Description
<実施例1−1>
バクテリオファージのスクリーニングおよび単一バクテリオファージの単離
韓国忠清道清原郡にある養鶏農場付近のニワトリの糞便から得た試料50mLを4,000rpmで10分間遠心分離した後、上清を0.45μmのフィルターで濾過して試料液を用意し、これを用いて軟寒天オーバーレイ法を行った。軟寒天オーバーレイ法は、トップアガー(0.7%の寒天を用い、固体培地上に載置したもの)上で増殖した宿主細胞を用いてバクテリオファージの溶菌を観察する方法を言う。
バクテリオファージの大規模培養と精製
実施例1−1で得たバクテリオファージを、サルモネラ・センフテンベルグ(SS)を用いて大規模に培養し、そこからバクテリオファージを精製した。
ΦCJ26の形態観察
実施例1で精製したバクテリオファージΦCJ26を0.01%のゼラチン溶液で希釈し、2.5%グルタルアルデヒド溶液で固定した。これを炭素コーティングした雲母板(約2.5mm×2.5mm))に滴下して10分間順化させた後、蒸留水で洗浄した。
ΦCJ26の全ゲノムDNAサイズ分析
実施例1で精製したバクテリオファージΦCJ26からゲノムDNAを抽出した。
ΦCJ26のタンパク質パターン分析
精製したバクテリオファージΦCJ26の溶液(1011pfu/mLタイター)15μLと5×SDS試料溶液3μLとを混合して5分間沸騰させた後、12%SDS−PAGEを行った(図3)。
ΦCJ26の遺伝的特性分析
実施例1で精製したバクテリオファージΦCJ26の遺伝子的特性を調べるため、バクテリオファージΦCJ26のDNAを遺伝子分析装置であるFLXチタニウムシーケエンサー(Roche)を用いて分析した。マクロジェン社(Macrogen Inc.)にて、GSおよびデノボアセンブリソフトウェア(Roche)を用いて遺伝子の組換えを行った。GeneMArk.hmm、Glimmer v3.02およびFGENESBソフトウェアを用いてオープンリーディングフレームを特定し、BLASTPおよびインタープロスキャン(InterProScan)プログラムを用いてオープンリーディングフレームの注釈付けを行った。
ΦCJ26のpH安定性
バクテリオファージΦCJ26が胃の中と同じような低pHにおいて安定性を保つことができるか否かを確認するため、様々なpH(pH3.0、pH4.0、pH5.0およびpH5.5)においてΦCJ26の安定性を調べた。
ΦCJ26の温度安定性
バクテリオファージを、その種々の剤形のうちの飼料添加剤として用いる場合、製剤過程において熱が発生する可能性があるため、熱に対するバクテリオファージ安定性を確認する目的で下記の実験を行った。
ΦCJ26の乾燥安定性
バクテリオファージを、その種々の剤形のうちの飼料添加剤として用いる場合、製剤過程において乾燥が行われるため、乾燥条件に対するΦCJ26の安定性を確認する目的で下記の実験を行った。
単離された野生型サルモネラ菌株に対するΦCJ26の感染範囲の検討
バクテリオファージΦCJ26の溶菌活性について、本実験で用いたサルモネラ・センフテンベルグに加えて、サルモネラ・センフテンベルグ9株、サルモネラ・モンテビデオ14株、サルモネラ・ニューポート12株、サルモネラ・ケンタッキー10株、サルモネラ・ムバンダカ13株、サルモネラ・インファンティス11株、サルモネラ・ハダール4株、サルモネラ・ダービー5株、サルモネラ・コレラエスイス4株およびサルモネラ・トムソン13株に対して試験を行った。これらはいずれも、建国大学校獣医学部の農場から入手し、単離した。
Claims (12)
- サルモネラ菌に対する特異的死滅能を有し、配列番号1ないし3のいずれか一つのDNA塩基配列を含む、寄託番号がKCCM11464PであるバクテリオファージΦCJ26。
- 請求項1に記載のバクテリオファージΦCJ26を有効成分として含む、サルモネラ菌による感染症を予防または治療するための組成物。
- 前記感染症が食中毒であることを特徴とする、請求項2に記載のサルモネラ菌による感染症を予防または治療するための組成物。
- 請求項1に記載のバクテリオファージΦCJ26を有効成分として含む、抗生剤。
- 請求項1に記載のバクテリオファージΦCJ26を有効成分として含む、飼料添加剤。
- 請求項5に記載の飼料添加剤を含む、飼料。
- 請求項1に記載のバクテリオファージΦCJ26を有効成分として含む、飲用水添加剤。
- 請求項7に記載の飲用水添加剤を含む、飲用水。
- 請求項1に記載のバクテリオファージΦCJ26を有効成分として含む、消毒剤。
- 請求項1に記載のバクテリオファージΦCJ26を有効成分として含む、洗浄剤。
- 請求項1に記載のバクテリオファージΦCJ26または請求項2に記載の組成物をヒト以外の動物に投与する工程を含む、サルモネラ菌による感染症を予防または治療するための方法。
- 前記動物が家禽であることを特徴とする、請求項11に記載のサルモネラ菌による感染症を予防または治療するための方法。
