全てのDNA操作をSambrooket al.(Sambrook, et al., 1989)に記載される標準的な条件のもとで行った。簡潔には、テンプレートとしてH.ピロリ株J99に由来するゲノムDNAを使用するPCRによってFliD遺伝子を増幅した。以下のオリゴヌクレオチドをプライマーとして使用した:5’- CAT ATG GCA ATA GGT TCATTA A-3’(配列番号19)、及び5’- CTC GAG ATT CTT TTT AGCCGC TGC-3’(配列番号20)。このアプローチを使用して、NdeI部位をフォワードプライマーの5’末端に導入し、XhoI部位をリバースプライマーの5’末端に導入した。PCR増幅の後、生成物(2058 bp)をpTZ57R/Tクローニングベクター(リトアニアMBI FermentasのInsTAclone(商標)PCRクローニングキット)に連結した。その後、インサートをPCR及びシーケンシングにより確認し、NdeI及びXhoIの制限酵素を使用してPET-28a(+)発現ベクター(米国のQiagen)にクローニングした。
E.コリ(E. coli)BL21(米国のQiagen)コンピテント細胞をpET-28a(+)-fliDで形質転換し、抗生物質(カナマイシン、50 μg/ml)を含むLBブロスに播種した。0.6の光学密度(OD600)において1 mmol/Lのイソプロピルβ-D-1-チオガラクトピラノシド(IPTG)の添加により発現を誘導した。4時間後、細胞を回収し、ドデシル硫酸ナトリウム−ポリアクリルアミドゲル電気泳動(SDS-PAGE)により全溶解物のタンパク質分析を行った。親和性クロマトグラフィー(GEHealthcareのHisTrap crude)を使用して可溶性のヒスチジンタグ付タンパク質を精製した。第2の精製(polishing)工程として、またバッファー交換のため、サイズ排除クロマトグラフィー(GEHelthcareのSuperdex 75)を行った。関連する画分を収集し、10 kDaのカットオフを有する遠心濾過装置(Millipore)により濃縮し、-80℃で保存した。精製した組換えタンパク質を抗Hisタグ-HRP抗体、またマウス抗H.ピロリ-HRP抗体(米国ロックフォードのPierce)を使用するウェスタンブロットにより評価し、ECLシステム(スウェーデン、ウプサラのGE Healthcare)により検出した。
わずかに変更したHay et al.のプロトコル(Hay, etal., 2002)に従って、成熟した白色のニュージーランドウサギを精製したタンパク質で免疫した。簡潔には、同じ容量(0.5 ml)のフロイント完全アジュバントと共に250 μgの精製した組換えタンパク質(0.5 ml)の筋肉内注射によって免疫を行った。追加免疫のため、4週間後、6週間後、8週間後及び10週間後に同じ容量(0.5 ml)のフロイント不完全アジュバント中に調整した125 μgの精製タンパク質でウサギを追加接種した。陰性対照として、免疫の前に血清試料を採取した。最後に、最終免疫の2週間後に血液を収集し、血清を分離した。製造業者のプロトコル(Pharmacia, 1988)に従って作製したrFliD結合カラムを使用するセファロース-4BアフィニティークロマトグラフィーによってポリクローナルIgG抗体を精製した。タンパク質濃度50 μg/mlで超音波上清を使用するウェスタンブロットによりH.ピロリ(J99)のFliD発現を検出した。rFliDタンパク質に対して産生されたウサギポリクローナルIgG抗体を一次抗体(1:5000希釈)、ウサギIgGに対するHRP標識化ヒツジ抗体(イラン、テヘランのAvicenna ResearchInstitute)を二次抗体(1:3000希釈)として使用し、検出にはECLシステムを使用した(Chen, et al., 2001)。
ELISAプレートをPBS中1 μg/mlの濃度のrFliDタンパク質100μlにより被覆し、4℃で一晩インキュベートした。被覆したウェルを2.5 %のウシ血清アルブミン(SigmaのBSA)を含有するリン酸緩衝生理食塩水(PBS)により37℃で2時間に亘りブロックした。一次抗体として患者の血清の最適希釈物(1:100希釈)、二次抗体としてHRP結合抗ヒトIgG(ドイツ、マンハイムのPromega)(1:100希釈)、及び基質としてTMB(3,3',5,5'-テトラメチルベンジジン)を使用することによって、本研究で使用した全てのH.ピロリ陽性及び陰性の血清学試料をFliDに対する抗体についてスクリーニングした。さらに、各血清に対する対照としてウェルを被覆せずにおいた。患者の血清試料に対するELISAの結果は、そのOD450値が陰性血清試料の平均プラス3 SDを超えた場合に陽性とした(Chen, et al., 2001)。
ニトロセルロースに固定化した組換えH.ピロリタンパク質に基づくライン免疫アッセイを作製した。ELISAとは対照的に、上記試験の原理は、異なる単一の抗原の別々の適用によりH.ピロリの様々な抗原に対する特異的抗体の同定を可能とする。
上に規定される材料を使用して、ラテラルフローアッセイの設計に関して本明細書に開示される原理に基づきラテラルフローアッセイを開発した。
合計618名のヒト患者(男性308名、女性310名)を本研究に登録した。本研究の目的の説明を受けた後、各患者からインフォームドコンセントを得て、いかなる治療も開始する前に内視鏡検査の際に血液試料を採取した。血清を分離し、-20℃で保存した。感染の診断は、至適基準としての組織病理学的検査に基づいた。組織病理学的検査の結果が陽性であった場合に患者をH.ピロリ陽性とした。全ての患者をFliDラインアッセイによってスクリーニングし、246の血清の部分集合を上に記載されるFliD ELISA及び上に記載されるラインアッセイによって試験した。
バイオインフォマティックスの手段を用いて、H.ピロリG27株のFliDタンパク質を他の生物、主に原核生物と幅広く比較した。この分析は、200超のH.ピロリ株間で97 %超の相同性を示す。

試料
81のヒト患者に由来するH.ピロリ単離株を本研究に登録した。選択プレート上での従来の細菌培養によって上記試料を陽性と診断した。かかる試験において、微好気性条件下(10 % CO2、5% O2、8.5% N2、及び37℃)で36時間、ウイルキンス−チャルグレン血液アガープレートで細菌を生育し、オキシダーゼ、カタラーゼ、及びウレアーゼに対する陽性を生化学試験により確認した。培養細菌の一部をDNA単離に使用し、残部をウェスタンブロット分析用のタンパク質溶解物の作製にあてた。
PBSに再懸濁した、抗原として30 μgの組換えH.ピロリFliD及びアジュバントとして10 μgのCT(コレラ毒素)により3匹のC57BL6マウスを3回(週1回)免疫した。最終の免疫追加接種(immunization boost)の1週間後、マウスを出血させて血清を貯蔵した。貯蔵した血清の抗原性及び特異性をウェスタンブロット分析により試験した。
ウェスタンブロットアッセイの最適条件を確立するため、同じ条件下で作製され精製された種々の濃度の組換えFliDタンパク質、また同様に他の組換え対照タンパク質(Tig(トリガーファクター)(Tomb et al., 1997))及びgGT)を8 %SDSゲルにアプライした。ニトロセルロースメンブレンへのタンパク質のブロッティングの後(ドイツ、フライブルグのWhatman/GE Healthcare)、室温で1時間、5 %の脱脂乳でメンブレンをブロックし、一次抗体として抗血清の種々の希釈物と共に一晩インキュベートした。上記メンブレンのHRP標識化抗マウスIgGとのインキュベーションの後、ECLウェスタンブロッティング検出試薬の添加によりバンドを検出することができた。
配列番号2を対象としてFliDのDNA配列に基づいて4つのPCRを設計した。表5に示される各プライマー対の特異性を遺伝子バンクの全ての細菌ヌクレオチド配列に対するブラスト解析により確認した。H.ピロリDNAを陽性対照、10の他の微生物のゲノムDNAを陰性対照としてPCRを確立した。GoTaqポリメラーゼマスターミックス(Promega)、アニーリング温度56℃、及び30秒の伸長時間を使用してPCRを行った。

FliDのORFは全てのH.ピロリ患者分離株に存在する(患者の生検から分離された培養菌)。FliDのORFの存在は、このアッセイに使用した4つ全てのPCRによって確認することができた。81のH.ピロリ試料から単離されたDNAによって行われたPCR1、PCR2及びPCR3は、概して陽性であった。一方、PCR4は79の試料について陽性であった(図6)。P.アエルギノサ(P. aeruginosa)(ATCC 27813)、クレブシエラ・オキシトカ(Klebsiella oxytoca)(ATCC 700324)、カンジダ・アルビカンス(Candida albicans)(ATCC 90028)、エンテロコッカス・フェカリス(Entrococcus faecalis)(ATCC 29292)、A群連鎖球菌(Strep. Group A)(ATCC 19615)、S.ティフィムリウム(S. thyphimurium)(ATCC 13311)、S.アウレウス(S.aureus)(ATCC 25923)、S.エピデルミデス(S. epidermidis)(ATCC 18228)、H.インフルエンザ(H. influensae)(ATCC49247)、及びE.コリ(E.coli)(ATCC 25922)から単離されたDNAを適用することによって上記アッセイの特異性を確認した。

Claims (9)
- 被験体においてH.ピロリ感染を検出する方法であって、該方法が該被験体に由来する試料中でFliDに対する免疫応答を検出することを含み、該免疫応答が抗FliD抗体を含み、該方法は試料と全長FliD又はそのフラグメントとを反応させることを含み、該フラグメントはFliDのフラグメントとして該フラグメントを同定し、該フラグメントがFliDと異なるタンパク質又はポリペプチドのフラグメントを排除するのに十分な長いアミノ酸配列を有する、方法。
- 前記方法は試料と全長FliD又はそのフラグメントとを反応させることを含み、該フラグメントは全長FliDのアミノ酸配列よりも短いアミノ酸配列のフラグメントであり、該フラグメントは病原性因子としてなお活性がある、請求項1に記載の方法。
- 前記抗FliD抗体がIgG抗体及び/又はIgA抗体である、請求項1又は2に記載の方法。
- 前記検出がELISA、ラテラルフローアッセイ、又はラインアッセイによって生じる、請求項1〜3のいずれか一項に記載の方法。
- ヘリコバクター若しくはH.ピロリの1若しくは複数の抗原を検出することを更に含み、該1若しくは複数の抗原がCagA、VacA、GroEL、Hp 0231、JHp0940、及びHtrAを含む群から選択される、請求項1〜4のいずれか一項に記載の方法。
- 前記試料が血清、血漿、全血、及び便からなる群から選択される、請求項1〜5のいずれか一項に記載の方法。
- 請求項1〜6のいずれか一項に記載の方法における、FliD又はそのフラグメントと、少なくとも1つの更なる構成要素とを含むキットの使用であって、
該フラグメントはFliDのフラグメントとして該フラグメントを同定し、該フラグメントがFliDと異なるタンパク質又はポリペプチドのフラグメントを排除するのに十分な長いアミノ酸配列を有する、及び
該少なくとも1つの更なる構成要素は、バッファー、固相、及び使用説明書からなる群から選択される、使用。 - キットが全長FliD又はそのフラグメントを含み、該フラグメントは全長FliDのアミノ酸配列よりも短いアミノ酸配列のフラグメントであり、該フラグメントは病原性因子としてなお活性がある、請求項7に記載の使用。
- キットは全長FliD又はそのフラグメント及び少なくとも1つの更なる構成要素を含み、該フラグメントは全長FliDのアミノ酸配列よりも短いアミノ酸配列のフラグメントであり、該フラグメントは病原性因子としてなお活性がある、及び
該少なくとも1つの更なる構成要素は、バッファー、固相、及び使用説明書からなる群から選択される、請求項1〜6のいずれか一項に記載の方法の使用に適切なキット。
