ヒトアルファシヌクレインモノマー及び繊維の発現
ヒトアルファ-シヌクレインをコードする遺伝子を合成によって作製し、標準分子生物学技術を使用してベクターpMH 10His TEV(CMVプロモーターを含有する)中にサブクローニングして、N末端10His-TEVタグを有するアルファシヌクレインを生成するように遺伝子操作されたベクターを作出した。得られたベクターを、Expi293(商標)発現系(Invitrogen)を製造業者のプロトコールに従って使用して、Expi293F細胞中にトランスフェクトした。培養培地中に蓄積したアルファシヌクレインタンパク質を、固定化金属イオンアフィニティークロマトグラフィーHisTrapエクセルカラム(GE Healthcare)を使用してそこから回収した。25mM TrisHCl、300mM NaCl、pH8.0を用いてカラムを洗浄し、同バッファー中500mMイミダゾールの段階的勾配を用いてタンパク質を溶出した。10Hisタグを、TEVプロテアーゼを使用して除去した。次いで、サンプルを濃縮し、脱塩し、その後、切断されたタンパク質をHisTrapエクセルカラムに再度アプライし、切断されたアルファシヌクレインをフロースルー中に収集した。HiLoad 26/600 Superdex75カラム(GE Healthcare)でのゲル濾過によってアルファシヌクレインをさらに精製し、Proteus NoEndoカートリッジ(Generon)に通すことによって内毒素を除去した。精製されたSEC MALSによってアルファシヌクレインがモノマーであることを確認した(図1A)。
種々の種及び免疫原を使用する多数の免疫処置戦略を実施した。抗体5811は、50μgの上記のように発現された野生型のタグが付けられていないアルファシヌクレインモノマー(配列番号8)を用いて皮下免疫処置を施されていた雌Sprague Dawleyラット(>180g)に由来した。
B細胞培養物を、Tickleら、2015年 J Biomol Screen:第20巻(4号)、492~497頁によって記載されるものと同様の方法を使用して調製した。手短には、免疫処置された動物から得たB細胞に由来するリンパ節又は脾細胞を、ウェル当たりおよそ2000~5000個細胞の密度で、バーコード付き96ウェル組織培養プレート中で、200μl/ウェルの、10%FCS(Sigma Aldrich)、2% HEPES(Sigma Aldrich)、1% L-グルタミン(Gibco BRL)、1%ペニシリン/ストレプトマイシン溶液(Gibco BRL)、0.1% β-メルカプトエタノール(Gibco BRL)、1%の活性化ヒトPBMC上清(BSS)及びX線照射突然変異体EL4マウス胸腺腫細胞(5×104個/ウェル)を補給したRPMI 1640培地(Gibco BRL)を用いて、5% CO2の雰囲気中37℃で7日間培養した。培養は、免疫処置したすべての動物から得たB細胞を使用して設定し、合計で、およそ1.7×109個のB細胞をサンプリングした。
B細胞培養上清中のヒトアルファシヌクレイン特異的抗体の存在を、標的抗原の供給源としてビオチン化組換えヒトアルファシヌクレイン全長モノマーを用いてコーティングされたSuperavidin(商標)ビーズ(Bangs Laboratories)を使用する均一蛍光ベース結合アッセイを使用して調べた。本明細書に記載されるような組換えヒトアルファシヌクレインを、3倍モル過剰のビオチンを使用してビオチン化した。アルファシヌクレイン分子内の7個のリシン残基の完全修飾を避けるために低モル過剰のビオチンを使用した。アルファシヌクレインモノマーをビオチンとともに40℃で一晩インキュベートし、翌日、Zeba(商標)スピン脱塩カラムを使用して遊離ビオチンを除去した。スクリーニングは、Agilent Bravo液体ハンドラーを使用する、バーコード付き96ウェル組織培養プレートから、Superavidinビーズ(10ul/ウェル)上に固定化されたビオチン化組換えヒトアルファシヌクレインモノマーを含有するバーコード付き384ウェル黒色壁アッセイプレート中への10μlの上清の移動を含んでいた。結合は、ヤギ抗ラットIgG Fcγ断片特異的Alexafluor647コンジュゲート(Jackson)を用いて示した。アルファシヌクレイン特異的IgGを含有するウェルを同定するためにTTP Labtech Mirrorballでプレートを読み取った。
一次スクリーニング後、Beckman Coulter BiomekNXPヒットピッキングロボットを使用して、陽性上清を96ウェルバーコード付きマスタープレート上に統合し、細胞培養プレート中のB細胞を-80℃で凍結した。次いで、ビオチン化組換えヒトアルファシヌクレインモノマー又はビオチン化組換えヒトアルファシヌクレイン繊維を使用するストレプトアビジン捕獲ELISAアッセイにおいてマスタープレートをスクリーニングした。これは、モノマー及び繊維状組換えヒトアルファシヌクレイン療法との結合を与えたウェルを同定するために、またSuperavidin(商標)ビーズとのオフターゲット結合を示す何らかの偽陽性ウェルを排除するために実施した。繊維の不溶性を考慮すると、溶液中のタンパク質とともに使用される従来のELISAコーティングプロトコールは好都合ではなかった。繊維状構造を保つため、また、ストレプトアビジンを用いて事前コーティングされたELISAプレート上での繊維の効率的なコーティングを促進するために、最小のビオチン化プロトコールが使用されることが決定された。
対象の上清の選択からの抗体可変領域遺伝子の回収を可能にするために、B細胞の不均一な集団を含有していた所与のウェルにおける抗原特異的B細胞の同定を可能にするための逆重畳積分ステップを実施しなくてはならなかった。これは、蛍光焦点法(Clargoら、2014年MAbs:第6巻(1号)、143~159頁)を使用して達成した。手短には、陽性ウェルから得た免疫グロブリン分泌B細胞を、ビオチン化組換えヒトアルファシヌクレイン繊維でコーティングされたストレプトアビジンビーズ(New England Biolabs)(上記のように1:50ミックスを使用して作製された)及びヤギ抗ウサギFcγ断片特異的FITCコンジュゲート(Jackson)の1:1200最終希釈物と混合した。37℃で1時間の静的インキュベーション後、B細胞の周囲の蛍光ハロの存在のために抗原特異的B細胞を同定できた。Olympus顕微鏡を使用して同定されたいくつかのこれらの個々のB細胞クローンを、次いで、Eppendorfミクロマニピュレーターを用いて選び取り、PCR管中に蓄積させた。
次いで、精製されたラット-マウスキメラ抗体を、ELISAによるさらなるスクリーニング付した。ビオチン化組換えヒトアルファシヌクレインモノマー及び繊維を、炭酸含有バッファー(dH2O+0.16% Na2CO3+0.3% NaHCO3)中のストレプトアビジンを用いてコーティングされた384ウェルMaxisorpプレート(ThermoScientific/Nunc)上に捕獲した。別個のプレートはまた、配列番号8によるヒトアルファシヌクレインの残基117~126(ペプチド PVDPDNEAYE)に対応するビオチン化ペプチドを用いてコーティングして、一過性のものが、これ又は分子上の異なる領域に結合したか否かを調べた。プレートを、1% w/v PEG/PBSを用いてブロッキングし、次いで、精製一過性上清のいくつかの希釈物とともにインキュベートした。二次HRPコンジュゲート型ヤギ抗マウスIgG Fc抗体(Stratech Scientific Ltd/Jackson ImmunoResearch)をプレートに添加し、続いて、TMB基質(3,3’,5,5’-テトラメチルベンジジン、EMD Millipore製;10μl/ウェル)を用いて結合を可視化した。光学濃度は、BioTek Synergy2マイクロプレートリーダーを使用して630nMで測定した。IgG Fcγ1(配列番号34及び35を含む)としての、及びFab(配列番号34及び36を含む)としての抗体5811についてのデータは、図3A及び3Bに示されている。見られるように、5811抗体は、モノマー及び繊維状組換えヒトアルファシヌクレイン両方との結合を示し、PVDPDNEAYEペプチドとの結合も示す。
抗体特性決定
Biacore動態
Biacore T200機器で表面プラズモン共鳴技術を使用することによって相互作用動態を決定した。本明細書に記載されるように調整した、組換え全長ヒトアルファシヌクレインモノマー、精製組換えヒトアルファシヌクレイン繊維及び精製組換えマウスアルファシヌクレイン繊維を含む3種の異なるリガンドを、アミンカップリング化学を使用してCM5チップ表面の3種の異なるフローセル上に各々固定化した。3種のリガンドを10mM NaAc、pH3.5中で調製し、10μl/分の流速で、それぞれ、アルファシヌクレインモノマーについての約30反応単位(RU)、ヒトアルファシヌクレイン繊維についての約40RU及び、マウスアルファシヌクレイン繊維についての約300RUの固定化レベルに到達するように別個のフローセル表面に固定化させた。リガンド固定化及び動態アッセイ両方のためのランニングバッファーとして、バッファーHBS-EP+(GE healthcare Bio-Sciences AB)を使用した。次いで、抗アルファシヌクレインモノクローナル5811 mIgG1抗体及びモノクローナル5811 mFab抗体の、3種のリガンドに対する結合を測定した。モノクローナルmIgG1又はmFab抗体を、800nM~0.195nMの7種の異なる濃度で、3つのフローセルにわたって、3分の接触時間及び30分の解離時間を用い、100μl/分の流速で注入した。10μl/分で90秒間の50mM HClの1回の注入及び10μl/分で60秒間の50mM HClの別の注入によって表面を再生した。データは、バルク寄与なし(RI=0)及びmIgG1形式のためのグローバルRmaxを仮定した二価解析物モデル並びに柔軟なバルク寄与(ローカルRI)及びグローバルRmaxを用いる1:1モデルを使用してBiacore T200評価ソフトウェア(バージョン3.0)を使用して解析した。
ヒトアルファシヌクレインに対して産生された抗体のヒトベータシヌクレインとの結合を、rペプチドベータシヌクレインを使用するウエスタンブロットによって試験した。1マイクログラムのシヌクレインを4~12% Bis/Tris ゲルに流し、PVDFメンブレン上にブロットした。メンブレンを、3% BSA及び0.1% Tween20を有するPBS中でブロッキングした。ブロッキングされたブロットに抗体5811mIgG1を添加し、室温で1時間インキュベートし、PBS、0.1% Tween20を用いて洗浄し、二次抗体-HRPコンジュゲート(抗ウサギH+L HRPコンジュゲート、Bethyl、A120-101P)とともに1時間インキュベートした。ブロットを、0.1% Tween20を有するPBS、PBS及び水で十分に洗浄した。ECLウエスタンブロット基質(Pierce)の添加後に化学発光を測定した。図4(A)レーン3に示されるように、抗体5811mIgG1は、ヒトベータ-シヌクレインと結合しない。
NMR
ヒトアルファ-シヌクレインを、タンパク質が任意のタグを伴わずに発現されるようにpET28a発現ベクター中にクローニングした。構築物を大腸菌BL21(DE3)細胞(Stratagene)中に形質転換し、細胞を、重水(D2O)の存在下及び不在下でC13標識されたDL-グルコース及びN15標識された硫酸アンモニウムを有する規定の培地で増殖させた。OD600nm=1で300mM IPTGを用いて発現を誘導し、培養物を30℃で4時間インキュベートした。細胞をペレットにし、100ml溶解バッファー(20mM Tris/HCl pH8.0、25ユニットのベンゾナーゼ(Merck Millipore)、完全EDTA不含プロテアーゼ阻害剤カクテル(2錠、Roche)及び10mgリゾチーム(Sigma))中での3回の凍結解凍サイクルによって溶解した。溶解物を18000rpmでの遠心分離によって清澄化し、清澄化された溶解物を0.22μmフィルター(Stericup、Millipore)に通した。滅菌溶解物を、20mM Tris/HCl pH8.0、5CVを用いて平衡化したMonoQ 10/100GL(GE Healthcare)の頂部にロードし、同バッファー中500mMへのNaCl勾配を用いてタンパク質を溶出させた。最も純粋な画分のさらなる精製を、20mM Tris/HCl pH8.0での5倍希釈後MonoQ 10/100GLカラムで反復した。最も純粋な画分をプールし、10kDa MWCO遠心濃縮機(Centriprep、Millipore)を用いて濃縮し、HiLoad 26/600 Superdex75カラム(GE Healthcare)でのサイズ排除によって精製し、25mMリン酸ナトリウムバッファー、100mM NaCl(pH6.4)で溶出した。Superdex 75カラムから得た画分をプールし、ナトリウムアジド(0.02%最終濃度)及びAEBSF(10、μM最終濃度)を添加した。最終タンパク質濃度は、およそ5mg/mlであった。
NMRサンプルは、通常、5mm Shigemi管中の360μM 13C/15N標識された、又は430μM 2H/13C/15N標識されたヒトアルファシヌクレインのタンパク質濃度を有する、容量350μlとした。バッファー条件は、100mM NaCl、25mMリン酸ナトリウムpH6.4、10μM AEBSF、0.02% NaN3とした。すべての実験は、低温貯蔵用冷却プローブが取り付けられた600MHz Bruker AVIII又は800MHz Bruker AVIIスペクトロメーターのいずれかで20℃で記録した。タンパク質、HN(i)-N(i)-N(i±1)中の残基の骨格NMRシグナル間の連続接続は、それぞれ15N、15N及び1H次元において、28、28及び10ppmのスペクトル幅並びに117(F1)、117(F2)及び140(F3)ミリ秒の獲得時間を用い、増分当たり8回スキャン及び1.5秒の緩和遅延を用いて記録された、3D(H)N(CA)NNH実験(Weisemannら、1993年「NHの連続割当のための3D三重共鳴NMR技術並びに15N-及び13C-標識タンパク質における15N共鳴」(3D Triple-resonance NMR techniques for the sequential assignment of NH and 15N resonances in 15N- and 13C-labelled proteins.)J.Biomol.NMR 3)を使用して行った。10%(40000のハイパーコンプレックス(hyper-complex)点のうち4000)のサンプリング密度を有する均一でないサンプリングを採用して、2.75日の総取得時間をもたらした。TROSY-HNCA(Grzesiek及びBax、1992年「31kDaのタンパク質に適用された改善された3D三重共鳴NMR技術」(Improved 3D triple-resonance NMR techniques applied to a 31 kDa protein.)J.Magn.Reson.第96巻、432~440頁;Salzmannら、1998年「三重共鳴実験におけるTROSY:大きなタンパク質の連続NMR割当の新規展望」(TROSY in triple-resonance experiments:new perspectives for sequential NMR assignment of large proteins.)Proc.Natl.Acad.Sci.USA.第95巻、13585~90頁)及びTROSY-HNCACB(Wittekind及びMueller、1993年「HNCACB、アミド-プロトン及び窒素共鳴を、タンパク質中のアルファ-及びベータ-炭素共鳴と関連づけるための高感度3D NMR実験」(HNCACB,a High-Sensitivity 3D NMR Experiment to Correlate Amide-Proton and Nitrogen Resonances with the Alpha- and Beta-Carbon Resonances in Proteins.)J.Magn.Reson.Ser.B 第101巻、201~205頁;Salzmannら、1999年「大きなタンパク質の連続NMR割当のTROSY型三重共鳴実験」(TROSY-type Triple Resonance Experiments for Sequential NMR Assignment of Large Proteins.)J.Am.Chem.Soc.第121巻、844~848頁)実験を使用して、連続接続が確認され、残基種類が同定された。TROSY-HNCA実験は、それぞれ13C、15N及び1H次元において23、28、10ppmのスペクトル幅及び12.1(F1)、21.7(F2)及び100(F3)ミリ秒の取得時間(増分当たり8回スキャン、1.5秒の緩和遅延、1日の総取得時間)を用いて記録したが、TROSY-HNCACBは、それぞれ13C、15N及び1H次元において56、28、10ppmのスペクトル幅及び8.2(F1)、21.7(F2)及び100(F3)ミリ秒の取得時間(増分当たり8回スキャン、1.5秒の緩和遅延、1.7日の総取得時間)を用いて記録した。骨格カルボニル割当は、それぞれ13C、15N及び1H次元において10、29、10ppmのスペクトル幅及び80(F1)、21.7(F2)及び150(F3)ミリ秒の取得時間(増分当たり8回スキャン、1.5秒の緩和遅延)を用いて記録されたTROSY-HNCOスペクトルから得た(Grzesiek及びBax、1992年「31kDaのタンパク質に適用された改善された3D三重共鳴NMR技術」(Improved 3D triple-resonance NMR techniques applied to a 31 kDa protein.)J.Magn.Reson.第96巻、432~440頁;Salzmannら、1998年「三重共鳴実験におけるTROSY:大きなタンパク質の連続NMR割当の新規展望」(TROSY in triple-resonance experiments: new perspectives for sequential NMR assignment of large proteins.)Proc.Natl.Acad.Sci.USA.第95巻、13585~90頁)。15%(8050のハイパーコンプレックス(hyper-complex)点のうち1208)のサンプリング密度を有する均一でないサンプリングを採用して、19時間の総取得時間をもたらした。NMRスペクトルは、NMRPipe(Delaglioら、1995年「NMRPipe:a multidimensional spectral processing system based on UNIX(登録商標)pipes.」J.Biomol. NMR 第6巻、277~93頁)を使用して処理し、線形予測を使用して、最大1倍まで窒素における有効な取得時間を拡大した。均一でないサンプリングされたデータを、Harvard反復性ソフト閾値法を使用して再構築し(Hybertsら、2012年「多次元ポアソンギャップスケジューリングを用いて均一でなくサンプリングされたNMRデータの迅速な再構築のための反復性ソフト閾値の適用」(Application of iterative soft thresholding for fast reconstruction of NMR data non-uniformly sampled with multidimensional Poisson Gap scheduling.)J Biomol NMR 第52巻、315~27頁)、データを次のフーリエ数に再構築し、間接取得時間を最大60%まで増大した。データ解析はSparky(カリフォルニア大学、サンフランシスコ中、Goddard及びKneller、D.G. SPARKY 3.)を使用して実施し、133残基のアミドプロトン及び窒素共鳴の割当が得られ、これは、プロリン残基及びN末端メチオニンを除く残基の99%に相当した。割り当てられなかったアルファシヌクレインの唯一のその他の残基は、位置2のアスパラギン酸であった。
10%モル過剰の未標識5811Fabを含有する2H/13C/15N標識されたヒトα-シヌクレインの150μMのサンプルを使用して、5811の結合部位のマッピングを実施した。α-シヌクレインの骨格割当について上記で記載されたものと同一のバッファーでサンプルを調製した。アルファシヌクレイン/Fab複合体で記録されたTROSY-HNCO(Grzesiek及びBax、1992年「31kDaのタンパク質に適用された改善された3D三重共鳴NMR技術」(Improved 3D triple-resonance NMR techniques applied to a 31 kDa protein.)J.Magn.Reson.第96巻、432~440頁;Salzmannら、1998年「三重共鳴実験におけるTROSY:大きなタンパク質の連続NMR割当の新規展望」(TROSY in triple-resonance experiments:new perspectives for sequential NMR assignment of large proteins.)Proc.Natl.Acad.Sci.USA.第95巻、13585~90頁)スペクトルの、遊離アルファシヌクレインで記録された等価対照スペクトルとの比較によって、1H、15N及び13C化学シフト変化を決定した。遊離アルファシヌクレインの対照TROSY-HNCO実験は、それぞれ13C、15N及び1H次元において10、28及び10ppmのスペクトル幅及び80(F1)、1.7(F2)及び150(F3)ミリ秒の取得時間(増分当たり16回スキャン、1.4秒の緩和遅延)を用いて記録した。25%(8050のハイパーコンプレックス(hyper-complex)点のうち2013)のサンプリング密度を有する均一でないサンプリング(NUS)を採用して、2.5日の総取得時間をもたらした。α-シヌクレイン/Fab複合体のTROSY-HNCO実験は、それぞれ13C、15N及び1H次元において10、28及び10ppmのスペクトル幅及び80(F1)、21.7(F2)及び80(F3)ミリ秒の取得時間(増分当たり32回スキャン、1.5秒の緩和遅延)を用いて記録した。25%(4477のハイパーコンプレックス(hyper-complex)点のうち1119)のサンプリング密度を有する均一でないサンプリングを採用して、2.8日の総取得時間をもたらした。NMRスペクトルは、NMRPipe(Delaglioら、1995年「NMRPipe:a multidimensional spectral processing system based on UNIX(登録商標) pipes.」J.Biomol.NMR 第6巻、277~93頁)を使用して処理し、NUSデータの再構築は、mddnmrを使用して実施した。(OrekhovとJaravine、2011年「多次元分解を用いる均一でないサンプリングスペクトルの解析」(Analysis of non-uniformly sampled spectra with Multi-Dimensional Decomposition.)Prog.Nucl.Magn.Reson.Spectrosc.、第59巻、271~292頁)。窒素次元の有効な取得時間は、データ再構築の際に最大1倍まで増大された。
[式中、ΔδHN、ΔδN及びΔδCは、それぞれ1H、15N及び13C化学シフトにおける相違である]。αN及びαCは、それぞれ0.2及び0.35のスケーリング係数に対応し、アミドプロトン、窒素及びカルボニル化学シフトの化学シフト範囲における相違を説明するために使用される。
抗体5811mFabによって結合されるエピトープのさらなる特性決定を、ヒトアルファシヌクレインのC末端領域を代表する、及びそれを覆う短い(通常、9マー又は10マー)ペプチドを使用することによって実施した。これらは、比較表面プラズモン共鳴アッセイにおいて使用して、任意のものが、抗体のBiacoreチップ上に固定化されたモノマーアルファシヌクレイン又は事前形成アルファシヌクレイン繊維のいずれかとの結合を阻害可能か否かを試験した。次いで、最大レベルの阻害を示すペプチドを、正確なエピトープを確認するための抗体との共結晶化研究のために選択した。
ヒトアルファシヌクレイン(Hisタグが付けられた)の単一アミノ酸突然変異体を調製し、製造業者の説明書に従ってExpi293系(Thermo Fisher Scientific)において発現させた。アラニン残基を、すでにアラニンであり、セリンに変更した残基124を除く、位置118~128に配置した(表4)。遠心分離(4200rpm、2時間)とそれに続く滅菌濾過(Stericup、Millipore)後に得られた上清から突然変異体タンパク質を精製した。上清を、予備平衡化したHisTrap Excelカラム(GE Healthcare)にロードし、25mMリン酸ナトリウム及び500mM NaClを用いて洗浄した。同バッファー中、最大500mMのイミダゾールの勾配を用いて結合しているタンパク質を溶出した。対象のタンパク質を有する画分をNuPageゲル電気泳動によって同定し、プールし、Centriprep 10kDa MWCO(Millipore)を使用して濃縮し、PD-10カラムでPBSにバッファー交換した。タンパク質画分を、Ultracel 3KDa MWCO 遠心スピンコンセントレーター(Millipore)を使用して濃縮した。濃縮物を0.22mM滅菌フィルター(Millex GV、Millipore)に通し、-20℃で保存した。すべての突然変異体は、野生型ヒトアルファシヌクレインと同様の量で発現された(図6A)。
抗体ヒト化
ラット抗体5811を、ラットV領域からヒト生殖系列抗体V領域フレームワークにCDRをグラフトすることによってヒト化した。抗体の活性を回復させるために、ヒト化配列中でラットV領域に由来するいくつかのフレームワーク残基も保持した。これらの残基は、Adairら(WO91/09967)によって概説されるプロトコールを使用して選択した。ラット抗体(ドナー)V領域配列の、ヒト生殖系列(アクセプター)V領域配列とのアラインメントは、図7及び8に、設計されたヒト化配列と一緒に示されている。ドナーからアクセプター配列にグラフトされたCDRは、CDR-H1を除いてKabat(Kabatら、1987年)によって定義されるとおりであるが、CDR-H1では、組み合わされたChothia/Kabat定義が使用されている(Adairら、1991年「ヒト化抗体」(Humanized Antibodies.)WO91/09967を参照のこと)。
免疫組織化学
免疫組織化学は、Asterand Bioscience(英国、ロイストン)によって実施された。凍結切片(10μm)を、自動化加熱及び冷却を用いて97℃で20分間Dako PT Link及びEnVision FLEX標的検索溶液(pH6)を使用する抗原検索手順に最初に付した。以下のインキュベーションステップすべてを室温で実施した。凍結切片を30分間風乾し、1×PBSで調製した4%パラホルムアルデヒド中で10分間固定化し、Dako EnVision(商標)FLEX洗浄バッファー(Dako)中で洗浄し、次いで、Dako Autostainer Plusにロードした。切片をDakoペルオキシダーゼブロック(Dako)とともに5分間インキュベートすることによって内因性ペルオキシダーゼ活性を遮断した。次いで、切片を1×PBSを用いて2回洗浄し、次いで、Dako CSA IIタンパク質ブロック(Dako)とともに10分間インキュベートした。タンパク質ブロック溶液をエアージェットによって除去し、切片を30分間、Dako抗体希釈剤(Dako)で希釈した(0.05μg/ml)5811ラット-マウスIgG1抗体(配列番号34及び35を含む)とともにインキュベートした。インキュベーション後、切片を1×PBSを用いて2回洗浄し、次いで、抗マウスDako Flexポリマー-HRP基質(Dako)とともに20分間インキュベートし、2回洗浄し、次いで、ジアミノベンジジン基質(Dako)とともに10分間インキュベートした。スライドを蒸留水ですすぐことによって発色反応を停止させた。発色後、切片をDako Autostainer Plusから回収し、ヘマトキシリンを用いて手作業で対比染色し、上昇系列のエタノールで脱水し、キシレンの3回の変更で清澄化し、DPX封入剤(Sigma-Aldrich)下にカバーガラスを付けた。Aperio ScanScope AT Turboシステム(Leica Biosystems)を使用して染色した切片のデジタル画像を得た。抗体5811mIgG1を、5個体の異なるpS129-アルファ-シヌクレイン陽性及び3個体の異なるpS129-アルファ-シヌクレイン陰性ドナーに由来する脳切片(1切片/ドナー)で試験した。抗体5811mIgG1は、PD患者の側頭皮質及び黒質においてニューロピル及び時折レビー小体様特徴を標識した(図9A~E)。非PD脳組織では、抗体5811mIgG1は、側頭皮質においてニューロピルを標識したが、皮質又は黒質においてレビー小体様構造は観察されなかった(図9F~H)。これらの観察結果は、抗体5811mIgG1が、PD及び非PD患者から得た脳組織のニューロピル中の正常アルファシヌクレインと結合するが、一方で、PD患者のみにおいてレビー小体中に存在する病理学的α-シヌクレインと結合することを示唆する。
ヒト化抗体の特性決定
3種のAb5811ヒト化IgG4P抗体(5811gL5gH4、5811gL8gH4 5811gL14gH4、表1中の配列)を試験して、熱安定性(Tm)、実験pI、疎水性、溶解度(PEG沈殿アッセイ)、空気/液体界面での凝集安定性及びCDR-L3上のAsn93の脱アミド化傾向に関する化学安定性を含むその生化学的及び生物物理学的特徴を評価した(5811gL14gH4は、N(93)Aモチーフを有し、5811gL8gH4及びgL5gH4の両方ともN(93)Gモチーフを有する)。
Thermofluorアッセイを使用して、融解温度(Tm)又はアンフォールディングの中点の温度を決定した。この方法では、蛍光色素SYPRO(登録商標)Orangeを使用して、温度が高まるにつれ、露出するようになる疎水性領域と結合することによって、タンパク質アンフォールディングプロセスをモニタリングした。
5811gL14gH4、5811gL8gH4及び5811gL5gH4の実験pIは、全キャピラリー画像化cIEF iCE3(商標)システム(ProteinSimple)を使用して得た。サンプルは、以下を混合することによって調製した:30μLのサンプル(HPLC等級水中、1mg/mLストックから)、35μLの1%メチルセルロース溶液(Protein Simple)、4μLのpH3~10両性電解質(Pharmalyte)、0.5μLの4.65及び0.5μLの9.77合成pIマーカー(Protein Simple)、12.5μLの8M尿素溶液(Sigma-Aldrich(登録商標))。HPLC等級水を使用して最終容量を100μLに構成した。混合物を短時間ボルテックス処理して、完全混合を確実にし、解析の前に10,000rpmで3分間遠心分離して気泡を除去した。サンプルを1.5kVで1分間、続いて、3kVで5分間集束させ、ProteinSimpleソフトウェアを使用してキャピラリーのA280像をとった。得られた電気泳動図をiCE3ソフトウェアを使用して最初に解析し、pI値を割り当てた(pIマーカー間の線形関係)。次いで、較正された電気泳動図をEmpower(登録商標)ソフトウェア(Waters)を使用して統合した。
ヒト化抗体5811gL14gH4、5811gL8gH4及び5811gL5gH4を、疎水性相互作用クロマトグラフィー(HIC)によってその疎水性挙動について評価した。HICでは、抗体は、高濃度の極性塩の存在下で疎水性固定相と結合し、塩の濃度が低下するにつれ移動相中に放出される。保持時間が長いほど、大きい疎水性と一致する。

ポリエチレングリコール(PEG)沈殿アッセイを使用して、ヒト化抗体5811gL14gH4、5811gL8gH4及び5811gL5gH4のコロイド安定性を解析した。PEGを使用して、PEGの濃度(w/v)を増大すること及び溶液中に残存するタンパク質の量を測定することによって量的に定義可能な方法でタンパク質溶解度を低減した。このアッセイは、従来の濃度法を使用せずに高濃度溶解度の効果を模倣するように働いた。
このアッセイは、製造(例えば、限外濾過)の際に抗体が付されるであろうストレスを模倣するように働く。抗体などのタンパク質は、空気-液体界面に曝露されるとアンフォールディングする傾向があり、疎水性表面が疎水性環境(空気)に、親水性表面が親水性環境(水)に提示される。タンパク質溶液を撹拌すると、凝集を駆動し得る大きな空気-液体界面が得られる。
3種の抗体:5811gL5gH4、5811gL8gH4及び5811gL14gH4のすべてを使用して加速化ストレス研究を実施して、1つの同定された潜在的部位:CDR3の軽鎖中のAsn(93)の脱アミド化傾向を調べ、5811gL14gH4は、Asn(93)Alaモチーフを有し、5811gL8gH4及びgL5gH4は両方ともAsn(93)Glyモチーフを有する。
細胞ベースの凝集アッセイ
HEK Freestyle 293F細胞(懸濁液細胞)を、Freestyle293発現培地(Invitrogen(商標))中、0.7×106個細胞/mlで調製し、300×106個細胞/mlに培養した。トランスフェクションを製造業者の説明書に従って実施し、手短には、600μgの、アルファ-シヌクレイン遺伝子を組み込むpcDNA3.1(+)を、20mlのOptiMEM培地中に混合し、一方で、293FectinをOptiMEM培地(Invitrogen(商標))で希釈し、室温で5分間インキュベートした。希釈したDNAを添加し、室温で20分間インキュベートし、その後、細胞に1滴ずつ滴下した(フラスコ当たり20ml)。細胞を37℃、125rpm、8%CO2で24時間インキュベートした。細胞を直ちに使用するか、又はFBS+10% DMSO中500万個細胞/mlの濃度で凍結した。
一次ニューロン凝集アッセイ
E17マウス胚から得た海馬を、解剖バッファー(カルシウムを含まない、マグネシウムを含まないHBSS、0.6% D-(+)-グルコース、20mM Hepes)中で解剖した。次いで、解剖バッファーを除去し、解離溶液(カルシウムを含まない、マグネシウムを含まないHBSS、0.6 %D-(+)-グルコース、20mM HEPES、40U/mパパイン、1mg/ml DNアーゼ、1mM L-システイン、0.5mM EDTA)によって置き換えた。37℃で30分インキュベートした後、解離バッファーを除去し、海馬を、プレーティング培地(Neurobasal(商標)培地、2% B27上清、1mM GlutaMAX、2.5% FBS、50ユニット/mlペニシリン-ストレプトマイシン)で3回洗浄した。組織凝集塊を1mlピペットを用いてトリチュレートして、単細胞懸濁液を得た。細胞をプレーティング培地で適当な濃度に希釈した。PDLコーティング384ウェルプレートの各ウェルにおいて約15000個細胞をプレーティングした。次いで、37℃、5% CO2、95%湿度で細胞培養インキュベーター中で細胞を維持した。
in vivoでのVR5811有効性の評価
抗体5811gL5gH4 IgG4P(配列番号14及び配列番号26を含み、本明細書において以下、簡単にVR5811と呼ばれる)を、ヒトアルファシヌクレインを発現するα-シヌクレインノックアウトマウスのトランスジェニックモデルにおいて(以下SNCA-OVXと名付ける;Charles River、フランス)試験した。
マウスにおける抗体5811の薬物動態
雄C57/Bl6マウス(薬物あたりn=3)に、抗体5811gL14gH4 IgG4P(5811)を用いて2mg/kgの単回用量として静脈内注射した。
Claims (24)
- アルファシヌクレインと結合する抗体又はその抗原結合断片であって、
該抗体又はその断片が、
a.配列番号1のアミノ酸配列からなるCDR-L1、配列番号2のアミノ酸配列からなるCDR-L2及び配列番号3のアミノ酸配列からなるCDR-L3を含む軽鎖可変領域、並びに
b.配列番号4のアミノ酸配列からなるCDR-H1、配列番号5のアミノ酸配列からなるCDR-H2及び配列番号6のアミノ酸配列からなるCDR-H3を含む重鎖可変領域
を含む、抗体又はその抗原結合断片。 - 配列番号3における位置6のアミノ酸残基グリシン(Gly;G)が、アラニン(Ala;A)によって置き換えられる、請求項1に記載の抗体又はその抗原結合断片。
- 配列番号8における位置113から129の間のアルファシヌクレインの2個以上のアミノ酸残基と結合し、配列番号8における少なくともアミノ酸残基D119、N122及びY125と結合する、請求項1又は請求項2に記載の抗体又はその抗原結合断片。
- アルファシヌクレイン繊維によって誘導されるアルファシヌクレインの凝集を防止する、請求項1から3までのいずれか一項に記載の抗体又はその抗原結合断片。
- モノマーとしてのアルファシヌクレイン、及び繊維状のアルファシヌクレインと結合することが可能である、請求項1から4までのいずれか一項に記載の抗体又はその抗原結合断片。
- モノマーとしてのアルファシヌクレインと比較して、繊維状のアルファシヌクレインに対してより高い結合親和性を有する、請求項1から5までのいずれか一項に記載の抗体であって、
繊維状のアルファシヌクレインに対してよりも、モノマーアルファシヌクレインに対して少なくとも10倍高い解離定数(KD)を特徴とする、抗体。 - 60pM以下の繊維状のアルファシヌクレインに対する解離定数(KD)を有する、請求項1から5までのいずれか一項に記載の抗体。
- 前記抗体が、キメラ又はヒト化抗体である、請求項1から7までのいずれか一項に記載の抗体又はその抗原結合断片。
- IgG1、IgG4又はIgG4Pから選択される、請求項1から8までのいずれか一項に記載の抗体。
- 前記抗原結合断片が、Fab、Fab’、F(ab’)2及びscFvから選択される、請求項1から5および8までのいずれか一項に記載の抗原結合断片。
- 前記抗体又はその断片が、
a.配列番号13のアミノ酸配列からなる軽鎖可変領域及び配列番号25のアミノ酸配列からなる重鎖可変領域、又は
b.配列番号17のアミノ酸配列からなる軽鎖可変領域及び配列番号25のアミノ酸配列からなる重鎖可変領域、又は
c.配列番号21のアミノ酸配列からなる軽鎖可変領域及び配列番号25のアミノ酸配列からなる重鎖可変領域
を含む、請求項1から10までのいずれか一項に記載の抗体又はその抗原結合断片。 - 前記抗体が、
a.配列番号14のアミノ酸配列からなる軽鎖及び配列番号26のアミノ酸配列からなる重鎖、又は
b.配列番号18のアミノ酸配列からなる軽鎖及び配列番号26のアミノ酸配列からなる重鎖、又は
c.配列番号22のアミノ酸配列からなる軽鎖及び配列番号26のアミノ酸配列からなる重鎖
を含む、請求項1から9までのいずれか一項に記載の抗体。 - 請求項1から12までのいずれか一項に記載の抗体又はその抗原結合断片をコードする単離ポリヌクレオチド。
- a.軽鎖可変領域をコードするポリヌクレオチドが、
i.配列番号15、19若しくは23に対して少なくとも90%同一である、若しくは
ii.配列番号15若しくは19若しくは23を含む、若しくは
iii.配列番号15、19若しくは23からなり、且つ
b.重鎖可変領域をコードするポリヌクレオチドが、
i.配列番号27に対して少なくとも90%同一である、
ii.配列番号27を含む、若しくは
iii.配列番号27からなり、あるいは
c.軽鎖をコードするポリヌクレオチドが、
i.配列番号16、20若しくは24に対して少なくとも90%同一である、若しくは
ii.配列番号16、20若しくは24を含む、若しくは
iii.配列番号16、20若しくは24からなり、且つ
d.重鎖をコードするポリヌクレオチドが、
i.配列番号28に対して少なくとも90%同一である、若しくは
ii.配列番号28を含む、若しくは
iii.配列番号28からなる、
請求項13に記載の単離ポリヌクレオチド。 - 請求項13又は14に記載の1つ又は複数のポリヌクレオチドを含むクローニング又は発現ベクター。
- a.請求項13若しくは14に記載の1つ若しくは複数のポリヌクレオチド、又は
b.請求項15に記載の1つ若しくは複数の発現ベクター
を含む、宿主細胞。 - 請求項1から12までのいずれか一項に記載の抗体又はその抗原結合断片の生産方法であって、請求項16に記載の宿主細胞を、抗体又はその抗原結合断片を生産するのに適した条件下で培養すること及び抗体又はその抗原結合断片を単離することを含む、方法。
- 請求項1から12までのいずれか一項に記載の抗体又はその抗原結合断片と、1つ又は複数の薬学的に許容される担体、賦形剤又は希釈剤とを含み、任意選択で、1つ又は複数のさらなる有効成分を含む、医薬組成物。
- 治療において使用するための、請求項18に記載の医薬組成物。
- 1つ又は複数のシヌクレイノパチーの処置において使用するための、請求項18または19に記載の医薬組成物。
- 前記シヌクレイノパチーが、パーキンソン病(PD)(パーキンソン病の特発性及び遺伝性形態を含む)、レビー小体型認知症(DLB)、びまん性レビー小体病(DLBD)、アルツハイマー病のレビー小体異型(LBVAD)、アルツハイマー病及びパーキンソン病の合併、多系統萎縮症(MSA)及び脳内鉄沈着を伴う神経変性1型(NBIA-1)から選択される、請求項20に記載の医薬組成物。
- 前記シヌクレイノパチーがパーキンソン病である、請求項21に記載の医薬組成物。
