X−は、陰イオン化学種を表す。]
で表される化合物を含む、カタラーゼ阻害剤、
[2]式中、R1〜R4は、各々独立して、水素原子、又は炭素数1〜4の炭化水素基を表し、前記炭化水素基は、ハロゲン、アミノ基、ヒドロキシル基及びカルボキシル基からなる群より選択される少なくとも一種の置換基を有していてもよく、X−は、ハロゲンイオン、ハロゲンを含む化合物のイオン、水酸化物イオン、カルボン酸イオン、硝酸イオン、亜硝酸イオン、酢酸イオン、炭酸水素イオン、リン酸二水素イオン、硫酸水素イオン、硫化水素イオン、シュウ酸水素イオン、シアン酸イオン、及びチオシアン酸イオンからなる群から選択されるいずれか1つを表す、[1]記載のカタラーゼ阻害剤、
[3]式中、R1〜R4が、各々独立して、水素原子、又は非置換の炭素数1〜4の直鎖又は分枝鎖の飽和炭化水素基を表し、X−が、ハロゲンイオン、又はハロゲンを含む化合物のイオンを表す、[1]記載のカタラーゼ阻害剤、
[4]式(I)で表される化合物が2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファートである、[1]記載のカタラーゼ阻害剤、
[5]過酸化水素の生成に基づくアナライトの測定方法であって、下記の工程を含む方法:
(A)アナライト及びカタラーゼが含まれる試料と、[1]〜[4]のいずれかに記載のカタラーゼ阻害剤を混合する工程、
(B)第1酵素により前記アナライトを酸化させて過酸化水素を生成させる工程、及び、
(C)前記過酸化水素を測定する工程、
[6]前記工程(B)は、前記工程(A)の後、又は、前記工程(A)と同時に行われる、[5]に記載のアナライトの測定方法、
[7]過酸化水素の生成に基づくアナライトの測定方法であって、下記の工程を含む方法:
(a)アナライト及びカタラーゼが含まれる試料と、[1]〜[4]のいずれかに記載のカタラーゼ阻害剤を混合する工程、
(b−1)第2酵素により前記アナライトから基質を生成させる工程、
(b−2)第1酵素により前記基質を酸化させて過酸化水素を生成させる工程、及び、
(c)前記過酸化水素を測定する工程、
[8]前記工程が(b−1)、(a)、(b−2)、次いで(c)の順で行われるか、又は前記工程(b−1)の後、前記工程(a)と(b−2)が同時に行われ、次いで前記工程(c)が行われるか、又は前記工程(a)と(b−1)が同時に行われ、次いで前記工程(b−2)の後、前記工程(c)が行われる、[7]に記載のアナライトの測定方法、
[9]前記工程(C)又は(c)において、前記過酸化水素は、比色法、化学発光法又は電気化学的手法により測定される、[5]〜[8]のいずれかに記載のアナライトの測定方法、
[10]カタラーゼが含まれる試料中のアナライトを測定する測定キットであって、
前記アナライトを酸化させて過酸化水素を生成させる反応を触媒する第1酵素、及び、
[1]〜[4]のいずれかに記載のカタラーゼ阻害剤、
を含む、測定キット、
[11]カタラーゼが含まれる試料中のアナライトを測定する測定キットであって、
基質を酸化させて過酸化水素を生成させる反応を触媒する第1酵素、
[1]〜[4]のいずれかに記載のカタラーゼ阻害剤、及び、
前記アナライトから前記基質を生成させる反応を触媒する第2酵素、
を含む、測定キット、
[12]ペルオキシダーゼをさらに含む、[10]又は[11]に記載のキット、
[13][1]〜[4]のいずれかに記載のカタラーゼ阻害剤を含む、電気化学的手法による測定のための試料前処理用試薬、及び
[14]第1酵素での基質の酸化による過酸化水素の生成に基づいて、カタラーゼが含まれる試料中のアナライトを電気化学的手法により測定するための、試料前処理用キットであって、
[1]〜[4]のいずれかに記載のカタラーゼ阻害剤、及び、
前記アナライトから前記基質を生成させる反応を触媒する第2酵素、
を含む、キット、
[15]第1酵素が酸化酵素であり、第2酵素が分解酵素である、[5]〜[12]及び[14]のいずれかに記載の測定方法又はキット。
本願の第一の態様に係るカタラーゼ阻害剤(以下、「本願のカタラーゼ阻害剤」ともいう)は、下記の式(I):

X−は、陰イオン化学種を表す。]
で表される化合物(以下、化合物(I)ともいう)を含む。
本願のカタラーゼ阻害剤は、例えば、過酸化水素の生成に基づくアナライトの測定方法に用いることができる。したがって、本願発明の第二の態様として、本願のカタラーゼ阻害剤を用いる過酸化水素の生成に基づくアナライトの測定方法(以下、「本願の測定方法」ともいう)が提供される。
ase、EC番号;1.7.3.1)、尿酸オキシダーゼ(Uric Acid Oxidase、EC番号;1.7.3.3)、3−aci−ニトロプロパンオキシダーゼ(3−aci−Nitropropanoate Oxidase、EC番号;1.7.3.5)、亜硫酸オキシダーゼ(Sulfite Oxidase、EC番号;1.8.3.1)、チオールオキシダーゼ(Thiol Oxidase、EC番号;1.8.3.2)、グルタチオンオキシダーゼ(Glutathione Oxidase、EC番号;1.8.3.3)、メタンチオールオキシダーゼ(Methanethiol Oxidase、EC番号;1.8.3.4)、プレニルシステインオキシダーゼ(Prenylcysteine Oxidase、EC番号;1.8.3.5)、ファルネシルシステインリアーゼ(Farnesylcysteine lyase、EC番号;1.8.3.6)、3−ヒドロキシアントラニル酸オキシダーゼ(3−Hydroxyanthranilate Oxidase、EC番号;1.10.3.5)、キサンチンオキシダーゼ(Xanthine Oxidase、EC番号;1.17.3.2)、6−ヒドロキシニコチン酸デヒドロゲナーゼ(6−Hydroxynicotinate Dehydrogenaze、EC番号;1.17.3.3)、レチクリンオキシダーゼ(Reticuline Oxidase、EC番号;1.21.3.3)等が挙げられる。
(A)アナライト及びカタラーゼが含まれる試料と、化合物(I)を含むカタラーゼ阻害剤を混合する工程、
(B)第1酵素により前記アナライトを酸化させて過酸化水素を生成させる工程、及び、
(C)前記過酸化水素を測定する工程、
を含む方法である。該測定方法において、アナライトは、第1酵素の基質である。
(a)アナライト及びカタラーゼが含まれる試料と、化合物(I)を含むカタラーゼ阻害剤を混合する工程、
(b−1)第2酵素により前記アナライトから基質を生成させる工程、
(b−2)第1酵素により前記基質を酸化させて過酸化水素を生成させる工程、及び、
(c)前記過酸化水素を測定する工程、
を含む方法である。
次に、本願の第三の態様に係るカタラーゼが含まれる試料中のアナライトを測定する測定キット(以下、「本願の測定キット」ともいう)について説明する。
次に、本願の第四の態様に係る、化合物(I)を含む、電気化学的手法による測定のための試料前処理用試薬(以下、「本願の試料前処理用試薬」ともいう)について説明する。
次に、本願の第五の態様に係るカタラーゼが含まれる試料中のアナライトを電気化学的手法により測定するための、試料前処理用キット(以下、「本願の試料前処理用キット」ともいう)について説明する。
さらに、本願の第六の態様として、アナライトを測定するためのセンサであって、作用極及び対極を含む電極を含み、第1酵素及び本願のカタラーゼ阻害剤が前記電極に担持されているセンサを提供する。前記電極は、さらに第2酵素を担持していてもよい。第1酵素及び第2酵素の例については上述したとおりである。電極の構成材料については、特に限定されず、白金、金、銀、パラジウム、カーボン、イリジウム、酸化イリジウム等の過酸化水素の電気化学的定量に通常用いられているいずれの材料であってもよい。好ましくは、白金、又はパラジウムが電極材料として用いられる。前記センサは、さらに参照極を含むものであってもよい。前記酵素、及びカタラーゼ阻害剤を電極上に担持させる方法は、所望の酵素、及びカタラーゼ阻害剤を電極上に担持させることが可能ないずれの方法であってもよく、特に限定されない。当該分野で既知のいずれかの方法を用いることができる。例えば、インクジェット方式を用いてもよい。
(方法)
2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファート(0.001%(w/v))又はアジ化ナトリウム(0.001%(w/v)と、プロテアーゼ(アクチナーゼE、18000U/mL)、陰イオン性界面活性剤(N−ドデカノイルサルコシン酸ナトリウム、0.5%(w/v))、グルタミン酸カリウム(300mM)、ビストリス緩衝液(10mM、pH6.5)、及びヒト血液(25倍希釈相当)を混合した。血液添加後4分30秒後に、過酸化水素1%(v/v)を前記混合液中に投入した。過酸化水素投入後の発泡(酸素発生)の有無を目視により確認した。なお、各成分の濃度は反応時終濃度を示す。
2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファート又はアジ化ナトリウムのいずれを添加した場合も、過酸化水素投入後の発泡が見られなかった。これは、血液中のカタラーゼを抑制できたことを示す。
(方法)
プロテアーゼ(アクチナーゼE、2250U/mL)、陰イオン性界面活性剤(N−ドデカノイルサルコシン酸ナトリウム、5.0%(w/v))、グルタミン酸カリウム(300mM)、リン酸カリウム緩衝液(30mM、pH8)、カタラーゼ阻害剤、及びHbA1c測定用実試料一次標準物質(検査医学標準物資機構より入手、JCCRM 411、レベル1レベル3、及びレベル5のそれぞれの25倍希釈相当を使用)を混合し、測定溶液を調製した。前記カタラーゼ阻害剤として、アジ化ナトリウム(350μM、又は1750μM)、又は2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファート(350μM、又は1750μM)を使用した。
パラジウム電極からなる作用極及び対極を備えたセンサに、フルクトシルペプチドオキシダーゼ(229U/ml)、カルボキシメチルセルロース(0.125%(w/v))、リン酸カリウム緩衝液(25mM、pH8)、グルタミン酸カリウム(10mM)及びトレハロース(0.5%(w/v))を担持させた。
前記センサに前記測定溶液を点着し、0.62Vを1秒間作用極と対極の間に印加し、15秒静止後、0.25Vを作用極と対極に印加し、10秒後の電流値を測定した。
なお、各成分の濃度は測定液中の終濃度を示す。
結果を図2に示す。図2は、アジ化ナトリウム又は2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファートをそれぞれ350μM又は1750μM添加した時のHbA1c濃度に対する電流応答を示す。また、各測定結果の線形近似曲線の式を表1に示し、線形近似曲線の傾きを切片で割った値を表2に示す。
なお、表中、「NaN3」はアジ化ナトリウムを示し、「AIP」は2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファートを示す。
(方法)
実施例2と同じ方法で、HbA1c濃度に対する電流値を測定した。但し、カタラーゼ阻害剤として、2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファート(1750μM、又は3500μM)を使用した。
結果を図3に示す。図3は、2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファートを1750μM又は3500μM添加した時のHbA1c濃度に対する電流応答を示す。また、各測定結果の線形近似曲線の式を表3に示し、線形近似曲線の傾きを切片で割った値を表4に示す。
なお、表中、「AIP」は2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファートを示す。
(方法)
実施例2と同じ方法で、HbA1c濃度に対する電流値を測定した。但し、カタラーゼ阻害剤として、2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファート(1750μM、又は250μM)を使用した。
結果を図4に示す。図4は、2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファートを1750μM又は250μM添加した時のHbA1c濃度に対する電流応答を示す。また、各測定結果の線形近似曲線の式を表5に示し、線形近似曲線の傾きを切片で割った値を表6に示す。
なお、表中、「AIP」は2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファートを示す。
Claims (11)
- 2−アジド−1,3−ジメチルイミダゾリニウムヘキサフルオロホスファートを含む、カタラーゼ阻害剤。
- 過酸化水素の生成に基づくアナライトの測定方法であって、下記の工程を含む方法:
(A)アナライト及びカタラーゼが含まれる試料と、請求項1記載のカタラーゼ阻害剤を混合する工程、
(B)第1酵素により前記アナライトを酸化させて過酸化水素を生成させる工程、及び、
(C)前記過酸化水素を測定する工程。 - 前記工程(B)は、前記工程(A)の後、又は、前記工程(A)と同時に行われる、請求項2に記載のアナライトの測定方法。
- 過酸化水素の生成に基づくアナライトの測定方法であって、下記の工程を含む方法:
(a)アナライト及びカタラーゼが含まれる試料と、請求項1記載のカタラーゼ阻害剤を混合する工程、
(b−1)第2酵素により前記アナライトから基質を生成させる工程、
(b−2)第1酵素により前記基質を酸化させて過酸化水素を生成させる工程、及び、
(c)前記過酸化水素を測定する工程。 - 前記工程が(b−1)、(a)、(b−2)、次いで(c)の順で行われるか、又は前記工程(b−1)の後、前記工程(a)と(b−2)が同時に行われ、次いで前記工程(c)が行われるか、又は前記工程(a)と(b−1)が同時に行われ、次いで前記工程(b−2)の後、前記工程(c)が行われる、請求項4に記載のアナライトの測定方法。
- 前記工程(C)又は(c)において、前記過酸化水素は、比色法、化学発光法又は電気化学的手法により測定される、請求項2〜5のいずれか1項記載のアナライトの測定方法。
- カタラーゼが含まれる試料中のアナライトを測定する測定キットであって、
前記アナライトを酸化させて過酸化水素を生成させる反応を触媒する第1酵素、及び、
請求項1記載のカタラーゼ阻害剤、
を含む、測定キット。 - カタラーゼが含まれる試料中のアナライトを測定する測定キットであって、
基質を酸化させて過酸化水素を生成させる反応を触媒する第1酵素、
請求項1記載のカタラーゼ阻害剤、及び、
前記アナライトから前記基質を生成させる反応を触媒する第2酵素、
を含む、測定キット。 - ペルオキシダーゼをさらに含む、請求項7又は8に記載のキット。
- 請求項1記載のカタラーゼ阻害剤を含む、カタラーゼが含まれる試料中のアナライトの電気化学的手法による測定のための試料前処理用試薬。
- 第1酵素での基質の酸化による過酸化水素の生成に基づいて、カタラーゼが含まれる試料中のアナライトを電気化学的手法により測定するための、試料前処理用キットであって、
請求項1記載のカタラーゼ阻害剤、及び、
前記アナライトから前記基質を生成させる反応を触媒する第2酵素、
を含む、キット。
