Claims (1)
- 【特許請求の範囲】 1.細菌中で異種ポリペプチドを産生する方法であって、(a)DsbAまたはD sbCタンパク質をコードする核酸、該異種ポリペプチドをコードする核酸、Dsb AまたはDsbCタンパク質および異種ポリペプチドの両者の分泌のためのシグナ ル配列、およびDsbAまたはDsbCタンパク質をコードする核酸および該異種ポ リペプチドをコードする核酸の両者の誘発可能なプロモーターを含む細菌細胞を 、DsbAまたはDsbCタンパク質をコードする核酸の発現を誘発した後に異種ポ リペプチドをコードする核酸の発現を誘発する条件下および該異種ポリペプチド およびDsbAまたはDsbCタンパク質の両者を細菌のペリプラズムに分泌させる かまたは該異種ポリペプチドを該細菌細胞を培養する培地に分泌させる条件下で 培養する;および(b)該異種ポリペプチドを該ペリプラズムまたは該培養培地 から回収することを含む方法。 2.該異種ポリペプチドが哺乳動物のポリペプチドである請求項1に記載の方法 。 3.該哺乳動物のポリペプチドがヒトのポリペプチドである請求項2に記載の方 法。 4.該異種ポリペプチドがインシュリン様増殖因子である請求項2に記載の方法 。 5.該細菌細胞が真正細菌細胞である請求項1に記載の方法。 6.該真正細菌細胞がエンテロバクテリアセエ細胞である請求項5に記載の方法 。 7.該真正細菌細胞が大腸菌細胞である請求項6に記載の方法。 8.DsbAをコードする核酸を発現する請求項1に記載の方法。 9.該ポリペプチドを該細菌細胞のペリプラズムから回収する請求項1に記載の 方法。 10.該ポリペプチドの回収後、DsbAまたはDsbCタンパク質を該ポリペプチ ドから分離する請求項1に記載の方法。 11.該細菌細胞をDsbAまたはDsbCタンパク質をコードする核酸および該異 種ポリペプチドをコードする核酸を含む1つまたは2つの発現ベクターで形質転 換する請求項1に記載の方法。 12.該細菌細胞をDsbAまたはDsbCタンパク質をコードする核酸および該異 種ポリペプチドをコードする核酸を各々含む2つのベクターで形質転換する請求 項11に記載の方法。 13.DsbAまたはDsbCタンパク質をコードする核酸および該異種ポリペプチ ドをコードする核酸を、該細菌細胞を形質転換する1つのベクターに含む請求項 11に記載の方法。 14.DsbAまたはDsbCをコードする核酸の発現の誘発を培養培地にインデュ ーサーを添加することによって行う請求項1に記載の方法。 15.該インデューサーがIPTG、ラクトースまたはL−アラビノースである 請求項14に記載の方法。 16.該培養がグルタチオンの不在下で行われる請求項1に記載の方法。 17.総ポリペプチドの収率が該方法の結果として増加し、可溶性ポリペプチド の収率が変化しないかまたは減少する請求項1に記載の方法。 18.該培養を熱ショック転写因子をコードする核酸の発現レベルが該核酸の発 現の内在レベルを越えて増強しないで行う請求項1に記載の方法。
