물질 및 방법

랫트에서 내독소-유도된 포도막염: 포도막염은 인간에서 유해한 안구 염증성 병태이다. 전측 포도막염은 반복되는 염증성 질환이고 잠재적으로 눈이 부신 결과를 낳을 수 있다. 질환의 발병은 저조하게 이해되고, 사용된 항-염증성 요법은 비-특이적이고 유의미한 합병증과 연관된다. 동물 모델은 질환의 이해 및 신규 요법의 시험의 핵심이다. 발바닥에서 단일 저용량의 지질 다당류 (LPS)는 랫트에서 전측 포도막염을 유도한다. 내독소-유도된 포도막염으로서 공지된 이러한 모델은 인간 전측 포도막염의 유용한 패러다임으로 쓰인다. 루이스 랫트 수컷 (260 ± 25 그램)은 Charles River Laboratory로부터 구매했다. (멸균된 0.9% 염수 중) 100 μl의 1 mg/ml LPS (List Biological Labs) 용액이 랫트의 발바닥 (뒷쪽 좌측)에 주입되었다. 시험 화합물은 인산나트륨, 2염기성 헵타히드레이트, 염, CMC 및 멸균수로 구성된 비히클에서 제형화되었다. 화합물은 LPS 2 시간 후 국소로 (0.1-1%) 또는 피하로 (10mg/kg) 복용되었다. 동물은 LPS 주사 24 시간에 희생되었다. 눈방수는 수집되고 염증 세포 수 및 총 단백질 농도를 결정하기 위해 부석되었다.
토끼의 알칼리 화상: 각막 상피는 각막 기능 및 온전성의 유지에 중요한 역할을 한다. 장기적인 각막 상피성 결함은 각막 불투명, 신생혈관형성, 박테리아 감염 및 시각 손실을 야기한다. 각막 상피성 치유는 부상에 대한 염증성 반응, 세포 증식 및 이동을 수반하는 복합적인 과정이다. 각막 부상의 동물 모델은 신규 항-염증성 및 프로-상처 치유 요법을 시험하는데 아주 유용하다. 2.1 내 2.5 kg로 칭량된 뉴질렌드 화이트 토끼는 케타민/크실라진 (35/5 mg/kg)로 피하로 전신 마취되었고 프로파라카인 (0.5%)로 국소 마취되었다. 한쪽 눈의 각막 상피성 상처는 1.0 N NaOH를 함유하는 NaOH 포화된 여과지로 30 초 동안 유도되었다. 눈은 멸균된 PBS로 헹구었다. 각막 상처는 10% 나트륨 플루오레신으로 플루오레신 염색으로 확인되었다 (Science Lab Com) 및 슬릿 램프 사진술. 시험 화합물은 상기에서 기재된 비히클에서 제형화되었다. 초기 연구를 위해 화합물은 국소로 3 회/1일 복용되었다. 각막 상처 부위의 정량화는 Image J 소프트웨어를 사용하여 수행되었고, 여기서 형광 염색 녹색 부분이 추적되었고 총 픽셀로 전환되었다.
아래의 화합물은 많은 상이한 유형의 안구 염증에서 유의미한 효과를 갖는 것으로 기대되지만, 랫트의 내독소-유도된 포도막염에서 항-염증 활성을 실증하여 예시되었다 (도 1 및 2). 이러한 모델에서 항-염증성 활성은 하기 FPR2 작용제와 함께 예시되었다:
이러한 모델에서 본 화합물은 중성구 및 단백질의 전방으로의 침윤의 차단에서 강한 항-염증성 활성을 보여준다. 또한 FPR2 작용제는 화합물 {[(2S,3S)-2-{[(4-브로모페닐)카바모일]아미노}-3-메틸펜타노일]아미노}아세트산에 의해 예시된 바와 같이 각막 상처의 마우스 모델에서 가속화된 치유 및 재-상피화를 보여준다 (도 3). 이들 데이타는, FPR2 작용제가 안구 염증의 상이한 모델에서 안구용에 적합한 강력한 및 유효한 항-염증제라는 것을 실증한다.
