Claims (17)
- (a) 원핵세포 복제 기원;(b) 진핵세포 프로모터(promoter)의 조절하에 영양요구성 원핵세포의 생존에 필요한 산물을 합성하는데 요구되는 효소를 암호화하는 하나 이상의 진핵세포 영양요구성 표지 유전자;(c) 원핵세포 프로모터의 직접적인 조절을 받아 mRNA가 생성되는 외부 유전자; 및(d) 하나 이상의 전사 종결유전자를 포함하고, 숙주 세포 서열의 연속된 염기쌍을 50개 이하로 포함하는, 숙주 세포에서 사용하기 위한 원핵세포 발현 벡터.
- 삭제
- 숙주 세포 서열의 연속된 염기쌍을 50개 이하로 포함하고, 추가로 (a) 원핵세포 복제 기원, (b) 진핵세포 프로모터의 조절하에 영양요구성 원핵세포의 생존에 필요한 산물을 합성하는데 요구되는 효소를 암호화하는 하나 이상의 진핵세포 영양요구성 표지 유전자, 및 (c) 하나 이상의 전사 종결유전자를 포함하는 원핵세포 발현 벡터에서 원핵세포 프로모터의 직접적인 조절하에 재조합 단백질을 암호화하는 유전자를 발현시킴을 포함하는 재조합 단백질의 생산 방법.
- 제 1 항에 있어서,리보솜 결합 부위 및 다중 클로닝(cloning) 부위를 추가로 포함하는 원핵세포 발현 벡터.
- 제 1 항에 있어서,영양요구성 표지 유전자 및 외부 유전자가 반대 방향으로 배향되고, 전사 종결유전자가 상기 영양요구성 표지 유전자 및 외부 유전자 사이에 배치되어 양 유전자의 전사를 종결시키는 원핵세포 발현 벡터.
- 제 1 항에 있어서,영양요구성 표지 유전자가 효모 유전자인 원핵세포 발현 벡터.
- 제 5 항에 있어서,영양요구성 표지 유전자가 효모 유전자인 원핵세포 발현 벡터.
- 제 7 항에 있어서,영양요구성 표지 유전자가 URA3 또는 TRP1인 원핵세포 발현 벡터.
- 제 8 항에 있어서,영양요구성 표지 유전자가 URA3 또는 TRP1 유전자의 5' 측위 영역(flanking region)으로부터 발현되는 원핵세포 발현 벡터.
- 제 9 항에 있어서,원핵세포 프로모터가 T5-PN25/03/04인 원핵세포 발현 벡터.
- 제 10 항에 있어서,전사 종결유전자가 박테리오파지 fd 또는 λ-T0으로부터 유래한 원핵세포 발현 벡터.
- 제 11 항에 있어서,복제 기원이 pBR 또는 pUC 플라스미드로부터 유래한 원핵세포 발현 벡터.
- 제 1 항의 원핵세포 발현 벡터를 포함하며, 이 발현 벡터의 영양요구성 표지 유전자에 의해 발현되는 산물을 발현할 수 없도록 1회 이상 돌연변이된 원핵세포 영양요구성 숙주 세포.
- 제 13 항에 있어서,에스케리치아 콜라이(Escherichia coli) 세포인 원핵세포 영양요구성 숙주 세포.

- 제 15 항에 있어서,영양요구성 표지 유전자가 URA3 또는 TRP1이고, 영양요구성 표지 유전자와 외부 유전자가 반대 방향으로 배향되어 있으며, 전사 종결유전자가 상기 영양요구성 표지 유전자와 외부 유전자 사이에 배치되어 양 유전자의 전사를 종결시키는 제 1 항의 원핵세포 발현 벡터를 포함하는 원핵세포 영양요구성 숙주 세포.
- 제 16 항에 있어서,URA3 유전자 산물 또는 TRP1 유전자 산물을 최소 배지상에서 생존하기에는 충분한 양으로 생산하지만, 세포에서 생산되는 단백질 총량의 1.0% 이상을 차지하는 양으로는 생산하지 않는 원핵세포 영양요구성 숙주 세포.
