Claims (1)
- 【特許請求の範囲】 1. 分析方法であって、規定の位置のヌクレオチドを含むポリヌクレオチド 標的、および支持体に固定されたオリゴヌクレオチドプローブであって、該標的 に対して相補的であり且つ該規定の位置でまたはその近くで終結する該プローブ を与え;そして (a)該標的を該プローブと一緒にインキュベートして二重鎖を形成し、 (b)該二重鎖を、該標的に対して相補的な標識オリゴヌクレオチドと一緒に 連結反応条件下でインキュベートし、そして (c)該標的中の規定の位置での点突然変異を示すものとして(b)の連結反 応を監視する 工程を行うことを含む上記方法。 2. 分析方法であって、可変数タンデム反復部分および隣接部分を有するポ リヌクレオチド標的、並びに該標的の反復部分および隣接部分に対して相補的な 部分を有するオリゴヌクレオチドプローブを与え;そして (a)該標的を該プローブと一緒にインキュベートして二重鎖を形成し、 (b)該二重鎖を、標識オリゴヌクレオチドおよび/または少なくとも一つの 標識ヌクレオチドと一緒に鎖延長条件下でインキュベートし、そして (c)該標的の可変数反復部分の長さを示すものとして鎖延長を監視する 工程を行うことを含む上記方法。 3. プローブが支持体に固定されている請求項2に記載の方法。 4. ポリメラーゼおよび/またはリガーゼを工程(b)で用いる請求項2ま たは請求項3に記載の方法。 5. ポリヌクレオチド標的が、可変数タンデム反復部分および二つの隣接部 分を有し、そして工程(b)において、標識オリゴヌクレオチドを二重鎖のプロ ーブ鎖に対して連結させる請求項2〜4のいずれか1項に記載の方法。 6. ポリヌクレオチド標的が、可変数タンデム反復部分および二つの隣接部 分を有し、そして工程(b)において、二重鎖のプローブ鎖を、標識ヌクレオチ ドの付加によって鎖延長させる請求項2〜4のいずれか1項に記載の方法。 7. 2種類またはそれ以上の異なったプローブが、アレイの形で支持体の異 なった場所に固定されている請求項1〜6のいずれか1項に記載の方法。 8. 可変配列を有するポリヌクレオチド標的を分析するためのオリゴヌクレ オチドのアレイであって、成分オリゴヌクレオチドそれぞれが、(i)該標的の 予想される変異体を含めた、該標的に対して相補的な配列を含み、そして(ii) (a)標的との二重鎖形成を可能にし且つ(b)該オリゴヌクレオチドの配列が 該標的の可変配列に対合する場合にのみ鎖延長を可能にする化学方向で固体支持 体に固定されている上記オリゴヌクレオチドのアレイ。 9. 可変数タンデム反復配列を有するポリヌクレオチド標的を分析するため のオリゴヌクレオチドのセットまたはアレイであって、成分オリゴヌクレオチド それぞれが、(i)該反復配列のすぐ隣の該標的の一部分に対して相補的な配列 を含み、(ii)該標的の反復配列に対して相補的で且つ該標的中に予想される多 数の反復を有する配列を含み、そして(iii)(a)標的との二重鎖形成を可能 にし且つ(b)該オリゴヌクレオチド中の反復数が、該標的中の反復数と等しい かまたはそれより少ない場合にのみ鎖延長を可能にする方式で立体配置されてい る上記オリゴヌクレオチドのセットまたはアレイ。 10.成分オリゴヌクレオチドそれぞれが、(iii)(b)該オリゴヌクレオ チド中の反復数が標的中の反復数と等しい場合にのみ連結反応によって、または 該オリゴヌクレオチド中の反復数が標的中の反復数より少ない場合にのみ重合に よって鎖延長を可能にする方式で立体配置されている請求項9に記載のセットま たはアレイ。 11.成分オリゴヌクレオチドそれぞれが、固体支持体に固定されている請求 項9または請求項10に記載のオリゴヌクレオチドのセットまたはアレイ。 12.異なったオリゴヌクレオチドが異なった場所を占有し且つそれぞれのオ リゴヌクレオチドが、共有結合によってそれを支持体に固定する3′ヌクレオチ ド残基およびリン酸化されている5′ヌクレオチド残基を有するオリゴヌクレオ チドのアレイ。 13.支持体の異なった場所に固定された異なったオリゴヌクレオチドのアレ イを製造する方法であって、支持体に固定された第一中間体オリゴヌクレオチド および溶液中の第二中間体オリゴヌクレオチド、並びに該第一および第二中間体 オリゴヌクレオチド両方に対して相補的である第三オリゴヌクレオチドを与え、 該第一および第二中間体オリゴヌクレオチドと一緒に該第三オリゴヌクレオチド の二重鎖を形成し、そして該第一中間体オリゴヌクレオチドを該第二中間体オリ ゴヌクレオチドと連結する工程;並びに該支持体の異なった場所に固定されたオ リゴヌクレオチドを用いて該工程を反復することを含む上記方法。
