Description
[1] 変性凝固により固体化された血漿由来タンパク又は血清由来タンパクを含む担体、及び該担体に担持されたペルオキシダーゼ様活性を有するタンパクを含む複合体。
[2] ペルオキシダーゼ様活性を有するタンパクが実質的に未変性の状態又は変性した状態で担持されている上記[1]に記載の複合体。
[3] ペルオキシダーゼ様活性を有するタンパクが血漿由来タンパク又は血清由来タンパクとともに変性凝固により固体化された状態で担持されている上記[1]に記載の複合体。
[4] ペルオキシダーゼ様活性を有するタンパクがヘモグロビン又はミオグロビンである上記[1]ないし[3]のいずれか1項に記載の複合体。
[5] さらに1種又は2種以上の免疫刺激物質を担持させた上記[1]ないし[4]のいずれか1項に記載の複合体。
[6] 免疫刺激物質が精製ツベルクリン、BCG菌抽出物、サイトカイン、トル様受容体リガンド、NOD様受容体アゴニスト、RIG様受容体アゴニスト、Cタイプレクチン受容体アゴニスト、環状GMP-AMPシンターゼに結合する外因性DNA、アラーミン、抗原、及び抗体よりなる群から選ばれる1ないし2以上の物質である上記[5]に記載の複合体。

[19] ヒトを含む哺乳類動物において疾病、好ましくは悪性腫瘍を治療及び/又は予防する方法であって、上記[7]に記載の免疫刺激剤又は上記[8]に記載の免疫刺激性組成物と、免疫反応抑制作用阻害物質との組み合わせの有効量をヒトを含む哺乳類動物に投与する工程を含む方法。

本発明における用語や概念は、当該分野において慣用的に使用される用語の意味に基づくものであり、本発明を実施するために使用する技術は、特にその出典を明示した技術を除いては、公知の文献等に基づいて当業者であれば容易かつ確実に実施可能である。また、各種の分析などは、使用した分析機器又は試薬、キットの取り扱い説明書、カタログなどに記載の方法を用いて行っている。
ヒトマクロファージ様細胞株THP-1は、培養下でPhorbol 12-Myristate 13-Acetate(PMA)を作用させて分化誘導すると、貪食能を示すのみならず(非特許文献18)、抗原提示能も獲得し、抗原提示細胞となることが知られている(非特許文献19)。また、この細胞をサイトカインであるヒトインターフェロンガンマ(IFNg)で前処理すると、T細胞への抗原提示能が増強される(非特許文献20)。分化し貪食をしたTHP-1細胞はTNFαを産生する(非特許文献21)。TNFαの産生はマクロファージ(ないし、抗原提示細胞)が活性化したことを示しており、体内におけるマクロファージ(ないし、抗原提示細胞)の活性化は、それに続く炎症反応及び免疫反応の起点になっていることが知られている(非特許文献22、23)。
成人のボランティアより常法によりヘパリン添加血液として採血し、常法の遠心法によって血漿を得た。この2.25mLを110℃、5分間熱処理し凝固させた後、スパーテルで大まかに破砕した。ここに市販の10%中性緩衝ホルマリン液を20mL加え、3日間室温で放置しホルマリン固定した。固定破砕物を生理食塩液で洗浄後、マイクロチューブをセットできる汎用の組織破砕機(Tissue Lyser II, Qiagen社製)にて10分間破砕処理した。微粒子化した断片を70μmの孔径のメッシュを通過させ、十分量の生理食塩液で遠心洗浄し、さらにエタノールに漬けて殺菌、再び十分量の生理食塩液で遠心洗浄した。卓上遠心機で3000rpm(最大加速度1580G)で15分間遠心沈殿させた断片の容量割合が25v/v%になるように、生理食塩液を適量加えて懸濁し、バイオアッセイ用サンプル2「変性凝固固体化血漿担体懸濁液」とした。
常法に従って維持培養しているヒトマクロファージ様細胞株THP-1を、培養液にて50万個/mLに調整した。培養液は、常法による加熱処理済みウシ胎児血清を3%含むRPMI1640培養液である(以下、「3%medium」という)。これにphorbol 12-myristate 13-acetate(PMA)溶液 (SIGMA社製PMAをdimethylsulfoxideに1.62mMとなるように溶解したもの)を終濃度0.16μMになるように添加し、24穴プレートに1穴あたり0.5mL播種して4日間培養、THP-1細胞を抗原提示細胞に分化誘導した。以後の実施例でも、抗原提示細胞と記載しているのは、この方法により分化誘導した細胞である。
サンプル 1. 生理食塩液、32μL(このサンプルのTNFα産生量をブランク値とした)

サンプル 4. 未変性ミオグロビン吸着変性凝固固体化血漿担体懸濁液、32μL
アッセイ培養後の培養液を冷却微量高速遠心機で、4℃、12000rpm(最大加速度11000G)、5分間遠心、上清を回収し、その中のTumor necrosis factor alpha (TNFα)濃度を測定した。1つのサンプルについて2又は3ウエルを用い、平均値をデータとした。
市販のELISA法によるTNFα測定用キット(BD Biosciences社製OptEIA Set Human TNF, Cat. No. 555212)を使用した。ELISAの詳細方法はこのキット添付の取り扱い説明書に従った。この操作過程では、界面活性剤Tween 20を含む大過剰の洗浄液をELISA用の各ウエルに添加して洗浄する工程が繰り返されるが、本ELISAでは、捕捉用1次抗体でコーティングしてあるELISA用ウエルに回収培養液遠心上清を添加後では5回、検出用2次抗体に追加添加するstreptavidin-horseradish peroxidase (このキットに含まれている、SAv-HRPと略す)又はstreptavidin-alkalinephosphatase (Promega社製、Cat. No. V5591、SAv-ALPと略す)添加後では7回、計12回の洗浄工程を繰り返した。そのため、回収培養液遠心上清中に、仮に変性凝固固体化血漿担体の微粒子が混入していたとしても、十分に洗浄除去される。実際、回収培養液遠心上清中のTNFα測定の際、SAv-HRPをあえて添加せずに本TNFα測定用キットによるELISAを実行すると、TNFα測定値がブランクレベルになってしまうことから、回収培養液遠心上清は十分に洗浄除去されていることを確認した。
例1では、未変性ヘモグロビンを変性凝固固体化血漿担体に吸着させたものに抗原提示細胞の刺激効果があることを示したが、血清タンパクの代表例であるアルブミンと、赤血球由来のヘモグロビンをあらかじめ混合し、その混合物を変性凝固固体化した場合の抗原提示細胞の刺激活性を調べた。
(1)バイオアッセイ用サンプルの調製方法
市販の生物学的製剤基準 人血清アルブミン(化学及血清療法研究所製、献血アルブミン25“化血研”、以後「25%HSA」という)に対し、以下に示す割合でヒトヘモグロビン(SIGMA社製、H7379-5G、生理食塩液に溶解・懸濁)を混合した液を作製し、110℃、5分間熱処理し凝固させた後、スパーテルで大まかに破砕した。ここに10%中性緩衝ホルマリン液を20mL加え、3日間室温で放置しホルマリン固定した。固定破砕物を生理食塩液で洗浄後、マイクロチューブをセットできる汎用の組織破砕機(Tissue Lyser II, Qiagen社製)にて10分間破砕処理した。破砕断片を70μmの孔径のメッシュを通過させ、十分量の生理食塩液で遠心洗浄し、さらにエタノールに漬けて殺菌し、再び十分量の生理食塩液で遠心洗浄した。卓上遠心機で3000rpm、15分間遠心沈殿させた断片の容量割合が25v/v%になるように、生理食塩液を適量加えて懸濁し、バイオアッセイ用サンプル「変性凝固固体化ヘモグロビン・アルブミン微粒子懸濁液」とした。このサンプルには、ヘモグロビンの割合を変えた以下の種類がある。
サンプル 2. 25%HSA 2.4mL + 100mg/mL Hemoglobin 0.6mL
サンプル 3. 25%HSA 1.84mL + 220mg/mL Hemoglobin 0.909mL + 生理食塩液0.251mL
サンプル 4. 25%HSA 1.04mL + 220mg/mL Hemoglobin 1.818mL + 生理食塩液0.141mL
サンプル 5. 220mg/mL Hemoglobin 3.0mL
サンプル 6. 生理食塩液 3.0mL(このサンプルのTNFα産生量をブランク値とした)
これらのサンプルは、アッセイ培養時には、0.5mLの3%mediumを含むウエルに、いずれも1ウエルあたり32μL添加した。1サンプルについて3ウエルを用い、平均値をデータとした。
前述の例1、及び上記のバイアッセイ用サンプルにエンドトキシンが混入していると、結果に影響する可能性がある。そこで、エンドトキシン含有量を測定した。測定には、Kinect-QCL 192 Test Kit(Lonza社製)、及び、標準エンドトキシンとしてE. coli O55:B5を用い、測定方法はこのキットの使用説明書に従った。その結果、前述の例1のバイアッセイ用サンプル、今回のバイアッセイ用サンプルでは、すべてについてエンドトキシン含量は検出限界以下(<0.500EU/mL)であった。この結果から、図1及び図2におけるエンドトキシンの影響はないと判断した。
ヘモグロビンやミオグロビンのようなタンパクにはペルオキシダーゼ様の強い活性があることは広く知られている。この活性は、ホルマリン固定しパラフィン包埋処理してから作製した病理組織切片でも発揮されるため、病理組織切片中に例えば大量のヘモグロビンが含有されていると、ペルオキシダーゼ標識抗体による免疫染色では望ましからぬ染色像が出ることがある。この活性を消去できるのが過酸化水素水処理である(非特許文献26)。上記HAMPのペルオキシダーゼ様活性も非常に強いと考えられる。
例2(1)のバイオアッセイ用サンプル中で、ヘモグロビンの割合が最も少ないサンプル2をHAMPの代表例とした。これを遠心し、沈殿の重量を測定してから、沈殿含量を生理食塩液で40w/v%に合わせた懸濁液とした。この20μLを、ペルオキシダーゼによる発色反応に汎用されている市販のTMB基質液200μLに添加したところ、無色のTMB基質液が数秒で濃い青に変化した。これにより、HAMPのペルオキシダーゼ様活性も非常に強いことを確認した。
ホルマリン固定・パラフィン包埋後に作製された病理組織切片中のペルオキシダーゼ様活性は、過酸化水素水処理により消失することが広く知られている。例2におけるバイオアッセイ用サンプル2に過剰の過酸化水素水を添加して一夜放置後、生理食塩液で繰り返し遠心洗浄すると、サンプル2による市販のTMB基質液の着色反応は観察されなかった。一方、例2(1)のバイオアッセイ用サンプル中で、ヘモグロビンの割合が最も多いサンプル5のペルオキシダーゼ様活性は最も強いことから、サンプル5の150μLに対し、1)精製水、又は、2)3%過酸化水素水を900μL加え、一夜室温で撹拌した。冷却微量高速遠心機で4℃、12000rpm(最大加速度11000G)の条件下に5分間遠心して沈殿を得て、これを十分量の生理食塩液で遠心洗浄後、沈殿に対して112.5μLの生理食塩液を添加して懸濁し、バイオアッセイ用サンプルをそれぞれ1. Hemoglobin-MP、2. H2O2-Hemoglobin-MPとした。抗原提示細胞によるバイオアッセイ方法、及び、TNFαの測定方法は例1と同じである。
精製ツベルクリンは、IV型アレルギー反応(すなわち細胞性免疫反応)を誘導する代表的な免疫刺激物質である。精製ツベルクリン本体そのものは安定な物質であるが、溶解後はガラス瓶内壁に吸着し見かけ上の失活を起こしやすいこと、及び、その吸着はヒト血清アルブミンで防止可能であることが知られている(非特許文献16)。そこで、HAMPが精製ツベルクリンを吸着するか否かを検討した。
一般診断用精製ツベルクリン(PPD)(1μg/バイアル、日本ビーシージー製造株式会社)に含まれているMycobacterium tuberculosis由来のタンパクの一つ、DnaK(非特許文献24)に対するHuCAL-Fab抗体(ジーンフロンティア株式会社、千葉県柏市)を得た。これを1次抗体とし、Jackson Immuno Research 社の抗体#109-035-097を2次抗体として、精製ツベルクリンのELISA定量法を策定した。抗原としての精製ツベルクリン(PPD)、1次抗体、2次抗体以外は当業者に周知されている通常のELISA定量法である。最終的な発色反応には例1(3)に示したhorseradish peroxidaseに対する汎用のTMB基質液と反応停止液を用い、結果はA450値として出した。本法では、生理食塩液をブランクとした。ブランク値を差し引いたとき、上記PPDの濃度が1.25μg/mL(ELISA反応時の液量はELISA用96ウエルマイクロプレートの1ウエル当たり200μL)のときのA450値は0.063であり、定量可能な範囲であった。
プラスチック製マイクロチューブにHAMP(卓上遠心機で3000rpm(最大加速度1580G)で15分間遠心沈殿させた微粒子の容量割合が40v/v%になるように生理食塩液を適量加えて懸濁した)を終濃度20v/v%となるように、上記PPDの濃度が1.25μg/mLとなるように加え、室温で撹拌した。撹拌時間は30分~6時間である。撹拌後、微量高速遠心機で、4℃、12000rpm(最大加速度11000G)、5分間遠心、上清を得た。この上清150μLを用い、上記のELISA定量法により(生理食塩液をブランクとして)、上清中のPPD量を測定した。
(3)結果
上清中に残存するPPD量を、ブランク値を差し引いたA450値として図4に示した。この結果は、室温にて30分の撹拌で92%のPPDがHAMPに吸着することを示している。2時間以上の撹拌では(最小のA450値が-0.004という若干の測定誤差があるが)ほぼ全量がHAMPに吸着していると考えられる。また、この結果は、抗原となり得る異種由来タンパクもHAMPに吸着し得ることを示している。
例4に示したように、HAMPにはPPDに対する強い吸着力がある。そこで、吸着したPPDの抗原提示細胞の刺激活性はどうか、及び生体内で重要な作用をするサイトカインを吸着させた場合にサイトカイン吸着HAMPが抗原提示細胞の刺激活性を示すか、について検討した。
(1)吸着操作
50-mL遠心チューブに生理食塩液に懸濁したHAMP沈殿とPPDの濃度がそれぞれ25v/v%、286ng/mLとなるように加え、室温で2時間攪拌した。これをPPD-HAMP懸濁液とした。
ヒト顆粒球マクロファージコロニー刺激因子(hGM-CSF, Leukine(sargamostim)、sanofi-aventis U.S. LLC社製)を水で溶解、100μg/mLとした。この100μLとHAMP (25w/v%、変性凝固固体化ヘモグロビン・アルブミン微粒子沈殿の重量をもとに調製した)200μLと合わせ、室温で2時間攪拌した。攪拌後、微量高速遠心機で、4℃で12000rpm(最大加速度11000G)の条件下に5分間遠心して沈殿を得た。この沈殿を750μLの生理食塩液にて懸濁、同様に遠心して沈殿を得る遠心洗浄を計3回繰り返し、最終的な沈殿に150μLの生理食塩液を加えて、hGM-CSF-HAMP懸濁液とした。

サンプル 2. 一般診断用精製ツベルクリン(PPD)1人用(0.25μg/バイアル、日本ビーシージー株式会社)を1.2mLの生理食塩液で溶解したPPD溶液、32μL
サンプル 3. HAMP懸濁液、32μL
サンプル 4. PPD-HAMP(HAMPにPPDを吸着させたもの)懸濁液、32μL
サンプル 5. hGM-CSF-HAMP(HAMPにhGM-CSFを吸着させたもの)懸濁液、32μL
サンプル 6. hIL-2-HAMP(HAMPにhIL-2を吸着させたもの)懸濁液、32μL
サンプル 7. hIFNg-HAMP(HAMPにhIFNgを吸着させたもの)懸濁液、32μL
1サンプルについて3ウエルを用いた。22時間のアッセイ培養後に回収した培養液は、新鮮な3%mediumで1/2に希釈し、TNFαのELISAによる測定に供した。
ブランク値を差し引いたA450値について3ウエルの平均値を図5に示した。ただし、サンプル4では、サンプル2に比べ2倍量のPPDが添加した32μL中に含まれているため、実測A450値は0.374だが、図では測定値の半分0.187にしてある。
この結果では、溶液状態のPPD(サンプル2)はブランクとした生理食塩液(サンプル1)の場合と同レベルで(A450値は0.004、例3の場合と同様に測定誤差の範囲である)、抗原提示細胞の刺激効果は認められなかった。また、HAMPのみ(サンプル3)では、例2の場合と同様に、本来のブランク値よりTNFαの産生量が抑制されていたが、HAMPにPPDを吸着させたPPD-HAMP(サンプル4)では、刺激効果が観察された。従って、溶解性のPPDを固体化した断片であるHAMPに吸着固定した方が抗原提示細胞をより強く活性化できることが判明した。
前述の例1(2)項に記載したように、THP-1細胞を抗原提示細胞に分化誘導した場合、TNFα産生には、トリガーとして微量のIFNgを培養液に添加しておく必要があるが、HAMPのみではこの培養液中の微量IFNgを吸着してしまうために、例2のサンプル1、2、及び本実施例のサンプル3、6では、A450値がマイナスになったものと考えられる。
例5の結果から、HAMPに溶解性のPPDを吸着させた方が抗原提示細胞をより強く活性化できることが判明したが、他の免疫刺激物質でもこのような効果が期待できるか検討した。ここでは、多様な低分子が入り混じっているBCG菌抽出物(BCGext)を用いた。
(1)BCGextの作製方法
乾燥BCGワクチンアンプル1本(日本ビーシージー製造株式会社、12 mg入り)を110℃で5分間、オートクレーブにかけ、エタノール1 mlを加え、6時間以上攪拌した後、その懸濁液を採取して別の乾燥BCG製剤1本に加え、再び十分に攪拌し、その後、微量高速遠心器にて12000 rpm(最大加速度11000G)で5分間遠心し、この上清を回収してBCGextとした。
HAMP(40w/v%)に、エタノールにて1/5に希釈したBCGextを等量加え、室温にて一夜撹拌した。これを十分量の生理食塩液で遠心洗浄し、もとのHAMP(40w/v%)となるように懸濁した。3%mediumにて5.65倍に希釈し、サンプルHAMP-BCGextとした。
(3)抗原提示細胞によるバイオアッセイ方法、及びTNFαの測定方法は、例1と同じで、TNFαの測定ではSAv-HRPを用いている。
ここでは、バイアッセイ用サンプルとして、1ウエルあたり、以下のものを添加した。
サンプル 1. 生理食塩液、30μL(このサンプルのTNFα産生量をブランク値とした)
サンプル 2. HAMP(40w/v%)を生理食塩液にて5.65倍に希釈したもの、30μL(アッセイ培養時の終濃度0.4w/v%)
サンプル 3. BCGextを3%mediumにて28.3倍に希釈したもの、30μL(アッセイ培養時の終濃度0.2v/v%)
サンプル 4. HAMP-BCGext、30μL(アッセイ培養時のHAMP終濃度0.4w/v%, 元のBCGextが全部HAMPに吸着したと仮定すれば終濃度0.18v/v%となるはずのもの)
結果を図6に示す。この結果はBCGextをそのまま添加するよりも、HAMPにBCGextを吸着させた方がより強力に抗原提示細胞を刺激できることを示している。すなわち、PPDに限らず、溶解性の免疫刺激物質をHAMPに吸着させることができれば、より強力な免疫刺激剤となることから、HAMPは免疫刺激物質の効果的な担体であることがわかる。
結果を図7に示す。この結果から、TNFαに比べ、1/40量程度のhGM-CSFが同時に産生されていることが判明した。
例5のサンプル5で見られたように、hGM-CSFは直接HAMPに吸着し、吸着hGM-CSFは抗原提示細胞を刺激し活性化できる。従って、BCGextを担持させたHAMPによる刺激をきっかけに、抗原提示細胞からhGM-CSFが産生され、それが抗原提示細胞自身にも作用して活性化するというオートクライン機構が作動することが判明した。産生されたhGM-CSFは、産生した抗原提示細胞自身に作用するだけではなく、別種の抗原分子について提示を行う抗原提示細胞であっても、近隣にいるならばhGM-CSFはそれにも作用し活性化できるというパラクライン作用もあることは、当業者にとっては周知である。従って、本発明の免疫刺激剤では、刺激を受けた抗原提示細胞から分泌されるhGM-CSFが、近隣の抗原提示細胞に対する作用を通じて一般的な免疫反応を強化できるので、同時に他の特定の抗原を投与せずともBRMとしても使用できる。
抗体は、その種類によっては免疫刺激作用があることが知られている。前項の実施例までに、HAMPは各種の分子を吸着する能力があると推定された。そこで抗体も吸着するか、検討した。
(1)吸着操作
市販のヤギ抗マウスIgG(H+L)抗体(horseradish peroxidase標識)(Cat. No. W4021、Promega社製)をDulbeccoのCa+Mg+不含リン酸緩衝液(PBS(-))で1/25000に希釈し、この20μLをとり、HAMP 60μL及び生理食塩液920μLを含む懸濁液に加え、0~60分間室温で撹拌した。撹拌後、直ちに冷却微量高速遠心機で、4℃、12000rpm(最大加速度11000G)、5分間遠心、その上清100μL中に残存する抗体のペルオキシダーゼ活性を測定した。ペルオキシダーゼ活性は、例1(3)TNFαの測定方法のTNFα測定用キットに付随しているTMB基質液による発色反応を利用してA450値で表した。
(2)結果
図8に示すように、上清中のペルオキシダーゼ活性は撹拌時間とともに減少した。ヤギ抗マウスIgG(H+L)抗体にHAMP を添加していない場合、抗体の希釈液は1時間程度の間では活性に変化がないことが知られているため、この減少は抗体がHAMP沈殿に経時的に吸着していくことを示している。また、この結果は、もしヤギ抗体を抗原タンパクとして利用したい場合、抗体溶液としてではなく、抗体をHAMPに吸着させた固体化抗原として利用できることを示している。
抗体吸着が別種の抗体でも同様に起こるかを検討した。
(1)吸着操作
市販のウサギ抗ヤギIgG(全分子)抗体(alkaline phosphatase標識)(A4187、Sigma-Aldrich社製)をPBS(-)で1000倍に希釈し、1次抗体とした。この100μLをとり、HAMP 60μL及び生理食塩液840μLを含む懸濁液に加え、0min~60min室温で撹拌した。撹拌後、直ちに冷却微量高速遠心機で、4℃で12000rpm(最大加速度11000G)の条件下に5分間遠心し、得られた沈殿を十分量の生理食塩液で遠心洗浄した。この洗浄後の沈殿を100μLの生理食塩液に懸濁し、その半量に、アルカリ性ホスファターゼ用緩衝液(TRACP & ALP Assay Kit、Code No. MK301、タカラバイオ社製)とこのキットの反応基質液を説明書に従って190μL加えた。室温で10分間撹拌し、反応停止液(0.5N NaOH)30μLを加えた後、冷却微量高速遠心機で、4℃で12000rpm(最大加速度11000G)の条件下に5分間遠心し、上清200μLをとって405nmの吸光度を測定した。
図9に示すように、沈殿中のアルカリ性ホスファターゼ活性は、1次抗体との撹拌時間の長さに従って増加した。すなわち、ウサギ抗ヤギIgG(全分子)抗体もHAMPに吸着することを示している。従って、抗体を吸着させたHAMPも、効果的な免疫刺激剤の組成物となり得ることが例7及び8の結果から分かる。また、抗体を吸着させたHAMPを免疫刺激剤として使用する場合、抗体が投与局所ですぐに溶解拡散してしまうことはないため、投与局所に限局した効果的な抗体適用法にすることができることがこの結果から明らかである。
例5では、HAMPに吸着したサイトカインによる抗原提示細胞の刺激効果が、hGM-CSF及びhIFNgでは認められたが、hIL-2では認められなかった。これらサイトカインは、いずれもヘパリンに結合しやすく、特にhIL-2はヘパリンに結合した状態で活性を示すことが知られている(非特許文献25)。そこで、HAMPとは別種の固体化担体であるヘパリン-アルブミンコアセルベート沈殿(微粒子状になっている)(特許文献4、ただし、この文献ではPPDを含むヘパリン-アルブミンコアセルベート沈殿について記載されているが、本実施例では、PPDを含まないヘパリン-アルブミンコアセルベート沈殿(nega-TuMPと命名)を用いた)にhIL-2を吸着させた微粒子(hIL-2-MP)とし、抗原提示細胞の刺激効果があるか否かを検討した。
5週令のC3H/HeN(オス)マウス14匹を2群にわけ、対照群には1匹あたり50μLの生理食塩液を、処置群には1匹あたり、PPD-HAMP(3000rpm(最大加速度1580G)で15分間遠心した時の沈殿量として19.1v/v%)とnega-TuMP懸濁液(容量割合で8.7 %)を含む懸濁液50μLを、それぞれ毎週1回、計3回、尾根部皮内に注射した。また、翌週よりそれぞれ毎週1回、計6回、大腿部皮下に注射し、体重を測定した。また、最終注射の7日後にマウスの脾臓、腎臓、肝臓の重量を測定した。
(2)結果
図10にマウスの体重変化を、図11に臓器重量を示す。これらの結果は、PPD-HAMPにnega-TuMPを混合した懸濁液には、成長期のマウスに体重減少をもたらすような強い毒性はなく、フロイントの完全アジュバント(FCA)を投与した場合に見られるような当業者に周知されている安全性に関する大きな問題がないことを示している。
例4に示したように、HAMPはPPDを速やかに吸着し、溶解性のPPDを不溶性にでき、また例5に示したように抗原提示細胞を刺激できる。そのため、PPD-HAMPは効果的な免疫アジュバントとなると推定される。また、HAMPは例5に示したようにサイトカインを吸着できる。一方で、サイトカインはヘパリンに良く吸着することが当業者には周知されており、ヘパリンとアルブミンのコアセルベーションにより生じたnega-TuMPにもサイトカインは良く吸着する。そこで、nega-TuMPにサイトカインを混合し吸着させ、PPD-HAMPに添加したときの免疫アジュバントとしての効果を検討した。
(1-1)サイトカイン吸着nega-TuMPの作製
マウスGM-CSF(Miltenyi Biotec社製, premium grade)100μgを1mLの精製水に溶解した。この50μLを100μLのnega-TuMP懸濁液と混合、一夜、室温で撹拌して吸着させ、これをmGM-CSF-MPとした。また、イムネース注35(シオノギ製薬)を注射用水625μLに溶解、この50μLをとって100μLのnega-TuMP懸濁液と混合、一夜、室温で撹拌し、これをhIL-2-MPとした。
卵白アルブミン(Calbiochem社製, カタログ番号32467、OVAと略す)1.12mgを1mLの生理食塩液に溶解した。これを抗原原液とし、下表のような液量の割合で、マウスのG1~G4群に合わせて、サイトカイン吸着(又は非吸着)nega-TuMPを免疫アジュバントとして添加した組成液を作製した。
6週齢のC57BL/6マウス(メス、日本クレア社より購入)に、上表の組成液を1匹あたり50μL、皮内注射した。一つのマウスの群は10匹である。注射開始日をday 0とした。このday 0のときの50μL中にはOVA原液を含んでいる。以後、OVAを含まない上表の組成液を、それぞれの群に1匹あたり50μL、day 4, 11, 18, 25, 32(計5回)に追加皮内注射した。
OVAタンパク断片を腫瘍抗原として発現しているマウスリンパ腫細胞株E.G7-OVAをアメリカンタイプカルチャーコレクションより入手、常法に従って培養した。この細胞を無血清のMEM培養液で洗浄、さらに無血清MEM培養液で2時間培養後に、もう一度無血清のMEM培養液で洗浄した。この細胞50万個を0.1mLの生理食塩液に懸濁し、(1-3)項で述べたマウスの右下肢皮下にday 15に注射、腫瘍細胞チャレンジとし、以後、増殖してくる腫瘍の容積を常法に従って測定した。腫瘍容積が5000 mm3に達してもなお生存しているマウスは測定打ち切りとし安楽死させた。
結果を図12に示す。マウスの生存期間中央値は、E.G7-OVA腫瘍細胞チャレンジ後、G1(対照)群で21日(組成液注射開始日からはday 36となる)、G2(PPD-HAMP-nega-TuMP)群で24日、G3(PPD-HAMP-mGM-CSF-MP)群で26日、G4(PPD-HAMP-mGM-CSF-MP-hIL-2-MP)群では31日以上となった。チャレンジ後31日目終了時点で生存しているマウスの割合は、G1群で1/10、G4群で5/10であり、この2群間の母比率の差の検定(カイ二乗検定)を実施したところ、両側p=0.051となり、G4群で延命効果がある強い傾向が観察された。他のG2群、G3群では、G1群に対する統計学的有意差はないが、それぞれ生存期間中央値がG1群よりも延びていることから、それぞれで延命効果がある傾向が認められた。
例10で用いた抗原は卵白アルブミンであり、免疫学においては典型的モデル抗原として汎用されているが、自然発生した腫瘍細胞に含まれる腫瘍抗原ではない。そこで、ここではマウス肺がん細胞のライセートを腫瘍抗原とし、生きている同じ肺がん細胞をチャレンジしたとき、添加剤としてサイトカイン吸着nega-TuMPを加えたPPD-HAMP懸濁液の免疫アジュバント効果を検討した。
(1-1)抗原と免疫アジュバントの組成液
サイトカイン吸着nega-TuMP懸濁液の作製法は例10の(1-1)項と同じである。マウス肺がんであるLLC細胞(理化学研究所・理研バイオリソースセンターより入手、細胞番号RCB0558)を、常法に従って増殖培養後、2時間無血清培養し、PBS(-)で十分洗浄後、常法のEDTA処理により細胞をはがしてからPBS(-)中で懸濁液とした。細胞数を2x107/mLに合わせてから、凍結融解を繰り返し、ライセートとした。ライセートは、3%トリパンブルー液で染色し、生残しているLLC細胞がいないことを確認した。これを腫瘍抗原とし、下表のような液量の割合で、マウスの各群に合わせて、サイトカイン吸着nega-TuMP(mGM-CSF-MP及びhIL-2-MP)を免疫アジュバントとして添加した組成液を作製した。ここではPPD-HAMP懸濁液にはnega-TuMPそのものは加えていない。
6週齢のC57BL/6マウス(メス、日本クレア社より購入)に、上表の組成液を1匹あたり0.1mL、背部皮下に注射した。一つのマウスの群は10匹であるが、mGM-CSF-hIL-2-PPD-HAMP群では8匹である。注射開始日をday 0とした。このときの投与0.1mL中にはLLC細胞ライセートを含んでいる。以後、LLC細胞ライセートを含まない上表の組成液を、それぞれの群に1匹あたり0.1mL、day 4, 10, 18, 25, 32, 39, 46に追加皮下注射した。
上述のライセートを作製した細胞の元株である生きているLLC細胞を、無血清のMEM培養液0.1mL中に懸濁し、1x104個をday 15に大腿部皮下に注射、腫瘍細胞チャレンジとし、以後、触診により初めて腫瘍塊が発生した日を記録した。チャレンジ後、この日までがtumor-freeの期間である。
LLC細胞のチャレンジ後、53日目まで観察をした結果、両群でtumor-freeのまま残ったマウスは図13に示すとおりであった。対照(saline)群では腫瘍細胞チャレンジ後17日目から腫瘍塊の発生が観察されたのに対し、PPD-HAMP群では腫瘍塊の発生が24日目からと遅れており腫瘍塊の発生に抑制効果が認められたが、両群ともチャレンジ後50日以降でtumor-freeのままで残るマウスは6/10(60%)で同じであった。しかし、mGM-CSF-hIL-2-PPD-HAMP投与群では全く腫瘍塊の発生は観察されず(8/8、100%)、対照群との間で2群間の母比率の差の検定(カイ二乗検定)を実施したところ、両側p=0.0425となり、統計学的に有意な差があった。
Claims (8)
- 変性凝固により固体化された血漿由来タンパク又は血清由来タンパクを含む担体、及び該担体に担持されたペルオキシダーゼ様活性を有するタンパクを含み、ペルオキシダーゼ様活性を有するタンパクがヘモグロビン又はミオグロビンである複合体を含む免疫刺激剤と、以下の(1)及び/又は(2):
(1)顆粒球マクロファージコロニー刺激因子、インターフェロンガンマ、精製ツベルクリン、及びBCG菌抽出物から選ばれる1又は2以上の物質;
(2)アルブミン-ヘパリンコアセルベート沈殿であって、アルブミン-ヘパリンコアセルベート沈殿がサイトカインを吸着させたヘパリン-アルブミンコアセルベート沈殿であり、かつサイトカインが顆粒球マクロファージコロニー刺激因子、インターロイキン-2、及びインターフェロンガンマよりなる群から選ばれる1又は2以上の物質、
とを含む免疫刺激性組成物。 - 複合体がペルオキシダーゼ様活性を有するタンパクが実質的に未変性の状態又は変性した状態で担持されている複合体である請求項1に記載の免疫刺激性組成物。
- 複合体がペルオキシダーゼ様活性を有するタンパクが血漿由来タンパク又は血清由来タンパクとともに変性凝固により固体化された状態で担持されている複合体である請求項1に記載の免疫刺激性組成物。
- 複合体がさらに1種又は2種以上の免疫刺激物質を担持させた複合体である請求項1ないし3のいずれか1項に記載の免疫刺激性組成物。
- 免疫刺激物質がトル様受容体リガンド、NOD様受容体アゴニスト、RIG様受容体アゴニスト、Cタイプレクチン受容体アゴニスト、環状GMP-AMPシンターゼに結合する外因性DNA、アラーミン、抗原、及び抗体よりなる群から選ばれる1ないし2以上の物質である請求項4に記載の免疫刺激性組成物。
- 請求項1ないし5のいずれか1項に記載の免疫刺激性組成物と、免疫チェックポイント阻害剤との組み合わせを含む、悪性腫瘍の治療及び/又は再発若しくは転移の予防に用いるための医薬。
- 免疫チェックポイント阻害剤が、抗PD-1抗体、抗PD-L1抗体 、抗PD-L2抗体 、抗CTLA-4抗体、抗CD40抗体、抗CD137抗体、抗OX40抗体、抗TGF-β抗体、抗LAG3抗体、抗B7-H3抗体、抗B7-H4抗体、抗TIM3抗体、抗CD96抗体、及び抗TIGIT抗体よりなる群から選ばれる1又は2以上の抗体である、請求項6に記載の医薬。
- 請求項1ないし5のいずれか1項に記載の免疫刺激性組成物と、腫瘍抗原との組み合わせを含む、悪性腫瘍の治療及び/又は再発若しくは転移の予防のために用いるワクチン。
