エンデュラシジン(図1)は、エンラマイシンとも呼ばれている、土壌細菌S.fungicidicus B-5477(ATCC21013)によって産生される17アミノ酸リポデプシペプチド抗生物質である。このペプチドは、発酵ブロス及び菌糸体から、主に、結合した脂質鎖の長さが1炭素異なるエンデュラシジンA及びBの混合物として単離される。構造的には、エンデュラシジンは、アスパラギン酸残基へのアミド結合によって結合されたC12又はC13 2Z、4E分岐脂肪酸部分、及びエンデュラシジン(End)、4-ヒドロキシフェニルグリシン(Hpg)、3,5-ジクロロ-4-ヒドロキシフェニルグリシン(Dpg)、シトルリン(Cit)及びオルニチン(Orn)などの多数の非タンパク質性アミノ酸残基の存在によって区別される(図1参照)。17個のアミノ酸のうち7個はD立体配置を有し、残基の6個はHpg又は塩素化誘導体Dpgである。

実施形態では、Streptomyces fungicidicusの組換え株は、少なくとも1つのエンデュラシジン産生増強オープンリーディングフレームを含む組換えプラスミドの、Streptomyces fungicidicusの親株の染色体への組込みによって構築してもよい。統合的コンジュゲートベクターは、強力な構成的ストレプトミセス菌プロモーターを有するか、有するように遺伝子操作され得る。いくつかの実施形態では、プラスミドは連鎖球菌レプリコンを欠いていてもよく、部位特異的シングルクロスオーバー相同組換えによって染色体に組み込まれてもよい。他の実施形態では、プラスミドは遊離プラスミドとして存在してもよい。いくつかの実施形態では、プラスミド挿入物が、部分的又は完全に欠失した目的の遺伝子、及び二重交差相同組換え後に染色体に組み込まれてフレーム内欠失変異体を生成し得るそのフランキング領域を有する接合ベクターを遺伝子操作し得る。
本開示によって提供されるStreptomyces fungicidicusの遺伝子工学組換え株は、増強されたレベルのエンデュラシジンを産生する方法を提供する。当技術分野におけるこの技術的進歩により、エンデュラシジンの製造に関連する大幅なコスト削減が可能になる。いくつかの例では、エンデュラシジンを産生する方法は、開示されたStreptomyces fungicidicusの組換え株をエンデュラシジンを産生するのに十分な条件下で培養することを含む。いくつかの例では、方法は、培養後に培地からエンデュラシジンを単離することをさらに含む。いくつかの例では、方法は、微生物を示すものとして黄色ブドウ球菌ATCC29213又は枯草菌(Bacillis subtilis)ATCC6633を使用するHPLC分析又はバイオアッセイなどによって、生成されたエンデュラシジンの抗菌活性を判定することをさらに含む。
特徴付けられた確立された実用的な種培養物が発酵バッチを開始するために使用される。凍結種子バイアルの1~5バイアルを低温貯蔵庫から取り出し、内容物が解凍されるまで自然に解凍するか28℃~32℃の水浴中に入れる。解凍した(1又は複数の)培養物を室温に保った滅菌水に無菌的に移し、穏やかに混合して培養物を再懸濁する。
再懸濁培養物を無菌的に0.005m3~0.05m3の種の培地に移す。種の培地は、グルコース(0.1~1.0g/L)、デキストリン(0.1~3g/L)、コーンスティープリカー(0~5.0mL/L)、大豆粉末(1~5.0g/L)、硫酸アンモニウム(0.1~0~0.5g/L)、リン酸一カリウム(0.13~0.54g/L)、硫酸第一鉄(0.00~0.5g/L)、水酸化カリウム(0.13mL/L)、炭酸カルシウム(1~2g/L)、シリコーン系消泡剤(0.1mL/L)、水、適量で構成される。培地を125℃~128℃で30~45分間滅菌し、次いで28℃~32℃に冷却する。培地の量は滅菌水を用いて所望の作業量に調整される。pHを6.5~7.0に調整する。
産生発酵槽培地(10m3~250m3)組成物は、トウモロコシの粉(13.0~15.0w/v%)、コーングルテンミール(3.0~6.0w/v%)、綿実粉(0.1~0.3w/v%)、コーンスティープリカー(0.1~0.6v/v%)、塩化ナトリウム(0.3w/v%)、硫酸アンモニウム(0.25~0.6w/v%)、乳酸(0~0.5v/v%)、塩化亜鉛(0.01w/v%)、硫酸第一鉄(0.0~0.02w/v%)、水酸化カリウム(0.20~0.5v/v%)、硫酸カルシウム(0.0~0.5w/v%)、カルシウム炭酸塩(0.5w/v%)、アミラーゼ(0.02~0.06w/v%)、水酸化カリウム(0.05v/v%)、植物油(0.5~2.0v/v%)、消泡剤、及び水、適量などの天然成分及び化学成分を含む。成分a~eは記載された順序に従って添加した。成分に水を加え、次に70~90℃に加熱して酵素に複合炭水化物を15分間その温度で分解させる。残りの成分を加え、pHを6.6~6.8に調整し、水を適量加える。125℃~128℃で25~50分間殺菌して培地を殺菌する。培地を25℃~32℃に冷却し、水を適量、作業量に加える。

[実施例1]
増強されたエンデュラシジン産生のための材料及び方法
この実施例は、増強されたエンデュラシジン産生のための代表的な方法を提供する。
Streptomyces fungicidicus B-5477(ATCC21013)及び大腸菌(Escherichia coli)S17-1(ATCC47055)はATCCから購入した。S.fungicidicus株BM38-2(ATCC PTA-122342)ならびにエンデュラシジンA及びBの標準は、Intervet/Merck Animal Health(MAH)により提供された。大腸菌株DH5α(Life Technologies,Inc.)、EPI300(Epicentre)及びXL10-Gold(Stratagene)を、大腸菌プラスミド、フォスミド及び大腸菌-ストレプトミセス菌シャトルベクターの宿主として使用した。プラスミドpSET152(Biermanら.,Gene 116:43-49,1992、これはその全体が参照により本明細書に組み入れられる)及びpIJ773は、Keith Chater教授(JIC、Norwich、UK)によって提供された。ermE*pプロモーターを有するプラスミドpWHM860は、Bradley Moore教授(UCSD、San Diego)によって提供された。ISP2(Difco(商標)ISP培地2)、ISP4及びTSB(Bacto(商標)トリプチックソイブロス)は、VWRから購入した。PCR及びDNA配列決定に使用したプライマーは、Fisher及びSigma-Aldrichから合成した。S.fungicidicusを増殖させるための培地及び培養条件は、Higashideら(Journal of Antibiotics,21:126-137,1968)に記載されている。すべての大腸菌の処置は標準的なプロトコルに従って実施した。
配列決定、フォスミドライブラリーの構築、サブクローニング及びPCRのためのS.fungicidicus B-5477、BM38-2(ATCC PTA-122342)ならびに誘導体組換え及び変異株からゲノムDNAを調製するために、個々の株から新たに収穫した胞子を接種し、5mMのMgCl2及び0.5%グリシンを補充した100mLのTSB液体培地で増殖させた。代表的な培養は、500mLの三角フラスコ中、回転振盪インキュベーターで、225rpm及び30℃で48から72時間実施した。菌糸細胞を4000rpm、4℃で15分間の遠心分離により回収した。上清を捨て、ペレットを10.3%スクロースで1回、10mMトリス-HCl及び1mMエチレンジアミン四酢酸二ナトリウム(EDTA)、pH8.0(TE緩衝液)で2回連続して洗浄した。水80μLの容量に相当する湿った細胞を1.5mLの無菌の微量遠心管に分布させた。200μLの10mMのTris-HCl及び1mMのEDTA、pH8.0及び0.3Mのスクロース(TES緩衝液)、50μLの0.5MのEDTA、50μLのリゾチーム(50mg/mL)を含む300μLの溶解溶液を添加した後、溶液が粘性になるまで、チューブを37℃で30~60分間インキュベートした。次に、5μLのプロテイナーゼK(20mg/mL)及び180μLの10%ドデシル硫酸ナトリウム(SDS)を各チューブに加えた。穏やかだが十分に混合した後、溶液を37℃で90分間インキュベートした。次いで、80μLの10%臭化セチルトリメチルアンモニウム(CTAB)を加えた。十分に混合した後、チューブを65℃で10分間インキュベートした。溶液をフェノール/クロロホルム/イソアミルアルコール(25/24/1)600μLで2回抽出した。上部水相中のゲノムDNAを回収し、0.6容量のイソプロパノールで沈殿させた。採取したゲノムDNAを70%エタノールで2回洗浄した。室温で10分間乾燥した後、ゲノムDNAを50~100μLの滅菌水に溶解した。良質のゲノムDNA調製物は、0.8%アガロースゲル電気泳動による完全な消化及び未消化のゲノムDNAの分解を示さないHindIII及びSau3AIによる消化によって確認された。プールしたゲノムDNAをRNaseでさらに消化してRNAの混入を除去した。ゲノムDNAの純度及び量は、Nanodrop分光光度計を用いて判定した。アガロースゲル電気泳動を含む一般的な連鎖球菌DNA操作を行い、QIAprep Spin Miniprepキット(Qiagen)を用いて大腸菌株からプラスミド及びフォスミドを調製した。制限エンドヌクレアーゼ、DNAリガーゼ、DNAポリメラーゼ、トランスポザーゼ、クレノウ酵素、アルカリホスファターゼ及びリガーゼは、Biolabs、Invitrogen、Epicentre及びRocheから購入し、製造元の推奨に従って使用した。QIAquick Gel Extractionキットを用いてDNA断片を精製した。
コロニーPCRは以下のように実施した:独立した変異体候補コロニーからの胞子をTSB液体培養物に接種した。48~72時間増殖させた後、菌糸体を遠心分離により収穫し、TE緩衝液(10mMのTris、1mMのEDTA)、pH8.0で2回洗浄した。菌糸体を滅菌H2Oに再懸濁し、60μLの菌糸体、150pmolの各プライマー、20μLの5×AccuPrime GCリッチ緩衝液A(Invitrogen)、及びExpand long template PCRシステム(Roche)から得た1μLのPolymix(80℃で添加)を含有する100μLの最終容量でPCR反応混合物中の鋳型として使用した。PCRは以下のようにして行った:1サイクルを95℃で3分間、30サイクルを95℃で1分間、55℃で1分間、及び72℃で2分間とした。1回の延長サイクルで72℃で10分間反応を終了させた。PCR産生物をゲル精製し、配列決定した。事前の精製なしに菌糸体コロニーから放出されたDNAを直接使用する代わりに、単離されたゲノムDNA、プラスミド/フォスミドDNAを鋳型として使用したことを除いて、上記と同様に一般的なPCRを行った。
S.fungicidicus野生型及びBM38-2(ATCC PTA-122342)株においてエンデュラシジン遺伝子クラスターから推定上の調節遺伝子orf24を異所的に発現させるために、orf24をpXY152aR20由来の組込み型プラスミドpXY152にクローニングした(Yinら,J.Natural Products,73:583-589,2010、その全体が参照により本明細書に組み入れられる)。orf24は、フォワードプライマー(End24Ndpf:
pXY300-orf18ifdは、orf18に隣接し、欠失の予定である2つの断片を、S.fungicidicusにおける選択のためのチオストレプトン耐性遺伝子(tsr)を含む大腸菌-ストレプトミセス菌シャトル接合体温度感受性ベクターであるpXY300にクローニングすることによって構築した。orf18に隣接する、それぞれorf18ifdNP及びorf18ifdPHと呼ばれる、「上流」2kb及び「下流」2kbのフランキング配列を、鋳型としてS.fungicidicusゲノムDNA及び2組のプライマーを使用するPCRによって生成した。断片orf18ifdPHは、フォワード及びリバースプライマー(Ifdenorf18pf1、
pXY300-orf18ifdの挿入断片をNheI及びHindIIIで消化して切り出し、ゲル精製した後、SpeI及びHindIII線状化ベクターpBluescript II KSと連結してプラスミドpKS-orf18ifdを得た。oriT断片をフォワードプライマー(Oritnhexbahd3f、
遺伝子破壊プラスミドを形質転換により大腸菌S17-1に個別に導入し、次いで接合を介してS.fungicidicus又はその誘導体に移した。手短に言えば、新たに採取したS.fungicidicus胞子を前発芽させ、大腸菌S17-1細胞をTerrificブロス中で37℃で一晩増殖させた。発芽胞子懸濁液の連続希釈液を作製し、各希釈液100mLを、pXY300ベースの破壊プラスミドを有する等容量の大腸菌S17-1と混合した。溶液を10mMのMgCl2を添加してISP4寒天プレート上にプレーティングし、30又は37℃のいずれかで22時間インキュベートした。各プレートに、ナリジクス酸ナトリウム及びアプラマイシン(0.5mg/mL)を含有する3mLの軟質栄養寒天を重層し、さらに30℃で約1週間インキュベートした。抗生物質選択を生き残った単離された接合体を、ナリジクス酸ナトリウム及びアプラマイシン(各50μg/mL)を補充したISP4寒天プレート上に画線することによって精製した。
orf18及びそのフランキング領域の欠失を有するアプラマイシン耐性変異株においてorf24を異所的に発現させるために、組込み発現プラスミドpXY152-endorf24-blatsrを設計した。このプラスミドを構築するために、クロラムフェニコール耐性遺伝子及びチオストレプトン耐性遺伝子(tsr)を有するカセット(camtsr)をSacI及びNheIでの消化によりプラスミドpUC57誘導体から切り出した。次にcamtsrカセットをSacI及びNheI線状化プラスミドpXY152-endorf24と連結して新しい構築物pXY152-endorf24-camtsrを得た(図7;配列番号20)。アンピシリン耐性遺伝子(bla)を、フォワードプライマー(amp2956SwaIpf、
Tn5ATカセットは、トランスポゾンTn5、oriT、及びaac3(IV)の3つの遺伝的要素を組み合わせるように設計されている。Tn5は、Tn5トランスポザーゼ(Epicentre)によって特異的かつ独特に認識され、大腸菌プラスミド及びフォスミド中にクローニングされた高G+Cストレプトミセス菌DNA中に容易に挿入される(米国特許第8,188,245号にて言及、これもまた本明細書中に参考として援用される)。oriTは大腸菌S17-1からストレプトミセス菌へのDNAの接合伝達に必要であり、aac(3)IVはアプラマイシン耐性を付与する大腸菌-ストレプトミセス菌二機能選択マーカーである。oriT及びaac3(IV)の両方を、XbaI断片としてプラスミドpIJ773から切り出し、次に予めXbaIで線状化したトランスポゾンドナープラスミドpMOD(商標)-2(MCS)(Epicentre)にクローニングした。得られたプラスミドpXYTn5ATa及びpXYTn5ATbは、XbaI断片の配向のみが異なり、製造者の仕様書に従ってPvuIIで消化することによってTn5ATカセットを調製するために使用した。
遺伝子置換研究のためのエンデュラシジン生合成クラスターのセグメントを保有するランダム変異誘発フォスミドのライブラリーを作製するために、フォスミドpXYF24及びpXYF148のインビトロトランスポゾン挿入変異研究を行った。2つの推定エンデュラシジン生合成調節遺伝子、すなわちorf18及びorf24は、それぞれフォスミドpXYF24及びpXYF148のインサート上に存在する(GenBank登録番号DQ403252)。10μL(0.5μg)のフォスミドテンプレートDNA、2μL(20ng)のTn5ATカセットDNA、2μLの10×反応緩衝液、1μLのTn5トランスポザーゼ、及び5μLの滅菌水を混合した後、37℃で2時間インビトロトランスポゾン反応を行った。トランスポゾン反応混合物を用いた大腸菌コンピテント細胞EPI300(商標)-T1R(Epicentre)の形質転換をエレクトロポレーションによって行った。100μg/mLのアプラマイシンを補充したLB寒天プレート上で変異誘発フォスミドを選択した。プレートを37℃で一晩インキュベートし、生き残ったコロニーを無作為に選び、100μg/mLのアプラマイシンを添加したLB液体培地中で増殖させた。これらのコロニーからの突然変異誘発フォスミドDNA及び対照のフォスミドpXYF24又はpXYF148をHindIIIで消化し、1%アガロースゲル上での電気泳動により分析した。Tn5ATカセットは単一のHindIII部位を含み、これはフォスミドインサート上の単一に対する複数の破壊事象についてスクリーニングするときに有用である。pXYF24又はpXYF148のフォスミドインサート中にはHindIII部位は存在せず、フォスミドベクター中にはただ1つのHindIII部位が存在する。それゆえ、HindIIIによる消化は、ゲル中に2つのバンドが存在することによって、Tn5ATの単一挿入のフォスミドを容易に同定する。単一のトランスポゾン挿入の変異誘発フォスミドを有するコロニーを無作為に選択し、LB液体培養中で増殖させて、フォスミド単離及び破壊された遺伝子の同定を可能にした。スクリーニングは、アプラマイシン耐性遺伝子の領域に対応するプライマー5’-AAGGAGAAGAGCCTTCAGAAGGAA-3’(配列番号24)を用いた配列分析によって行った。このようにして、フォスミドpXYF24D3及びpXYF148D12は、ヌクレオチド位置26386のorf18及びヌクレオチド位置34333(GenBank登録番号DQ403252)のorf24にそれぞれTn5ATが挿入されていることが見出された。
S.fungicidicus野生型、BM38-2(ATCC PTA-122342)及びその派生株におけるエンデュラシジンの産生のための実験室用振盪フラスコ発酵条件は、Higashideら.(J.Antibiotics,21:126-137,1968)によって記載された通りであった。ただし、特許(米国特許第4465771号)に開示されたエンデュラシジン産生培地は除かれる。実験室規模の発酵のために、5mLのTSBを、新たに収穫したストレプトミセス菌胞子を種の培養に接種するために使用した。典型的には5~10mLの種の培養物を回転式振盪機上で225rpm及び30℃で48時間インキュベートし、次いで10日間連続発酵のために50mLのエンデュラシジン産生培地に移した。厳密に制御された条件下での野生型及び誘導体株によるエンデュラシジンの産生もまた10リットル自動発酵槽中で行われた。
エンデュラシジン産生のHPLC分析のため代謝産物を抽出するために、新鮮な菌糸体を遠心分離によって収穫し、脱イオン水で洗浄し、5×容量の(水性菌体重量(g)に対する水性メタノール(mL)の比)70%水性メタノール(1NのHClでpHを3.5に調整した)に再懸濁した。懸濁液を室温で200rpmで一晩振盪し、次いで4000rpm及び4℃で20分間遠心分離した。次に、各サンプルから得た1.4mLの上清を個々の1.5mL微量遠心用チューブに移し、室温で13,000rpmで10分間遠心分離した。濾液を0.22μmのシリンジフィルターに通した後、HPLCで分析した。10Lの発酵槽で産生された菌糸体からの代謝産物の抽出が、実験室での発酵と同等の小規模で行われた。
上記のように調製した50μLのHPLCサンプルを、Shimadzu HPLCに接続したGemini C18カラム(5μm、4.6×150mm、Phenomenex、Torrance、カリフォルニア州)に注入した。溶媒A:水+0.1%TFA及び溶媒B:アセトニトリルを用いた18分の段階的線形勾配を用いて、分離を達成した。流速は、10%Bから始めて1mL/分であり、10分かけて40%Bまで上げ、次いで8分かけて95%Bまでさらに上げた。200から300nmのUV領域をSPD M20Aフォトダイオードアレイ検出器でスキャンした。70%メタノール中のエンデュラシジン標準のストック溶液から作成した標準曲線と比較することにより、エンデュラシジンの収量を計算した。2、4、6、8、10及び12μgのエンデュラシジンを含む一連の注射を用いて、230nmにおけるエンデュラシジンA及びBの吸光度面積の合計を用いて標準曲線を作成した。標準曲線から回帰式を生成し、これを用いてエンデュラシジンの収量を計算した。
黄色ブドウ球菌(ATCC29213)をバイオアッセイにおける指標の微生物として利用した。細胞を用いてLBブロスに接種し、37℃で一晩増殖させた後、100μLの培養物を5mLの上層寒天(等量の栄養寒天と栄養ブロスの混合物)と混合した。上層寒天を栄養寒天プレート上に重層し、寒天プラグを切り取ることによって、適切な間隔でウェルを作った。エンデュラシジン標準及び一定分量の培養抽出物を50%のMeOHに20μg/mLの濃度で溶解又は希釈し、100μLの各溶液をウェルに入れた。プレートを37℃で16時間インキュベートした後、阻害領域を観察及び比較し、プレートを写真撮影するか4℃で保存した。

pKS由来遺伝子不活化ベクターpKS-T-orf18pfrd-AmRシリーズの開発
本実施例では、pKS由来遺伝子不活化ベクターpKS-T-orf18pfrd-AmRシリーズの開発について説明する。
pSET152由来組込み遺伝子発現ベクターpXY152-endorf24-camtsr及びpXY152-endorf24-blatsrの開発
この実施例は、pSET152由来の組込み遺伝子発現ベクターpXY152-endorf24-camtsr(配列番号20)及びpXY152-endorf24-blatsr(配列番号23)の開発を記載する。
菌株改良のためのストレプトミセス菌調節及び生合成遺伝子の遺伝子操作
多数の天然産生物の微生物産生体のうち、既知の微生物抗生物質の約75%が放線菌によって産生されている。ストレプトミセス菌、グラム陽性糸状土壌細菌は、放線菌科のメンバーであり、構造的に多様な、薬理学的及び生物学的に活性な二次代謝産物の多様な配列を産生するそれらの比類のない能力で知られている。ポリケチドシンターゼ(PKS)によって産生されるポリケチド及び非リボソームペプチドシンテターゼ(NRPS)によって産生されるペプチド天然物が代表的である。
GenBankデータベースに対するOrf24タンパク質配列を用いたBLASTクエリは、数百のヒットを明らかにした(GenBank登録番号DQ403252)。多くは非常に高いアミノ酸類似性(60%から99%の同一性)を示し、アミノグリコシド系抗生物質ストレプトマイシンの生合成における転写制御因子として注釈されている。しかし、このグループの遺伝子のどれも実験的に検証された機能を有していなかった。BLASTの結果の分析は、機能的に特徴付けられたOrf24に対してより低い類似性(40%を超えるが60%未満のアミノ酸同一性)を共有するいくつかの関連タンパク質を同定した。これらには、Orf24とは低いが重大な類似性(311アミノ酸重複における54%アミノ酸同一性)を共有する、よく特徴付けられているタンパク質StrRが含まれる。StrRは、ストレプトマイセス・グリセウス(Streptomyces griseus)におけるストレプトマイシン生合成遺伝子の発現の経路特異的な陽性アクチベーターとして機能することが遺伝的及び生化学的に実証されている。StrRは、経路特異的アクチベーターのファミリーを表し、その一握りは遺伝子操作又は生化学的研究のいずれかによって特徴付けられている。図11は、6つの機能的に確認された放線菌StrR様タンパク質とのOrf24のアラインメントを示す。図11に下線を引いたように、典型的で高度に保存されたヘリックス・ターン・ヘリックス(HTH)DNA結合ドメインが7つすべてのタンパク質に存在する。Orf24はまた、非リボソーム的に生成された糖ペプチド抗生物質テイコプラニンの生合成を支配する経路特異的活性化因子であるTeil15*と重大な配列類似性(54%のアミノ酸同一性)を共有する。Tei15*は、テイコプラニンクラスター中の少なくとも17個の遺伝子の転写を正に調節する。野生型Actinoplanes teichomyceticusは、約100mg/Lのテイコプラニンを産生するが、親A.teichomyceticus由来で、異なるプロモーターの制御下で発現されるtei15*を保有する遺伝子組換え株は、テイコプラニンの収量を、ermE*pプロモーターの場合は1g/Lに、天然アプラマイシン耐性遺伝子プロモーターの場合には約4g/Lに増加させた。
ストレプトミセス菌種における抗生物質の産生は、大規模で費用対効果の高い工業産生に必要な収量を生み出すことから、多くの野生型抗生物質産生体の能力を制限する複雑な遺伝的ネットワークによって厳しく調節されている。1つの重要な調節機構は二成分シグナル伝達系である。二成分系は、センサーキナーゼ及び同族応答調節因子を含む。センサーキナーゼは、ストレス、栄養及び化学物質などのような特定の外部環境刺激/シグナルに応答し、次いで標的遺伝子の転写を誘発及び活性化する細胞質応答制御因子にシグナルを中継する。センサーキナーゼと対になっていない応答制御因子は、オーファン応答制御因子と呼ばれる。
変異体BM38-1.18pfrd-AmR株の欠失領域は、orf18の3つの遺伝子、orf18の下流に位置するor17のN末端部分をコードする領域、及びorf18の上流に位置するorf19全体を含む(図5及び図10)。orf17は、エンデュラシジンの生合成又は調節に関連する機能を明らかに持たないリボヌクレアーゼをコードすると予測される。また、orf18及びそのフランキング領域を置換するアプラマイシン耐性遺伝子はorf17と多岐に転写され、アプラマイシン耐性遺伝子プロモーターからいずれかのリードスルー事象を生じさせるべきではない。それゆえ、orf17の部分的欠失から生じる極性の影響はないはずである。
増強されたエンデュラシジン産生株のためのorf24及び/又はorf18のさらなる適用及び操作
上記で提供された例に加えて、エンデュラシジン産生を改善するためにorf24及びorf18の調節的役割を利用するための他の可能な方法がある。
pXY152-endorf24(図2に示す)は、広く使用されているストレプトミセス菌の強力な構成的発現プロモーターであるermE*pの制御下でのorf24の統合的異所性発現のために構築された。orf24の過剰発現はまた、他の構成的又は誘導的プロモーターによっても駆動され得る。tipAプロモーターはチオストレプトン誘導性の過剰発現ストレプトミセス菌プロモーターである。ストレプトミセス菌遺伝子の過剰発現に首尾よく使用されてきた、マルチコピーtipAプロモーター含有大腸菌-ストレプトミセス菌シャトルプラスミド、pXY200が開発された。本開示に関連する用途では、tipAプロモーターをpXY200から切り出し、pXY152にクローニングしてermE*pを置換し、orf24の発現を駆動することができる。同様に、プラスミドベースの発現のために、orf24はpXY152-endorf24からpXY200に容易に移すことができる。他のプロモーターの選択肢には、P(nitA)-NitRシステム及びストレプトミセス菌プロモーターSF14が含まれるが、これらに限定されない。最近、統合型プラスミドpKC1139及びアプラマイシン耐性遺伝子の天然プロモーターが、ペプチド抗生物質テイコプラニンの過剰産生のための調節遺伝子を発現するために首尾よく使用された。調節遺伝子sanGは、ニッコマイシン産生の経路特異的活性化因子をコードする。5つの異なるプロモーター(PhrdB、Ptcp830、PSF14、PermE*及びPneos)の制御下でsanGの余分なコピーを発現させると、ニッカマイシン収量がそれぞれ69%、51%、26%、22%、及び13%増加した(Duら,Applied Microbiology and Biotechnology97:6383-6396,2013参照)。
増加したエンデュラシジン産生を示すorf18欠失変異体及びorf24過剰発現株の両方を用いて、両方を含有する二重変異体を作製することができ、このペプチド抗生物質の収量に対する付加的な効果があるかどうか観察された。二重変異体は、過剰発現プラスミドpXY152-endorf24-blatsr(図8)を変異体BM38-2.18pfrd-AmRに導入することによって作製することができる。pXY152-endorf24-blatsrは、ストレプトミセス菌における選択のためのチオストレプトン耐性遺伝子(tsr)及び大腸菌における選択のためのアンピシリン耐性遺伝子(bla)を保有する接合型組込み型プラスミドである。接合に使用された大腸菌株S17-1はクロラムフェニコール(cam)に対して自然に耐性があるので、pXY152-endorf24-camtsr(上記参照)のクロラムフェニコール耐性マーカーは、S17-1形質転換体を選択するためにアンピシリン耐性(bla)で置き換えられた。あるいは、pXY152-endorf24-camtsr及び誘導体は、異なる接合大腸菌株ET12567/pUZ8002を使用することによってストレプトミセス菌に導入することができる。
Claims (18)
- 配列番号26のアミノ酸配列をコードする増強型オープンリーディングフレーム-24(orf24)、減少型オープンリーディングフレーム-18(orf18)、並びに、増強型orf24及び減少型orf18の両方からなる群から選択される1つ又は複数の修飾遺伝子を含む、増強されたエンデュラシジン産生のためのStreptomyces fungicidicusの組換え株であって、
対照としての野生型Streptomyces fungicidicus株で得られるものと比較して、前記増強されたエンデュラシジン産生が前記Streptomyces fungicidicusの組換え株で得られる、組換え株。 - 前記減少型orf18は、ヌル化されたために減少している、請求項1に記載の組換え株。
