Description
Claims (22)
- 1.(a)核酸構築体の複製物を調製する工程、(b)該核酸構築体複製物を生 物学的に不活注な担体粒子上に被覆する工程、(c)未成熟の稲胚を単離する工 程、 (d)培養した鼓胚を標的表面上に配置する工程、(e)該培養した胚に該核酸 で被覆した担体粒子による衝撃を与える工程、ここで該粒子を該標的表面に向け て物理的に加速して、該粒子の幾つかを該胚細胞の少なくとも幾つかの内部に包 含させる、(f)該衝撃を与えた胚を栽培して、該脛から苗条を発生させる工程 、(g)工程(f)で形成した該苗条を性的に成熟した完全な植物に育成する工 程、および (h)工程(f)で形成した該苗条または工程(g)で形成した該植物中におけ る該遺伝子構築体の存在を明らかにする工程、を含む形質転換された稲植物の製 法。
- 2.更に、(i)該核酸構築体の遺伝性を確認する工程をも含む請求の範囲第1 項に記載の方法。
- 3.該胚が上記工程(e)の前に、予備的条件付けされる請求の範囲第1項に記 載の方法。
- 4.該予備的条件付けが、オーキシンの存在下で該胚を培養することを含む請求 の範囲第1項に記載の方法。
- 5.切り取られる該胚の長さが約0.5〜1.5mmである請求の範囲第1項に 記載の方法。
- 6.該切り取られる胚がインディカ変種の胚である請求の範囲第1項に記載の方 法。
- 7.該切り取られる胚かジャポニカ変種の胚である請求の範囲第1項に記載の方 法。
- 8.該工程(f)が、各胚から胚形成カルス培養物を誘発し、かつ該胚形成カル スからの該胚を培養して苗条とすることを含む請求の範囲第1項に記載の方法。
- 9.該工程(f)が、選別剤に暴露することにより形質転換される再生苗条の割 合を富化する工程を更に含む請求の範囲第1項に記載の方法。
- 10.該選別剤か除草剤ビアラホスである請求の範囲第9項に記載の方法。
- 11.請求の範囲第1項に記載の方法により生成される形質転換された稻植物。
- 12.請求の範囲第1項に記載の方法により、稲植物の親に挿入した外来遺伝子 構築体を含むトランスジェニック稲植物。
- 13.(a)核酸構築体の複製物を調製する工程、(b)該核酸構築体複製物を 生物学的に不活性な担体粒子上に被覆する工程、(c)稲苗から分裂性盤状組織 を単離する工程、(d)該分裂性盤状組織を標的表面上に配置する工程、(e) 該分裂性盤状組織に該核酸で被覆した担体粒子による衝撃を与える工程、ここで 該粒子を該標的表面に向けて物理的に加速して、該粒子の幾つかを該分裂性細胞 の少なくとも幾つかの内部に包含させる、(f)該衝撃を与えた分裂性盤状組織 を栽培して、少なくとも幾つかの該衝撃を与えた分裂性盤状組織から苗条を発生 させる工程、(g)工程(f)で形成した該苗条を性的に成熟した完全な植物に 育成する工程、および (h)工程(f)で形成した該苗条または工程(g)で形成した該植物中におけ る該遺伝子構築体の存在を明らかにする工程、を含む形質転換された稲植物の製 法。
- 14.更に、(i)該核酸構築体の遺伝性を確認する工程をも含む請求の範囲第 13項に記載の方法。
- 15.該分裂性盤状組織が上記工程(e)の前に、予備的条件付けされる請求の 範囲第13項に記載の方法。
- 16.該予備的条件付けが、オーキシンによる処理を含む請求の範囲第13項に 記載の方法。
- 17.切り取られる該分裂性盤状組織の長さか約0.5〜1.5mmである請求 の範囲第13項に記載の方法。
- 18.該切り取られる胚がインディカ変種の胚である請求の範囲第13項に記載 の方法。
- 19.該切り取られる胚がジャポニカ変種の胚である請求の範囲第13項に記載 の方法。

- 22.ゲノム中に、稲植物にグルタミンシンターゼ阻害性除草剤耐性を付与する タンパクを発現する遺伝性外来遺伝子構築体を含むトランスジェニック稲植物。
