Claims (18)
- 配列番号1のアミノ酸配列において56位のアミノ酸残基AsnをAla、Cys、MetおよびSerから選択されるアミノ酸残基で置換してなるアミノ酸配列、または配列番号7のアミノ酸配列において56位のアミノ酸残基Asnをアミノ酸残基Alaで置換してなるアミノ酸配列を有し、野生型フルクトシルアミノ酸オキシダーゼと比較して減少されたオキシダーゼ活性を有し、かつ野生型フルクトシルアミノ酸オキシダーゼと比較して増加したデヒドロゲナーゼ活性を有する変異体フルクトシルアミノ酸オキシダーゼ。
- 配列番号1のアミノ酸配列において56位のアミノ酸残基AsnをAla、Cys、Phe、Met、SerおよびValから選択されるアミノ酸残基で置換してなるアミノ酸配列、または配列番号2もしくは7のアミノ酸配列において配列番号1記載のアミノ酸配列の56位に対応する位置のアミノ酸残基Asnを、アミノ酸残基Alaで置換してなるアミノ酸配列を有し、野生型フルクトシルアミノ酸オキシダーゼの30%以下のオキシダーゼ活性を有し、かつ野生型フルクトシルアミノ酸オキシダーゼの50%以上のデヒドロゲナーゼ活性を有する変異体フルクトシルアミノ酸オキシダーゼ。
- 配列番号1のアミノ酸配列において56位のアミノ酸残基AsnをAla、Cys、MetおよびSerから選択されるアミノ酸残基で置換してなるアミノ酸配列、または配列番号7のアミノ酸配列において56位のアミノ酸残基Asnをアミノ酸残基Alaで置換してなるアミノ酸配列を有し、野生型フルクトシルアミノ酸オキシダーゼと比較して増加したデヒドロゲナーゼ活性を有する変異体フルクトシルアミノ酸オキシダーゼ。

- 試料中のHbA1cを消化してフルクトシルバリンまたはフルクトシルバリルヒスチジンを生成すること、該フルクトシルバリンまたはフルクトシルバリルヒスチジンを請求項1〜9のいずれか1項に記載のフルクトシルアミノ酸オキシダーゼと接触させること、および酸化されたフルクトシルバリンまたはフルクトシルバリルヒスチジンの量を測定することを含むHbA1cをアッセイする方法。

- 電極上に固定された請求項1〜9のいずれか1項に記載のフルクトシルアミノ酸オキシダーゼを有する酵素電極。
- フルクトシルバリン、フルクトシルバリルヒスチジン、HbA1cまたはフルクトシルヘキサペプチドをアッセイするための酵素センサーであって、請求項17記載の酵素電極を作用電極として含む酵素センサー。
