(a)任意選択により、少なくとも1つのキナーゼ阻害剤を含むレジメンで患者を前処置するステップ;

Claims (23)
- 末梢血から末梢血リンパ球(PBL)を拡大培養する方法であって、
(a)イブルチニブ又は別のインターロイキン-2誘導性T細胞キナーゼ(ITK)阻害剤で前処置され、且つイブルチニブ又は当該別のITK阻害剤での処置に抵抗性である患者の末梢血から得られた末梢血単核細胞(PBMC)のサンプルを、IL-2及び抗CD3/抗CD28抗体を有する第1の培養培地を含む培養物で培養し、それにより前記PBMCから末梢血リンパ球(PBL)の拡大培養を行うステップ;
(b)ステップ(a)の培養物から前記PBLを回収するステップ、を含み、
前記方法が9日間、10日間、11日間、12日間、13日間及び14日間から成る群から選択される期間にわたって実施され、拡大培養されたPBLが、イブルチニブ又は前記別のITK阻害剤で前処置されなかった患者に由来する拡大培養された細胞より増加したレベルのIFNγ産生を含む、方法。 - ステップ(a)の前に、前記PBMCがCD19+選択に供されて、前記PBMCがCD19+PBMC及び非CD19+PBMCに分離され、ステップ(a)が非CD19+PBMCを培養することにより実施される、請求項1に記載の方法。
- ステップ(a)において、抗CD3/抗CD28抗体が磁気ビーズに結合されており、前記ビーズと細胞との比が培養物において3:1である、請求項1又は2に記載の方法。
- ステップ(a)において、前記PBMCの培養の4日目に追加のIL-2が培養物に添加され、第1の培養培地が培養物中で交換される、請求項1又は2に記載の方法。
- ステップ(a)における磁気ビーズが、抗CD3/抗CD28抗体で固定化されたビーズである、請求項3に記載の方法。
- 第1の培養培地が培養物中で第2の培養培地に交換される、請求項4に記載の方法。
- 第1の培養培地が第2の培養培地と異なる、請求項6に記載の方法。
- 第1の培養培地がCM-2であり、第2の培養培地がCM-4である、請求項6に記載の方法。
- 末梢血が血液悪性腫瘍に罹患している患者の末梢血に由来する、請求項1~8のいずれか一項に記載の方法。
- 血液悪性腫瘍が液性腫瘍である、請求項9に記載の方法。
- 液性腫瘍が白血病である、請求項10に記載の方法。
- 液性腫瘍が慢性リンパ性リンパ腫である、請求項11に記載の方法。
- 第1の培養培地が3000IU/mlのIL-2を含み、そして/あるいは培養物が37℃、5%CO2でインキュベートされる、請求項1又は2に記載の方法。
- 前記方法が9日間にわたり実施されるか、又は前記方法が11日間にわたり実施される、請求項1又は2に記載の方法。
- 前記患者がイブルチニブで前処置され、且つイブルチニブでの処置に抵抗性である、請求項1又は2に記載の方法。
- 前記患者がイブルチニブで少なくとも3ヶ月前処置される、請求項15に記載の方法。
- イブルチニブ又は別のITK阻害剤で前処置される前に少なくとも1ヶ月イブルチニブ又は前記別のITK阻害剤での処置を受けていない、請求項1又は2に記載の方法。
- 血液悪性腫瘍の治療における使用のため末梢血リンパ球(PBL)であって、
PBLが、末梢血単核細胞(PBMC)から拡大培養する方法であって、
(a)イブルチニブで前処置され、且つイブルチニブでの処置に抵抗性である患者の末梢血から得られた末梢血単核細胞(PBMC)であって、IL-2及び抗CD3/抗CD28抗体を有する第1の培養培地を含む培養物中にある、以前に凍結保存されたPBMCのサンプルを入手し、それにより前記PBMCから末梢血リンパ球(PBL)の拡大培養を行うステップ;
(b)IL-2及び抗CD3/抗CD28抗体を有する第1の培養培地を含む培養物中で前記PBLを培養するステップであって、前記方法が9日間、10日間、11日間、12日間、13日間及び14日間から成る群から選択される期間にわたって実施され、それにより前記PBMCから末梢血リンパ球(PBL)の拡大培養を行うステップ;及び
(c)ステップ(b)における前記培養物からPBLを回収するステップ、
を含む、
方法によって得られ、拡大培養されたPBLが、新鮮なPBMCから拡大培養された細胞より増加したレベルのIFNγ産生を含む、末梢血リンパ球(PBL)。 - 末梢血リンパ球(PBL)が、イブルチニブ又は別のITK阻害剤で前処置される前に少なくとも1ヶ月イブルチニブ又は前記別のITK阻害剤での処置を受けていない患者への投与のためのものである、請求項18に記載の末梢血リンパ球(PBL)。
- 血液悪性腫瘍が急性骨髄性白血病(AML)、マントル細胞リンパ腫(MCL)、濾胞性リンパ腫(FL)、びまん性大細胞型B細胞リンパ腫(DLBCL)、活性化B細胞(ABC)DLBCL、胚中心B細胞(GCB)DLBCL、慢性リンパ性白血病(CLL)、小リンパ球性白血病(SLL)、非ホジキンリンパ腫(NHL)、ホジキンリンパ腫、再発性及び/又は難治性ホジキンリンパ腫、B細胞急性リンパ芽球性白血病(B-ALL)、成熟B-ALL、バーキットリンパ腫、ワルデンストローム型マクログロブリン血症(WM)、多発性骨髄腫、骨髄異形成症候群、骨髄線維症、慢性骨髄性白血病、濾胞中枢リンパ腫、無痛性NHL、ヒト免疫不全ウイルス(HIV)関連B細胞リンパ腫並びにエプスタイン-バーウイルス(EBV)関連B細胞リンパ腫からなる群から選択される、請求項18又は19に記載の末梢血リンパ球(PBL)。
- 血液悪性腫瘍が液性腫瘍である、請求項18~20のいずれか一項に記載の末梢血リンパ球(PBL)。
- 液性腫瘍が白血病である、請求項21に記載の末梢血リンパ球(PBL)。
- 液性腫瘍が慢性リンパ性リンパ腫である、請求項22に記載の末梢血リンパ球(PBL)。
