ig을 코딩하는 유전자는 Ig G1의 Fc 부위를 코딩하는 DNA 단편이고, 상기 Ig에 융합하는 단백질의 세포외역 가용성 부위를 코딩하는 유전자는 TNFR의 세포외역 가용성 부위를 코딩하는 DNA단편으로서, 상기 연쇄체 형태의 이량체 Ig 융합 단백질은 TNFR/Fc 융합단백질인 연쇄체 형태의 Ig 융합 단백질을 제조하는 방법.
- 제 4항에 있어서, 상기 연쇄체 형태의 이량체 Ig 융합 유전자는 서열 5, 7, 9, 및 11에 기재된 염기서열을 갖는 것을 특징으로 하는 연쇄체 형태의 Ig 융합 단백질을 제조하는 방법.
- 제 4항에 있어서, 상기 연쇄체 형태의 이량체 TNFR/Fc 융합 유전자를 벡터에 삽입하는 벡터는 플라스미드 pTR11-Top10'(기탁번호: KCCM 10288), 플라스미드 pTR12-Top10'(기탁번호: KCCM 10289), 플라스미드 pTR21-Top10'(기탁번호: KCCM 10290), 플라스미드 pTR22-Top10'(기탁번호: KCCM 10291)인 것을 특징으로 하는 연쇄체 형태의 Ig 융합 단백질을 제조하는 방법.
- 제 4항에 있어서, 상기 형질전환체는 TR11Ig-CHO (기탁번호: KCLRF-BP- 00046), TR12Ig-CHO (기탁번호: KCLRF-BP-00047), TR21Ig-CHO (기탁번호: KCLRF-BP-00048), TR22Ig-CHO (기탁번호: KCLRF-BP-00049)이고, 상기 정제된 연쇄체 형태의 이량체 TNFR/Fc 융합 단백질은 서열 6, 8, 10, 및 12에 기재된 전체 아미노산 서열을 갖는 것을 특징으로 하는 연쇄체 형태의 Ig 융합 단백질을 제조하는 방법.
- 서열 6에 기재된 아미노산 서열을 갖는 것을 특징으로 하는 단백질.
- 서열 6에 기재된 아미노산 서열을 갖는 단백질을 암호화하는 DNA.
- 제 9항에 있어서, 상기 DNA는 서열 5에 기재된 염기서열을 갖는 것을 특징으로 하는 DNA.
- 제 10항의 DNA 서열을 포함하는 플라스미드 pTR11-Top10'(기탁번호 : KCCM 10288).
- 제 11항의 플라스미드 pTR11-Top10'로 형질전환된 형질전환체 TR11Ig-CHO (기탁번호 : KCLRF-BP-00046).
- 서열 8에 기재된 아미노산 서열을 갖는 것을 특징으로 하는 단백질.
- 서열 8에 기재된 아미노산 서열을 갖는 단백질을 암호화하는 DNA.

- 제 19항에 있어서, 상기 DNA는 서열 9에 기재된 염기서열을 갖는 것을 특징으로 하는 DNA.
- 제 20항의 DNA 서열을 포함하는 플라스미드 pTR21-Top10'(기탁번호 : KCCM 10290).
