Claims (1)
- 【特許請求の範囲】 1.ペプチドフラグメント質量スペクトルを配列データベース由来のアミノ酸 配列に相関させるための方法であって: 少なくとも第一のペプチドの複数のフラグメントの第一の質量スペクトルを表 すデータを記憶し; 前記配列データベース内の複数の前記配列の少なくとも一部の複数の予測質量 スペクトルを計算し;そして 前記複数の予測質量スペクトルの各々について少なくとも第一の測定値を計算 することからなり、前記第一の測定値が前記第一の質量スペクトルと前記複数の 質量スペクトルの各々との間の一致度を示すものであるような方法。 2.前記第一の質量スペクトルが、タンデム型質量分析計装置から与えられる 、請求項1に記載の方法。 3.タンデム型質量分析計が、三重−四重質量分析計、フーリエ変換サイクロ トロン共鳴質量分析計、タンデム型飛行時間質量分析計及び四重極イオントラッ プ質量分析計のうちの一つである、請求項2に記載の方法。 4.前記配列データベースが、複数のタンパク質のアミノ酸配列のデータベー スである、請求項1に記載の方法。 5.前記配列データベースが、ヌクレオチドデーである、請求項1に記載の方 法。 6.更に、前記配列データベースから第一の複数のサブ配列を選択するステッ プも含み、複数の予測質量スペクトルを計算する前記ステップが、選択した前記 第一の複数ののサブ配列の各々について少なくとも一つの予測質量スペクトルを 計算する操作を含む、請求項1に記載の方法。 7.第一の測定値を計算する前記ステップが、所定の閾値より大きい強度を有 する値を前記第一の質量スペクトルから選択する操作を含む、請求項1に記載の 方法。 8.更に、少なくとも第一の測定値を計算する前記ステップの前に前記第一の スペクトルを正規化するステップも含む、請求項1に記載の方法。 9.複数の予測質量スペクトルを計算する前記ステップが、前記配列データベ ースの一部のみについて予測質量スペクトルを計算する操作を含む、請求項1に 記載の方法。 10.前記第一のペプチドが、第一の生体から得たタンパク質に由来し、前記配 列データベースの前記タンパク質が、前記第一の生体に存在するタンパク質の配 列を含む部分である、請求項9に記載の方法。 11.前記タンデム型質量分析計装置の第一の質量分析計を第一の質量を有する 成分の分離に使用し、前記質量分析計装置の活性化装置を前記第一の成分のフラ グメント化に使用し、前記タンデム型質量分析計装置の第二の質量分析計を前記 第一の質量スペクトルを与えるために使用する、請求項2に記載の方法。 12.前記第一のペプチドをクロマトグラフィーによって分離する、請求項1に 記載の方法。 13.前記第一の質量スペクトルを表す前記データが複数の質量/電荷数対を含 む、請求項1に記載の方法。 14.予測質量スペクトルを計算する前記ステップが: 前記複数の配列の部分から複数の質量を導出し、但し各質量は前記複数の配列 内のある配列の一部に対応するペプチドフラグメントの質量に等しく; 前記複数の質量から、前記第一のペプチドの質量の所定の質量許 容差内にある質量を選択し、選択した前記質量の各々がどの配列のどの部分に対 応するかという指示を記憶して、複数の候補配列部分を与え;そして 前記候補配列部分の各々について複数の質量/電荷数対を計算し、前記質量/ 電荷数対の各々が前記候補配列部分のうちの一つの一部に対応するペプチドフラ グメントの質量に実質的に等しい質量を有する;ことから成る請求項1に記載の 方法。 15.前記第一の測定値が相関係数からなる、請求項1に記載の方法。 16.第一の測定値を計算する前記ステップが: 前記複数の候補配列部分の各々について予備得点を計算し; 主候補部分として識別されない少なくとも一つの候補配列より大きい予備得点 を有する複数の主候補部分を識別し;そして 前記主候補部分の各々について相関係数を計算する、ことからなる請求項1に 記載の方法。 17.前記複数の質量スペクトル及び前記第一の質量スペクトルの各々が複数の 質量/電荷数対を含んでおり、各質量/電荷数対が強度値を有し、更に、前記複 数の質量スペクトルの各々について、前記第一の質量スペクトル内で対応するフ ラグメントから所定の差より小さい差を有する一組の一致フラグメントを識別す るステップも含み、前記予備得点が前記一組の一致フラグメントの中で予測質量 スペクトルを有するフラグメントの数に、前記一致フラグメントに対応する質量 /電荷数対の強度値の合計を掛けたものである請求項8に記載の方法。 18.オリゴヌクレオチドの質量スペクトルを解読するための方法であって: ヌクレオチド配列ライブラリーを提供し; 前記ライブラリーからの複数のヌクレオチドサブ配列、即ちn量体より小さい 配列を総て含むヌクレオチドサブ配列をデータベース内に記憶し; 前記オリゴヌクレオチドの複数のフラグメントの第一の質量スペクトルを表す データを記憶し; 前記複数のヌクレオチドサブ配列の各々について予測質量スペクトルを計算し ;そして 前記予測質量スペクトルの各々について、前記第一の質量スペクトルに対し、 少なくとも第一の一致度測定値を計算する、ことからなる方法。 19.nが10である請求項18に記載の方法。 20.タンパク質混合物中のペプチドが配列データベース内のアミノ酸配列によ って特定される複数のタンパク質のうちのいづれかの一部に相応するかどうかを 調べるための方法であって: タンデム型質量分析計を用いて、前記タンパク質混合物から得られた複数ペプ チドを受容し、前記ペプチド混合物から少なくとも第一のペプチドを選択し、前 記第一のペプチドをフラグメント化し、ペプチドフラグメント質量スペクトルを 形成し; 前記第一の質量スペクトルを表すデータを記憶し;そして 前記質量スペクトルを前記配列データベース内のアミノ酸配列と相関させて、 前記配列データベース内で特定されたタンパク質と前記タンパク質混合物中のタ ンパク質との相似を調べる、ことからなる前記方法。 21.前記相関ステップが、前記アミノ酸配列から少なくとも一つの質量スペク トルを予測する操作を含む請求項20に記載の方法。 22.タンパク質混合物を、同定すべき細胞又は微生物から得る請求項20に記載 の方法。 23.タンパク質混合物を、細胞タンパク質のタンパク質分解消化によって得る 請求項22に記載の方法。 24.細胞タンパク質が細胞外タンパク質である請求項23に記載の方法。 25.同定すべき生体又はその成分から得たペプチドの一つ又は複数の質量スペ クトルが既知の生体のスペクトルライブラリーに含まれているかどうかを調べる ことにより所期の生体を同定する方法であって: タンデム型質量分析計を用いて、同定すべき前記生体から得たタンパク質混合 物から得られた複数のペプチドを受容し、前記複数のペプチドから少なくとも第 一のペプチドを選択し、前記第一のペプチドをフラグメント化し、ペプチドフラ グメント質量スペクトルを形成し; 前記第一の質量スペクトルを表すデータを記憶し;そして 前記質量スペクトルを既知の生体の前記スペクトルライブラリー内の質量スペ クトルに相関させて、前記スペクトルと、同定すべき生体から得たペプチドから 得られたスペクトルとの相似を調べ、それによって前記生体を同定することから なる方法。 26.同定すべき生体が細菌、真菌又はウイルスである請求項25に記載の方法。 27.タンパク質混合物を生体のタンパク質の酵素消化によって得る請求項25に 記載の方法。 28.炭水化物混合物から所期の炭水化物構造の特徴を決定するための方法であ って: タンデム型質量分析計を用いて、炭水化物混合物から得た複数の炭水化物を受 容し、タンデム型質量分析計の第一の質量分析計で炭水化物混合物から少なくと も第一の炭水化物を選択し、前記第一の 炭水化物をフラグメント化し、炭水化物フラグメント質量スペクトルを形成し; 前記第一の質量スペクトルを表すデータを記憶し;そして 前記質量スペクトルを既知の炭水化物のスペクトルデータベースと相関させて 、前記炭水化物データベース内で特定された炭水化物と、前記炭水化物混合物中 の炭水化物との相似を調べ、それによって所期の炭水化物の構造の特徴を決定す ることからなる方法。 29.炭水化物混合物を所期のグリコシル化タンパク質から得る請求項28に記載 の方法。 30.炭水化物混合物をタンパク質からの化学的又は酵素的遊離によって所期の グリコシル化タンパク質か得る請求項29に記載の方法。
