各々のハイブリッドポリペプチドおよび個々のダイマーペプチドは、別々に精製された。Hisタグハイブリッドポリペプチドを発現するE.coli JM105の細胞ペーストは、マイクロフルデーション(microfluidation)(Microfluidics,Inc.,Newton,MA)によって、リン酸緩衝化生理食塩水(PBS)中で溶解された。遠心分離後、清澄化された溶解物を、ニッケル装填アフィニティーキレート樹脂(Tosoh Biosep,Montgomeryville,PA)にバッチ吸収し、洗浄し、PBS中の段階的勾配のイミダゾールを用いて溶出した。溶出されたハイブリッドポリペプチドを含む画分をプールし、5mlのHITRAP(登録商標)Qアニオン交換カートリッジ(Amersham Pharmacia Biotech,Piscataway,NJ)を通してポンプ上げし、スターセル(stir cell)(Millipore,Bedford,MA)において濃縮した。濃縮したハイブリッドポリペプチドを、PBSを用いて平衡化したSuperdex 200カラム(600cm長、Amersham Pharmacia Biotech)を使用して、サイズ排除クロマトグラフィーによってさらに精製した。画分純度を、SDS−PAGEを使用してモニターし、そして純粋なハイブリッドポリペプチドを含む画分をプールし、そして使用まで、−20℃で保存した。ハイブリッドポリペプチドの純度、同一性および濃度を、逆相HPLC、アミノ酸分析、およびエレクトロスプレー質量分析によってさらに評価した。

(免疫ウサギからの血清を使用するELISA)

免疫ウサギ由来の血清を使用するオプソン化アッセイ)
