本明細書において、疾患についての治療の有効性の判定を、それを必要とする対象において行う方法が提供される。方法は、対象からの試料を本明細書に詳述されるコンジュゲートと、そのコンジュゲートと試料中の標的との間で複合体が形成されることを可能にする条件下で接触させること、試料中の複合体のレベルを判定することであって、複合体のレベルが試料中の標的のレベルを示す、判定すること、および試料中の標的のレベルと標的の対照レベルとを比較することをさらに含み得、標的のレベルが対照レベルと異なる場合、治療は、その疾患の治療において有効または無効であると判定される。
材料および方法
クローニング。ZiPPのための合成遺伝子を化学合成オリゴマー(IDT Inc.;Coralville,IA)からアセンブルした。オリゴマーを、プラスミド再構築回帰的方向性ライゲーション(plasmid reconstruction recursive directional ligation)(Pre−RDL)技術(McDaniel,J.R.,et al.Biomacromolecules,2010,11,944−952)を使用して大腸菌(E.coli)中でpET発現ベクター中にクローニングした。
ZiPPの発現および精製
ZiPPの熱応答特性により、ITCを使用する簡易な非クロマトグラフィー精製が可能となった。銅染色SDS−PAGEゲル中の単一バンドの出現により、産物の純度が確認された(図2A)。2つの異なるZiPP、(VPKEG)80(MW=44kDa)および(VPREG)80(MW=42kDa)を精製産物の代表例として図7Aに示す。50kDaラダーをゲル中の参照MWとして標識するが、SDSゲル中で使用されたラダーは、球状タンパク質からのものであり、したがって非構造化ZiPPと直接比較可能でない。純度およびMWを確認するため、精製産物をMALDI−TOFにより分析した。MALDIスペクトルは、(VPKEG)80について21kDaおよび42kDa(ピーク1および2)のm/z値ならびに(VPREG)80について22kDaおよび44kDa(ピーク3および4)のm/z値におけるイオンの存在を示した(図7B)。MALDIスペクトルは、(VPKDG)120について20kDa、30kDa、および60.5kDaのm/z値および(VPRDG)120について21kDaおよび32kDaおよび63.8kDaのm/z値におけるイオンの存在を示し(図2B)、それにより精製ZiPP構築物の分子量(それぞれMW=60.5kDa、MW=63.8kDa)が確認された。本発明者らは、CDスペクトルを使用することによりZiPPの固有の無秩序性質も確認した。図2Dおよび図7CのCDスペクトルは、低波長で負の楕円率およびより高い波長でわずかに正の楕円率を示し、それはELPのような無秩序構造の典型であるランダムコイルの特徴であり、それによりポリマーの非構造化性質が確認された。動的光散乱を使用して流体力学的半径を計測し、それはELP対照と比較して十分に水和したZiPPを示した(図2C)。ネイティブPAGEゲルは、ZiPPがアルブミンと相互作用しないことを示した(図2E)。
インビボ薬物動態試験
ZiPPの薬物動態パラメータを判定するため、尾静脈注射または皮下注射を介するマウスにおける全身投与後の72時間の期間にわたり血漿濃度を追跡した。合致するアミノ酸長を有する非荷電ポリマーを対照として使用した。実験設計を図3Aおよび図4Aに示す。ポリマーをAlexa488により蛍光標識し、マウスに注射した。血液試料を72時間までの種々の時点で回収した。ELP120(VPGAG)120を長さ対照として使用した一方、ELP160(VPGAG)160を分子量対照として使用した。図3Bおよび図4Bは、注射後の時間に応じた血漿ポリマー濃度を示し、それは、双性イオン性モチーフ(具体的には、例として、それぞれX1およびX2についてK、D、およびE)の取り込みがステルス特性を付与し、それが次いで非荷電ELPと比較してポリマーの循環時間を増加させることを示した。注射後の時間に応じた血漿濃度は、ZiPPがELPよりも良好に機能することを示した。これらのポリマーは、2つのコンパートメンタルモデルに従い、したがってこのモデルを使用して半減期および曲線下面積を計算した。2つのコンパートメントモデルを血漿ポリマー濃度にフィットさせ、薬物動態パラメータの曲線下面積(AUC)(図3Cおよび図4C)および排出半減期(図3D)を生じさせた。AUCを実験の時間経過にわたる全ポリマー曝露の尺度として計算した。AUCは、VPKEGおよびVPKDGが非荷電ELP長さ対照および分子量対照よりも有意に良好に機能することを示した。データは、図3Cについて平均±SE、n=5を表し、図4Cについて平均±SE、n=3〜4を表す。ZiPPの最終半減期は、VPGAG構築物のものと比較して6時間だけ増加した(図3D)。さらに、最も記述的な薬物動態パラメータの、ZiPPについての血漿濃度曲線下面積(AUC)により計測されるポリマーの全累積血液曝露は、同一鎖長の非荷電ポリペプチドVPGAGのものよりも約3倍高かった(図3C)。結果は、ペプチドポリマー中への双性イオン性モチーフ、より具体的には荷電残基の取り込みが、ポリマーの薬物動態の改善において重要な役割を担うことを示した。
パクリタキセル−ZiPPコンジュゲートの特徴付け
パクリタキセル(PTX)を、(VPGXG)16(ここで、Xが1:1の比のVまたはCである)にVPKEGペンタペプチド単位の120個のリピートのC末端におけるトレーラーに化学的にコンジュゲートさせた。パクリタキセルを、pH感受性リンカーを介して8つのC末端残基に化学的にコンジュゲートさせた。この設計を図5Aに示す。Amicon Ultra−15 Centrifugal Filter Units(MWCO:10kDa;Millipore;Billerica、MA)を使用してポリマー薬物コンジュゲートを精製し、精製産物をHPLC上でランして未反応遊離薬物の不存在を確認した。HPLCクロマトグラムにより、無視可能な量の遊離薬物でのポリマー−薬物コンジュゲートの純度が確認された。精製ZiPP−PTXコンジュゲートは、MALDI−TOFスペクトルを使用して親ZiPPポリマー鎖とZiPP−PTXコンジュゲートとの間で計算されたMWの差異により確認されるとおり、1ポリマー鎖当たり3.2〜4つの薬物を有した(図5B)。さらに、動的光散乱計測は、PTXコンジュゲーション後、ZiPPが実際に58nmの流体力学的半径(Rh)のナノ粒子に自然に自己集合することを示した。これらは、58nm半径のミセルに自己集合し、1ミセル当たりの凝集数は26であった。Rg/Rhとして計算された形状因子(ρ)は0.82であり、それは球状ミセルの形成を示す。
ZiPP−PTXコンジュゲートのインビトロ抗腫瘍効力
時間に応じた一連の濃度にわたる細胞生存率を検査することにより、ZiPP−PTXのインビトロ細胞毒性を計測した。ヒト三重陰性乳癌細胞系MDA−MB−231をモデルとして使用した。薬物処理の72時間後、MDA−MB−231細胞の増殖は、対照(薬物なし)と比較して阻害された(図5C)。さらに、阻害は、遊離薬物のものと同等であった。遊離薬物についてのIC50値は約2nMであった一方、ZiPP−PTXのものは12.4nM(薬物に関する濃度)であった。ZiPP−PTXについてのIC50値は、遊離薬物のものよりも6倍高かったが、そのような結果は、遊離薬物が薬物トランスポーターを介して細胞内外に容易に拡散し得る一方、薬物−ポリマーコンジュゲートからのPTXがエンドソーム内側に存在する場合にのみ放出されるインビトロ環境で予測される。このプロセスは緩慢である。なぜなら、ナノ粒子がエンドサイトーシスを介して取り込まれ、pHが低い後期エンドソームにナノ粒子が移動した後に薬物が放出されるためである。この低いpHは、ZiPPからのPTXの放出を誘発する。これらの結果は有望である。なぜなら、これらは、PTX−ポリマーコンジュゲートがインビボプラットフォームに順応するほど十分に安定であり、強力であることを示すためである。IC50値は、細胞生存率を50%だけ低減させる薬物の濃度を表した。
ZiPP−Tn3コンジュゲート
多価足場タンパク質(Tn3)は、TNF関連アポトーシス誘導リガンド受容体2(TRAIL2)の超アゴニストであり、ZiPPに付着させるためのタンパク質として選別した。融合タンパク質の設計の概要を図6Aに示す。TRAIL2の超アゴニストを選択した。なぜなら、TRAILR2の活性化は種々のヒト癌でアポトーシスを誘導し得、したがって癌治療薬についての潜在性を有するためである。(Tn3)6は、MedImmuneにより高親和性でTRAIL2に結合するように遺伝子操作されたモノマーTn3単位の6つのタンデムリピートを表す。種々の長さのZiPPを有する(Tn3)6を大腸菌(E.coli)中で組換え発現させた。親和性精製試料のSDS−PAGE分析を図6Bに示す。Colo205(結腸癌細胞)に対する細胞毒性アッセイは、図6Cに示されるとおり、融合タンパク質が高度に細胞毒性であり、それらの効力は遊離タンパク質(付着ZIPPを有さない(Tn3)6)と同等であることを示した。IC50値は、細胞生存率を50%だけ低減させる薬物の濃度を表した。

前記対象からの試料を前記コンジュゲートと、前記薬物分子と前記試料中の標的との間で複合体が形成されることを可能にする条件下で接触させること;
前記試料中の前記標的のレベルを判定することであって、前記複合体のレベルが前記試料中の前記標的の前記レベルを示す、判定すること;および
前記試料中の前記標的の前記レベルと前記標的の対照レベルとを比較することであって、前記対照レベルと異なる前記標的のレベルが前記対象における疾患を示す、比較することを含む、キット。 - 前記対照レベルが、前記対象が治療を開始した期間の前またはその期間中の時点での前記対象におけるレベルに対応し、前記試料が後の時点で前記対象から採取される、請求項20に記載のキット。
- 前記疾患または障害が、乳癌、結腸直腸癌、結腸癌、肺癌、前立腺癌、精巣癌、脳癌、皮膚癌、直腸癌、胃癌、食道癌、非上皮性悪性腫瘍、気管癌、頭頸部癌、膵臓癌、肝臓癌、卵巣癌、リンパ癌、子宮頸癌、外陰癌、黒色腫、中皮腫、腎臓癌、膀胱癌、甲状腺癌、骨肉腫、上皮性悪性腫瘍、非上皮性悪性腫瘍、および軟部組織癌から選択される癌である、請求項19〜21のいずれか一項に記載の組成物またはキット。
