Description
背景

本发明人出人意料地发现,包含与肽-MHC结合部分融合的PD-1激动剂的分子导致对PD-1信号传导的有效抑制。
不受理论的束缚,本发明人假设对T细胞活化的有效抑制需要使PD-1激动剂定位至免疫突触。PD-1激动剂连接至与疾病特异性肽-MHC结合的部分(例如TCR或TCR样抗体),会将激动剂引导至免疫突触,从而提供了更安全且更有效的策略来调节PD-1途径。
T细胞受体(TCR)由CD4+和CD8+ T细胞自然表达。TCR旨在识别与主要组织相容性复合物(MHC)分子(在人类中,MHC分子也称为人白细胞抗原或HLA)复合的抗原呈递细胞表面上展示的短肽抗原(Davis,等人,(1998),Annu Rev Immunol 16:523-544.)。CD8+ T细胞也称为细胞毒性T细胞,特异性识别与I类MHC结合的肽,通常负责发现感染细胞或癌细胞以及介导对感染细胞或癌细胞的破坏。

发明内容
作为第一方面,本发明提供了包含pMHC结合部分和PD-1激动剂的双功能结合多肽。pMHC结合部分可以包含TCR可变结构域和/或抗体可变结构域。pMHC结合部分可以是T细胞受体(TCR)或TCR样抗体。pMHC结合部分可以是异二聚体α/βTCR多肽对或单链α/βTCR多肽。PD-1激动剂可以是PD-L1的可溶性胞外形式或其功能片段,PD-L1可以包含以下序列或由以下序列组成:FTVTVPKDLYVVEYGSNMTIECKFPVEKQLDLAALIVYWEMEDKNIIQFVHGEEDLKVQHSSYRQRARLLKDQLSLGNAALQITDVKLQDAGVYRCMISYGGADYKRITVKVNAPY。PD-1激动剂可以是全长抗体或其片段,例如scFv抗体。
PD-1激动剂可以与pMHC结合部分的C末端或N末端融合,并且可以通过接头与pMHC结合部分融合。接头的长度可以多达25个氨基酸。优选地,接头的长度为2、3、4、5、6、7或8个氨基酸。
当pMHC结合部分是TCR时,TCR可以在α链的恒定区和β链的恒定区之间包含非天然二硫键,并且可以与肽抗原特异性结合。

当pMHC结合部分是TCR时,TCR可以是非天然存在的和/或纯化和/或工程化的。相对于天然TCR,一个以上突变可以存在于α链可变结构域和/或β链可变结构域中。优选在CDR区域内进行突变。通常引入这样的突变以改善结合部分(例如TCR)对特定肽抗原HLA复合物的结合亲和力。
pMHC结合部分可以是TCR样抗体。TCR样抗体是本领域中用于被赋予了对MHC呈递的肽抗原的TCR样特异性的抗体分子的术语,并且通常TCR样抗体对抗原的亲和力高于天然TCR(Dahan等人,Expert Rev Mol Med 14:e6,2012)这些抗体可以包含重链和轻链,该重链和轻链各自包含可变区和恒定区。本发明中涵盖这些抗体的功能片段,例如如本领域所熟知的scFv、Fab片段等。

本发明的双功能结合多肽将对肽抗原和PD-1具有合适的结合亲和力。确定结合亲和力(与平衡常数KD成反比)和结合半衰期(表示为T1/2)的方法是本领域技术人员已知的。在优选的实施方式中,使用表面等离子体共振(SPR)或生物层干涉法(BLI),例如分别使用BIAcore仪器或Octet仪器来确定结合亲和力和结合半衰期。应当理解,结合多肽的亲和力加倍导致KD减半。T1/2通过ln2除以解离速率常数(koff)来计算。因此,T1/2加倍导致koff减半。通常针对多肽的可溶形式测量KD和koff值。为了考虑独立测量之间的变化,尤其对于解离时间超过20小时的相互作用,可以使用相同的测定方案测量给定多肽的结合亲和力或结合半衰期数次(例如3次或更多次),并且取结果的平均值。为了比较两个样品(即两个不同多肽和或相同多肽的两种制剂)之间的结合数据,优选使用相同的测定条件(例如温度)进行测量。

还提供了包含这样的核酸或这样的载体的宿主细胞,其中编码双功能结合多肽的核酸可以以单个开放阅读框或分别编码TCR的α链和β链的两个不同的开放阅读框存在。

实施例
实施例1
以下实施例证明,与可溶性TCR融合的PD-1激动剂在靶向免疫突触时有效抑制T细胞活化。
在该双功能结合多肽中使用的可溶性TCR是天然TCR的亲和力增强版本,该版本特异性识别源自人前胰岛素原的HLA-A*02限制性肽(WO2015092362中描述了这样的分子)。PD-1激动剂是包含PD-1相互作用位点的PD-L1胞外区的截短版本(Zak等人,Structure 23:2341-2348,2015)。PD-L1通过标准的5个氨基酸接头与TCRα链的N末端融合。
使用Jurkat NFAT荧光素酶PD-1报告物测定,在存在HEK293T抗原呈递靶细胞的情况下,测量TCR-PD1激动剂融合分子介导的对T细胞NFAT活性的抑制。
方法
TCR-PD1激动剂融合分子的表达、重折叠和纯化
使用基于悬浮适应的中国仓鼠卵巢(CHO)细胞的高产率瞬时表达系统(ExpiCHO表达系统,Thermo Fisher)进行TCR-PD1激动剂融合分子的表达。使用含有与PD-1激动剂融合的TCR链的哺乳动物表达质粒,根据制造商的说明来进行细胞共转染。收获后,通过在冷冻离心机中将上清液以4000-5000×g离心30分钟来进行细胞培养上清液的澄清。上清液通过0.22μm过滤器过滤并收集以进行进一步纯化。
可替代地,使用大肠杆菌作为宿主生物进行TCR-PD1激动剂融合分子的表达。将含有α链和β链的表达质粒分别转化到BL21pLysS大肠杆菌菌株中,并在含有100μg/mL氨苄青霉素的LB琼脂平板上铺板。从每个转化中挑选环状菌落,并在LB培养基(含100μg/mL氨苄青霉素和1%葡萄糖)中在37℃下生长,直到OD600达到约0.5至1.0。然后将LB起始培养物添加到自诱导培养基(Foremedium)中,细胞在37℃生长约3小时,然后30℃过夜。通过离心收集细胞,并在Bugbuster(Novagen)中裂解。通过进行两次Triton洗涤(50mM Tris pH 8.1、100mM NaCl,10mM EDTA、0.5%Triton)去除细胞碎片和膜,提取包涵体(IB)。每次通过以10000g离心5分钟来收获IB。为了除去去污剂,将IB用50mM Tris pH8.1、100mM NaCl和10mMEDTA洗涤。最终将IB重悬在50mM Tris pH8.1、100mM NaCl和10mM EDTA的缓冲液中。为了测量蛋白质产率,将IB溶解在8M尿素缓冲液中,并通过280nM的吸光度确定浓度。
对于重折叠,以1:1的摩尔比混合α链和β链,并在37℃下在6M胍盐酸盐、50mM TrispH8.1、100mM NaCl、10mM EDTA、20mM DTT中变性30分钟。然后将变性链添加到由4M尿素、100mM Tris pH 8.1、0.4M L-精氨酸、2mM EDTA,1mM胱胺和10mM半胱胺组成的重折叠缓冲液中,并在持续搅拌下孵育10分钟。将含有变性链的重折叠缓冲液在Spectra/Por 1膜中对10x体积的H2O透析约16小时,对10x体积的10mM Tris pH 8.1透析约7小时,并对10x体积的10mM Tris pH8.1透析约16小时。
使用POROS 50HQ(Thermo Fisher Scientific)阴离子交换柱,用20mM Tris pH8.1作为上样缓冲液且20mM Tris pH8.1与1M NaCl作为结合和洗脱缓冲液,在AKTA pure(GE Healthcare)上纯化从哺乳动物或大肠杆菌表达系统获得的可溶性蛋白。将蛋白质上样到柱子上,并用0-50%的洗脱缓冲液梯度洗脱。合并含有蛋白质的级分,并在20mM MESpH6.0中稀释20倍(体积/体积),以在POROS 50HS(Thermos Fisher Scientific)柱上、使用20mM MES pH6.0以及20mM MES pH6.0、1M NaCl分别作为结合缓冲液以及洗脱缓冲液,进行第二步阳离子交换色谱法。使用0-100%的洗脱缓冲液梯度洗脱阳离子交换柱上的结合蛋白。合并包含蛋白质的阳离子交换级分,并使用PBS作为运行缓冲液在Superdex 200HR(GEHealthcare)凝胶过滤柱上进一步纯化。来自凝胶过滤的阳性级分合并,浓缩并在-80℃下储存直至需要。
Jurkat NFAT Luc-PD-1报告物测定
将HLA-A*02阳性的HEK293T靶细胞用TCR活化剂质粒(BPS Bioscience,目录号:60610)进行瞬时转染,并用TCR-PD1激动剂融合分子识别的相关肽进行冲击。然后将靶细胞与不同浓度的TCR-PD1激动剂融合分子一起孵育,使得与同源肽-HLA-A2复合物结合。将组成型表达PD-1的Jurkat NFAT Luc PD-1效应细胞添加到靶细胞中,并在18至20小时后确定NFAT活性。在洗去或未洗去的情况下进行实验(TCR-PD1激动剂融合分子结合后)。使用未受冲击的靶细胞进行进一步的对照。包括了TCR活化剂/PD-L1转染的HEK293T A2B2M靶细胞作为阳性对照。
结果
图1所示的数据表明,在存在肽冲击的靶细胞的情况下,无论洗去或未洗去,通过TCR-PD1激动剂融合分子观察到对NFAT报告物活性的剂量依赖性抑制。至关重要的是,未受冲击的靶细胞观察到最小抑制,表明靶向免疫突触对于PD-1激动剂活性至关重要。
实施例2
以下实施例提供了与可溶性TCR融合的PD-1激动剂在靶向免疫突触时可有效抑制T细胞活化的进一步证据。
该实施例中使用的实验系统和方法与实施例1中所描述的相同,除了在该情况下,TCR-PD1激动剂融合分子的PD-1激动剂部分是scFv抗体片段,例如WO2011110621中所述。
使用实施例1中所述的Jurkat NFAT荧光素酶PD-1报告物测定,在存在HEK293T抗原呈递靶细胞的情况下,测量TCR-PD1激动剂融合分子介导的对T细胞NFAT活性的抑制。
结果
如图2a所示,在用100nM TCR-PD1激动剂融合分子处理的肽冲击的细胞(标记为+PPI)中观察到对NFAT活性的显著抑制(>60%);而在用TCR-PD1激动剂融合分子处理的未受冲击的靶细胞(标记为-PPI)中观察到最小抑制。单独使用可溶性TCR或单独使用PD-1激动剂(以scFv或IgG4形式)的对照实验均未显示报告物活性受到抑制,这表明PD-1激动剂活性需要使PD-1激动剂靶向免疫突触。图2b进一步显示了对NFAT活性的剂量依赖性抑制。同样,只有TCR-PD1激动剂融合分子形式能够抑制NFAT活性。非靶向PD-1激动剂抗体不能抑制活性。
综上所述,这些结果证明,使PD-1激动剂靶向免疫突触对于PD-1激动剂活性至关重要。
实施例3
以下实施例提供了与可溶性TCR融合的PD-1激动剂在靶向免疫突触时可有效抑制T细胞活化的进一步证据。
本实施例中使用的TCR-PD1激动剂融合分子与实施例2中描述的相同,其中PD1激动剂是scFv抗体片段。
在这种情况下,使用替代测定来评估TCR-PD1激动剂融合分子对原代人T细胞功能的影响。
方法
原代人T细胞测定
使用pan-T细胞分离试剂盒(Miltenyi,目录号:130-096-535)从新鲜制备的PBMC中分离原代人T细胞。用葡萄球菌肠毒素B(SEB,100ng/ml,Sigma S4881)预加载(pre-load)HLA-A*02阳性Raji B细胞(Raji A2B2M)1小时,然后用33Gy照射。对于预活化,将原代人T细胞与加载SEB的Raji A2B2M靶细胞以1:1的比例孵育,在24孔细胞培养板中使用1x10E6个细胞/ml的每种细胞类型。将原代人T细胞与加载SEB的Raji A2B2M细胞孵育10天,并在第3天和第7天添加IL-2(50U/ml)。在第10天,将预活化的T细胞洗涤并重悬于新鲜培养基中。将新鲜的Raji A2B2M细胞用TCR-PD1激动剂融合分子识别的20μM相关肽冲击2小时,或不进行冲击2小时。在肽冲击的最后1小时中,用SEB(10ng/ml)加载Raji A2B2M细胞,然后用33Gy照射。将Raji A2B2M细胞以1x10E5个细胞/孔在96孔细胞培养板中铺板,然后与TCR-PD1激动剂融合分子滴定液预孵育1小时。将预活化的T细胞以1x10E5细胞/孔添加到Raji A2B2M靶细胞中,并孵育48小时。收集上清液,并使用MSD ELISA测定IL-2水平。
结果
图3所示的数据表明,在存在肽冲击的靶细胞的情况下,TCR-PD1激动剂融合分子对原代人T细胞IL-2的产生进行剂量依赖性抑制,而非靶向TCR-PD1激动剂融合分子(即用未受冲击的靶细胞)或单独的PD-1激动剂scFv则不能。这些数据证明,将PD-1激动剂靶向免疫突触导致原代细胞中的PD-1激动剂活性。
实施例4
以下实施例证明,使用识别替代抗原的TCR观察到相同的技术效果。
本实施例中使用的实验系统和方法与实施例2中描述的相同。在这种情况下,PD-1激动剂抗体与两种不同的可溶性TCR融合。
使用实施例1中所述的Jurkat NFAT荧光素酶PD-1报告物测定,在存在HEK293T抗原呈递靶细胞的情况下,测量TCR-PD1激动剂融合分子介导的对T细胞NFAT活性的抑制。
结果
如图4所示,通过两种TCR-PD1激动剂融合分子(包含与TCR1或TCR2融合的PD-1激动剂scFv抗体片段),在存在用各自的肽(肽1或2)冲击的靶细胞的情况下,观察到强的剂量依赖性抑制。对于两种TCR-PD1激动剂融合分子,在不存在靶向肽的情况下进行研究时,观察到最小活性。
这些结果证明,使用对不同的pMHC具有特异性的可溶性TCR,可以将TCR-PD1激动剂融合分子导向不同的组织,并且TCR-PD1激动剂融合分子可以促进对T细胞活性的靶向抑制。
Claims (22)
1.一种双功能结合多肽,包含pMHC结合部分和PD-1激动剂。
2.根据权利要求1所述的双功能结合多肽,其特征在于,所述pMHC结合部分包含TCR可变结构域和/或抗体可变结构域。
3.根据权利要求1所述的双功能结合多肽,其特征在于,所述pMHC结合部分是T细胞受体(TCR)或TCR样抗体。
4.根据前述权利要求中任一项所述的双功能结合多肽,其特征在于,所述pMHC结合部分是异二聚体α/βTCR多肽对。
5.根据前述权利要求中任一项所述的双功能结合多肽,其特征在于,所述pMHC结合部分是单链α/βTCR多肽。
6.根据权利要求3至5中任一项所述的双功能结合多肽,其特征在于,所述TCR在α链的恒定区和β链的恒定区之间包含非天然的二硫键。
7.根据权利要求3至6中任一项所述的双功能结合多肽,其特征在于,所述TCR与肽抗原特异性结合。
8.根据前述权利要求中任一项所述的双功能结合多肽,其特征在于,所述PD-1激动剂是PD-L1或其功能片段。
9.根据权利要求8所述的双功能结合多肽,其特征在于,所述PD-L1包含以下序列或由以下序列组成:

10.根据权利要求1至7中任一项所述的双功能结合多肽,其特征在于,所述PD-1激动剂是全长抗体或其片段。
11.根据权利要求10所述的双功能结合多肽,其特征在于,所述PD-1激动剂是scFv抗体。
12.根据前述权利要求中任一项所述的双功能结合多肽,其特征在于,所述PD-1激动剂与所述pMHC结合部分的C末端或N末端融合。
13.根据前述权利要求中任一项所述的双功能结合多肽,其特征在于,所述PD-1激动剂通过接头与所述pMHC结合部分融合。
14.根据权利要求13所述的双功能结合多肽,其特征在于,所述接头的长度为2、3、4、5、6、7或8个氨基酸。
15.一种药物组合物,其包含根据权利要求1至14中任一项所述的双功能结合多肽。
16.一种核酸,其编码根据权利要求1至14中任一项所述的双功能结合多肽。
17.一种表达载体,其包含权利要求16所述的核酸。
18.一种宿主细胞,其包含根据权利要求16所述的核酸或根据权利要求17所述的载体,任选地,其中编码所述双功能结合多肽的所述核酸以单个开放读码框或分别编码α链和β链的两个不同的开放阅读框存在。
19.一种制备根据权利要求1至14中任一项所述的双功能结合多肽的方法,所述方法包括在表达核酸的任选条件下维持根据权利要求18所述的宿主细胞,并分离所述双功能结合肽。
20.根据权利要求1至14中任一项所述的双功能结合多肽、根据权利要求15所述的药物组合物、根据权利要求16所述的核酸和/或根据权利要求17所述的载体,用于医学,特别是用于治疗自身免疫疾病的用途,或者用于治疗或预防疼痛特别是与炎症有关的疼痛的用途。
21.根据权利要求20所述的双功能结合多肽、药物组合物、核酸和/或载体的用途,其特征在于,所述自身免疫疾病是斑秃、强直性脊柱炎、特应性皮炎、格雷夫斯病、多发性硬化、牛皮癣、类风湿性关节炎、系统性红斑狼疮、1型糖尿病和白癜风、炎性肠病、克罗恩氏病、溃疡性结肠炎、乳糜泻、眼部疾病(例如葡萄膜炎)、皮肤性狼疮和狼疮性肾炎以及由PD-1/PD-L1拮抗剂引起的癌症患者的自身免疫疾病中的一种。
22.一种治疗自身免疫病症的方法,所述方法包括将权利要求1至14中任一项所述的双功能结合多肽、根据权利要求15所述的药物组合物、根据权利要求16所述的核酸和/或根据权利要求17所述的载体给药至有需要的患者。
