Description
本願は、2017年3月30日付け出願の米国仮特許出願番号62/478,789の利益を主張するものであり、その開示のすべてを出典明示により本明細書に取り込むものとする。
本発明は、一般に、乾癬などの自己免疫性および自己炎症性疾患の治療用の臨床試験中にある、Tyk2阻害剤である、6-(シクロプロパンアミド)-4-((2-メトキシ-3-(1-メチル-1H-1,2,4-トリアゾール-3-イル)フェニル)アミノ)-N-(メチル-d3)ピリダジン-3-カルボキシアミドの製造方法に、ならびに該方法にて用いられる新規な中間体に関する。

分離反応器に水(7.5Kg)を入れ、5℃に冷却した。反応物を、内部温度を5℃より下に維持しながら、該水溶液にゆっくりと添加した。該反応器を濯ぎ、移すのを助けるために、さらなる水(0.5Kg)を使用した。得られた混合物を5℃で3時間かき混ぜ、次にMTBEで3回(4x4.5Kg)抽出した。有機層を合わせ、pH7の緩衝水溶液(5.0L/Kg、15重量%KH2PO4/K2HPO4)および水(2.5Kg)で連続して洗浄した。該粗製物を、総容量がほぼ3L/Kgとなるまで、真空下で蒸留させた。ACN(2x6.3Kg)を加え、つづいてさらに蒸留を行って約3L/Kgに戻した。該粗製物を20℃に冷却し、化合物2を30-36重量%溶液として収率90-95%で得た。
20℃で、トルエン(3.46Kg、4.0L/Kg)およびACN(1.57Kg、2.0L/Kg)を、窒素でフラッシュ処理に付したガラスライニング反応器に入れた。化合物8(1.00Kg)を、つづいてDBU(0.39Kg、1.00当量)を添加した。反応器のラインをトルエン(0.43Kg、0.5L/Kg)で濯いだ。化合物10(0.54Kg、2.5当量)およびK2CO3(325メッシュ等級、0.70Kg、2.0当量)を該反応混合物に添加し、つづいてトルエン(1.30Kg、1.5L/Kg)およびACN(0.79Kg、1.0L/Kg)を加えた。プリホームの触媒溶液を該反応混合物に移し、次にそれを75℃に加熱し、反応が完了するまで、かき混ぜた。

必要に応じて、生成物は次のように任意の再結晶操作に供することができる。
NMP(6.2kg、6.0L/Kg)および化合物I(1.0Kg)をガラスライニング反応器に入れた。バッチを70℃に加熱して淡黄色の溶液を形成させ、次にそれを70℃でポリッシュフィルターを介してきれいな容器に移した。2-プロパノール(2.4Kg、3L/Kg)を加え、つづいて化合物Iの種(0.005Kg、0.005Kg/Kg)を添加した。1時間放置した後、さらなる2-プロパノール(4.8kg、6L/Kg)を2時間にわたって入れた(3L/Kg/時間)。そのスラリーを70℃で1時間放置し、0℃にゆっくりと冷却し、0℃でさらに12時間放置した。生成物を濾過で単離した。湿ったケーキを2-プロパノール(2x3.1kg、2x4L/Kg)で洗浄し、その後で高真空下にて65℃で乾燥させた。化合物Iを>99.9%の純度および収率83%にて単離した。
分離反応器において、水(12Kg/Kg)およびメタノール(6.5Kg/Kg、8.3L/Kg)を20℃でよく混合した。該ニトロ化粗製物をメタノール-水の混合液にゆっくりと移した。反応器のラインをメタノール(0.5Kg/Kg)で濯いだ。該粗製物を40-45℃に加熱した。水性水酸化アンモニウム(25重量%、7.4Kg/Kg)をゆっくりと添加した。得られたスラリーを20℃に冷却し、3時間かき混ぜた。生成物を濾過により単離した。ケーキを水(2x6L/Kg)で洗浄し、真空下の45℃で乾燥させた。化合物6を99APおよび収率95%において単離した。
必要に応じて、生成物は次のように任意の再結晶操作に供することができる。メタノール(5.6Kg、8.3L/Kg)および化合物13(1.0Kg)をガラスライニング反応器に入れた。DBU(0.1Kg)をゆっくりと添加した。粗製物を1時間かき混ぜた。THF(12.4Kg、13.9L/Kg)をゆっくりと添加し、得られたスラリーを2時間放置した。生成物を濾過で単離した。ケーキをTHF(2.6Kg、2.9L/Kg)で洗浄し、真空下の45℃で乾燥させた。化合物13を収率60%にて単離した(第1収量)。母液を、総容量が約1L/Kgになるまで、真空下で蒸留した。メタノール(2x2.8Kg、2x3.6L/Kg)とのプット/テイク蒸留を2回行い、該溶液を濃縮して約1L/Kgに戻した。該粗製物を20℃に冷却した。THF(4.8Kg、5.4L/Kg)を加え、得られたスラリーを2時間放置した。生成物を濾過で単離した。ケーキをTHF(1.0Kg)で洗浄し、真空下の45℃で乾燥させた。化合物13を収率25%にて単離した(第2収量)。
