実施例1.1.− 抗CD3抗体抗HER2 Fynomer(登録商標)融合タンパク質の発現および精製
15アミノ酸リンカー(配列番号162)を介してHER2結合Fynomer(登録商標)C12(配列番号4)を抗CD3結合抗体(配列番号2および3)に遺伝子融合して、本発明に係る二重特異的抗体Fynomer(登録商標)融合タンパク質を生成した。COVA420では、Fynomer(登録商標)C12を抗CD3抗体(配列番号2および3)の重鎖のN末端に融合した。COVA422では、Fynomer(登録商標)C12を抗CD3抗体(配列番号2および3)の重鎖のC末端に融合した。
本発明に係る抗体Fynomer(登録商標)融合タンパク質および対照抗体は、単一工程で発現および精製を行うことが可能であった。図1および2に示されるように、SDS−PAGE解析により>95%の純度を実証可能であった。表1には、CHO細胞への一過性トランスフェクションおよび37℃で6日間のタンパク質発現を行った後の発現収量がまとめられている。
PBS/1%BSA/0.2%アジ化ナトリウム中、(i)1×105Jurkat E6−1細胞(CD3陽性細胞)、(ii)1×105BT474 HER2陽性乳癌細胞、(iii)1×105HER2またはCD3陰性MDA MB468細胞のいずれかを含有する100μlの全体積で、タンパク質COVA420(配列番号163および164)、COVA422(配列番号165および166)、およびCOVA446(配列番号167)を100nMの濃度でインキュベートした。陰性対照として、タンパク質の代わりにPBS/1%BSA/0.2%アジ化ナトリウムのみを用いて、同一の細胞をインキュベートした(PBS対照)。
フローサイトメトリーを介してFynomer(登録商標)−抗体融合タンパク質の結合性を評価した。図4に示されるように、(COVA420(配列番号163および164)、COVA422(配列番号165および166)、および二重特異的scFv対照(COVA446、配列番号167)は、HER2発現BT474細胞(パネルA)およびCD3発現Jurkat E6細胞(パネルB)に特異的に結合した。CD3またはHER2のいずれも発現しない細胞系では、非特異的結合では観測されなかった(パネルC)。
Jaeger et al(2009)Cancer Research,69 10):4270−6から採用したプロトコルを用いて、リダイレクトT細胞媒介細胞傷害性アッセイで、ポリペプチドCOVA420(配列番号163および164)および二重特異的scFv対照(配列番号167)を試験した。
生存%=(値−100%溶解)/(自然溶解−100%溶解)*100
を用いて細胞生存パーセントを計算した。次いで、細胞生存%をエフェクター分子濃度に対してプロットし、Prism 5(GraphPad Software)を用いて用量−反応シグモイドに当てはめることにより、データを評価した。
HER2発現SKBR3細胞に対するCOVA420(配列番号163および164)の用量依存的細胞傷害性を観測可能であることを実証可能であった(図5)。この代表的なアッセイでは、COVA420(配列番号163および164)は、87pMのEC50値を有していた。重要なこととして、図5に示されるようにHER2陰性MDA−MB−468では細胞傷害性が観測されなかったため、細胞傷害作用は抗原依存的であった。
抗原陽性標的細胞およびCD8+富化T細胞の存在下および不在下で、COVA420(配列番号163および164)および対照として抗CD3抗体(配列番号2および3)のインキュベーション時の細胞培養培地中へのグランザイムBの放出を評価した。Haas A.et.al.Immunobiology,2009,214(6):441−53により記載されるように、グランザイムBの放出は、BiTE(登録商標)様作用剤により誘導されるT細胞媒介細胞傷害活性の主要インジケーターである。Fynomer(登録商標)−抗体融合タンパク質の細胞傷害活性の評価に対して以上に記載したように、サンプルをインキュベートした。インキュベーションの終了時、細胞培養上清を捕集し、製造業者の推奨に従ってグランザイムB ELISAキット(研究開発システム)を用いて、グランザイムBの濃度を評価した。
図7は、指定の濃度でCD8+T細胞の存在下ならびに抗原陽性標的細胞の存在下および不在下でCOVA420(配列番号163および164)のインキュベーションを行った3日後のグランザイムBの発現レベルを示している。標的細胞の不在下で50nMのCOVA420(配列番号163および164)のみを用いてT細胞をインキュベートした場合、グランザイムBの非特異的な放出が検出されうる。標的細胞COVA420(配列番号163および164)およびT細胞が存在した場合、グランザイムB発現の顕著な増加が観測された。細胞傷害作用が検出できない濃度でCOVA420(配列番号163および164)を用いた場合、実質的なグランザイムB発現を検出できなかったことから、細胞傷害活性とグランザイムB放出(0.5pM)との間の相関が示唆される。対照抗CD3抗体(配列番号2および3)(50nM)は、予想どおり、腫瘍標的およびエフェクター細胞の存在下でグランザイムB放出をトリガーしなかった。
C57BL/6マウス(Charles River)においてCOVA420(配列番号163および164)の薬動学的プロファイルを調べた。5匹のC57BL/6マウスに200μgのCOVA420(配列番号163および164)を静脈内注射した。10分後、6、24、48、96、120、144、および168時後、EDTA被覆microvettes(Sarstedt)中に血液を捕集し、9300gで10分間遠心分離し、そしてCOVA420(配列番号163および164)の血清中レベルをELISAにより決定した。黒色maxisorpマイクロタイタープレート(Nunc)を50nM HER2 ECD(ベンダーMedSystems)で被覆した。PBS中2%BSA(Sigma)でブロックした後、40μlのPBSおよび適切な希釈度の10μlの血清を適用した。1時間のインキュベーション後、ウェルをPBSで洗浄し、結合されたCOVA420(配列番号163および164)を抗−hIgG−HRP(Jackson ImmunoResearch)で検出した。アッセイをQuantaRed蛍光原基質(Pierce)で発色させ、544nm(励起)および590nm(発光)で5分後に蛍光強度を測定した。COVA420(配列番号163および164)(333〜0.5ng/mlに希釈)の標準曲線を用いて、COVA420(配列番号163および164)の血清中レベルを決定した。さまざまな時間点で決定された血清中に濃度および排出相(片対数スケールの中でプロットされた)の得られた傾きkから、式t1/2=ln2/−kを用いて、半減期を計算した。
図8は、マウスにおける静脈内ボーラス注射後のCOVA420(配列番号163および164)の血清中濃度を示している。排出相(β相、時間点24h〜168h)から決定されるCOVA420(配列番号163および164)の半減期値は、140時間であった。半減期がマウスにおける他のヒト抗体で得られる半減期に匹敵するため、この知見からCOVA420(配列番号163および164)は、IgG様in vivo PK特性を有することが示される(たとえば、アダリムマブ:102〜193時間、Humira(登録商標)Drug Approval Package(Drug Approval Package、Humira(登録商標)、FDA Application No.:125057s0110,Pharmacology Review,01/18/2008)。
HER2発現ヒトSKOV−3腫瘍異種移植片を有する照射NOD.CB17 PrkdcマウスにおいてCOVA420の抗腫瘍活性を調べた。
3×106SKOV−3(ATCCカタログ番号HTB−77(商標))細胞を2Gy照射NOD.CB17 Prkdcマウス(Charles River)に皮下注射(s.c.)した。ヒトT細胞注射の前日、腫瘍が約50mm3の平均サイズに達した時、動物に抗アシアロGM1ウサギ抗体(WAKO,Germany)の単回静脈内(i.v.)ボーラス注射を施した。1名の健常ドナーのバフィーコートから単離されたin vitro活性化拡大(22日間)ヒトT細胞(Miltenyi Biotech,Germany)を腹膜腔注射した(1.6×107/マウス)。T細胞注射の3日後、マウスをランダム化し、合計15日間にわたり側方尾静脈に静脈内ボーラス注射を行うことにより、0.5mg/kg COVA420(配列番号163および164、n=8)、媒体(PBS、n=7)処理、2回/週(6、9、13、15日目)、または1日1回等モル用量(=0.16mg/kg)のCOVA446(配列番号167、n=8)に施した。腫瘍成長阻害により治療有効性を評価した。腫瘍サイズは、外側カリパス測定および標準的半楕円式:体積=(幅)2×長さ×0.5を用いて計算される体積により測定した。6日目(療法開始)に対する相対腫瘍体積(RTV)を平均±SEMとして表す。GraphPad Prism 6ソフトウェア、バージョン6aを用いて統計解析を行った。対応のないノンパラメトリックt検定(マン・ホイットニー)を用いて、抗腫瘍有効性の統計的有意性を計算した。処理対媒体対照比(T/C):T/C(%)=RTV(16日目)/RTV(6日目)として表される、媒体対照と対比した腫瘍体積阻害として、16日目のCOVA420対媒体処理の抗腫瘍有効性をさらに評価した(Wu(2010),Journal of Biopharmaceutical Statistics,20:5,954−964)。
COVA420は、T細胞を腫瘍に積極的に動員することにより、SKOV−3腫瘍成長を有意に低減する(図9)。COVA420処理では、0.5mg/kg COVA420を4回投与した後、16日目、媒体対照と比較して、有意な成長阻害をもたらした(P=0.0059)。COVA420処理ではまた、1日1回注射0.16mg/kg COVA446対照と比較しても、有意により有効であった(P=0.04)。同一日でのT/C比は、媒体処理と比較して、COVA420の成長阻害では55%およびCOVA446では79%に等しい。COVA420は、薬理学的に活性であり、ヒトT細胞を効率的に腫瘍に動員することにより、その抗腫瘍活性を発揮し、その結果、媒体処理対照と比較して腫瘍成長阻害をもたらすことが、結果からが実証される。
Jaeger et al(2009)Cancer Research,69 10):4270−6から採用したプロトコルを用いて、リダイレクトT細胞媒介細胞傷害性アッセイで、ポリペプチドCOVA420(配列番号163および164)および二重特異的scFv対照COVA446(配列番号167)を試験した。簡潔に述べると、実施例1.3に記載したように、前日に、新鮮なバフィーコートからヒトPBMCを単離し、実験日にCD8+T細胞を単離した。実験の前夜、Accutase処理により適切な数の標的細胞を取り出し、96ウェルプレート中の10%FCSが追加された150μlの適切な成長培地中に5000細胞/ウェルの細胞密度で標的細胞を接種した。37℃および5%CO2でアッセイプレートを一晩インキュベートした。使用した標的腫瘍細胞は、高レベルのHER2(約1.7×106HER2分子/細胞)を発現するSKOV−3(ATCC(登録商標)HTB−77(商標))、および低レベルのHER2(約1×104HER2分子/細胞)を発現するMCF−7(ATCC(登録商標)HTB−22(商標))であった。製造業者の推奨に従ってQifikit(Dako K0078)を用いて、HER2表面発現を定量した。簡潔に述べると、飽和濃度の抗HER2抗体(R&D MAB1129)またはアイソタイプが一致した対照IgG、続いて、抗マウスFITC二次抗体で細胞を染色し、そしてフローサイトメトリー分析に付した。同時に、さまざまな明確に規定された量のマウスモノクローナル抗体分子で被覆されたビーズを二次抗体で染色し、分析し、分子/細胞の参照として標準曲線をプロットした。

実施例2.1.− 抗CD3抗体抗EGFR Fynomer融合タンパク質の発現および精製
Grabulovskiら(Grabulovski et al.(2007)JBC,282,p.3196−3204)に記載されるように、得られる構築物がC末端myc−ヘキサヒスチジンタグを有するように、配列番号169および187〜197に示されるポリペプチドをコードするDNAを細菌発現ベクターpQE12中にクローニングした。200μlスケールの培養でE.コリ(E.coli)細菌のサイトゾル中でポリペプチドを発現させた。10μg/mlの抗mycマウス抗体9E10(Roche)を含有するPBS/1%FCS/0.2%アジ化ナトリウム緩衝液中に、ポリペプチドを含有する透明化溶解液を5:1に希釈し、1×105MDA−MB−468細胞(ATCC(登録商標)HTB−132(商標))に添加した。氷上で60分間インキュベートした後、細胞を洗浄し、結合された配列を抗マウスIgG−Alexa488コンジュゲート(Invitrogen)により検出した。次いで、染色された細胞をFACSアナライザーで分析した。DNA配列決定により特異的バインダーのDNA配列を検証した。FynSH3由来EGFRバインダーのアミノ酸配列は、配列リスト中に添付されるように配列番号169および187〜197で示される。
本発明に係る抗体Fynomer融合タンパク質および対照抗体は、単一工程で発現および精製を行うことが可能であった。表1には、CHO細胞への一過性トランスフェクションおよび37℃で6日間のタンパク質発現を行った後の精製収量がまとめられている。
COVA489、COVA493、COVA494、COVA497、COVA499、およびCOVA445を、PBS/1%BSA/0.2%アジ化ナトリウム中で、(i)30nMの濃度で、100μlの全体積で、1×105Jurkat E6−1細胞(CD3陽性細胞、ATCC(登録商標)TIB−152(商標))と共に、または(ii)100nMの濃度で、100μlの全体積で、1×105EGFR過剰発現MDA−MB−468乳癌細胞(ATCC(登録商標)HTB−132(商標))と共に、インキュベートした。陰性対照として、タンパク質の代わりにPBS/1%BSA/0.2%アジ化ナトリウムのみを用いて、同一の細胞をインキュベートした(PBS対照)。
構築物はすべて、ヒトCD3を発現するJurkat E6−1細胞に結合し、かつFynomer−抗体融合体さらには抗CD3×抗EGFR scFv対照COVA445は、MDA−MB−468に結合した(図12)。予想どおり、抗CD3抗体COVA489では、MDA−MB−468に対する特異的結合は、観測されなかった。
Jaeger et al(2009)Cancer Research,69(10):4270−6から採用したプロトコルを用いて、リダイレクトT細胞媒介細胞傷害性アッセイで、ポリペプチドCOVA493、COVA494、COVA497、COVA499、二重特異的scFv対照COVA445、および抗CD3抗体(COVA489、配列番号171および172)を試験した。簡潔に述べると、実験の前夜、Accutase処理により適切な数の標的細胞を取り出し、96ウェルプレート中の10%FCSが追加された150μlの適切な成長培地中に3000〜5000細胞/ウェルの細胞密度で標的細胞を接種した。37℃および5%CO2でアッセイプレートを一晩インキュベートした。使用した標的腫瘍細胞は、高レベルのEGFR(約1.5×106EGFR分子/細胞)を発現するMDA−MB−468(ATCC(登録商標)HTB−132(商標))、および低レベルのEGFR(約5×104EGFR分子/細胞)を発現するHT−29(ATCC(登録商標)HTB−38(商標))であった。実施例1.7に記載されるようにEGFR表面発現を定量した。ただし、飽和濃度の抗EGFR抗体(Millipore MABF120)を使用した。
生存%=(値−100%溶解)/(自然溶解−100%溶解)*100
を用いて細胞生存パーセントを計算した。次いで、細胞生存%をエフェクター分子濃度に対してプロットし、Prism 5(GraphPad Software)を用いて用量−反応シグモイドに当てはめることにより、データを評価した。
高レベルのEGFRを発現するMDA−MB−468標的細胞に対するCOVA493、COVA494、COVA497、およびCOVA499の用量依存的リダイレクトT細胞傷害性を観測可能であった(図13。表5には、試験したタンパク質で得られたEC50値がまとめられている。
実施例3.1.− 抗CD3抗体抗CD33 Fynomer融合タンパク質の発現および精製
Grabulovskiら(Grabulovski et al.(2007)JBC,282,p.3196−3204)に記載されるように、得られる構築物がC末端myc−ヘキサヒスチジンタグを有するように、配列番号174および177〜186に示されるアミノ酸をコードするDNAを細菌発現ベクターpQE12中にクローニングした。200μlスケールの培養でE.コリ(E.coli)細菌のサイトゾル中でポリペプチドを発現させた。モノクローナル透明化ライセートをELISAに使用した。すなわち、ヒト組換えCD33(ヒトFcγ1への融合体としてR&Dからまで購入した)をMaxiSorpウェル(Nunc)上に固定し、PBS中の4%ミルク(Rapilait,Migros,Switzerland)でブロックした後、50μg/mlのマウス抗mycマウス抗体9E10(Roche)を含有するPBS中の12.5μlの10%ミルクと50μlの透明化ライセートとを適用した。1時間インキュベートした後、結合されていないFynomerを洗浄除去し、結合されたFynomerを抗マウスIgG−HRP抗体コンジュゲート(Sigma)で検出した。BM blue POD基質(Roche)を添加することによりペルオキシダーゼ活性の検出を行い、1M H2SO4を添加することにより反応を停止させた。ヒトIgG(Sigma)およびコーティングされていないMaxiSorpウェルに対する交差反応性の不在に関して、同一のELISAによりELISA陽性クローンを試験した。さらに、Grabulovski et al.(Grabulovski et al.(2007)JBC,282,p.3196−3204)に記載されるように、固定化金属アフィニティークロマトグラフィーカラムを用いて、配列番号174および177〜186に示されるポリペプチドをコードするDNAを細菌ライセートから精製した。23nMマウス抗myc抗体9E10を含有する100μlのPBS/1%FCS/0.2%アジ化ナトリウム中で、93nM精製Fynomerを1×105U937細胞(ATCC CRL−1593.2(商標))と共にインキュベートした。氷上で60分間インキュベートした後、細胞を洗浄し、結合された配列を抗マウスIgG−Alexa488コンジュゲート(Invitrogen)により検出した。次いで、染色された細胞をFACSアナライザーで分析した。DNA配列決定により特異的バインダーのDNA配列を検証した。FynSH3由来CD33バインダーのアミノ酸配列は、配列リスト中に添付されるように配列番号174および177〜186で示される。

CD33発現U937細胞に対するCOVA467(配列番号2および175)の用量依存的細胞傷害性を観測可能であることを実証可能であった(図15B)。この代表的なアッセイでは、COVA467(配列番号2および175)は、2.1pMのEC50値を有していた。重要なこととして、抗CD3抗体(COVA419、配列番号2および3)では細胞傷害性が観測されなかったため、細胞傷害作用は、抗CD33 Fynomerの存在に依存した。同一のアッセイ条件下で、二重特異的抗CD33×抗CD3 scFvscFv対照分子(配列番号176)では、1.6pMのEC50値が得られた。驚くべきことに、2価および全長IgGベースのFynomer−抗体融合タンパク質は、現在使用されているscFvscFvタンパク質と同一範囲内のリダイレクト死滅アッセイの効力を示すが、最適とは言えない生物物理学的性質および短いin vivo半減期という欠点を有していないことが見いだされた。
本発明は以下の態様を包含し得る。
[1] (i)CD3と、(ii)癌細胞上の表面標的抗原と、に特異的に結合する二重特異的結合分子であって、
(a)CD3に特異的に結合する抗体と、
(b)癌細胞上の表面標的抗原に結合するFyn−SH3由来ポリペプチドと、を含み、
前記Fyn−SH3由来ポリペプチドが、配列番号1のアミノ酸配列からなり、ただし、
(i)配列番号1のアミノ酸位置10〜19内の少なくとも1つのアミノ酸が、置換、欠失、または付加されていること、かつ
(ii)配列番号1のアミノ酸位置29〜36内の少なくとも1つのアミノ酸が、置換、欠失、または付加されていること、を条件とし、
前記ポリペプチドが、配列番号1の前記アミノ酸配列に対して少なくとも85%の配列同一性を有し、同一性の決定では、配列番号1のアミノ酸位置12〜17および31〜34が除外されること、を条件とする、二重特異的結合分子。
[2] CD3に特異的に結合する前記抗体が、テプリズマブ、オテリキシズマブ、Cris−7、またはSP34である、上記[1]に記載の結合分子。
[3] EGFR、HER2、CD33、CD19、CEA、EPHA2、黒色腫関連コンドロイチン硫酸プロテオグリカン、IGFR1、線維芽細胞活性化タンパク質α、前立腺幹細胞抗原(PSCA)、c−MET、EpCAM、またはPSMAに特異的に結合する2つのFyn−SH3由来ポリペプチドを含む、上記[1]または[2]に記載の結合分子。
[4] 前記結合分子が、HER2に特異的に結合する2つのFyn−SH3由来ポリペプチドを含み、前記2つのポリペプチドの第1が、前記抗体の2つの重鎖の第1のN末端に結合され、かつ前記2つのポリペプチドの第2が、前記抗体の前記2つの重鎖の第2のN末端に結合される、上記[1]〜[3]のいずれか一項に記載の結合分子。
[5] 前記結合分子が、EGFRに特異的に結合する2つのFyn−SH3由来ポリペプチドを含み、(i)前記2つのポリペプチドの第1が、前記抗体の2つの重鎖の第1のN末端に結合され、かつ前記2つのポリペプチドの第2が、前記抗体の前記2つの重鎖の第2のN末端に結合されるか、(ii)前記2つのポリペプチドの前記第1が、前記抗体の前記2つの重鎖の第1のC末端に結合され、かつ前記2つのポリペプチドの前記第2が、前記抗体の前記2つの重鎖の第2のC末端に結合されるか、(iii)前記2つのポリペプチドの前記第1が、前記抗体の2つの軽鎖の第1のN末端に結合され、かつ前記2つのポリペプチドの前記第2が、前記抗体の前記2つの軽鎖の第2のN末端に結合されるか、または(iv)前記2つのポリペプチドの前記第1が、前記抗体の前記2つの軽鎖の第1のC末端に結合され、かつ前記2つのポリペプチドの前記第2が、前記抗体の前記2つの軽鎖の第2のC末端に結合される、上記[1]〜[3]のいずれか一項に記載の結合分子。
[6] 前記結合分子が、CD33に特異的に結合する2つのFyn−SH3由来ポリペプチドを含み、前記2つのポリペプチドの第1が、前記抗体の2つの軽鎖の第1のC末端に結合され、かつ前記2つのポリペプチドの第2が、前記抗体の前記2つの軽鎖の第2のC末端に結合される、上記[1]〜[3]のいずれか一項に記載の結合分子。
[7] CD3に特異的に結合する前記抗体と前記Fyn−SH3由来ポリペプチドとがリンカーにより結合される、上記[1]〜[6]のいずれか一項に記載の結合分子。
[8] 前記リンカーがペプチドリンカーである、上記[7]に記載の結合分子。
[9] 前記リンカーが、配列番号162、160、および161からなる群から選択される配列を有する、上記[8]に記載の結合分子。
[10] 前記二重特異的結合分子が配列番号163および/または164を含む、上記[1]〜[4]、[8]、または[9]のいずれか一項に記載の結合分子。
[11] 前記二重特異的結合分子が配列番号171および/または198を含む、上記[1]〜[3]、[5]、[8]、または[9]のいずれか一項に記載の結合分子。
[12] 前記二重特異的結合分子が配列番号2および/または175を含む、上記[1]〜[3]、[6]、[8]、または[9]のいずれか一項に記載の結合分子。
[13] 上記[1]〜[12]のいずれか一項に記載の結合分子をコードする核酸分子。
[14] 上記[13]に記載の核酸分子を含むベクター。
[15] 上記[14]に記載のベクターで形質転換された宿主細胞または非ヒト宿主。
[16] 上記[1]〜[12]のいずれか一項に記載の結合分子、上記[13]に記載の核酸分子、上記[14]に記載のベクター、および/または上記[15]に記載の宿主細胞もしくは非ヒト宿主、を含む医薬組成物。
[17] 癌を治療または予防する方法における使用のための、上記[16]に記載の医薬組成物。
[18] 前記癌が、乳癌、血液学的悪性疾患、たとえば、NHL、B−ALL、AML、他の癌、たとえば、前立腺癌、黒色腫、肺癌、胃癌、卵巣癌、頭頸部癌、結腸直腸癌、膵臓癌、甲状腺癌、神経膠腫、膀胱癌、腎臓癌、肝臓癌、または悪性腹水からなる群から選択される、上記[17]に記載の使用のための医薬組成物。
[19] 前記癌が、上記[1]〜[12]のいずれか一項に記載の結合分子により特異的に結合される、上記[13]に記載の核酸分子によりコードされる結合分子により特異的に結合される、上記[14]に記載のベクターにより発現される結合分子により特異的に結合される、および/または上記[15]に記載の宿主細胞もしくは非ヒト宿主により発現される結合分子により特異的に結合される、表面標的抗原を、非腫瘍細胞上に発現される量の少なくとも2倍の量で発現する、上記[17]または[18]に記載の使用のための医薬組成物。
Claims (23)
- (i)CD3と、(ii)癌細胞上の表面標的抗原と、に特異的に結合する二重特異的結合分子であって、
(a)CD3に特異的に結合する抗体と、
(b)癌細胞上の表面標的抗原に結合するFyn−SH3由来ポリペプチドと、
を含み、
前記Fyn−SH3由来ポリペプチドが、配列番号1のアミノ酸配列からなり、ただし、
(i)配列番号1のアミノ酸位置10〜19内の少なくとも1つのアミノ酸が、置換、欠失、または付加されていること、かつ
(ii)配列番号1のアミノ酸位置29〜36内の少なくとも1つのアミノ酸が、置換、欠失、または付加されていること、
を条件とし、
前記ポリペプチドが、配列番号1の前記アミノ酸配列に対して少なくとも90%の配列同一性を有し、同一性の決定では、配列番号1のアミノ酸位置12〜17および31〜34が無視されること、を条件とする、二重特異的結合分子。 - CD3に特異的に結合する前記抗体が、Fc部分を含む、請求項1に記載の結合分子。
- EGFR、HER2、CD33、CD19、CEA、EPHA2、黒色腫関連コンドロイチン硫酸プロテオグリカン、IGFR1、線維芽細胞活性化タンパク質α、前立腺幹細胞抗原(PSCA)、c−MET、EpCAM、またはPSMAに特異的に結合する2つのFyn−SH3由来ポリペプチドを含む、請求項1または2に記載の結合分子。
- 前記Fyn−SH3由来ポリペプチドがHER2に特異的に結合し、配列番号4および9〜159からなる群から選択されるアミノ酸配列のいずれか一つを含む、請求項1〜3のいずれか一項に記載の結合分子。
- 前記Fyn−SH3由来ポリペプチドがHER2に特異的に結合し、配列番号4のアミノ酸配列を含む、請求項4に記載の結合分子。
- 前記Fyn−SH3由来ポリペプチドがEGFRに特異的に結合し、配列番号169、187〜197、および201〜203からなる群から選択されるアミノ酸配列のいずれか一つを含む、請求項1〜3のいずれか一項に記載の結合分子。
- 前記Fyn−SH3由来ポリペプチドがEGFRに特異的に結合し、配列番号169または187のアミノ酸配列を含む、請求項6に記載の結合分子。
- 前記Fyn−SH3由来ポリペプチドがCD33に特異的に結合し、配列番号174、177〜186、および207からなる群から選択されるアミノ酸配列のいずれか一つを含む、請求項1〜3のいずれか一項に記載の結合分子。
- 前記Fyn−SH3由来ポリペプチドがCD33に特異的に結合し、配列番号174のアミノ酸配列を含む、請求項8に記載の結合分子。
- 前記結合分子が、HER2に特異的に結合する2つのFyn−SH3由来ポリペプチドを含み、前記2つのポリペプチドの第1が、前記抗体の2つの重鎖の第1のN末端に結合され、かつ前記2つのポリペプチドの第2が、前記抗体の前記2つの重鎖の第2のN末端に結合される、請求項1〜5のいずれか一項に記載の結合分子。
- 前記結合分子が、EGFRに特異的に結合する2つのFyn−SH3由来ポリペプチドを含み、(i)前記2つのポリペプチドの第1が、前記抗体の2つの重鎖の第1のN末端に結合され、かつ前記2つのポリペプチドの第2が、前記抗体の前記2つの重鎖の第2のN末端に結合されるか、(ii)前記2つのポリペプチドの前記第1が、前記抗体の前記2つの重鎖の第1のC末端に結合され、かつ前記2つのポリペプチドの前記第2が、前記抗体の前記2つの重鎖の第2のC末端に結合されるか、(iii)前記2つのポリペプチドの前記第1が、前記抗体の2つの軽鎖の第1のN末端に結合され、かつ前記2つのポリペプチドの前記第2が、前記抗体の前記2つの軽鎖の第2のN末端に結合されるか、または(iv)前記2つのポリペプチドの前記第1が、前記抗体の前記2つの軽鎖の第1のC末端に結合され、かつ前記2つのポリペプチドの前記第2が、前記抗体の前記2つの軽鎖の第2のC末端に結合される、請求項1〜3、6および7のいずれか一項に記載の結合分子。
- 前記結合分子が、CD33に特異的に結合する2つのFyn−SH3由来ポリペプチドを含み、前記2つのポリペプチドの第1が、前記抗体の2つの軽鎖の第1のC末端に結合され、かつ前記2つのポリペプチドの第2が、前記抗体の前記2つの軽鎖の第2のC末端に結合される、請求項1〜3、8および9のいずれか一項に記載の結合分子。
- CD3に特異的に結合する前記抗体と前記Fyn−SH3由来ポリペプチドとがリンカーにより結合される、請求項1〜12のいずれか一項に記載の結合分子。
- 前記リンカーがペプチドリンカーである、請求項13に記載の結合分子。
- 前記ペプチドリンカーが、配列番号162、160、および161からなる群から選択される配列を有する、請求項14に記載の結合分子。
- 請求項1〜15のいずれか一項に記載の結合分子をコードする核酸分子。
- 請求項16に記載の核酸分子を含むベクター。
- 請求項17に記載のベクターで形質転換された宿主細胞または非ヒト宿主。
- 請求項1〜15のいずれか一項に記載の結合分子、請求項16に記載の核酸分子、請求項17に記載のベクター、および/または請求項18に記載の宿主細胞もしくは非ヒト宿主、を含む医薬組成物。
- 癌を治療または予防する方法における使用のための、請求項19に記載の医薬組成物。
- 前記癌が、乳癌、血液学的悪性疾患、および他の癌からなる群から選択される、請求項20に記載の使用のための医薬組成物。
- 前記血液学的悪性疾患が、NHL、B−ALL、およびAMLからなる群から選択される、請求項21に記載の使用のための医薬組成物。
- 前記他の癌が、前立腺癌、黒色腫、肺癌、胃癌、卵巣癌、頭頸部癌、結腸直腸癌、膵臓癌、甲状腺癌、神経膠腫、膀胱癌、腎臓癌、肝臓癌、および悪性腹水からなる群から選択される、請求項21または22に記載の使用のための医薬組成物。
