Abstract
CD6 항체를 인식하는 단일세포 항체에서 약제 제조법은 건선과 같은 형태의 병을 갖고 있는 환자들에게 효과적으로 임상적이고 조직학적으로 치료할 수 있다.
Description
도 1 : ior t1A mAb의 헤비 체인의 변수 영역을 인간에게 적용함으로서 변형된 분석.
도 2 : ior t1A mAb의 라이트 체인의 변수 영역을 인간에게 적용함으로서 변형된 분석.
도 3 : 건선 벌거를 갖는 환자의 장애 부위에서 염증이 침투한 특성을 갖는 림프구의 안티 CD6 항체를 표현한 결과표.
도 4 : 잇따르는 84일 이후 건선 벌거를 갖는 환자의 안티 CD6 단일세포 항체(ior t1)로 국부 치료를 한 임상 반응을 평가한 도면.

(3.14). 시간 0에서 스케일(scale)의 디멘션(dimension)은 100%이고, 스케일의 디멘션에서 다음 퍼센트를 평가하는 것은 비례적으로 이루어진다.
PASI(psoriasis area and severity index)(Freddiksson, T. et al. (1978) Dermat ologica 157:238)와 유사한 PSORIASIS SEVERITY SCORE(PSS)는 다음의 식에 의해 만들어진다.
치료에 대한 반응은 평가시간이 완전해질 때 PSS 내의 변화와 일치하여 추출되고, 다음의 카테고리에 따라 평가할 수 있다(Perkins, W. et al. (1993) Br. J. Dermatol. 129:584):
-소거 (PSS 내에서 90% 이상의 향상)
-적응 (PSS 내에서 50% 이상의 향상)
-비-적응 (PSS 내에서 50% 이하의 향상 또는 악화)
-악화 (PSS 내에서 50% 이상 증진)
응답을 위한 평가시간은 치료약을 처음 투약한 날부터 12주로 설정할 수 있다(사전 처리는 1, 2, 3, 4, 6, 8, 12주).
실시예1: ior t1A 모노클로니얼(MONOCLONAL) 항체 DNA 시퀀싱의 쥐 변수 영역 세포질 RNA는 Faloro et al(Foloro, J. et al. (1989) Enzimology의 방법 65:718)에서 상술된 바와 같이 106T1의 하이브리도마 세포로부터 추출된다.
-cDNA 합성 반응은 50μl의 전체 양 중에서 5μg의 RNA, 50 mM의 Tris-HCL, pH 7.5, 75 mM의 KCL, 10 mM의 DTT, 3 mM의 MgCl2, 헤비(heavy) 체인 변수 영역을 위한 25pmol의 CG2AFOR 프리머 (5'GGAAGCTTAGACCGATGGGGCC TGTTG TTTTG 3') 또는 라이트 (light) 체인 변수 영역을 위한 CK2FOR (5'GGAAGCTT GAAGATGGATACAGTTGGTGCAGC 3'), 각각 250 uM 씩의 dTP, aTTP, aCTP, aGTP, 15 u의 리보뉴클레아제 억제물(RNA 가드, 제약자)로 구성된다.
샘플은 70℃에서 10분 동안 가열되고, 30분 동안 서서히 37℃로 떨어지게 한다. 그러면 100 유니트의 MMLV 역 전사효소(BRL)는 37℃에서 1시간 동안 연속적으로 추가되고 인큐베이션된다.
VH 및 VK cDNAs는 Orlandi et al(Orlandi, R. et al. Proc. Natl. Acad. Sci. USA 86:3833-3837, (1989))에서 상술된 바와 같이 PCR을 사용함으로써 증폭된다. VH의 PCR 증촉을 위해서, DNA/프라이머 혼합물은 5μl의 cDNA, (5'GGAAGCTTA GACCGATGGGGCCTGTTG TTTTG 3')을 위한 25 pmoles의 CG2A 및 VH1 BACK 프라이머 (5'AGGT(G/C)(A/C)A(A/G)CTGCAG(G/C)AGTC (A/T)GG 3')로 구성된다.
VK의 PCR 증폭을 위해서, DNA/프라이머 혼합물은 5 μl의 cDNA, (5'GGAAG CTTGAAGATGGATACAGTTGGTGCAGC 3')을 위한 25 pmoles의 CK2 및 VK10 BACK 프라이머 (5' TTGAATTCCAGTGATGTTTTGATGACCCA 3')로 구성된다.
이러한 혼합물은 50 μl의 최종량에서 각각 2.5 mM의 dATP, dCTP, dTTP 및 dGTP와, 5 μl의 10X 완화 서모라제 및 1 유니트의 서모라제(IBI)가 추가된다. 샘플은 94℃에서 30초동안 25번의 열사이클을 수행하고; 50℃에서 30초 동안 수행하며; 72℃에서 1분 동안 수행하고; 그리고 72℃에서 5분 동안 마지막 인큐베이션을 수행한다. 증폭된 VH 및 파 움는 Prep 상에서 정제된다. 유전제 정제 기구(BioRad).
정체된 VH 및 VK DNA는 M13 벡터로 복제된다. 복제물은 T7 DNA Pol(제약자)를 사용한 디디옥시 방법에 의해 연속적으로 복제된다. 도 1 및 도 2를 보라.
실시예2: 예견된 T-세포 항체 시퀀싱을 사람에게 적용하기 위한 ior t1A 쥐 세포 항체의 변수 영역 시퀀스의 변경.
ior t1A의 헤비 및 라이트 체인의 변수 영역 시퀀스는 T-세포 항체 시퀀스를 위해 분석된다. 이는 AMPHI 컴퓨터 알고리즘을 사용하여 만들어지고, 이 컴퓨터 알고리즘은 길이의 형태가 앰피패틱 나선형 구조인 시퀀스 11 아미노산의 세그먼트를 예견할수 있으며, 이는 MHC II 분자와 결합된 한측은 소수성이고 한측은 친수성을 갖는다.
헤비 체인의 변수 영역 시퀀스는 다음과 같은 3 세그먼트를 예상할 수 있다:(이는 Kabat's 넘버링을 사용한 것이다.)
1. 아미노산 2-21 간의 FR1
2. 아미노산 29-43 간의 CDR1 및 FR2
3. 아미노산 95-111 간의 CDR3 및 FR4
도 1은 헤비 체인과 일치하는 시퀀스를 도시한 것이다.
이러한 쥐 시퀀스는 유전자 은행 및 EMBL 데이터베이스 내에 포함된 면역글로블린과 비교한다. 대부분의 이체동형의 인간 변수 영역 시퀀스는 결정되고, 또한 쥐의 시퀀스와 아주 닮은 인간의 서브그룹도 정의된다. 이러한 경우에 발견된 인간 시퀀스는 Kabat의 서브그룹 III에 속하는 IgM이다.
양쪽의 변수 영역 시퀀스에서, 인간과 쥐는 잔류물을 위해 잔류물을 비교하게 되고, Vernier 영역에 포함되지 않거나 또는 캐노니컬한 구조를 갖지 않는 FR 영역에서 이러한 잔류물을 선택하게 된다. 그러므로 양쪽의 변수 영역 시퀀스는 인간의 시퀀스 상에 동일하게 위치한 이러한 잔류물에 의해 변화될 수 있다. 위치 13 및 19는 아미노산 19는 아미노산 Lys가 동일한 서브그룹에 속한 다른 인간의 면역글로블린의 동일 위치에 존재하기 때문에 변경되지 않는다.
쥐 iro t1A의 헤비 체인을 위해 네가지의 대체물을 제안한다:
1. ALA에 의해 위치 40을 THR로 대체.
2. GLY에 의해 위치 42를 GLU로 대체.
3. LEU에 의해 위치 108을 THR로 대체.
4. VAL에 의해 위치 109를 LEU로 대체.
동일 처리과정은 라이트 체인(도2)에 적용되고, 아미노산 2에서부터 아미노산 69까지의 세그먼트를 중첩하여 셋팅하게 된다.
이러한 분석을 실행한 이후에, 위치에서 FRs 1 및 2를 위한 7가지의 대체물을 제안한다: 3, 11, 12, 15, 17, 41 및 43.
1. GLN에 의해 위치 3을 LYS로 대체.
2. LEU에 의해 위치 11을 MET로 대체.
3. SER에 의해 위치 12을 TYR로 대체.
4. VAL에 의해 위치 15를 LEU로 대체.

실시예3 : PCR 변이유전자에 의해 ior t1A의 쥐 헤비 및 라이트 체인 변수 영역의 구축.
쥐 헤비 및 라이트 체인 변수 영역의 아미노산의 변화는 PCR 변이유전자(Kammann, M. et al. (1989) Proc.Natl.Acad.Aci. USA, 86: 4220)를 사용함으로써 구축되어진다.
요약: PCR에 의한 두 개의 증폭: 반응작용 혼합은 M13은 복제된 단일 성분 DNA인 0.5μl의 VH 상청액, 25 pmoles의 돌연변이 유전자 결핍 1 또는 2, 25 poles의 돌연변이 유전자 결핍 3 또는 4 프라이머(다음에 설명할 프라이머 시퀀스를 참조하라)로 이루어진다. 이러한 혼합물은 50 μl의 전체량 중에서 각각 2.5 mM의 dATP, Dttp, dCTP 및 dGTP와, 5 μl의 10X 벤트 폴리메라아제 버퍼(NEB) 및 1 유니트의 벤트 DNA 폴리메라아제(NEB)로 이루어진다. 샘플은 94℃에서 30초 동안 12-15의 열사이클을 수행하고; 50℃에서 30초 동안 수행하며; 75℃에서 1분 동안 수행하고; 75℃에서 5분 동안 마지막 인큐베이션을 수행한다. 양쪽의 PCRs를 생성한 것은 단지 (3 및 4) 바깥 프라이머를 사용함으로써 제2 PCR 내에 결합되어진다. 증폭된 VHDNA는 Prep 상에서 정제된다. 유전자 정제 기구(BioRad).

ior t1 mAB는 정제되고, 4.50㎎의 염기성 인산나트륨, 10.88㎎의 2염기성 인산나트룸, 86.00㎎의 염화나트륨, 80 1.852 uL의 폴리소르베이트, 주입을 위한 물로 이루어진 불임 버퍼 용해제(10 mL pH 7.0 +/- 0.5) 내에 용해된다(50.00㎎). mAb를 포함한 용해제는 젤리 주약에 추가된다.
치료제로 쓰이는 젤리는 mAb (pH 7.0 +/- 0.5)을 유사하게 사용할 수 있도록 하는 구성을 갖는 버퍼 용해제로 동화된다.
실시예 5: 건선 벌거로 피부장애를 갖는 환자에 대한 조직학적 연구.
건선 벌거로 피부장애를 갖는 환자의 면역조직화학적 연구는 피부 조직의 저온 형성 장치 부분에서 이루어지고, T 림프세포의 CD에 직접 대항한 다른 mAbs와 유사하게 있는 mAb ior t1을 사용함으로써 이루어진다. mAbs ior t3 (안티 CD3), ior t4(안티 CD4), ior t8(안티 CD8), ior L3 (안티 CD45) 및 DAKO CD6 (안티 CD6)은 ior egf/r3 (안티 상피 성장 요소 감각기관)에서와 같이 제어되고 사용된다. 그리고 비오틴화된 안티 쥐 면역글로블린(즉, sheep, DAKO로부터)과 Avidin 비오틴 과산화제 복합물(즉, DAKO)를 사용한 조직 부분의 인큐베이션에 의해 수행된다. 최종적인 반응은 색원체 (Sigma)로써 3-아미노-9-에틸-카본졸레(carbazole)를 사용함으로써 이루어진다. 생체검사법은 두 전문가에 의해 실험되었고, CD6의 평가는 포인트의 크기에 적합하도록 조정된다.

환자는 초기 치료시 이전에 형성된 장애를 생체검사한 안티 CD6 단일세포 항체로 치료되고, 초기 생체검사를 수행한 다음 3주의 끝까지 치료를 계속 수행한다. 분류된 치료응답의 크기는 퍼센트로 질을 나타내고, 전체 CD45+ 세포 상에서 CD6 세포의 인덱스에 의해 수행되며, 각각의 생체검사에 의해 평가된다. 피부의 다른 층 내에 있는 안티 상피 성장 요소 감각기관을 표현하는 퍼센트율은 질병의 조직 특성형태의 변형에 따라 결정되어진다.

실시예 6: 건선 벌거를 갖는 환자에게 ior t1 mAb로 국부 치료를 하는 임상 반응.
이러한 질병의 장애 특성이 도지는 건선 벌거를 진단할 수 있도록 환자에 대한 임상 실험이 수행되었다. 이 연구는 국부 치료(ior t1 mAb 또는 부형약)를 위해 사용된 젤리와 일치하도록 한정된 두 개의 그룹을 구성하고 그룹 당 40명의 환자들을 포함시켜 임상 실험이 계획되었다. 국부 치료 공식은 mAb가 이 결합된 젤리 주약 또는 부형약(나트륨 탄화메틸셀룰로제 v/v, 프로필렌클리콜, 메틸다라베노, 삼중에탄올라미네)에 의해 수행되었다. 치료는 장애 치료를 중단하지 않고 21 동안 하루에 두 번씩 실행하였다.
건선 플라그 장애는 ior t1 mAb(도4)로 치료한 모든 환자들을 희게 만들었다. 이러한 결과는 mAb를 처음에 적용한 이후 21일 째에 수행된 생체검사법에 의한 결과와 일치한다. 질병의 조직 부호 특성의 복귀와 마찬가지로 각질의 T 림파구 침투의 감소와 EGF-R 표현의 감소는 획득되어졌다.
실시예 7: ior t1 단일세포 항체로 스케일링-다운 국부 치료를 수행한 이후에 환자의 건선 벌거의 임상 반응.
10% 내지 25%의 만성 건선 벌거를 갖는 환자에게 19번 실행하여 확인된 안내적인 임상 실험은 환자의 피부에 대해 수행되었다. 세 개의 다른 그룹 6, 7 및 6 환자들은 각각이 젤리 형태인 0.3, 1 및 3 ㎎의 ior t1A, mAb/gram을 함유한 국부 치료 방식과 나트륨 탄화메틸셀룰로제 A/V, 프로필렌글리콜, 메틸파라베네, 프로필파라베네 및 트리에탄올라미네를 함유한 부형약으로 치료를 받았다.
PASI(Psoriatic Area and Severity Index; 건선 부분 및 시버러티 인덱스)는 기록 되어지고 분석되어졌으며, 또한 환자들의 질병 혈청과 일치하는 인간 안티 쥐 항체(HAMA; human anti mouse antibody)는 연구되어졌다. 임상 반응(PASI) 및 질병의 자유로운 간격과 관련된 최고의 결과는 ior t1 mAb (0.3㎎)의 최저량을 사용하여 처방한 그룹 뿐만 아니라 환자 그룹에 HAMA Human Anti-Mouse Antibody) 적정제의 양을 투여한 그룹에서도 얻어졌다. 더욱이 블록킹 ELISA (Enzyme Linked Immunosorbent Assay) 및 FACS (Fluorescent Activating Cell Sorter)를 사용하여 연구된 환자들의 혈정에서 안티-유전자형 항체의 존재는 젤리 그람 당 0.3㎎을 최저 약량으로 치료한 환자들보다 훨씬 더 자주 그리고 훨씬 더 높게 나타났다.`
실시예 8: 일반적인 건선에 의해 심한 형태를 갖는 한 명의 환자를 ior t1 mAb로 치료한 임상 반응.
여성 환자는 약 17세부터 건선 관절증으로 진단되어 지금까지 건선을 앓고 있는 56세의 사람이다. 최근 5년간 이 환자는 자주 그리고 심한 위험으로 고통받아왔으며, 자주 입원 치료를 받아야했다. 이러한 위험은 에리세마토스퀘아모스(erithematosquamous)로 일반적인 장애를 갖고서 시작되고, 관절과 근육의 고통, 열병, 일반화된 부종 및 말라리아로 고통받아왔다.
이러한 환자의 일반적인 상태는 지속되었고, 21일 이후에는 목과 가슴주변, 궤양과 열병에 의해 감염된 세로세 (serose) 분비와 같은 새로운 장애가 나타났다. 질병의 무감각한 전개와 스테로이드 크림을 포함한 이전의 모든 치료를 더 이상 참을 수 없는 것 때문에, 메토트렉사트에 의한 치료는 15 ㎎의 전체 분량으로 세 번의 사이클 동안 투여하도록 지시하였다. 어떠한 임상 반응도 관찰되지 않았다.
신체 무게에서 Kg 당 0.6 ㎎/Kg으로 ior t1 mAb의 단일 엔도베네우스(endovenous)분량을 염류성의 용해제 0.9%인 200 mL에 희석하여 서서히 투약하였다.
동시에 치료제로 사용되는 젤리의 mAb/g의 3㎎으로 농축한 ior t1 mAb를 포함한 젤리는 이틀 동안 모든 장애 부위에 하루에 두 번 투약하였다.
환자는 그녀의 일반적인 상태를 향상시키기 시작했고, 치료한 지 6일 정도 되면서 피부의 모습이 향상되기 시작했다. 치료한지 21일 째 되는 날 평가한 것은 질병의 임상적 표시가 개선되었음을 보여주는 피부의 잔여부분과 신체 피부에서 60%의 장애 부위가 없는 결과가 나왔다. 환자는 관절이 양호한 상태를 갖게 되어 관절의 증상도 개선되었고, 정상적인 생동적인 모습과 정기적인 실험 테스트를 받았다.
30일 후에 환자는 질병의 증상으로부터 완전히 벗어나 정상상태를 유지하였다.

본 발명은 면역학에 관한 것으로서 특히 CD6 백혈구 분화 항원(CD6 leukocyte differentiation antigen)을 인식하는 모노클로널 항체(monoclonal antibody)를 포함하는 제약 혼합물을 얻는 것에 관한 것이다.
[종래의 기술]
모노클러널 항체(mAbs)는 세포면에 나타나는 생리적 중요성을 갖는 분자들의 특성 기술을 가능하게 했다.
면역체계의 세포들에 한정된 벽혈구 분화군 또한 항원(CD) 참조(Scholossman, S.F. 등(1994) 면역학 Today 15(3) ; 98). 개체발생중 임파세포의 분화 및 성숙에 있어서 CD들의 역할에 대한 정의는 면역반응 동안 세포적 인식 및 착생의 메카니즘에서 그리고 활성화와 증식의 메카니즘에서 유망한 결과와 함께 진단 및 면역제 치료법 분야에서 개개의 mAbs를 사용을 시행해 왔다. (Dental, J.등(1991) Curr. Opin. Immunol. 3;740).
인간의 T임파구의 세포막에 나타난 분자들에 대향하는 뮤린 mAbs는 이러한 세포들의 기능장애가 존재하는 임상 실체의 진당을 향상시키는데 기여해 왔다. 더욱이 mAbs는 이들의 기능적 활동을 정형화하기 위한 목적을 가진 새로운 치료적 접근 방법을 개척하기 위해 사용되어져 왔는데, 이는 이식 거부반응(Transplant Rejection) 대숙주성 이식편성(Graft Versus Host Disease; GvHD)과 자기면역병(Autoimmune Dise ases)(Waldman, T.A.(1991) Science 252; 1657; Waldman, T.A(1992) Annu. CD6는 충분히 특허와된 분자는 아니다. CD6는 동적 평형에서 유지되는 두 개의 분자형태로 존재하며 포스포릴레이션의 등급에서만 차별되는 그리코프로테인으로 알려져 있다. 휴먼중의 T임파구에 있어서 CD6는 105 kda의 포포릴레이티드 분자이며, 반면 활동중인 세포에 있어서 CD6는 하치퍼스포릴레이티드 형태의 130kda이다. 독특한 구조를 가진 막 수용체군 및 분비단백질의 멤버. 말초혈액의 T 임파구에 있어서, CD6는 성숙된 인체티모사이트의 표면에 나타나며, B임파구의 아종(subtyupe) 및 뇌피질(brain cortex)의 뉴런에 있어서는 CD3+ 세포 개체군의 대다수를 이룬다.
말초혈액의 T임파구에 있어서 CD6는 세포 활동황의 메카니브에 참여한다.
상이한 병인학(etiologg)외관 병리학에 있어서의 가능한 역할 및 T세포 개체발생에 있어서의 cd6분자의 역할은 미지이다.
최근에 cd6리간드(ligand)는 흉성(thymus), 비장(spleen), 림포노드(lymph nod es), 및 피부와 같은 정상 조직에서 광범위한 세포분포를 갖는 것으로 식별되었으며, 또한 그렇게 특성화 되었다.
ALCAM(활성 백혈구-세포 착생분자)로 명명된 이 분자는 모노사이트 뿐만 아니라 활성 T 및 B 임파구에 나타나기 때문에 면역 글로불린의 그것들에 유사한 다섯개의 비세포 도메인을 갖는 100kDa 분자량 형태의 I막 크리코프로테인이다. 그것은 2가의 양이온에 따라 서로 다른 활동 수준을 나타내며 중재적 헤토필릭 및 호포필릭 상호작용을 할 수 있다.

IgG2a 아이소타입 모토클로널 항체는 제4차 국제 백혈구 분화항원 워크샵에서 항 CD6로 분류되어 있다. 이 mAb는 다른 항 CD6 mAbs에 의해 인식되는 것들과는 다른 항원요소를 정의한다. 항원요소는 안정적 형태를 가지고 있으며, 감소 에이전트에 비민감하며, CD6 분자의 주구조에 위치 가능하다.
이 모노크로널 항체는 건강한 기중자의 말초 단핵세포에서 다른 CD6 mAbs 보다 더 낮은 인식도를 갖고 있다. T임파구 오리진의 인체 세포층 라인에서의 ior t1 mAbs의 인식패턴은 쥬캇(Jurkat) 세포에서 47%이고, 몰트-4(Molt-4)세포에서 23%이며, CCRF-CEM세포에서 인식이 없다. 건서의 동시적 소강(remission)은 만약 피부의 T세포의 수가 감소한다면 예견될 수 있다. 따라서, 병의 임상적발로(clinical manifestations)에 책임이 있는 케라티노사이트(Keratinocytes)의 증식을 감소시킬 수 있는 면역반응의 해결중개자(soluble med iators)를 해제함으로서 병의 영속화에 중요한 역할을 하는 것으로 암시된다.
이러한 고려는 알로제닉 본 매로우 이식(allogeneic bone marrow transplant ation)에 따르는 병치료와 같은 사실에 의해 지지되며 또한 HLA와 스테로이드(steroids) 와의 결합, 시클로포린(cyclosporine)과 체방향상과의 결합, 그리고 항 T 세포 처방적 모노클로널 항체들과의 결합에 의해 지지된다. 모노클로널 항체들에 대한 면역요법의 성공은 타겟분자의 선택에 달려 있는데, 타켓분자는 중요한 세포 기능에 참여하거나 mAbs의 선택에 참여한다(Dental, Je,d(1991) urr. Opin. Immunol. 3; 740). CD3에 대항하는 전선에서 평가된 mAbs(Weinshenker, B.G. 등(1989) J.Am. Acad. Dermatol. 20:1132) 및 CD4에 대항하는 전선에서 평가된 mAbs(Poizot-Martin, I. 등(1991) Lancet 337:1477; Prinz J. 등(1991) Lancet 338; 320; Nicolas, J.F 등(1991) Lancet 338:321) 은 환자에게서 처방 향상을 발생시켰는데 그러한 항상은 짧은 기관의 부분적 소강 및 모든 경우에의 병의 증세의 조기 헤제와 함께 약 엔도비너스(emdovenous)를 적용함으로써 나타났다.
다중약에서 뮤린 mAbs의 처방적 응용은 환자에게서 몇가지 이차적 효과와 제한적인 임상용도와 연결되며, 이러한 것은 인체 항-마우스 항체 반응(HAMA)을 유도하는 뮤린 단백질의 제노제니시티(xenogenicity)와 관련된다. 프로테인 엔지니어링 (Proetin engineering)에 의해 발생된 인체적용 항체(humanized)는 이문제니시티(immunogenicity)를 감소시켜 파마코키네틱스(phaamacokinetics)와 임상효과를 향상시켰다.
지금까지 전선에서 피부의 염증성 침윤병소의 T 임파구에서의 CD6 분자의 표현 뿐만 아니라 이분자를 병의 발전과 결합시키는 가능성에 대해 기술하고 있는 연구 논문은 없었다. 더욱이 항CD6 mAbs의 이 병에 처방적으로 사용하는 것은 이전에는 평가된 적이 없다.
본 발명의 신규성은 항 CD6 모노크로널 제공하는 것이며 상이한 관리경로 및 이병의 상이한 임상적 형태를 사용하여 전선 벌거를 가진환자에 적용하기 위해 인체화된 모노크로널 항체 CD6의 옵텐션을 제공하는 것이다.
항원 배열 속에 T세포 항원의 펩타이드, 그리고, 제노제닉이나 알로제닉 항체의 면역원은 다른 포유류를 근거로 이들과 다른 T세포 항원 배열을 포함하는 잔여물을 대산함으로써 감소할 수 있다.
물론, 잉여 대체물은 표준구조나 또는 버니어존에서 포함된 이들을 포함하지 않는다.
잔여물의 바람직한 대체물은 구조적인 유전자나 인터도메인 접촉자에서는 효과가 없다.
따라서 특성을 묶는 리갠드는 여러 부위의 뼈대의 잉여물이 제한되는 교체 결과에 영향을 받지 아니한다.
-여러 기관의 분석-
이러한 절차는 1994년 베쓰다 마리랜드 의학잡지 5판 인체의 면역기구의 단백질 배열 캐벳에 의해 만들어진 다양한 부분의 인간항체를 위해 여러 배열에 관한 데이터를 사용할 수 있다.
첫 번째 단계에서 쥐의 다양한 영역은 이들의 인간배열의 여러가지 영역과 비교된다.

가장 구조가 비슷한 인간의 부분은 쥐의 영역과 일치하는 잔여물과 비교된다.
이것은 각각의 쥐의 배열인 인간의 서브그룹에 한정한다.
-T- 에피토프(epitopes) 예상-
두 번쩨 단계에서 두 가지의 여러 범위의 호르몬, 쥐와 인간은 T항원의 예상으로 분석된다.
알고리즘 AMPHI(베소프스키 (1987), 제이 이뮤놀 138; 2213)는 α 헬리컬 배열이 예상된다.
알고리즘 SOHHA는 헤릭스 소수성의 소모가 예상된다.
이러한 알고리즘은 항원 단백질의 T세포 조각이 예상된다.
-이뮤노제너서티(Immunogenicity) 감소에 대한 분석-
인간의 상응물과 구조가 다른 쥐의 구조에 있어서 잔여물은 인간의 상응물에 의해 대처된다.
이러한 전환은 T항원 배열인 잔여물을 생성한다.
마지막으로, 표준구조나 또는 버니어 존에 포함되어 반응하는 잔여물의 대처는 제3의 구조에서 효과가 있다.
또한 그들은 대처물을 포함할 수 없다.
제3의 구조에서 제안되는 대체물의 영향에 대한 추가 정보나 결합 사이트는 여러부위의 모델로부터 얻을 수 있다.
-개조항체 구성하고 개조하기 위한 방법-
다음의 절차는 유전자 재조합형 DNA배열을 대비하여 사용된다. 상기 DNA는 가볍고 무거운 사슬 두가지 모두 쥐의 mAb CDR을 포함한다.
a) 뮤타제네닉과 여러부분의 조직은 CDR과 FR의 영역을 포함한다. PCR 뮤타제네닉 방법은 다른 위치에서 변화를 개재하여 사용할 수 있다.
b) 표현의 벡터준비는 여러 가지 영역을 포함하고 인간의 대응은 불변의 영역을 포함한다. 인간의 영역은 결단과 성질에 대해 분비물에서 세포를 복제한다.
c) 무겁고 가변운 사슬의 복제는 세포라인을 충당한다.
2주후 세포 상청액은 인간의 IgG 생성물에 대한 ELISA에 의해 분석된다.
그 샘플은 다음에 뚜렷한 항원으로 결속이 가능한 인간 IgG에 의해 다른 방법으로 분석된다.
바이트로와 바이보의 특수 증상에서 형성된 공식을 연구한다.
바이트로와 바이보 중상에 사용될 수 있는 앤티 CD6 mAbs는 전선의 다른 임상형태를 제공한다.
이러한 mAbs는 소디움 아지드(0.01-0.2%)를 포함하는 용제(ph 7.0 +/- 0.5)에서 정제되고 용해되며, 보바인 서럼 알부민(0.05-0.2%)는 CD6+T 림포사이트 양을 재되어 사용할 수 있고, 또한 4℃에서 20내지 30분 동안 용액 0.1과 3㎎/mL 사이의 농도에서 mAb의 10에서 30mL로 이루어진 샘플의 50에서 200mL을 담은 바이오로지컬 용액에서 림포이드 세포의 표면상에 이 미립자를 사용할 수 있다.
이후 완충용액에서 세척을 하고 침해 물질로 변화되는 다른 동물계의 면역글로부린으로 부화한다.
이 저항 CD6 mAbs는 다른 방법에 의해 침해물질로 직접 변화시킬 수 있고 5내지 30㎎/mL 사이에서 농도로 유사하게 사용된다.
프소리아시스 벌가로 이루어진 환자의 조직에 다른 영향을 받거나 도는 피부를 해치는 손상에 대한 이 미뮤노히스토체미컬 연구는 차가운 아세톤에서 할 수 없고 고정된 저온유지 분위기에서 형성된다.
앤티 CD6 mAb는 소지움 아지드(0.05-0.2%)를 포함하는 완충용액(pH 7.0 +/- 0.5)에서 2-10㎎/mL 사이에서 용해된다. 그리고 30분 동안 보바인 서럼 알부민(0.05-0.2%)을 용해한다.
이후 아비딘 바이오틴 퍼록시다이스는 룸 온도에서 30분동안 혼합한다.
마지막으로 반응물은 크로모텐으로 3-아미노-9-에틸-카바졸로 사용이 제한된다.
생체검사법은 10%(+/-), 10-25%(+), 25-50%(++), 50-90%(+++), 90-100%(++++) 점의 번위에서 조절되는 CD6의 값과 두가지의 특별값으로 시험되어 진다.
T 림포사이트에서 다른 항체는 앤티 CD3(iot t3), 앤티 CD4(ior t4), 앤티 CD8(ior t8)과 앤티 CD45(ior t3)로 사용되며, 또한 케레이션사이트 액티베션의 제조(Mozzanica, N. et)에피더말 그로스 팩터 립셉터 mAb(ior egf/r3)로 사용된다.
이들은 질병의 치료제 과정중 손상에 스며드는 염증의 특징의 상세한 값을 허용한다.
앤티 CD6 모토크로날 항체로 처방되는 환자는 조직으로 사용하거나 또는 국부 조직의 효능 값으로 초기 생체검사 다음에 항상 치료초기, 치료중, 치료가 끝난 후에 이전에 형성된 손상을 생체검사할 수 있다.
손상 응답 등급에 대한 범위는 퍼센트로 구분되고 각 생체검사에서 결정된 전체 CD45+ 세포에서 CD6+ 세포의 색인으로 확정된다.
CD6+ 세포 인덱스 = CD6+ 세포 × 100의 수 / CD45+ 세포의 수
CD3+ 세포, CD4+ 세포와 CD8+ 세포의 인덱스는 항상 CD45+ 세포 전체와 CD4+의 인덱스와 CD6+세포의 전체비율로 계산할 수 있다.
CD3+ 세포 인덱스 = CD3+ 세포 × 100의 수 / CD45+ 세포의 수
CD4+ 세포 인덱스 = CD4+ 세포 × 100의 수 / CD45+ 세포의 수
CD8+ 세포 인덱스 = CD8+ 세포 × 100의 수 / CD45+ 세포의 수
CD4+ 세포 인덱스 = CD4+ 세포 × 100의 수 / CD6+ 세포의 수
CD8+ 세포 인덱스 = CD8+ 세포 × 100의 수 / CD6+ 세포의 수
앤티 CD6 mAB는 국부적으로 그리고 조직의 공식 둘사이에서 전선의 다른 임상의 형태에서 제공될 수 있다.
Claims (11)
- 건선의 진단 및 치료에 사용되는 인간 CD6을 인식하는 단일세포 항체.
- 제1항에 있어서, 인간 CD6을 인식하는 단일세포 항체는,단일세포 항체가 어떠한 인간에게도 적용할 수 있도록 변형된 ior t1 및 ior taA 이라는 이름의 쥐 단일세포 항체의 서브클론인 것을 특징으로 하는 단일세포 항체.
- 제1항 및 2항에 있어서, 인간 CD6을 인식하는 단일세포 항체는,시퀀스의 헤비 체인과 라이트 체인에서 변수 영역을 갖는 동일명(정해둔 개수: 계루 중임)의 하이브리도마로부터 얻어진 서브클론 ior t1A이고, 다음과 같은 시퀀스를 갖는 것을 특징으로 하는 단일세포 항체.헤비 체인의 시퀀스는 다음과 같고:GLU VAL그리고 라이트 체인의 시퀀스는 다음과 같다:
- 제1항 내지 3항에 있어서, 인간 CD6을 인식하는 단일세포 항체는,서브클론 ior t1A의 인간에게 적용할 수 있는 변형이고, 이것의 헤비 체인과 라이트 체인의 변수 영역은 다음과 같은 시퀀스를 갖는 것을 특징으로 하는 단일세포항체.헤비 체인의 변수 영역은 다음과 같은 시퀀스를 갖는다:그리고 라이트 체인의 변수 영역은 다음과 같은 시퀀스를 갖는다:

- 제5항에 있어서, 인간에게 적용될 수 있는 단일세포 항체는,IgG2a 아이소타이프임을 특징으로 하는 단일세포 항체.
- 제1항 내지 4항에 있어서, 단일세포 항체의 사용은,일정한 비율로 치료하는 데 사용되는 약제의 제조법에 사용되는 것을 특징으로 하는 단일세포 항체.
- 제1항 내지 4항에 있어서, 단일세포 항체의 사용은, 약제의 in vitro 및 in vivo를 진단하는데 사용되도록 반응하는 제조법에 사용되는 것을 특징으로 하는 단일세포 항체.
- 제1항 내지 4항에 있어서,단일세포 항체를 포함한 약제를 치료제로 사용하도록 한 약제 제조법.
- 제1항 내지 4항에 있어서,단일세포 항체를 포함한 약제를 진단하는데 사용하도록 한 반응법.
