Claims (1)
- 【特許請求の範囲】 1.他のタンパク質を有する混合物中に酵素を含んでなる水性溶液から酵素を 分離する方法であって、該溶液から塩を浸出し、次いで酵素のpI付近のレベルに 溶液のpHを調節し、次いで引き続き酵素を結晶質の形態で回収することを含んで なる、前記方法。 2.ダイアフイルトレーション、透析、又は電気透析より又はゲル濾過により 、又はクロマトグラフィー法により、又は沈殿により塩を浸出する、請求の範囲 第1項記載の方法。 3.10mS/cm以下、好ましくは5mS/cm以下最も好ましくは2mS/cm以下のコ ンダクタンスが検出可能であるまで塩を浸出する、請求の範囲第1又は2項記載 の方法。 4.水性溶液のpHを酵素のpI±3付近の範囲内のpH値、より好ましくは酵素の pI±1.5付近の範囲内のpH値、更に一層好ましくは酵素のpI±1付近の範囲内のp H値、最も好ましくは酵素のpI±0.5付近の範囲内のpH値に調節する、請求の範囲 第1〜3項のいずれか1項に記載の方法。 5.水性溶液のpHをギ酸、クエン酸又は酢酸を用いて調節する、請求の範囲第 1〜4項のいずれか1項に記載の方法。 6.酵素がリパーゼである、請求の範囲第1〜5項のいずれか1項に記載の方 法。 7.水性リパーゼ浸有溶液が5%(w/w)以上、好ましくは10−20%(w/ w)のリパーゼ含量を有しそして10%超の総乾燥物質のリパーゼ含量、好ましく は30%超の総乾燥物質のリパーゼ含量に相当する純度を有する、請求の範囲第6 項記載の方法。 8.リパーゼが菌類起源でありそしてアスペルギルス(Aspergillus)(例え ばA.ニガー(niger))の菌株、カンジダ(Candida)( 例えばC.アンタルクチカ(antarctica))の菌株、フミコラ(Humicola)(例 えばH.ラヌギノザ(lanuginosa)およびH.インソレンス(insolens))の菌 株およびリゾムコーム(Rhizomucor)(例えばR.マイヘイ(miehei))の菌株 から得ることのできる請求の範囲第6又は7項記載の方法。 9.リパーゼが、細菌由来のものでありそしてシュードモナス(Pseudomonas )(例えば(Ps.セパシア(cepacia))の菌株から得ることのできる、請求の 範囲第6又は7項記載の方法。
