Abstract

Description

EP 0317957 에서, Senter 는 항체가 디술피드 벤질 카르바메이트 또는 카르보네이트 링커를 이용해 약물에 결합되고, 디술피드 결합의 환원으로 약물이 방출되는, 약물-항체 전구약물을 기재하고 있다. I. 정의

실시예 1 은 또한, 콘쥬게이트를 형성하기 위한 중간체 화합물로 작용할 수 있는, 오르토-디아실디글리세롤디티오벤즈알콜의 제조 반응 조건에 대해 상술한다.

III. 콘쥬게이트 함유 리포좀의 제조
또다른 측면에서, 본 발명에는 소포형성 지질 및 상기 기재된 콘쥬게이트를 함유하는 리포좀 조성물이 포함된다. 리포좀은 다양한 치료 목적, 특히 리포좀의 전신 투여에 의해 치료제를 표적 위치 또는 세포에 전달하기 위해 사용되는 폐 지질 소포이다. 특히, 친수성 중합체 사슬, 예컨대 폴리에틸렌 글리콜 (PEG) 이 표면에 코팅된 리포좀은, 이러한 리포좀이 중합체가 코팅되지 않은 리포좀에 비해 연장된 혈액 순환수명을 제공하므로, 약물 전달체로서 바람직하다. 중합체는 혈액 단백질에 대한 장벽으로 작용하므로, 단백질의 결합 및 세망내피계의 기타 세포 및 대식세포에 의한 흡수 및 제거를 위한 리포좀 인지를 방지한다.
본 발명에 따른 리포좀에는, 하나의 구현예에서 소포형성 지질인 지질, 및 임의로 기타 이중막 성분과 배합된 콘쥬게이트가 포함된다. "소포형성 지질" 이란수 중에서 자발적으로 이중막 소포를 형성하는 지질이다. 소포형성 지질은 바람직하게는 두 개의 탄화수소 사슬, 전형적으로 아실 사슬, 및 극성 머리기를 갖는다. 당분야에는 다양한 합성 소포형성 지질 및 천연 생성 소포형성 지질이 공지되어 있으며, 두 개의 탄화수소 사슬은 전형적으로 탄소수 약 12 내지 약 24 의 길이이고, 다양한 불포화도를 갖는다. 예에는 인지질, 예컨대 포스파티딜콜린 (PC), 포스파티딜에탄올아민 (PE), 포스파티드산 (PA), 포스파티딜이노시톨 (PI), 및 스핑고미엘린 (SM) 이 포함된다. 본 발명에 사용하기 바람직한 지질은 수소화 콩 포스파티딜콜린 (HSPC) 이다. 또다른 바람직한 지질군은 디아실글리세롤이다. 이러한 지질은 시판되거나, 공개된 방법에 따라 제조될 수 있다.
소포형성 지질은 일정도의 유동성 또는 강성을 얻기 위해, 혈청 중 리포좀의 안정성을 조절하기 위해, 그리고 리포좀 중 트랩핑된 약제의 방출율을 조절하기 위해 선택될 수 있다. 보다 단단한 지질 이중막, 또는 액체 결정성 이중막을 갖는 리포좀은 비교적 단단한 지질, 예컨대 비교적 높은 상전이 온도, 예컨대 약 80℃ 이하를 갖는 지질을 삽입하여 제조할 수 있다. 강성의, 즉 포화된 지질은 지질 이중막의 보다 큰 막 강성에 기여한다. 기타 지질 성분, 에컨대 콜레스테롤도 또한 지질 이중막 구조 내 막 강성에 기여하는 것으로 공지되어 있다.
액체 유동성은 비교적 유동적인 지질, 전형적으로 비교적 낮은 액체에서 액체-결정상으로의 전이 온도, 예컨대 실온 이하 (약 20-25℃) 를 갖는 지질 상을 갖는 것들을 삽입하여 얻어진다.
또한, 리포좀은 지질 이중막 내로 삽입될 수 있는 기타 성분, 예컨대 스테롤을 함유할 수 있다. 이러한 기타 성분은 전형적으로 이중막 내부의 소수성 영역과 접촉하는 소수성 부분, 및 막 외부의 극성 표면쪽으로 배열된 극성 머리기 부분을 갖는다.
본 발명의 리포좀 중 또다른 지질 성분은, 친수성 중합체로 유도체화된 소포형성 지질이다. 상기 지질 성분에서, 유도체화 지질은 지질 이중막 표면의 내부 및 외부 모두에 친수성 중합체 사슬의 표면 코팅을 형성시킨다. 전형적으로, 약 1-20 몰 퍼센트의 유도체화 지질이 지질 조성물 중에 함유된다.
소포형성 지질의 유도체화에 적합한 친수성 중합체에는 폴리비닐피롤리돈, 폴리비닐메틸에테르, 폴리메틸옥사졸린, 폴리에틸옥사졸린, 폴리히드록시프로필옥사졸린, 폴리히드록시프로필메트아크릴아미드, 폴리메트아크릴아미드, 폴리디메틸아크릴아미드, 폴리히드록시프로필메트아크릴레이트, 폴리히드록시에틸아크릴레이트, 히드록시메틸셀룰로오스, 히드록시에틸셀룰로오스, 폴리에틸렌글리콜, 및 폴리아스파르트아미드가 포함된다. 중합체는 동종중합체 또는 블록 또는 랜덤 공중합체로서 사용될 수 있다.
바람직한 친수성 중합체 사슬은, 바람직하게는, 약 500 내지 약 10,000 달톤, 바람직하게는 약 1,000 내지 약 5,000 달톤의 분자량을 갖는 PEG 사슬인, 폴리에틸렌글리콜 (PEG) 이다. PEG 의 메톡시- 또는 에톡시-캡핑된 유사체도 또한 바람직한 친수성 중합체이다. 이러한 중합체는 다양한 중합체 크기, 예컨대 약 12 내지 약 220,000 달톤으로 시판되고 있다.
본 발명의 리포좀에는 전형적으로 약 1 내지 약 30 몰 퍼센트, 바람직하게는약 5 내지 약 30 몰 퍼센트, 보다 바람직하게는 약 5 내지 약 20 몰 퍼센트의 지질-DTB-약물 콘쥬게이트가 포함된다. 본 발명을 지지하기 위해 수행된 연구에서, 소포형성 지질 수소화 콩 포스파티딜콜린 (HSPC), 메톡시-폴리에틸렌 글리콜로 유도체화된 디스테아로일 포스파티딜에탄올아민 (mPEG-DSPE), 및 도 9A 에 나타낸 콘쥬게이트인 파라-디스테아로일-DTB-미토마이신 C (화합물 XVIII) 를 함유하는 리포좀을 실시예 4A-4B 에 기재된 바와 같이 제조하였다. 한 리포좀 제형물은 지질 HSCP/콜레스테롤/mPEG-DSPE/파라-디스테아로일-DTB-미토마이신 C (화합물 XVIII) 가 60/30/5/5 의 몰비로 존재하며, 콜레스테롤을 함유하였다 (실시예 4A). 콜레스테롤을 함유하지 않는 두 번째 제형물을 제조하고 분석하였다 (실시예 4B). 상기 제형물에서, 지질 HSCP/mPEG-DSPE/파라-디스테아로일-DTB-미토마이신 C (화합물 XVII) 는 90/5/5 의 몰 비로 존재하였다.
IV. 콘쥬게이트 함유 리포좀의 시험관 내 특징분석
A. 시험관 내 약물 방출
리포좀을 실시예 4A-4B 에 기재된 바와 같이 제조하고 시험관 내 특징분석하여, 환원제에의 노출에 따른 미토마이신 C 의 방출율을 측정하였다. 시험관 내 연구를 위해, 시험 매질에, 전형적으로 약 150 μM 농도의 시스테인을 첨가하여 환원 조건을 유도하였다. 생체 내, 내인성 환원 조건이 약물 방출을 위한 지질-DTB-약물 콘쥬게이트의 가황분해에 영향을 미치기에 충분할 수 있음이 이해될 것이다. 생체 내 환원 조건을 적합한 환원제, 예컨대 시스테인 또는 글루타티온의 투여에 의해 인공적으로 유도할 수 있음도 더 예상된다.
리포좀 제형물, 예컨대 HSPC/콜레스테롤/mPEG-DSPE/콘쥬게이트인 화합물 XVIII (이하, "콜레스테롤 있는 제형물") 및 HSCP/mPEG-DSPE/콘쥬게이트인 화합물 XVIII (이하, "콜레스테롤 없는 리포좀 제형물") 을 24 시간 동안, 150 μM 시스테인의 존재 하 37℃ 에서 인큐베이션하였다. 시료를 선택한 시점마다 채취하고 고성능 액체 크로마토그래피 (HPLC) 로 분석하여, 콘쥬게이트 및 유리 미토마이신 C 의 양을 정량하였다. HPLC 조건을 실시예 5 에 기재하였다.
도 10A-10B 는 두 개 리포좀 제형물의 HPLC 크로마토그램을 나타낸다. 도 10A 에서는, 콜레스테롤 없는 리포좀 제형물의 결과를 나타낸다. 시점 0 에서는, 측정가능한 유리 미토마이신 C 가 없고, 측정가능한 모든 약물은 리포좀에 결합한 지질-DTB-약물 콘쥬게이트의 형태이다. 인큐베이션 시간이 증가할수록, 콘쥬게이트 결합 미토마이신 C 의 존재에서 상응하는 감소와 함께 리포좀에서 방출되어 유리 형태로 측정가능한 미토마이신 C 의 양이 증가한다.
도 10B 는 콜레스테롤 있는 리포좀 제형물의 결과를 나타낸다. 시점 0 에서 채취한 첫번째 시료에는, 측정가능한 유리 미토마이신 C 가 없었다. 150 μM 시스테인의 존재 하에서 1 시간 인큐베이션한 후, 리포좀 결합 지질-DTB-미토마이신 콘쥬게이트의 분해를 시사하는 소량의 유리 약물이 검출되었다. 도 10A 와 대조하여, 콜레스테롤 있는 리포좀은 보다 느린 콘쥬게이트 분해율을, 따라서 보다 느린 약물의 방출을 나타낸다.
도 11 은 도 10A-10B 의 크로마토그램에서 측정된 바와 같은, 두 개의 리포좀 제형물에서 방출된 미토마이신 C 의 퍼센트를 나타내는 그래프이다. 콜레스테롤 없는 리포좀 (검은 다이아몬드) 이 콜레스테롤 있는 리포좀 (검은 원) 보다 높은 방출율을 가졌다. 콜레스테롤 없는 제형물에서는, 2 시간 후 미토마이신 C 의 50% 초과가 리포좀 결합 콘쥬게이트로부터 방출되었다. 두 가지 제형물에 대해, 24 시간 인큐베이션 기간의 말기에는 약물의 80% 초과가 방출되었다.
또다른 연구에서는, 두 가지 리포좀 제형물을 1.5 mM 시스테인 중에 인큐베이션하였다. 실시예 5 에 기재된 바와 같이 분석을 수행하여, 결과를 도 12A-12B 에 나타낸다. 도 12A 는 콜레스테롤 없는 리포좀 내에 삽입된 지질-DTB-약물 콘쥬게이트 (HSPC/PEG-DSPE/지질-DTB-미토마이신 C) 로부터 방출된 미토마이신 C 의 퍼센트를 나타낸다. 대조를 위해, 150 μM 과의 인큐베이션 중 방출 퍼센트 (검은 다이아몬드) 를 또한 나타낸다. 보이는 바와 같이, 더 높은 농도의 환원제 (1.5 mM, 흰 다이아몬드) 와 인큐베이션하여, 콘쥬게이트 분해율 및 약물 방츌율이 증가한다.
도 12B 는 콜레스테롤 있는 리포좀 제형물의 결과를 나타낸다. 1.5 mM 에서 인큐베이션한 리포좀 (흰 원) 은 150 μM 시스테인에서 인큐베이션한 동일 리포좀 (검은 원) 보다 상당히 높은 분해율을 나타낸다.
B. 시험관 내 세포독성
지질-DTB-미토마이신 C 콘쥬게이트 (화합물 XVIII) 함유 리포좀의 시험관 내 세포독성을 마우스 폐 육종주인 M-109 세포를 이용하여 평가하였다. 실시예 6 에 기재된 바와 같이, M109 세포를 유리 미토마이신 C 또는 디스테아로일-DTB-미토마이신 C 콘쥬게이트 함유 리포좀의 존재 하에서 인큐베이션하였다. 실시예 6A 에명시된 몰 비로 실시예 4A-4B 에 기재된 바와 같이 제조된 리포좀을 시험하였다. 150 μM, 500 μM 및 1000 μM 농도의 시스테인을 일부 시험 세포에 첨가하여 콘쥬게이트의 가황분해 및 미토마이신 C 의 방출을 일으켰다.
실시예 6 에 기재된 바와 같이, IC50 값을 대조군 성장율을 50% 억제시키는 약물 농도 (IC50) 로 취하였다. 결과를 표 1 에 나타낸다.

세포독성 연구에서 결정된 M109 마우스 육종 세포의 성장율 퍼센트를 도 13 에 나타낸다. 성장율 퍼센트는 미토마이신 C 및 시스테인의 부재 하 M109 세포의 성장율을 기준으로 한 퍼센트로 표현되며, 미토마이신 C 농도 (nM) 의 함수로 나타낸다. 세포의 성장율은 실시예 6 에 기재된 바와 같이 결정하였다. 보이는 바와 같이, 콜레스테롤 있는 리포좀 (흰 원) 및 콜레스테롤 없는 리포좀 제형 (검은 사각형) 둘 다에서 시스테인의 농도가 증가함에 따라 세포 성장율 퍼센트는 감소한다. 또한, 시스테인은 유리 미토마이신의 활성에 영향을 미치지 않으며, 미토마이신 C 는 콘쥬게이트에서 방출되어 세포 성장을 효과적으로 억제함을 알 수 있다.
유리 형태의 미토마이신 C 또는 리포좀 결합 지질-DTB-약물 콘쥬게이트의 형태인 미토마이신 C 로 처리한 M109 마우스 육종 세포의 시험관 내 성장율을 도 14A-14B 에 나타낸다. 도 14A 에서는, 콜레스테롤 없는 리포좀 제형물의 결과를 나타낸다. 그래프에서, M109 세포의 성장율은 약물 및 시스테인의 부재 하 M109 세포의 성장을 기준으로 한 퍼센트로 표현되며, 미토마이신 C 농도 (nM) 의 함수로 나타낸다. 유리 형태의 미토마이신 C (흰 삼각형) 및 유리 형태의 미토마이신 C 에 더해 1000 μM 의 시스테인 (검은 삼각형) 을 처리한 세포는 유리 형태 약물의 독성으로 인해 성장율 감소를 나타낸다. 도 14B 는 콜레스테롤 있는 리포좀 제형물에 대한 유사 그래프이다. 콜레스테롤에 더해 150 μM (흰 다이아몬드), 500 μM (검은 원) 및 1000 μM (흰 사각형) 농도의 부가적 시스테인을 함유하는 리포좀 조성물로 처리한 세포에서 동일한 양식이 관찰되었다. 즉, 시스테인 농도가 증가함에 따라, 세포 성장율이 감소하였다. 이는 시스테인이 유도한 미토마이신 C 의 방출이 시스테인 농도와 직접적인 상관관계에 있음을 시사한다. 리포좀 제형물과는 대조적으로, 유리 형태의 미토마이신 C (흰 삼각형) 로 처리한 세포의 시험관 내 성장율은 유리 형태의 미토마이신 C 에 더해 1000 μM 시스테인 (검은 삼각형) 을 처리한 세포의 성장율과 동일하였다.
도 15 는 유리 미토마이신 C 및 리포좀 제형물의, 시스테인 농도 (μM) 의함수로서의 세포독성의 증가 퍼센트를 나타낸다. 세포독성의 증가는 IC50 의 감소 퍼센트, 예컨대 시스테인의 부재 하 IC50 에 대한 시스테인의 존재 하 IC50 곱하기 100 ((IC50시스테인 없는 경우/IC50시스테인 있는 경우)x100) 으로 측정되었다. 보이는 바와 같이, 세포독성 퍼센트는, 콜레스테롤 있는 리포좀 (흰 삼각형) 및 콜레스테롤 없는 리포좀 제형물 (검은 원) 둘 다에서, 시스테인의 농도가 증가함에 따라 현저히 증가한다. 유리 미토마이신 C (검은 사각형) 의 세포독성은 시스테인의 존재에 영향받지 않는다.
세포독성 데이타는 콜레스테롤 없는 리포좀 제형물이 시스테인에 의해 보다 많이 영향받음을 나타낸다. 특정 시스테인 농도에서의 콜레스테롤 없는 리포좀 제형물의 IC50 는 단독 유리 약물보다 단지 2 배 더 낮다. 콜레스테롤 있는 리포좀 제형물은 콜레스테롤 없는 리포좀 제형물보다 세포독성이 적다. 또한 데이타는, 시스테인이 종양 세포에 세포독성 효과를 갖지 않으며, 유리 미토마이신 C 의 세포독성에 영향을 미치지 않음을 나타낸다. 또한, 데이타로부터, 시스테인이 리포좀 결합 미토마이신 C 의 세포독성을 용량 의존적 방식으로 증가시킴이 현저하다. 따라서, 리포좀 제형물에서 관찰되는 세포독성 효과는 지질-DTB-약물 콘쥬게이트로부터 시스테인이 매개한 미토마이신 C 의 방출에 의해 대부분 설명된다.
C. 생체 내 약동역학
콜레스테롤 있는 리포좀 및 콜레스테롤 없는 리포좀 제형물의 생체 내 약동역학을 래트에서 측정하였다. 실시예 7 에 기재된 바와 같이, 동물들에 유리 형태의 미토마이신 C 또는 본 발명에 따른 지질-DTB-미토마이신 C 콘쥬게이트 형태의 리포좀에 삽입된 미토마이신 C 약 0.1 mg/mL 를 단회 볼루스 정맥주사하였다. 주사 후, 혈액 시료를 채취하여, 미토마이신 C 의 양을 분석하였다. 결과를 도 16A-16B 에 나타낸다.
도 16A 는 정맥 주사 후, 시간의 함수로서 래트의 혈액 중 미토마이신 C 의 농도 (㎍/mL) 를 나타낸다. 보이는 바와 같이, 유리 형태로 정맥 투여한 유리 미토마이신 C (흰 사각형) 는 혈액으로부터 신속히 제거된다. 리포좀 결합 지질-DTB-약물 콘쥬게이트의 형태인 미토마이신 C 는 상당히 더 긴 시기동안 순환하며 남아 있다. 콜레스테롤 있는 리포좀 (검은 다이아몬드) 및 콜레스테롤 없는 리포좀 (검은 원) 과 연합된 미토마이신 C 는 혈액 중에서 20-25 시간 동안 10 ㎍/mL 초과로 검출되었다.
도 16B 는 시험 제형물의 정맥 주사 후, 시간의 함수로서 혈액 중 잔여 주사 용량의 퍼센트를 나타낸다. 실제로 유리 미토마이신 C (흰 사각형) 의 용량은 약 5 분 초과 시점에서 혈액 중에 남아 있지 않았다. 그러나, 리포좀 제형물의 주사 20 시간 후, 미토마이신 C 용량의 약 15-18 퍼센트가 순환하며 남아 있었다. 이는 순환 중인 리포좀 내에서 미토마이신 C-DTB-지질 콘쥬게이트가 안정하게 남아있고, 혈장 중에서 최소한의 가황분해적 절단이 일어남을 시사한다. 따라서, 상기 시스템이 연장된 혈액 순환수명 및 종양 내 증가된 축적을 나타내는 장기 순환 리포좀 (스텔스리포좀) 과 상용성인 것으로 나타났다.
V. 실시예
하기 실시예는 본원에 기재된 본 발명을 보다 예시하며, 본 발명의 범위를 한정하는 것이 아니다.
재료
모든 재료는 상업적 공급자, 예컨대 Aldrich Corporation 에서 구매하였다.
실시예 1
파라-디아실디글리세롤디티오벤즈알콜 (화합물 IV) 및 오르토-디아실디글리세롤디티오벤즈알콜의 합성
A. 파라-디아실디글리세롤디티오벤즈알콜
본 반응은 도 1 에 예시되어 있다. Snyder, W.R. (Journal of Lipid Research, 28:949 (1987)) 의 방법에 따라 화합물 II 및 III 을 제조하였다.
물 5 ml 중 3-메르캅토-1,2-프로판디올 (화합물 I, 1 g, 9.26 mmol) 을 함유하는 100 ml 둥근바닥 플라스크를 얼음조에 위치시켰다. 상기 신속 교반 플라스크에, 온도를 30-40℃ 로 유지하면서 과산화수소 (정확히 0.5 몰 당량, 525 ㎕, 4.63 mmol) 을 적가하였다. 발열 단계의 말기에, 반응물을 실온에서 하룻밤동안 교반하였다. 아세토니트릴을 20 ml 양씩 연속 첨가하여, 회전 증발로 물을 공비반응시켰다. 아세토니트릴 첨가 공정을 3-4 회 또는 물이 모두 제거될 때까지 반복하여, 깨끗한 오일을 얻었다. 금속 스파툴라로 플라스크를 긁어내고 -20℃ 에서 하룻밤동안 냉각시킨 후, 오일성 산물을 고형화하였다 (화합물 II, rac-3,3'-디티오비스(1,2-프로판디올)). 백악질의 고체를 P2O5상에서 진공 건조하였다.
화합물 (980 mg, 4.6 mmol) 을 오븐 건조된 100 mL 둥근바닥 플라스크에 첨가하고 건조 염화메틸렌 (40 mL) 중에 용해시켜, rac-3,3'-디티오비스(1,2-프로판디올) 산물 (화합물 II) 을 아실화시켰다. 상기물에, 스테아르산 (4.92 g, 17.1 mmol) 및 4-디메틸아미노)피리디늄 4-톨루엔술포네이트 (1.38 g, 4.6 mmol) 를 촉매로서 첨가하여 실온 (25℃) 에서 20 분 동안 교반하였다. 이어서, 디이소프로필카르보디이미드 (3.1 mL, 20 mmol) 을 피펫팅하여 실온에서 하룻밤동안 반응시켰다. (rac-3,3'-디티오비스(1,2-프로판디올) Rf=0.60; rac-3,3'-디티오비스(1,2-프로판디스테아로일) Rf=0.35). 약염기성 이온교환 수지인 AmberlystA-21 (~3 g) 및 강산성 이온교환 수지인 Amberlyst15 (~ 3 g) 를 반응 혼합물에 첨가하였다. 30 분 동안 교반한 후, 수지를 여과하고 여과액을 건조시켰다. 잔류물을 이소프로판올로부터 3 회 (100 mL 씩) 재결정화시켰다. 고체 산물, rac-3,3'-디티오비스(1,2-프로판디스테아로일) (화합물 III) 을 수합하여 P2O5상에서 건조하였다.
다음 단계에서, rac-3,3'-디티오비스(1,2-프로판디스테아로일) (화합물 III) (2.97 g, 2.33 mmol) 의 용액을 톨루엔 (30 mL) 중에 용해시켜 얼음 조에 위치시켰다. 염화술푸릴 (1.9 mL, 23.2 mmol) 을 플라스크 내로 피펫팅하고 혼합물을 차가운 얼음조 온도에서 30 분 동안 교반하였다. 이어서, 플라스크를 실온에 위치시키고 다시 30 분 동안 교반하였다. 과량의 염화술푸릴을 회전 증발기로 제거하였다. 신선한 톨루엔 일정량 (20 mL) 을 반응 플라스크에 첨가하여 얼음조에 위치시켰다. 상기물에, 톨루엔 중 4-메르캅토벤즈알콜 (780 mg, 5.6 mmol) 용액을 느린 속도로 첨가하였다. 5 시간의 반응 시간 후, 모든 용매를 회전 증발로 증발시켜 건조하였다. 따뜻한 에틸 아세테이트 (10 mL) 를 반응 플라스크에 첨가하여 고체를 용해시키고 불용성 물질을 여과하였다. 에틸 아세테이트 용액에, 에테르 50 mL 을 첨가하여 침전시키고, 고체 산물 (파라-디아실-디글리세롤-디티오벤잘알콜, 화합물 IV) 을 여과로 수합하였다. 상기 공정을 2 회 반복하였다. 수율: 75%.
산물 (파라-디아실-디글리세롤-디티오벤잘알콜, 화합물 IV) 을 정제하기 위해, 클로로포름 중 실리카 겔 칼럼 (20 x 2.5 cm) 을 제조하였다. 시료를 최소량의 클로로포름 중에 용해시키고, 두 개의 상이한 이동상을 첨가하여 크로마토그래피하였다. 일차로, 100% CHCl3(100 ml) 를 용출하였다. 상기 분획에는 불순물인 디티오벤질 알콜이 함유된다.1HNMR 로 확인하였다. 이어서, 이동상을 클로로포름 중 15% 메탄올로 변화시켜, 순수한 산물을 플래시 크로마토그래피로 수합하였다. CH3OH:CHCl3(15:85) 500 ml 로 용출하여, 순수한 DGTBA (TLC 상에 한 점임) 를 수합하였다. 용매를 증발시킨 후, 고체를 t-BuOH 로부터 동결건조하여 P2O5상에서 진공 건조하였다. 최종 정제로 수율이 40%, 1.4 g 으로 감소하였다.

실시예 2
파라-디아실디글리세롤디티오벤잘-미토마이신 C (화합물 XVIII) 의 합성
본 반응을 도 6A 에 예시한다.
50 mL 둥근바닥 플라스크를 포스겐 (3.1 mmol) 및 톨루엔 (5 mL) 으로 충진하고 용액을 0℃ 로 냉각하였다. 톨루엔 (2.5 mL) 중 파라-디아실-디글리세롤-디티오벤잘-알콜 (화합물 IV, 실시예 1 에 기재된 바와 같이 제조함, 0.31 mmol) 의 용액을 제조하였다. 이어서, 알콜 용액을 포스겐 용액에 적가하였다. 혼합물을 하룻밤동안 실온으로 가온시켰다. 18 시간 후, 용액을 진공 농축하여 과량의 포스겐을 제거하였다. 조 염화아실를 톨루엔 (5 mL) 중에 재용해시켰다.
미토마이신 C (0.31 mmol), 디메틸아미노피리딘 (0.031 mmol) 및 DMF (1 mL) 의 용액을 제조하였다. 미토마이신 C 용액을 염화아실 용액에 적가하였다. 1 시간 후, 톨루엔을 증발제거하고 조 산물을 실리카 상에서 크로마토그래피하였다 (1:1 헥산: 에틸 아세테이트). 이어서, 정제 산물을 t-BuOH (50 mL) 에 취하여 동결건조하였다. 산물은 자주색 고체였다 (183 mg, 53%). Rf= 0.38 (50% 헥산:에틸아세테이트);
실시예 4
리포좀 제조
A. 콜레스테롤 있는 리포좀
1. 리포좀 제조
HSPC 59 mg, 콜레스테롤 14.4 mg, mPEG-DSPE 17.4 mg 및 파라-디스테아로일-DTB-미토마이신 C 7.4 mg (몰 비 60/30/5/5) 를 60-65℃ 에서 탈수 에탄올 1 mL 에 첨가하고 용해될 때까지, 약 10 분 동안 혼합하였다.
증류수 중 10 mM 히스티딘 및 150 mM NaCl 로 구성된 수화 배지를 70℃ 로 가온시켰다.
따뜻한 지질 용액을 혼합하면서, 따뜻한 (63-67℃) 수화 매질에 신속히 첨가하여, 불균질한 크기를 갖는 리포좀 현탁액을 형성하였다. 현탁액을 63-67℃ 에서 1 시간 동안 혼합하였다.
2. 압출
테플론을 붙인 스테인레스 스틸 용기 내에 장착된 폴리카르보네이트 필터 카트리지를 통해 조절 압출시켜, 리포좀을 원하는 평균 입자 지름으로 크기를 맞추었다. 6-8 시간의 기간인 압출 공정 동안, 리포좀 현탁액을 63-65℃ 에서 유지하였다.
3. 투석여과
투석여과에 의해, 에탄올을 리포좀 현탁액으로부터 제거하였다. 무균수에 히스티딘 (10 mM) 및 염화나트륨 (150 mM) 을 용해시켜, 히스티딘/염화나트륨 용액을 제조하였다. 용액의 pH 를 약 7 로 조절하였다. 용액을 0.22 ㎛ Durapore 필터를 통해 여과하였다. 리포좀 현탁액을 히스티딘/염화나트륨 용액으로 약 1:1 (v/v) 비로 희석하여, 폴리술폰 중공-섬유 울트라필터를 통해 투석여과하였다. 히스티딘/염화나트륨 용액에 대해 8 배 부피의 교환을 수행하여 에탄올을 제거하였다. 공정 유동 온도를 약 20-30℃ 로 유지하였다. 총 투석여과 시간은 약 4.5 시간이었다.
4. 무균 여과
리포좀 현탁액을 33-38℃ 로 가열하고, 0.2 ㎛ Gelman Supor 폴리에테르술폰 필터를 통해 여과하였다. 총 여과 시간은 약 10 분이었다.
각 공정 단계 (수화, 압출, 투석 및 여과) 후, 지질 농도 및 콘쥬게이트/약물 농도를 HPLC 로 측정하였다. 리포좀 입자 크기는 다이나믹 광산란으로 측정하였고, 외부 현탁액 매질 중 "유리", 비결합 미토마이신 C 의 양은 HPLC 로 측정하였다.

B. 콜레스테롤 없는 리포좀 제형물
상기 기재된 바와 같이, 리포좀을 90/5/5 의 몰비의 HSPC, mPEG-DSPE 및 파라-디스테아로일-DTB-미토마이신 C 지질 조성으로 제조하였다. 구체적으로, HSPC 88.5 mg, mPEG-DSPE (PEG MW 2000 달톤) 17.9 mg 및 콘쥬게이트 7.3 mg 을 에탄올 1 mL 에 용해시켰다. 리포좀 크기, 지질 및 약물 농도, 및 외부 현탁액 매질 중 유리 미토마이신 C 농도를 각 공정 단계 후에 측정하였다.

시험관 내 특징분석을 위한 HPLC 조건

실시예 6
세포독성 연구
A. 리포좀 제조
실시예 4A-4B 에 기재된 바와 같이 제조된 리포좀은 HSPC/mPEG-DSPE/디스테아로일-DTB-미토마이신 C (90/5/5) 또는 HSPC/콜레스테롤/mPEG-DSPE/디스테아로일-DTB-미토마이신 C (90/45/5/5) 로 구성되었다. 리포좀 제제를 0.45 ㎛ 셀룰로오스 막을 통해 무균 여과하고, 압출을 통해 크기를 줄이지 않았다. 리포좀을 제형화한 후, 미토마이신 C 농도를 이소프로판올에 10-20 배 희석시켜 가용화된 리포좀 중 360 nm 에서의 흡광도로 측정하고, 인지질 농도를 무기 포스페이트 분석으로 측정하였다.
콜레스테롤 있는 리포좀은 275 ±90 nm 의 평균 지름을 가졌다. 콜레스테롤 없는 리포좀은 150 ±50 nm 의 평균 지름을 가졌다. 두 가지 리포좀 제형물에서 인지질 농도는 10 μM/mL 이었고, 두 가지 제형물에서 미토마이신 C 의 농도는 120 ㎍/mL 이었다.
B. 화학민감성 분석 및 성장율 측정
유리 미토마이신 C 또는 리포좀 내에 삽입된 디스테아로일-DTB-미토마이신 C 콘쥬게이트 형태인 미토마이신 C 의 세포독성 효과를 이전 (Horowitz, A.T. 등, Biochem. Biophys. Acta, 1109:203-209 (1992)) 에 기재된 메틸렌 블루 염색 방법을 약간 변형한 발색측정으로 분석하였다. 분석 종료 시, 세포를 고정하고, 메틸렌 블루 염색 분석을 이용하여 평가하였다.
분석에서, 200 ㎕ 량의 배양액 (RPMI-1640 배지 + 10% 우 태아 혈청) 에서 지수적으로 성장하는 1500 개의 M109 마우스 육종 세포를 96 웰 평편바닥 마이크로타이터 플레이트 상에 플레이팅하였다. 세포가 부착하여 성장을 개시하는 동안인 배양 20 시간 후, 시험 제형물 (유리 미토마이신 C 또는 리포좀 제형물) 20 ㎕ 를 각 웰에 첨가하였다. 약물 농도가 10 배씩 증가할 때마다, 4 가지 약물 농도점을 시험하였다. 각 시험을 세 개 웰씩, 두 개의 평행 플레이트 상에서 수행하였다. 세포를 72 시간 동안 연속 처리하였다.
72 시간 처리 기간 후, 각 웰에 2.5% 글루타르알데히드 50 ㎕ 를 10 분 동안 첨가하여 배양물을 고정시켰다. 탈이온수로 3 회, 0.1 M 보레이트 완충액 (pH 8.5) 으로 1 회 플레이트를 세척한 후, 실온 (20-25℃) 에서 메틸렌 블루 (pH 8.5, 0.1 M 완충 보레이트 중의 1 %) 100 ㎕ 로 60 분 동안 염색하였다. 플레이트를 탈이온수의 5 개 조에서 헹구어, 세포에 결합하지 않은 염료를 제거한 후 건조하였다. 37℃ 에서, 0.1 N HCl 200㎕ 로 60 분 동안 염료를 추출하고, 마이크로플레이트 분광계를 이용하여 광학 밀도를 측정하였다.
세포 수는 분광계의 흡광도와 잘 연관되는 혈구계로 세포를 계수하여 측정하였다. 초기 세포 플레이팅 밀도는 연구 말기의 세포 수 및 흡광도 간 선형비례 관계가 확실시 되도록 선택하였다. 각 연구에서, 약물을 첨가하기 전에 6 개 웰을 고정시켜, 초기 평균 흡광도를 결정하였다. 상기 값을 사용하여, 하기 공식을 이용한 대조군 및 약물 처리 세포의 성장율 (GR) 및 배가 시간 (DT) 를 계산하였다: DT = ln2/ln[(ODt/ODc)/h]; 여기서, DT = 배가 시간 (시간); ODt= 연구 말기의 시험 웰의 광학 밀도; ODc= 연구 개시의 대조군 웰의 광학 밀도; h = 인큐베이션 기간 (시간) 이다.
성장율은 GR = (ln2/DT) 로 계산하였다. 성장 억제 퍼센트 또는 성장율 조절 퍼센트는, 약물 처리 세포의 성장율을 비처리한 대조군 세포의 성장율로 나누어 얻었다. 대조군 성장율의 50% 억제를 일으키는 약물 농도 (IC50) 는, 성장 억제 곡선의 가장 가까운 두 값을 내삽하여 계산하였다.
10-8- 10-5M 범위의 미토마이신 C 를 분석하였다. 10-8- 3 x 10-5M 범위의 콘쥬게이트 결합 리포좀 제형물을 분석하였다. 상호작용 연구를 위해, 시스테인 (SIGMA, St. Louis, MO) 을 미토마이신 C 또는 리포좀 제형물과 함께, 150, 500, 또는 1000 μM 의 최종 농도로 첨가하였다.
결과를 표 1 및 도 13, 14 및 15A-15B 에 나타낸다.
실시예 7
생체 내 약동역학 연구
A. 리포좀 제형물
콜레스테롤 있는 리포좀 및 콜레스테롤 없는 리포좀을 실시예 5A 및 5B 에 기재된 바와 같이 제조하였다.
에탄올 119 ㎕ 에 미토마이신 C 11.9 mg 을 용해시켜 유리 형태의 미토마이신 C 용액을 제조하였다. 용해 후, 10 mM 히스티딘/150 mM 식염수 용액 약 11.8 ㎕ 을 첨가하였다. 사용 전에, 미토마이신 C 용액을 히스티딘/식염수 용액 100 ㎍/mL 로 희석하여 여과하였다.
B. 동물
8 마리의 래트를 무작위로 하기와 같은 처리군으로 사용하였다:

시험 제형물의 단일 정맥 주사를 볼루스 용량으로 투여하였다. 혈액 시료를 주사 후 하기 시간에 각 동물로부터 채취하였다: 30 초, 15 분, 30 분, 1 시간, 2 시간, 4 시간, 8 시간, 24 시간, 48 시간, 72 시간 및 96 시간. 혈액 중 미토마이신 C 의 양을 하기 주어진 HPLC 절차에 의해 측정하였다. 7.5% EDTA 5.1 ml 중에 요오도아세트아미드 199.3 mg 을 넣어 200 mM 요오도아세트아민 용액을 제조하였다. 200 mM 요오도아세트아미드 용액 15 ㎕ 를 각 혈액 시료 1 ㎕ 에 넣었다.
C. 혈장 중 미토마이신 C 를 측정하기 위한 HPLC 방법
1. 용액 제조
탈이온수로 채운 1 L 부피측정 플라스크 내에 인산암모늄 1.321 g 을 넣어 pH=7, 10 mM 인산암모늄을 함유하는 수성 완충액을 제조하였다. 혼합물을 교반하고, o-인산으로 pH 를 7 로 조절하였다. 완충액을 사용 전에 0.45 ㎛ 나일론 필터를 통해 여과하였다.
메탄올 이동상 및 수성 완충액을 Waters Alliance 이중 펌프를 이용한 농도구배 프로그램을 통해 혼합하였다.
2. 표준 용액 및 품질 조절 시료의 제조
2 개 개별 중량의 미토마이신 C 및 미토마이신 C 콘쥬게이트를 표준 및 품질 조절 시료로서 제조하였다. 미토마이신 C 및 미토마이신 C 콘쥬게이트 1 mg 을 계량하여 개별적으로 희석제 (20% 클로로포름 및 80% 메탄올 혼합물) 1 mL 에 용해시켰다. 두 화합물의 스톡 용액 농도는 1 mg/mL 이었다. 희석제 중에 여러 희석물을 제조하여, 표준 및 품질 조절 시료용으로 5 ㎍/mL 내지 100 ㎍/mL 농도를 얻었다.
래트 혈장 0.1 mL 량에 적절한 부피 (10 ㎕-50 ㎕) 의 미토마이신 C 및 미토마이신 C 콘쥬게이트 표준 용액을 넣었다. 미토마이신 C 및 미토마이신 C 콘쥬게이트에 대한 농도 범위는 각각 0.05-5.0 ㎍/mL 및 0.1-5 ㎍/mL 이었다. 메탄올로 최종 부피를 1 mL 로 조절하였다. 유사한 절차를 따라, 품질 조절 시료를 제조하였다. 래트 혈장 중, 품질 조절 시료의 농도는 미토마이신 C 가 0.1, 0.5 및 5 ㎍/mL 및 미토마이신 C 콘쥬게이트가 0.1, 1 및 5 ㎍/mL 이었다. 시료를 실온에서 10 분 동안 3,000 rpm 에서 회전침전시켰다. 상청액 300 ㎕ 를 주사용 삽입물 300㎕ 가 들어있는 HPLC 바이알에 옮겼다.
3. 시료 제조
혈장 시료 100 ㎕ 를 메탄올 900 ㎕ 로 변성시킨 후, 3,000 rpm 에서 10 분 동안 원심분리하였다. 상청액 300 ㎕ 양을 주사용 삽입물 300 ㎕ 가 들어있는 HPLC 바이알에 옮겼다.
4. 크로마토그래피 조건
SupelcoC-8, 5 μ, 4.6 mm x 5 cm 칼럼을 사용하였다. 이동상 A 는 pH 7, 10 mM 인산암모늄이었다. 이동상 B 는 메탄올이었다. 유속은 1 mL/분이었고, 360 nm 에서 UV 로 검출하였다. 주사액 부피는 40 ㎕ 이었고, 통상 수행 시간은 15 분이었다. 농도구배 프로그램은 하기와 같았다:

5. 분석 및 계산
저농도에서 고농도로 제조된 직선상 표준물 (6 개 농도 레벨) 을 주사하였다. 이어서, 품질 조절 및 혈장 시료를 분석을 위해 주사하였다.
PE-Nelson Turbochrom (4.1 버전) 시스템으로, 피크 넓이 및 정체 시간을 측정하였다. 직선 회귀 프로그램을 이용하여 미토마이신 C 및 미토마이신 C 콘쥬게이트 의 농도를 계산하였다. 6 개 농도 레벨로부터의 표준 반응으로 직선화 방법을 평가하였다. 데이타는 1/x2의 중량 인자로, 직선 회귀 공식 y=B*x + A 로 맞춰졌다. 품질 조절 시료뿐만 아니라 표준물의 농도를 역계산하여, 상기 방법의 정확도 및 신뢰도를 평가하였다.
결과를 도 16A-16B 에 나타낸다.
본 발명을 특정 구현예에 대해 기재하였으나, 당업자에게는 본 발명에서 벗어나지 않는 다양한 변화 및 변형이 만들어질 수 있음이 자명할 것이다.

- 제 1 항에 있어서, 치료 약물이 우레탄, 아민, 아미드, 카르보네이트, 티오-카르보네이트, 에테르 및 에스테르로 구성된 군으로부터 선택되는 결합으로 공유 부착된 콘쥬게이트.
- 제 1 항에 있어서, L 이 콜레스테롤, 디아실글리세롤, 인지질 및 그의 유도체로 구성된 군으로부터 선택되는 콘쥬게이트.
