Description
Claims (1)
- 【特許請求の範囲】 1.試験試料中の核酸鎖を増幅する方法であって、 第一に、前記試験試料からの前記核酸鎖を、同時に少なくとも三つのオリゴヌ クレオチドプライマーと接触させ、ここで少なくとも一つのプライマーはプロモ ーター−プライマーであり、少なくとも一つの前記プライマーは前記核酸鎖に相 補的であり、および少なくとも一つの他の前記プライマーは前記核酸鎖に相補的 な鎖に相補的である;そして 第二に、前記核酸鎖およびプライマーを、RNA指向性および/またはDNA 指向性DNAポリメラーゼ活性、RNAポリメラーゼ活性およびRNAse H 活性を有する一つまたはそれ以上の蛋白質と、一定の温度で前記核酸鎖が増幅し うるプライマー長条件下で接触させる; の各工程を含む方法。 2.前記核酸鎖がDNA鎖であり、前記第二の接触工程の前に前記核酸鎖およ びプライマーを約60℃またはそれ以上で活性なDNAポリメラーゼ活性を有す る酵素と約60℃またはそれ以上で接触させる、請求項第1項に記載の方法。 3.前記第二の接触工程が、逆転写酵素の存在下、約42℃またはそれ以上で 行われる、請求項第1項に記載の方法。 4.前記核酸鎖および前記プライマーが前記第二の接触工程の前に、約95℃ またはそれ以上に加熱される、請求項第1項に記載の方法。 5.前記第一の接触工程において4つのプライマーがに使用される、請求項第 1−4項に記載の方法。 6.一つの前記プライマーが一つの他の前記プライマーと異なる濃度で提供さ れる、請求項第1項に記載の方法。 7.一つの前記プライマーが一つの他の前記プライマーと異なる濃度で提供さ れる、請求項第5項に記載の方法。 8.二つの前記プライマーが前記他のプライマーと異なる濃度で提供される、 請求項第5項に記載の方法。 9.二つの前記プライマーがプラス−センスプライマーであり、内側プラス− センスプライマーがプロモーター−プライマーである、請求項第1項に記載の方 法。 10.二つの前記プライマーがマイナス−センスプライマーであり、前記外側 プライマーがプロモーター−プライマーである、請求項第1項に記載の方法。 11.すべての前記酵素活性が逆転写酵素およびRNAポリメラーゼにより提 供される、請求項第1項に記載の方法。 12.前記酵素活性がDNAポリメラーゼ活性を有しないRNAse Hによ り補給される、請求項第11項に記載の方法。 13.前記DNAポリメラーゼが5’−3’エキソヌクレアーゼ活性を欠失し ている、請求項第2項に記載の方法。 14.前記DNAポリメラーゼが、5’−3’エキソヌクレアーゼ活性をその 天然形では保持しているDNAポリメラーゼから誘導される、請求項第13項に 記載の方法。 15.前記DNAポリメラーゼが、バシラス(Bacillus)種のDNA ポリメラーゼIである請求項第13または14項に記載の方法。 16.前記種がバシラス ステアロテルモフィラス(Bacillus st earothermophilus )またはバシラス カルドテナックス(Ba cillus caldotenax)である、請求項第15項に記載の方法。 17.二つの外側プライマーが前記核酸鎖またはその相補体に最大2000塩 基離れてハイブリダイズする、請求項第1項に記載の方法。 18.二つの外側プライマーが前記核酸鎖またはその相補体に最大500塩基 離れてハイブリダイズする、請求項第1項に記載の方法。 19.二つの外側プライマーが前記核酸鎖またはその相補体に最大350塩基 離れてハイブリダイズする、請求項第1項に記載の方法。 20.一つの前記プライマーが1から10μMの間の濃度で提供され、別の前 記プライマーが10から50μMの間の濃度で提供される、請求項第6項に記載 の方法。
