Description
TCRは、抗原提示細胞(APC)上にエピトープとして提示される特異的主要組織適合性複合体(MHC)−ペプチド複合体(「pMHC複合体」)の認識を媒介し、T細胞による該pMHCエピトープの認識を媒介する。このように、TCRは、免疫系の細胞性武器の機能化に必須である。
抗原提示細胞により提示されるpMHCエピトープに特異的に結合する抗体も公知である(例えば:Neethling FA.ら,Vaccine (2008) 26 (25):3092-102を参照)。pMHCエピトープに特異的に結合する、抗原結合性フラグメント(Fab)抗体(例えば:Chames P.ら,Proc Natl Acad Sci U S A (2000) 97 (14):7969-74;Willemsen RA.ら,J Immunol (2005) 174 (12):7853-8;Willemsen R.ら,Cytometry A (2008) 73 (11):1093-9を参照)又は単鎖抗体フラグメント(scFv)(例えば:Denkberg G.ら,J Immunol (2003) 171 (5):2197-207;Marget M.ら,Mol Immunol (2005) 42 (5):643-9を参照)が存在する。
WO 03/020763には、天然型リガンドを認識し得るように正確にリフォールディングされ、経時的に安定であり、妥当な量で作製することが可能な可溶性TCRが記載されている。このTCRは、それぞれの鎖の定常領域中に導入したシステイン同士間の鎖間ジスルフィド結合によりTCRβ鎖細胞外ドメインと二量体化したTCRα鎖細胞外ドメインを含んでなる。
当該分野におけるこの仮説に反して、本発明において、免疫エフェクター部分がpMHC結合パートナーのN末端に融合されている二機能性分子が、関連する免疫応答の誘導において、融合がpMHC結合パートナーのC末端に対してである対応する構築物より効果的であることが見出された。N融合構築物のこの増強された免疫応答は、N融合体及びC融合体のpMHC結合親和性が類似しているにも拘らず、達成される。
したがって、本発明は、所定のpMHCエピトープに特異的なポリペプチド結合パートナーと免疫エフェクターポリペプチドとを含んでなり、pMHC結合パートナーのN末端が免疫エフェクターポリペプチドのC末端と連結しており、但し、該ポリペプチド結合パートナーは配列番号7のα鎖及び配列番号9のβ鎖を含んでなるT細胞レセプターではないことを条件とする二機能性分子を提供する。
pMHC結合パートナーと免疫エフェクターポリペプチドとの連結は、直接的であってもよいし、リンカー配列を介する間接的であってもよい。リンカー配列は、可撓性を制限する可能性のある嵩高い側鎖を有さないアミノ酸、例えばグリシン、アラニン及びセリンから作製されるという点で、通常、可撓性である。リンカー配列の使用可能な又は適切な長さは、任意の所定のpMHC結合パートナー−免疫エフェクター構築物の場合で、容易に決定される。しばしば、リンカー配列は、約12未満、例えば10未満、又は5〜10アミノ酸長である。
本発明の他の実施形態において、pMHC結合パートナーは、Vα-L-Vβ、Vβ-L-Vα、Vα-Cα-L-Vβ又はVα-L-Vβ-Cβタイプ(ここで、Vα及びVβはそれぞれTCRのα及びβ可変領域であり、Cα及びCβはそれぞれTCRのα及びβ定常領域であり、Lはリンカー配列である)の単鎖αβ TCRポリペプチドである。
A1.エフェクターポリペプチドとして抗CD3抗体を有する可溶性NY-ESO TCR
本実施例の可溶性NY-ESO TCRは、HLA-A2分子上に提示されるとSLLMWITQVペプチドに結合する性質を有する。
配列番号1(図1)は、C162(配列番号1の番号付けを使用)がTRAC定常領域のT48から置換したNY-ESO TCRのα鎖のアミノ酸配列である。
配列番号2(図2)は、C170(配列番号2の番号付けを使用)がTRBC2定常領域のS57から置換したNY ESO-TCRのβ鎖のアミノ酸配列である。
配列番号3(図3)は、抗CD3 UCHT-1 scFv抗体のアミノ酸配列である(内部リンカー配列に下線を付した)。
配列番号14(図16)は、抗CD3 scFvのN末端がTCR β鎖のC末端に別のペプチドリンカー配列(下線を付した)を介して融合した図2のβ鎖のアミノ酸配列である。
配列番号15(図17)は、抗CD3 scFvのC末端がTCR β鎖のN末端に配列番号14と同じペプチドリンカー配列(下線を付す)を介して融合した図2のβ鎖のアミノ酸配列である。
(a)配列番号1のTCR α鎖と(b)配列番号2及び配列番号3の融合配列とをコードする合成遺伝子を、E.coli株BL21-DE3(pLysS)中での高レベル発現用のT7プロモーターを含有するpGMT7ベースの発現プラスミド中に別々にライゲートした(Panら,Biotechniques (2000) 29 (6):1234-8)。
この発現プラスミドをE.coli株BL21 (DE3) Rosetta pLysS中に別々に形質転換し、アンピシリン耐性の単一コロニーを37℃にてTYP(アンピシリン100μg/ml)培地中で0.6〜0.8のOD600まで増殖させた後、0.5mM IPTGでタンパク質発現を誘導した。誘導の3時間後、Beckman J-6B中4000rpmで30分間の遠心分離により細胞を採集した。MgCl2及びDNaseの存在下、25mlのBug Buster(NovaGen)で細胞ペレットを溶解させた。Beckman J2-21遠心分離器中13000rpmで30分間の遠心分離により封入体ペレットを回収した。次いで、3回の界面活性剤による洗浄を行って細胞残渣及び膜成分を除去した。各回ごとに、封入体ペレットをTriton緩衝液(50mM Tris-HCl pH8.0、0.5% Triton-X100、200mM NaCl、10mM NaEDTA)中でホモジナイズした後、Beckman J2-21中13000rpmで15分間の遠心分離によりペレット化させた。その後、下記緩衝液中での同様な洗浄により界面活性剤及び塩を除去した:50mM Tris-HCl pH8.0、1mM NaEDTA。最後に、封入体を30mgのアリコートに分け、−70℃で凍結させた。
約20mgのTCR α鎖及び約40mgのscFv−TCR β鎖の可溶化封入体を凍結ストックから解凍し、20mlのグアニジン溶液(6Mグアニジン-塩酸、50mM Tris HCl pH8.1、100mM NaCl、10mM EDTA、20mM DTT)中に希釈し、完全な鎖変性を確実にするため、37℃の水浴で30分〜1時間インキュベートした。次いで、完全に還元し変性したTCR鎖を含有するグアニジン溶液を、下記のリフォールディング緩衝液1リットルに注入した:100mM Tris pH8.1、400mM L-アルギニン、2mM EDTA、5M尿素。レドックスカップル(システアミン塩酸塩及びシスタミン二塩酸塩(それぞれ16mM及び1.8mMの最終濃度まで))を添加して約5分後、変性TCR α及びscFv−TCR β鎖を添加した。この溶液を〜30分間放置した。リフォールディングしたscFv−TCRを、透析チューブセルロース膜(Sigma-Aldrich;製品番号D9402)中10LのH2Oに対して18〜20時間透析した。この期間の後、透析緩衝液を新鮮な10mM Tris pH8.1(10L)に2回交換し、透析を5℃±3℃で更に〜8時間続けた。可溶性で正確にフォールディングしたscFv−TCRを、ミスフォールディング体、分解産物及び不純物から下記の3工程精製法により分離した。第2精製工程は、可溶性抗CD3 scFv−TCR融合体のpIに依存して、イオン交換クロマトグラフィー又はアフィニティークロマトグラフィーのいずれかであり得る。
透析したリフォールディング体(10mM Tris pH8.1中)をPOROS 50HQアニオン交換カラムにロードし、Akta精製装置(GE Healthcare)を用いて6カラム容量にわたる0〜500mM NaClのグラジエントで結合タンパク質を溶出させた。ピーク画分(導電率〜20mS/cmで溶出)を4℃で保存した。ピーク画分を、Instant Blue Stain(Novexin)染色SDS-PAGEにより分析した後、プールした。
イオン交換クロマトグラフィー
カチオン交換精製:
プールしたアニオン交換画分を、scFv−TCR融合体のpIに依存して、20mM MES pH6〜6.5での希釈により緩衝液交換した。希釈したプール画分(20mM MES pH6〜6.5中)をPOROS 50HSカチオン交換カラムにロードし、Akta精製装置(GE Healthcare)を用いて6カラム容量にわたる0〜500mM NaClのグラジエントで結合タンパク質を溶出させることにより、可溶性で正確にフォールディングしたscFv−TCRを、ミスフォールディング体、分解産物及び不純物から分離した。ピーク画分(導電率〜10mS/cmで溶出)を4℃で保存した。
プールしたアニオン交換画分を、10mM NaH2PO4 pH6.0での希釈により緩衝液交換した。希釈したプール画分(10mM NaH2PO4 pH6.0中)をヒドロキシアパタイトカラムにロードし、Akta精製装置(GE Healthcare)を用いて6カラム容量にわたる10〜500mM NaH2PO4/1M NaClのグラジエントで結合タンパク質を溶出させることにより、可溶性で正確にフォールディングしたscFv−TCRを、ミスフォールディング体、分解産物及び不純物から分離した。ピーク画分(導電率〜20mS/cmで溶出)を4℃で保存した。
6〜6.5に近いpIを有する幾つかのscFv−TCR融合体に対してはイオン交換工程を使用することはできないが、アフィニティークロマトグラフィー工程で置き換えることができる。プロテインLアフィニティークロマトグラフィーカラム(Pierce、製品番号89928)は、κ軽鎖を介して或る種の免疫グロブリンクラスを単離及び精製する。プロテインLはまた、単鎖可変フラグメント(scFv)に結合することができる。プールしたアニオン交換画分を、PBS/0.02%アジ化ナトリウムでの希釈により緩衝液交換した。希釈したプール画分をプロテインLカラムにロードし、Akta精製装置(GE Healthcare)を用いて3カラム容量にわたる0〜25mMグリシン(pH2.3)/0.02%アジ化ナトリウムのグラジエントで結合タンパク質を溶出させることにより、可溶性で正確にフォールディングしたscFv−TCRをミスフォールディング体、分解産物及び不純物から分離した。scFv−TCRはグラジエント中で非常に遅く溶出した。Tris pH8.1(100mM Tris pH8.1最終濃度)の添加により溶出画分のpHを中和した。ピーク画分を4℃で保存した。
第2精製工程からのピーク画分をInstant Blue Stain(Novexin)染色SDS-PAGEにより分析した後にプールした。次いで、プール画分を最終精製工程用に濃縮し、PBS緩衝液(Sigma)中で予め平衡化したSuperdex S200ゲル濾過カラム(GE Healthcare)を用いて可溶性scFv−TCRを精製し、特徴付けた。約78kDaの相対分子量で溶出するピークを、Instant Blue Stain(Novexin)染色SDS-PAGEにより分析した後、プールした。
図5は、配列番号1のα鎖及び配列番号2のβ鎖を有し、配列番号2のTCR β鎖のN末端にリンカー配列L2(すなわち、AHHSEDPSSKAPKAP(配列番号5)を介して融合した配列番号3の抗CD3 UCHT-1 scFv抗体を有する可溶性NY-ESO αβ TCRの構造をブロック図の形態で示す。
図6は、配列番号1のα鎖及び配列番号2のβ鎖を有し、配列番号2のTCR β鎖のN末端にリンカー配列L3(すなわち、GGEGGGSEGGGS(配列番号6)を介して融合した配列番号3の抗CD3 UCHT-1 scFv抗体を有する可溶性NY-ESO αβ TCRの構造をブロック図の形態で示す。
図4、5、6及び7の構築物について記載したものと同様の様式で、配列番号1のTCR α鎖及び配列番号14のTCR β鎖−抗CD3 scFvを有し、抗CD3 scFvがTCR β鎖のC末端に融合した融合タンパク質、又は配列番号1のTCR α鎖及び配列番号15のTCR β鎖−抗CD3 scFvを有し、抗CD3 scFvがTCR β鎖のN末端に融合した融合タンパク質を作製した。
配列番号7(図8)は、マウスH-2Kd複合体により提示されるマウスインスリン由来ペプチドLYLVCGERG(配列番号8)(LYLVCGERG−H-2Kd)に結合特性を有するTCRのα鎖のアミノ酸配列(C158(配列番号7の番号付けを使用)がTRAC定常領域のT48から置換している)である。
配列番号9(図9)は、マウスLYLVCGERG−H-2Kd複合体に結合する同じTCRのβ鎖のアミノ酸配列(C171(配列番号9の番号付けを使用)がTRBC2定常領域のS57から置換している)である。
配列番号7及び9のTCRは、各々がマウス可変領域及びヒト定常領域を含んでなるα及びβ TCR鎖からなるキメラTCRである。キメラ形のTCRは、完全なマウスTCRで遭遇したリフォールディングの問題を改善するために構築した。このキメラTCRは、マウスインスリン由来ペプチド−マウスH-2Kd複合体についてマウスTCRと同じ親和性を有することが示された。
配列番号11(図11)はマウスIL-13 ポリペプチドのアミノ酸配列である。
図12は、配列番号7のα鎖及び配列番号9のβ鎖を有し、配列番号9のTCR β鎖のN末端にリンカー配列L5(すなわち、GGEGGGP;配列番号12)を介して融合した配列番号10のマウスIL-4を有する可溶性キメラインスリンαβ TCRの構造をブロック図の形態で示す。
図13は、配列番号7のα鎖及び配列番号9のβ鎖を有し、配列番号9のTCR β鎖のC末端にリンカー配列L6(すなわちGSGGP;配列番号13)を介して融合した配列番号10のマウスIL-4を有する可溶性キメラインスリンαβ TCRの構造をブロック図の形態で示す。
図15は、配列番号7のα鎖及び配列番号9のβ鎖を有し、配列番号9のTCR β鎖のC末端にリンカー配列L6(すなわちGSGGP;配列番号13)を介して融合した配列番号11のマウスIL-13を有する可溶性キメラインスリンαβ TCRの構造をブロック図の形態で示す。
図12〜15の構築物は下記のとおりに作製した。

TCR α鎖及びサイトカイン−β鎖をそれぞれ含有する発現プラスミドをE.coli株BL21 (DE3) Rosetta pLysS中に別々に形質転換し、アンピシリン抵抗性の単一コロニーを37℃にてTYP(アンピシリン100μg/ml)培地中で0.6〜0.8のOD600まで増殖させた後、0.5mM IPTGでタンパク質発現を誘導した。誘導の3時間後、Beckman J-6B中4000rpmで30分間の遠心分離により細胞を採集した。MgCl2及びDNaseの存在下、25mlのBug Buster(NovaGen)で細胞ペレットを溶解させた。Beckman J2-21遠心分離器中13000rpmで30分間の遠心分離により封入体ペレットを回収した。次いで、3回の界面活性剤による洗浄を行って細胞残渣及び膜成分を除去した。各回ごとに、封入体ペレットをTriton緩衝液(50mM Tris-HCl pH8.0、0.5% Triton-X100、200mM NaCl、10mM NaEDTA)中でホモジナイズした後、Beckman J2-21中13000rpmで15分間の遠心分離によりペレット化させた。その後、下記緩衝液中での同様な洗浄により界面活性剤及び塩を除去した:50mM Tris-HCl pH8.0、1mM NaEDTA。最後に、封入体を30mgのアリコートに分け、−70℃で凍結させた。6Mグアニジン-HClで可溶化して封入体タンパク質の収量を定量し、OD測定をHitachi U-2001分光光度計で行った。次いで、理論的消光係数を用いてタンパク質濃度を算出した。
約20mgのTCR α鎖及び約40mgのサイトカイン−TCR β鎖の可溶化封入体を凍結ストックから解凍し、20mlのグアニジン溶液(6Mグアニジン-塩酸、50mM Tris HCl pH8.1、100mM NaCl、10mM EDTA、10mM DTT)中に希釈し、完全な鎖変性を確実にするため、37℃の水浴で30分〜1時間インキュベートした。次いで、完全に還元し変性したTCR鎖を含有するグアニジン溶液を、下記の冷(5〜10℃)リフォールディング緩衝液1リットルに注入した:100mM Tris pH8.1、400mM L-アルギニン、2mM EDTA、5M尿素。レドックスカップル(システアミン塩酸塩及びシスタミン二塩酸塩(それぞれ10mM及び2.5mMの最終濃度まで))を添加して約5分後、変性TCR α及びサイトカイン−TCR β鎖を添加した。この溶液を〜30分間放置した。リフォールディングしたサイトカイン−TCRを、透析チューブセルロース膜(Sigma-Aldrich;製品番号D9402)中10LのH2Oに対して18〜20時間透析した。この期間の後、透析緩衝液を新鮮な10mM Tris pH8.1(10L)に2回交換し、透析を5℃±3℃で更に〜8時間続けた。
下記のとおり、室温で、可溶性サイトカイン−TCR融合体を分解産物及び不純物から3工程精製法により分離した。
透析したリフォールディング体を、カラム精製の前に、Sartopore 0.2μmカプセル(Sartorius)を用いて濾過した。濾過したリフォールディング体をPOROS 50HQアニオン交換カラムにロードし、Akta精製装置(GE Healthcare)を用いて6カラム容量にわたる0〜500mM NaClの線形グラジエントで結合タンパク質を溶出させた。250mM NaClで溶出し、正確にフォールディングしたタンパク質からなるピーク画分を4℃で保存した。ピーク画分をInstant Blue Stain(Novexin)染色SDS-PAGEにより分析した後、プールした。
可溶性サイトカイン−TCRを含有するプール画分を等容量の50mM Tris/1M (NH4)2SO4 pH8と混合して、最終濃度0.5Mの(NH4)2SO4及び室温の導電率75〜80mS/cmを得た。このサンプルを予め平衡化した(50mM Tris/0.5M (NH4)2SO4 pH8)ブチル疎水性相互作用カラム(5ml HiTrap GE Healthcare)にロードし、Akta精製装置(GE Healthcare)を用いてフロースルーを採集することにより、可溶性サイトカイン−TCRを分解産物及び不純物から分離した。可溶性サイトカイン−TCRを含有するフロースルーサンプルをInstant Blue Stain(Novexin)染色SDS-PAGEにより分析した後、プールし、4℃で保存した。
プール画分を等容量の10mM Tris pH8で希釈し、10ml(濃度≦3mg/ml)に濃縮した。可溶性サイトカイン−TCRを、PBS緩衝液(Sigma)中で予め平衡化したSuperdex S200ゲル濾過カラム(GE Healthcare)を用いて精製した。約63kDaの相対分子量で溶出するピークを、Instant Blue Stain(Novexin)染色SDS-PAGEにより分析した後、プールした。
B1.エフェクターポリペプチドとして抗CD3抗体を有する可溶性NY-ESO TCR
a.NY-ESO ペプチド提示細胞に対する抗CD3抗体融合可溶性NY-ESO TCRによるCD8+ T細胞の再指向化及び活性化
特異的ペプチド−MHC複合体を介する抗CD3 scFv−TCR融合体による細胞傷害性Tリンパ球(CTL)の活性化を証明するために、下記のアッセイを行った。IFN-γ産生(ELISPOTアッセイを用いて測定)を、細胞傷害性Tリンパ球(CTL)の活性化及び該融合体の抗CD3 scFv部分の効力の評価に関する読取値(read-out)として使用した。
アッセイ培地:10% FCS(Gibco, Cat# 2011-09)、88% RPMI 1640(Gibco, Cat# 42401 )、1%グルタミン(Gibco Cat# 25030)及び1%ペニシリン/ストレプトマイシン(Gibco, Cat# 15070-063)。
ペプチド(SLLMWITQV):最初に4mg/mlでDMSO(Sigma, cat# D2650)中に溶解し、凍結させた。T2細胞を記載のペプチドでパルスし、標的細胞として使用した。
洗浄緩衝液:0.01M PBS/0.05% Tween 20
PBS(Gibco Cat# 10010)
ヒトIFNγ ELISPOT PVDF-Enzymatic kit(Diaclone, France;Cat#856.051.020)は、必要な他の全ての試薬を含有する(捕捉及び検出抗体、スキムミルク粉、BSA、ストレプトアビジン-アルカリホスファターゼ及びBCIP/NBT溶液並びにヒトIFN-γ PVDF ELISPOT 96ウェルプレート)。
標的細胞の調製
本方法で使用した標的細胞は、(1)天然エピトープ提示細胞(例えばMel624又はMel526細胞)又は(2)(試薬の項で記載した)目的のペプチドでパルスしたT2細胞のいずれかであった。十分な標的細胞(50 000細胞/ウェル)を、Megafuge 1.0(Heraeus)中1200rpmで10分間の3回の遠心分離により洗浄した。次いで、細胞をアッセイ培地に106細胞/mlで再懸濁した。
本方法で使用したエフェクター細胞(T細胞)は、CD8+ T細胞((CD8 Negative Isolation Kit, Dynal, Cat# 113.19を用いる)ネガティブ選択によりPBLから取得)、EBV株又はPBMCからのT細胞のいずれかであった。エフェクター細胞を解凍し、アッセイ培地中に配置した後、Megafuge 1.0(Heraeus)中1200rpmで10分間の遠心分離により洗浄した。次いで、細胞をアッセイ培地中に4×最終必要濃度で再懸濁した。
アッセイ培地中に希釈して4×最終濃度を得ることにより、種々の濃度の試験化合物(TCR−抗CD3融合体;10nM〜0.03pM)を調製した。
プレートを下記のように調製した:100μlの抗IFN-γ捕捉抗体をプレートあたり10mlの滅菌PBS中に希釈した。次いで、100μlの希釈捕捉抗体を各ウェル中に小分けした。次いで、プレートを一晩4℃にてインキュベートした。インキュベーション後、プレートを洗浄(プログラム1、プレートタイプ2、Ultrawash Plus 96ウェルプレートウォッシャー;Dynex)して捕捉抗体を除去した。次いで、滅菌PBS中2%スキムミルク100μlを各ウェルに添加し、プレートを室温にて2時間インキュベートすることにより、プレートをブロックした。次いで、スキムミルクをプレートから洗い流し(プログラム1、プレートタイプ2、Ultrawash Plus 96ウェルプレートウォッシャー, Dynex)、ELISPOTプレートをペーパータオル上で軽く叩くことにより、残った洗浄緩衝液を除去した。
次いで、アッセイの構成要素をELISPOTプレートに下記の順序で添加した:
50μlの標的細胞 106細胞/ml(合計50 000標的細胞/ウェルを得る)
50μlの試薬(抗CD3 scFv−TCR融合体;種々の濃度)
50μlの培地(アッセイ培地)
50μlのエフェクター細胞(1000〜50000 CD8+細胞/ウェル;500〜1000 EBV細胞/ウェル;1000〜50000 PBMC/ウェル)。
図4〜7の抗CD3 scFv−TCR融合構築物をELISPOTアッセイ(上記)により試験した。Prism(Graph Pad)を使用して、各ウェル中で観察されたELISPOTスポットの数を、試験構築物の濃度に対してプロットした。
これら用量-応答曲線から、EC50値を決定した(EC50は、最大応答の50%を誘導する抗CD3 scFv−TCR融合体の濃度で決定される)。
特異的ペプチド−MHC複合体を介するTCR−抗CD3 scFv融合体による細胞傷害性Tリンパ球(CTL)の活性化を証明し、CTLを活性化してIM9細胞を殺傷させる該融合体の抗CD3 scFv部分の効力を評価するために、下記のアッセイを行った。このアッセイは、51Cr放出細胞毒性アッセイに代替する比色アッセイであり、細胞溶解に際して放出される酵素である酪酸デヒドロゲナーゼ(LDH)を定量的に測定する。培養上清中の放出されたLDHは、テトラゾリン塩(INT)の赤色ホルマザン産物への変換を生じる、30分間のカップリング酵素アッセイで測定する。形成した色量は、溶解した細胞数に比例する。吸光度データは、標準の96ウェルプレートリーダーを用いて490nmで収集する。
− CytoTox96(登録商標)非放射性細胞毒性アッセイ(Promega)(G1780)は、基質ミックス、アッセイ緩衝液、溶解溶液及び停止溶液を含有する。
− アッセイ培地:10% FCS(熱不活化, Gibco, cat# 10108-165)、フェノールレッドを含まない88% RPMI 1640(Invitrogen, cat# 32404014)、1%グルタミン、200mM(Invitrogen, cat# 25030024)、1%ペニシリン/ストレプトマイシン(Invitrogen cat# 15070063)。
− Nuncマイクロウェル丸底96ウェル組織培養プレート(Nunc, cat# 163320)
− Nunc-Immuno plates Maxisorb(Nunc, cat# 442404)
標的細胞の調製
本アッセイで使用した標的細胞は、多発性骨髄腫患者に由来するIM9 EBV形質転換B細胞株(HLA-A2+ NY-ESO+)であった。Mel526メラノーマ細胞株をコントロールとして使用し。この細胞株はHLA-A2+ NY-ESO-である。標的細胞はアッセイ培地中に調製した:標的細胞濃度は、50μl中1×104細胞/ウェルとなるように2×105細胞/mlに調整した。
本アッセイで使用したエフェクター細胞はCD8+ T細胞であった。使用したエフェクター 対 標的比は10:1(50μl中1×105細胞/ウェルとなるように2×106細胞/ml)であった。
種々の濃度のNY-ESO TCR−抗CD3融合体(配列番号1のTCR α鎖及び配列番号14のTCR β鎖−抗CD3 scFv融合体を有するか、又は配列番号1のTCR α鎖及び配列番号15のTCR β鎖−抗CD3 scFv融合体を有する)は、実施例A1に記載のように調製し、アッセイ培地中への希釈(10-13〜10-8M最終濃度)によって本アッセイ用に調製した。
アッセイの構成要素をプレートに下記の順序で添加した:
− 各ウェルに、50μlの標的細胞IM9又はMel526(上記で説明したとおり調製)
− 各ウェル、50μlの試薬(上記で説明したとおり調製)
− 各ウェル、50μlのエフェクター細胞(上記で説明したとおり調製)
幾つかのコントロールを下記に説明するとおり調製した:
− エフェクター自発放出:50μlのエフェクター細胞単独
− 標的細胞自発放出:50μlの標的細胞単独
− 標的最大放出:50μlの標的細胞+80μg/mlのジギトニン(細胞を溶解させるためアッセイの開始時)
− アッセイ培地コントロール:150μlの培地単独
実験ウェルは3連(in triplicate)で、コントロールウェルは2連(in duplicate)で、150μlの最終容量で準備する。
プレートを250×gにて4分間遠心分離した。アッセイプレートの各ウェルからの37.5μlの上清を、平底96ウェルNunc Maxisorbプレートの対応するウェルに移した。基質ミックスを、アッセイ緩衝液(12ml)を使用して再構成した。次いで、37.5μlの再構成基質ミックスをプレートの各ウェルに添加した。プレートをアルミニウムホイルで覆い、室温にて30分間インキュベートした。37.5μlの停止溶液をプレートの各ウェルに添加して反応を停止させた。490nmでの吸光度を、停止溶液の添加後1時間以内にELISAプレートリーダーで記録した。
培養培地のバックグランド吸光値の平均を、実験、標的細胞自発放出及びエフェクター細胞自発放出並びに標的最大放出の全ての吸光値から減算した。
最初の2工程で得られた補正値を下記式で使用して、パーセント細胞毒性を計算した:
%細胞毒性=100×(実験−エフェクター自発−標的自発)/(標的最大放出−標的自発)
(i)配列番号1のTCR α鎖及び配列番号14のTCR β鎖−抗CD3 scFv融合体(C末端融合体)又は(ii)配列番号1のTCR α鎖及び配列番号15のTCR β鎖−抗CD3 scFv融合体(N末端融合体)を有するNY-ESO TCR−抗CD3 scFv融合構築物を、LDH放出アッセイ(上記)により試験した。Prism(Graph Pad)を使用して、各ウェルで観察された%細胞毒性を試験構築物の濃度に対してプロットした。これら用量-応答曲線からEC50値を決定した(EC50は、最大応答の50%を誘導するTCR融合体の濃度で決定する)。
a.エフェクターポリペプチドとしてのマウスIL-4サイトカイン
下記アッセイを使用して、図12〜13のマウスIL-4−TCR融合構築物のサイトカイン部分の生物学的活性を試験した。これは、マウス細胞株CTLL-2を使用するバイオアッセイである。この細胞株は、増殖に関してマウスIL-4に依存性であり、マウスIL-4−TCR融合体のサイトカイン部分の生物学的活性を証明するために本明細書で使用される。
CTLL-2細胞、Promega CellTiter-Glo(登録商標)発光細胞生存アッセイ(Cat# G7572)(CellTiter-Glo(登録商標)緩衝液及びCellTiter-Glo(登録商標)基質(凍結乾燥)を含む)
アッセイ培地:10%熱不活化胎仔ウシ血清を補充したRPMI(Gibco, cat# 10108-165)、88% RPMI 1640(Gibco, cat# 42401-018)、1%グルタミン(Gibco, cat# 25030-024)、1%ペニシリン/ストレプトマイシン(Gibco, cat# 15070-063)
下記アッセイを使用して、図14〜15のマウスIL-13−TCR融合構築物のサイトカイン部分の生物学的活性を試験した。
このアッセイは、サイトカイン−TCR融合体からのサイトカイン部分の活性、すなわちヒト単球によるIL-1β産生の阻害を証明するために行った。このアッセイを使用して、サイトカインがマウスIL-13であるサイトカイン−TCR融合体を試験することができる。
バフィーコート由来の単球(NBS Bristol Transfusion Serviceのバフィーコート)
未処理(untouched)ヒト細胞用のDynal Dynabeads MyPure Monocyte Kit 2(113.35)
アッセイ培地:10%胎仔ウシ血清(熱不活化, Gibco, cat# 10108-165)、88% RPMI 1640(Gibco, cat# 42401-018)、1%グルタミン(Gibco, cat# 25030-024)、1%ペニシリン/ストレプトマイシン(Gibco, cat# 15070-063)
洗浄緩衝液:0.01M PBS/0.05% Tween 20(1リットルの蒸留水中に溶解した1袋のTween 20(pH7.4)含有リン酸緩衝化生理食塩水(Sigma, cat# P-3563)により最終組成0.01M PBS、0.138M NaCl、0.0027M KCl、0.05% Tween 20が得られる)。
PBS(Gibco, cat# 10010-015).
HBSS Ca2+及びMg2+フリー(Gibco, cat# 1018-165)
Cytokine Eli-pair ELISAキット:IL-1β(Diaclone cat# DC-851.610.020)。これらキットは、必要な他の全ての試薬、すなわち捕捉抗体、検出ビオチン化抗体、ストレプトアビジン-HRP、IL-1β標準物、即時使用可能(ready-to-use)TMBを含有する。下記の方法は、各キットで供給される指示書に基づく。

BSA(Sigma, cat# A3059)
H2SO4(Sigma cat # S1526)
トリパンブルー(Sigma cat # T8154)
E.coli 0111:B4由来リポ多糖(LPS)(Sigma, cat# L4391)
組換えマウスIL-13(Peprotech, cat# 210-13)。マウスIL-13−TCR融合体試薬を試験するときに使用した標準物。
バフィーコートからPBMCを単離した:HBSS(Ca2+及びMg2+フリー)を用いてバフィーコートを2倍希釈し、希釈血液をlymphoprep上に積層し(15mlのlymphoprep上に35mlまでの血液)、15分間800×g(室温)にて遠心分離した(ブレーキオフ);界面の細胞を取り出し、HBSSで4回洗浄し、1200rpmで10分間遠心分離した。最終洗浄後、細胞を50mlのアッセイ培地中に再懸濁し、カウントして、トリパンブルー溶液を用いて生存を評価した。Dynal Dynabeads MyPure Monocyte Kit 2を使用して単球を単離した。PBMCをPBS/0.1% BSA中に107細胞あたり100μl緩衝液で再懸濁し、20μlのブロッキング試薬/107細胞及び20μlの抗体ミックス/107細胞を添加し、細胞を20分間4℃にてインキュベートした。細胞を洗浄し、107細胞あたり0.9mlのPBS/0.1% BSA中に再懸濁した。予め洗浄したビーズを添加し(100μl/107細胞)、混合し、穏やかに回転させながら更に15分間20℃にてインキュベートした。注意深くピペッティングすることでロゼットを再懸濁し、107細胞あたり1mlのPBS/0.1% BSAを添加した。試験管をDynalマグネット中に2分間置いた。ネガティブ単離された細胞を含有する上清を新たな試験管に移し、カウントした。細胞は直ぐに使用したか、又は更なる使用のために90% FCS/10% DMSO中で凍結させた。
ELISAプレートをPBS中のIL-1β捕捉抗体100μl/ウェルでコートし、4℃にて一晩放置した。単球を解凍し、アッセイ培地中で2回洗浄し、5×105細胞/mlで再懸濁した。単球を丸底96ウェルプレート中に播種した(100μl/ウェル、すなわち5×104/ウェル)。LPS、Peprotech組換えサイトカイン及び試験サイトカイン−TCR融合タンパク質を、4×最終濃度となるようにアッセイ培地中に希釈して調製した。LPSを各ウェルに添加し(10ng/ml最終)、続いて3連のウェルに50μlの滴定濃度(10倍系列希釈ごとに1点の6点)のPeprotech組換えIL-13(10-8〜10-13M最終)又は試験サイトカイン−TCR融合タンパク質(10-7〜10-13M最終)を添加した。プレートを37℃にて5% CO2と一晩インキュベートした。
抗体をコートしたIL-1β ELISAプレートを、洗浄緩衝液中で3回洗浄し、250μlのPBS/5% BSA/ウェルで室温にて少なくとも2時間(又は4℃にて一晩)ブロックした。ELISAプレートを洗浄緩衝液中で3回洗浄し、軽く叩いて乾燥させた。IL-1β標準物をPBS/1%BSA中に希釈した。細胞を含有するプレートを1200rpmにて5分間遠心分離した。次いで、各ウェルからの上清を、予めコートしたIL-1β ELISAプレートに移した。100μlの細胞上清(PBS/1% BSAで3倍希釈)又は標準物を該当するウェルに添加し、50μlの検出抗体/ウェル(キットの指示書に従って希釈)を添加した。プレートを室温にて2時間インキュベートした。プレートを洗浄緩衝液中で3回洗浄した。ウェルあたり100μlのストレプトアビジン-HRP(キットの指示書に従って希釈)を添加し、プレートを室温にて20分間インキュベートした。プレートを洗浄緩衝液中で3回洗浄した。ウェルあたり100μlの即時使用可能TMBを添加し、プレートを暗所(ホイル下)で5〜20分間(シグナル強度に依存)発色させた。100μl/ウェルの1M H2SO4を添加して反応を停止させた。
プレート吸光度を、450nmにてマイクロプレートリーダー及び650nmに設定した参照フィルターで読み取った。各滴定点についての阻害量は、単球及びLPSを含有し、サイトカイン−TCR融合タンパク質を含まないサンプル(最大シグナルが得られる)のパーセンテージとして算出する。こうして用量-応答曲線を作成する。
Claims (12)
- 所定のpMHCエピトープに特異的なポリペプチド結合パートナーと免疫エフェクターポリペプチドとを含んでなり、pMHC結合パートナーがへテロ二量体αβ TCRポリペプチド対又は単鎖αβ TCRポリペプチドであり、へテロ二量体TCRポリペプチド対のα若しくはβ鎖のN末端又はscTCRポリペプチドのN末端が免疫エフェクターポリペプチドのC末端アミノ酸と連結しており、但し、該ポリペプチド結合パートナーは配列番号7のα鎖及び配列番号9のβ鎖を含んでなるT細胞レセプターではないことを条件とする二機能性分子。
- pMHC結合パートナーが、α及びβポリペプチドの各々がTCRの可変領域及び定常領域を有するが、TCRの膜貫通領域及び細胞質領域を欠いているへテロ二量体αβ TCRポリペプチド対である請求項1に記載の二機能性分子。
- α及びβポリペプチドの定常領域が、TRAC1のエキソン1のThr 48及びTRBC1若しくはTRBC2のエキソン1のSer 57を置換したシステイン残基間のジスルフィド結合により又はTRAC1のエキソン2のCys4とTRBC1若しくはTRBC2のエキソン2のCys2との間の天然型ジスルフィド結合より連結している請求項2に記載の二機能性分子。
- pMHC結合パートナーが単鎖αβ TCRポリペプチドである請求項1に記載の二機能性分子。
- 免疫エフェクターポリペプチドが、T細胞により提示される抗原に特異的に結合する抗体である請求項1〜4のいずれか1項に記載の二機能性分子。
- 抗体がscFv抗体である請求項5に記載の二機能性分子。
- 抗体が抗CD3抗体である請求項5又は6に記載の二機能性分子。
- 抗体がOKT3である請求項7に記載の二機能性分子。
- 抗体がUCHT-1である請求項7に記載の二機能性分子。
- 抗体がBMA031である請求項7に記載の二機能性分子。
- 抗体が12F6である請求項7に記載の二機能性分子。
- エフェクターポリペプチドが、リンパ球の免疫抑制又は免疫刺激活性をモジュレートするサイトカインである請求項1〜4のいずれか1項に記載の二機能性分子。
