Description
Claims (16)
- 1.特定の周波数の光によって励起された時に蛍光を発する複数の着色剤によっ て着色された検体を観察する際、光学顕微鏡と共に使用される多色共焦点顕微鏡 システムであって、 単一のレーザ光源から、各々が前記着色剤のうちの1つの励起周波数に対応する 多数の励起線、を含んだ多線入射レーザ光を発生するレーザ手段と、前記入射レ ーザ光を前記顕微鏡に導き入れ、且つ、前記顕微鏡から発せられ、各々が前記着 色剤のうちの1つの励起周波数に対応する多数の蛍光、を含む光を取り込むため の取込み手段と、 前記顕微鏡から発せられる光を取り込むよう配置されていて、前記光を電気信号 に変換する検出手段と、前記検出手段に接続されていて、前記電気信号を累積し 、正確な焦点面に、各々が前記レーザの線の1つに対応する前記検体の複数の像 、を作る制御手段とを具備した多色共焦点顕微鏡システム。
- 2.前記レーザ手段に接続されていて、2つ以上の前記着色剤を励起することと なる前記多数の励起線の発生を抑制するための手段を含む、請求項1の多色共焦 点顕微鏡システム。
- 3.前記入射レーザ光の光路に配設されていて、前記レーザ手段からの前記励起 線のうち選択された励起線を選択的に通過させるフィルタ手段を含む、請求項1 の多色共焦点顕微鏡システム。
- 4.前記顕微鏡から発生される光の光路に配設されていて、前記蛍光のうち選択 された蛍光を前記検出手段に対して選択的に通過させるフィルタ手段を含む、請 求項1の多色共焦点顕微鏡システム。
- 5.前記フィルタ手段が、各前記蛍光を分離した光路に分離し、各前記光路を前 記複数の検出器のうちの1つに導き入れるため前記顕微鏡から発生される光の光 路に配設された二色ミラーを含む、請求項4の多色共焦点顕微鏡システム。
- 6.前記検出手段が、一度に1つの線を検出し、時分割多重式に多色の光学像を 作るための手段を含む、請求項3の多色共焦点顕微鏡システム。
- 7.前記取込み手段が、前記入射レーザ光により前記検体をラスタ走査するため の手段を含み、前記検出手段が、前記反射された光を走査するための手段を含む 、請求項1の多色共焦点顕微鏡システム。
- 8.前記制御手段が、複数の擬似色で前記検体を表示する表示手段を有するコン ピュータを含み、各前記擬似色が前記着色剤のうちの1つの着色剤の発光に係る 蛍光に対応するものである、請求項1の多色共焦点顕微鏡システム。
- 9.前記レーザ手段が、単一のクリプトン・アルゴンレーザを含む、請求項1の 多色共焦点顕微鏡システム。
- 10.前記クリプトン・アルゴンレーザは、各線がフルオレスセイン着色剤、リ ザミンロダミン着色剤およびシアニン5.18着色剤のうちのせいぜい1つを励 起するような、複数のレーザ線を発生するものである、請求項9の多色共焦点顕 微鏡システム。
- 11.前記検出手段が光電子増倍管を含むものである、請求項1の多色共焦点顕 微鏡システム。
- 12.複数の着色剤によって着色された検体を観察する光学顕微鏡と共に使用さ れる多色共焦点顕微鏡システムであって、 複数の線を有するレーザ光を発生する多線レーザであって、前記線のうちのある ものは前記レーザから発するのが選択的に阻止され、前記線のうちの他のものが 前記レーザから励起線として選択的に発せられるようになっており、各前記発せ られる励起線が前記複数の着色剤のうちのただ1つ着色剤の励起周波数に対応す るものである、多線レーザと、 各々が前記レーザ光の光路に選択的に配置された複数の励起フィルタと、 前記励起フィルタの後方において前記レーザ光の光路に配置されており、励起線 を反射し、発せられた蛍光を通過するよう選択された第1の二色フィルタと、前 記第1の二色フィルタから前記レーザ光を取り込み、前記レーザ光を前記顕微鏡 中にラスタ走査パターンで反射するよう配設されており、さらに、前記顕微鏡か ら前記蛍光を取り込み、前記ラスタ走査パターンを走査し、前記第1の二色フィ ルタを介して前記蛍光を反射するよう配設されている走査ユニットと、 前記第1の二色フィルタの後方において前記蛍光の光路に配置されており、前記 発せられる光の前記蛍光のいくつかの波長を反射し、その他の波長を通過するよ う選択された第2の二色フィルタと、 前記第2の二色フィルタを通過した前記蛍光を取り込むよう配設された第1の検 出器と、 前記第2の二色フィルタから反射された前記蛍光を取り込むよう配設された第2 の検出器と を具備した多色共焦点顕微鏡システム。
- 13.特定の周波数の光によって励起された時に蛍光を発する複数の着色剤によ って着色された検体を観察する方法であって、 単一のレーザ光源から、各々が前記着色剤のうちの1つの励起周波数に対応する 多数の励起線、を含んだ多線入射レーザ光を発生する工程と、 前記入射レーザ光を前記検体を保持している前記顕微鏡に導き入れる工程と、 前記顕微鏡から発せられ、各々が前記着色剤のうちの1つ着色剤の励起周波数に 対応する多数の蛍光、を含む光を取り込む工程と、 前記顕微鏡から発せられる光を取り込んで、該光を電気信号に変換する工程と、 前記電気信号を累積し、正確な焦点面に、各々が前記レーザの線の1つに対応す る前記検体の複数の像、を作る工程 とからなる方法。
- 14.2つ以上の前記着色剤を励起することとなる前記多数の励起線の発生を抑 制する工程を含む、請求項13の方法。
- 15.前記入射レーザ光をフィルタ処理し、前起励起線のうち選択された1つの 励起線を選択的に通過させる工程を含む、請求項13の方法。
- 16.前記発生される光をフィルタ処理し、前記蛍光のうち選択された蛍光を選 択的に通過させる、請求項13の方法。
