Abstract
Description
Claims (1)
- 【特許請求の範囲】 1.骨置換が起こる生理的環境下に移植された場合に移植後少なくとも約3日の 間物理的形状保持性を維持し、約7日から14日の間その多孔性を維持する、骨 置換のための、多孔性で生体内分解性のマトリックスであって、不溶性のバイオ ポリマー繊維、バインダーおよび固定化リン酸カルシウム無機質の結合されたネ ットワークを含む上記マトリックス。 2.前記のバインダーが、可溶性コラーゲン、ゼラチン、ポリ乳酸、ポリグリコ ール酸、乳酸とグリコール酸のコポリマー、ポリカプロラクトン、カルボキシメ チルセルロース、セルロースエステル、デキストロース、デキストラン、キトサ ン、ヒアルロン酸、フィコール、コンドロイチン硫酸、ポリビニルアルコール、 ポリアクリル酸、ポリプロピレングリコール、ポリエチレングリコール、水溶性 ポリアクリレートおよび水溶性ポリメタクリレートからなる群より選ばれる、請 求項1に記載のマトリックス。 3.前記のバイオポリマーが原繊維コラーゲンを含む、請求項1に記載のマトリ ックス。 4.前記の無機質がヒドロキシアパタイトを含む、請求項1に記載のマトリック ス。 5.前記の無機質が直径約5ミクロン以下の粒子からなる、請求項1に記載のマ トリックス。 6.前記の無機質が、骨によって置換される間、前記の生理的環境へ粒子として 持続的に放出され、前記の物理的形状保持性と多孔性を、それぞれについて前記 の期間維持する、請求項1に記載のマトリックス。 7.前記のコラーゲン及び固定化リン酸カルシウムが、コラーゲンを約30−8 0重量%含む無機化コラーゲンの形態である、請求項3に記載のマトリックス。 8.更に骨髄細胞を含む、請求項1〜7のいずれか1項に記載のマトリックス。 9.自家性骨を含む、請求項1〜8のいずれか1項に記載のマトリックス。 10.1以上の骨成長因子を含む、請求項1〜8のいずれか1項に記載のマトリッ クス。 11.生分解性で多孔性の骨移植用マトリックスの作成方法であって、 (a)生分解性の水不溶性バイオポリマー、水溶性バインダーおよび前記バイ ンダーまたはバイオポリマーによって固定化されているリン酸カルシウム無機質 微粒子を含む分散物を形成させる工程と、 (b)前記の分散物を多孔性マトリックスに形作る工程と、 (c)前記の多孔性マトリックスを架橋する工程とを含む方法。 12.前記の工程(b)が前記の分散物の凍結及び凍結乾燥を含む、請求項11に 記載の方法。 13.前記の分散物が更に滲出可能な粒子を含み、前記の工程(b)が前記の分散 物を乾燥すること及び前記の粒子を滲出して前記のマトリックスを形成させるこ とを含む、請求項11に記載の方法。 14.前記の分散物がさらに昇華可能な粒子を含み、前記の工程(b)が前記の分 散物を乾燥することおよび前記の粒子を昇華させて前記のマトリックスを形成さ せることを含む、請求項11に記載の方法。 15.前記の生分解性の水不溶性バイオポリマーが、約30−80重量%のコラー ゲンを含有する無機化コラーゲン繊維を含む、請求項11に記載の方法。 16.前記のバインダーが可溶性コラーゲンを含む、請求項11に記載の方法。 17.前記の無機質がヒドロキシアパタイトを含む、請求項11に記載の方法。 18.前記の無機質が直径約5ミクロン以下の粒子からなる、請求項11に記載の 方法。
