Abstract
Description
Claims (25)
- 1.以下に示す、 a.式 A1A2A3‥‥An−2An−1An3[式中、Anはその蛋白質もしくはそ の一部分のアミノ酸の位置nに存在するアミノ酸である] により表され、N変異型の集団を示すある蛋白質もしくはその一部分を選択する 段階、 b.N変異型における各アミノ酸の位置nに出現する各種類のアミノ酸の回数O n■■を決定する段階、 c.N変異型における各アミノ酸の位置nにおける各種類のアミノ酸の出現頻度 (On■■/N)nを算出する段階、およびd,式 A′1A′2A′3‥‥A′n−2A′n−1A′n[式中、A′nはN変異型 中において対応するアミノ酸の位置において選択された頻度を上回って出現する アミノ酸の種類として定義する]により実質的に表されるアミノ酸配列を有する ポリペプチド抗原のセットを作成する段階、 を含んでなる、ある蛋白質もしくはその一部分の変異型の集団から得られるアミ ノ酸配列を有するポリペプチド抗原のセットを産生させる方法。
- 2.ポリペプチド抗原のセットが8から150までのアミノ酸の範囲にわたる、 請求の範囲1に記載の方法。
- 3.選択された頻度が5%である、請求の範囲1に記載の方法。
- 4.選択された頻度が10%である、請求の範囲1に記載の方法。
- 5.蛋白質がある病原体の免疫原性構成成分である、請求の範囲1に記載の方法 。
- 6.病原体の免疫原性構成成分が一つもしくは複数の中和抗原エピトープを含む 、請求の範囲5に記載の方法。
- 7.蛋白質がウイルスの構成成分である、請求の範囲5に記載の方法。
- 8.蛋白質がHIV−1の構成成分である、請求の範囲7に記載の方法。
- 9.蛋白質が9P120もしくはその一部分である、請求の範囲8に記載の方法 。
- 10.ある蛋白質の一部分がgP120のV3ループである、請求の範囲9に記 載の方法。
- 11.ポリペプチド抗原のセットが、 i)各コドンの位置nにおいて最低数のヌクレオチド組合せ物を有し、そしてそ の組合せ物が少なくとも各種類のアミノ酸A′1からA′nまてをコードするコ ドンを含む、縮重オリゴヌクレオチド配列を合成し、そして ii)ある発現系内においてその縮重オリゴヌクレオチドを発現させてポリペプ チド抗原のセット A′1A′2A′3‥‥A′n−2A′n−1A′nを産生させることによりポ リペプチド抗原のセットを作製する、請求の範囲1に記載の方法。
- 12.請求の範囲1の方法により作製されるポリペプチド抗原のセット。
- 13.請求の範囲12のポリペプチド抗原のセットおよび薬剤学的に許容される 賦形剤を含んでなるワクチン組成物。
- 14.HIV−1変異型の集団のV3ループから得られるポリペプチド抗原のセ ットであって、そのポリペプチド抗原のセットはアミノ酸配列【配列があります 】 (配列番号11) [配列中、 Xaa1は、ThrおよびSerからなる群からより選択され、Xaa2は、A snおよびTyrからなる群より選択され、Xaa3は、AsnおよびLysか らなる群より選択され、Xaa4は、AsnおよびLysからなる群より選択さ れ、Xaa5は、ThrおよびIleからなる群より選択され、Xaa6は、A rgおよびIleからなる群より選択され、Xaa7は、LysおよびGlnか らなる群より選択され、Xaa8は、Ser、Gly、およびArgからなる群 より選択され、Xaa9は、Ile、Met、およびLeuからなる群より選択 され、Xaa10は、His、Asn、Arg、Ser、Pro、およびThr からなる群より選択され、 Xaa11は、Ile、Met、およびLeuからなる群より選択され、Xaa 12は、Arg、Lys、およびGlnからなる群より選択され、Xaa13は 、AlaおよびValからなる群より選択され、Xaa14は、Phe、Leu 、Ile、Met、およびValからなる群より選択され、 Xaa15は、Tyr、Phe、His、およびIeuからなる群より選択され 、 Xaa16は、Gly、Lys、Arg、およびGluからなる群より選択され 、 Xaa17は、Ile、Lys、およびArgからなる群より選択され、Xaa 18は、IleおよびThrからなる群より選択され、Xaa19は、Aspお よびTyrからなる群より選択され、Xaa20は、ArgおよびGlyからな る群より選択され、Xaa21は、HisおよびTyrからなる群より選択され る。]により表される、上記ポリペプチド抗原のセット。
- 15.請求の範囲14のポリペプチド抗原のセット、および生理学的賦形剤を含 んでなるワクチン組成物。
- 16.配列 【配列があります】 (配列番号10)により実質的に表される縮重オリゴヌクレオチドのセット。
- 17.請求の範囲16のオリゴヌクレオチドを含む発現ベクターのライブラリー 。
- 18.HIV−1変異型の集団のV3ループから得られるポリペプチド抗原のセ ットであって、そのポリペプチド抗原のセットが、一般的配列【配列があります 】 (配列番号7)[配列中、 Xaa1は、ThrおよびIleからなる群より選択され、Xaa2は、Asn およびSerからなる群より選択され、Xaa3は、ArgおよびLysからな る群より選択され、Xaa4は、ArgおよびLysからなる群より選択され、 Xaa5は、Arg、Gly、およびSerからなる群より選択され、Xaa6 は、Asn、Pro、Ser、Arg、Thr、およびHisからなる群より選 択され、 Xaa7は、MetおよびIleからなる群より選択され、Xaa8は、Lys およびArgからなる群より選択され、Xaa9は、ValおよびAlaからな る群より選択され、Xaa10は、IleおよびPheからなる群より選択され 、Xaa11は、HisおよびTyrからなる群より選択され、Xaa12は、 AlaおよびThrからなる群より選択され、Xaa13は、IleおよびTh rからなる群より選択され、Xaa14は、Arg、Asn、His、Gln、 Asp、およびGluからなる群より選択され、 Xaa15は、Gly、Arg、His、Gln、Asp、およびGluからな る群より選択され、 Xaa16は、PheおよびIleからなる群より選択され、Xaa17は、A la、Thr、Val、およびIleからなる群より選択され、 Xaa18は、AsnおよびAspからなる群より選択され、Xaa19は、L ysおよびGlnからなる群より選択される。]により表される、上記ポリペプ チド抗原のセット。

- 21.請求の範囲20のオリゴヌクレオチドを含む発現ベクターのライブラリー 。
