Abstract
Description
多くのタンパク質ベースの治療薬(抗体フラグメント及びd融合タンパク質を含む)は、投与後に身体から迅速に排出される。それらの短い循環半減期は、代表的には、小さなサイズ(これが腎濾過による効率的なクリアランスを可能にする)及び細胞内分解からの保護の欠如に帰せられる。この場合、頻回投与又は長い注入時間が、当該薬剤の有効濃度を長期間にわたって維持するために必要となる。投薬を改善するため、循環半減期を延長する幾つかのストラテジが用いられてきた。これらには、可撓性親水性分子(例えば、炭化水素又はPEG(ポリエチレングリコール))の付着によるタンパク質の流体力学半径の増加及び抗体Fcドメイン又は血清アルブミンの付着による胎児性Fcレセプター(FcRn)を介するリサイクリングの活用が挙げられる(Konnteman,Curr Opin Biotechnol. 2011 Dec;22(6):868-76)。
可溶性T細胞レセプターを抗CD3抗体フラグメントに融合して含んでなる融合体タンパク質は、6~8時間のインビボ半減期を有するT細胞結合性二重特異性融合タンパク質の新規カテゴリーである(Satoら,2018 J Clin Onc 2018 36, no. 15_suppl 9521-9521;Middletonら,J Clin Onc 2016 34, no. 15_suppl 3016-3016)。伝統的な抗体とは異なり、T細胞レセプターは、細胞内抗原に由来し、細胞表面にヒト白血球抗原によって提示される短いペプチド(ペプチド-HLA)を認識するように設計されている。抗原提示細胞上のペプチド-HLA複合体とT細胞との間での有効な免疫シナプス形成は、(膜内距離の増加により撹乱される)固定の相互作用幾何形状に依拠している(Choudhuriら,2005 Nature Jul 28;436(7050):578-82)。
半減期が増加した、有効な免疫シナプス形成を媒介することができるT細胞結合性二重特異性タンパク質に関する必要性が存在する。
i)T細胞結合性免疫エフェクターに連結された、ペプチド-主要組織適合性複合体(pMHC)結合性部分と、
ii)免疫グロブリンFc又はアルブミン結合性ドメインを含んでなる半減期延長ドメインと
を含んでなる複数ドメイン結合性分子が提供される。
i)T細胞結合性免疫エフェクターに連結された、ペプチド-主要組織適合性複合体(pMHC)結合性部分と、
ii)免疫グロブリンFc又はアルブミン結合性ドメインを含んでなる半減期延長ドメインと
を含んでなる複数ドメイン結合性分子が提供される。
TCRはヘテロ二量体α/β又はγ/δ TCRポリペプチド対であり得る。或いは、TCRは単鎖α/β又はγ/δ TCRポリペプチドであり得る。TCR可変ドメインのアミノ酸配列は、天然に見出されるものに相当してもよいし、天然TCRに関して1又は2以上の変異を含有してもよい。このような変異は、所与の抗原に関するTCRの親和性を増大させるようなものであってもよい。追加的に又は代替的に、安定性及び製造可能性を向上させる変異が組み込まれていてもよい。
TCRは、ペプチド抗原と複合体化したMHCに結合し得る。好ましくは、ペプチド抗原は任意の疾患関連抗原である。好ましくは、ペプチド抗原は任意の腫瘍関連抗原である。ペプチド抗原は、WO2011001152、WO2017109496、WO2017175006及びWO2018234319に記載されるようなGP100、NYESO、MAGEA4又はPRAMEに由来するペプチドであり得る。
TCRは、WO2011001152、WO2017109496、WO2017175006及びWO2018234319に記載されるようなアミノ酸配列を有していてもよい。
半減期延長ドメインは免疫グロブリンFcを含んでなってもよい。免疫グロブリンFcドメインは任意の抗体Fc領域であり得る。Fc領域は、細胞表面Fcレセプター及び補体系の幾つかのタンパク質と相互作用する抗体テイル領域である。Fc領域は、代表的には、共に2つ又は3つの重鎖定常ドメイン(CH2、CH3及びCH4と呼ぶ)及びヒンジ領域を有する2つのポリペプチド鎖を含んでなる。2つの鎖はヒンジ領域内でのジスルフィド結合により連結されている。免疫グロブリンサブクラスIgG1、IgG2及びIgG4のFcドメインは、FcRnに結合して、FcRnを介したリサイクリングを受け、長い循環半減期(3~4週間)を提供する。IgGとFcRnとの相互作用は、CH2及びCH3ドメインの部分をカバーするFc領域に局在化されている。本発明における使用に好適な免疫グロブリンFcドメインは、IgG1又はIgG4ののFcドメインを含むがこれらに限定されない。好ましくは、Fcドメインはヒト配列に由来する。Fc領域はまた、好ましくは、二量体化を容易にするKiH変異並びに活性化するレセプターとの相互作用を防止する変異を有していてもよい(すなわち、機能的にサイレントな分子であってもよい)。免疫グロブリンFcドメインは、その他のドメイン(すなわちTCR又は免疫エフェクター)のC又はN末端に融合されていてもよい。免疫グロブリンFcは、その他のドメイン(すなわちTCR又は免疫エフェクター)にリンカーを介して融合されていてもよいし、或いはリンカーが用いられていなくてもよい。リンカー配列は、通常、可撓性を限定する虞れがある嵩高い側鎖を有しないアミノ酸、例えばグリシン、アラニン及びセリンから主に作られている点で、可撓性である。或いは、より大きい剛性を有するリンカーが望ましくあり得る。リンカー配列の使用可能な又は最適な長さは、容易に決定され得る。多くの場合、リンカー配列は、約12アミノ酸長以下、例えば10アミノ酸長以下又は2~10アミノ酸長である。リンカーは、1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、25、26、27、28、29又は30アミノ酸長であり得る。本発明の複数ドメイン結合性分子に用い得る適切なリンカーの例として、GGGSGGGG、GGGGS、GGGSG、GGSGG、GSGGG、GSGGGP、GGEPS、GGEGGGP及びGGEGGGSEGGGS(WO2010/133828に記載のもの)が挙げられるが、これらに限定されない。免疫グロブリンFcは、TCRに融合される場合、α若しくはβ鎖のいずれか、又はα及びβ鎖の両方に、リンカー有り又は無しで融合され得る。更に、Fcの個々の鎖がTCRの個々の鎖に融合され得る。
Fc領域は、WT Fc配列に関して変異を含んでいてもよい。変異には、置換、挿入及び欠失が含まれる。このような変異は、望ましい治療特性の導入を目的としてなされたものであってもよい。例えば、ヘテロ二量体化を容易にするために、オブ・インツー・ホール(KiH)変異がCH3ドメイン中に工学的操作により導入されていてもよい。この場合、一方の鎖が嵩高い突出残基(すなわちノブ)、例えばYを含有するように操作され、他方の鎖が相補ポケット(すなわちホール)を含有するように操作されている。KiH変異の適切な位置は当該分野において公知である。追加的に又は代替的に、Fcyレセプターへの結合を消失させるか若しくは低減させる及び/又はFcRnへの結合を増大させる変異、及び/又はFabアーム交換を防止するか又はプロテアーゼ部位を除去する変異が導入されていてもよい。追加的に又は代替的に、製造上の理由のために、例えば翻訳語修飾(例えばグコシル化)に供され得るアミノ酸を除去又は置換する変異がなされていてもよい。

好ましくは、アルブミンはヒト血清アルブミン(HSA)である。アルブミン結合性ドメインのヒトアルブミンに関する親和性は、ピコモル~マイクロモルの範囲内であり得る。ヒト血清におけるアルブミンの極端に高い濃度(35~50mg/ml、約0.6mM)を考慮すれば、アルブミン結合性ドメインの実質的に全てがインビボでアルブミンに結合すると計算される。
1つの更なる観点において、第1の観点に従う複数ドメイン結合性分子を含んでなる医薬組成物が提供される。
別の1つの更なる観点において、第1の観点に従う複数ドメイン結合性分子をコードする核酸が提供される。この観点の核酸を含んでなる発現ベクターが提供される。加えて、この観点の核酸又はベクターを含んでなり、複数ドメイン結合性分子をコードする核酸が単一オープンリーディングフレーム又はα鎖及びβ鎖をそれぞれコードする2つの別個のオープンリーディングフレームとして存在する宿主細胞が提供される。
また、1つの更なる観点において、上記の宿主細胞を核酸の発現に最適な条件下で維持すること及び複数ドメイン抗原結合性ポリペプチドを単離することを含んでなる、第1の観点に従う複数ドメイン結合性分子を生産する方法が提供される。
1つの尚更なる観点において、第1の観点に従う複数ドメイン結合性分子を必要とする患者に投与することを含んでなる治療方法が提供される。
表現型上サイレントなバリアントは、あ1又は2以上の保存的置換及び/又は1又は2以上の寛容される置換を含有し得る。寛容される置換は、下記の保存的置換の定義には入らないにもかかわらず、表現上サイレントである置換を意味する。当業者は、種々のアミノ酸が類似する特性を有し、よって「保存的」であることを知っている。タンパク質、ポリペプチド又はペプチドの1又は2以上のこのようなアミノ酸は、多くの場合で、当該タンパク質、ポリペプチド又はペプチドの所望の活性を焼失することなく、1又は2以上の他のこのようなアミノ酸により置換することができる。
「同一性」は、当該分野において公知のように、配列比較により決定される、2若しくは3以上のポリペプチド配列間又は2若しくは3以上のポリヌクレオチド配列間の関係である。当該分野において、同一性はまた、妥当な場合、ポリペプチド配列間又はポリヌクレオチド配列間の、配列の並びの一致性によって決定される配列関連性の程度を意味する。2つのポリペプチド配列間又は2つのポリヌクレオチド配列間の同一性を測定する方法は幾つか存在するが、同一性の決定に一般に用いられる方法は、コンピュータプログラム化されている。2つの配列間の同一性を決定する好適なコンピュータプログラムとしては、GCGプログラムパッケージ(Devereuxら,Nucleic acid Research, 12, 387(1984)、BLASTP、BLASTN及びFASTA(Atschulら,J. Molec. Biol. 215, 403(1990))が挙げられるが、これに限定されない。
2つのアミノ酸配列又は2つの核酸配列のパーセント同一性は、最適比較を目的として配列を整列させ(例えば、最良整列のために第1の配列にギャップを導入することができる)、対応する位置でアミノ酸残基又はヌクレオチドを比較することにより決定する。「最良整列」は、最も高いパーセント同一性をもたらす2つの配列の整列である。パーセント同一性は、比較する配列中の同一アミノ酸残基又はヌクレオチドの数により決定する(すなわち、%同一性 = 同一位置の数/位置の総数×100)。
任意の適切な方法を用いることを条件に、変異(保存的及び寛容される置換、挿入及び欠失を含む)を配列に導入してもよい。このような方法としては、ポリメラーゼ連鎖反応(PCR)をベースにする方法、制限酵素ベースのクローニング又はライゲーション非依存性クローニング(LIC)手順が挙げられるが、これらに限定されない。これら方法は、多くの標準的な分子生物学の教科書に詳述されている。ポリメラーゼ連鎖反応(PCR)及び制限酵素ベースのクローニングに関する更なる詳細については、Sambrook及びRussell(2001),Molecular Cloning - A Laboratory Manual(第3版) CSHL Pressを参照。ライゲーション非依存性クローニング(LIC)手順についての更なる情報は、Rashtchian(1995),Curr Opin Biotechnol 6(1):30-6に見出すことができる。本発明により提供されるTCR配列は、固相合成又は当該分野において公知の任意の他の適切な方法から得てもよい。
本発明の分子は高収率精製が可能であり得る。収率は、精製プロセスの間に保持される材料の量に基づいて決定し得(すなわち、リフォールディング前に得られた可溶化した物質の量に対する、精製プロセスの終時に得られる正確にフォールディングした物質の量)、及び/又は収率は、元の培養体積に対する、精製プロセスの終時に得られる正確にフォールディングした物質の量に基づいて決定し得る。高収率は、1%以上、より好ましくは5%以上、又はより高い収率を意味する。高収率は、1mg/ml以上、より好ましくは3mg/ml以上、5mg/ml以上、又はより高い収率を意味する。
α及びβ鎖定常ドメイン配列は、TRACのエキソン2のCys4とTRBC1又はTRBC2のエキソン2のCys2との間の天然型ジスルフィド結合を欠失させるように短縮化又は置換により改変されていてもよい。α及び/又はβ鎖定常ドメイン配列は、例えばWO 03/020763及びWO06000830に記載されるように、それぞれの定常ドメインの残基同士間に導入されたジスルフィド結合を有していてもよい。α及びβ定常ドメインは、TRACのThr 48の位置及びTRBC1又はTRBC2のSer 57の位置でのシステイン残基の置換により改変され、該システインがTCRのα定常ドメインとβ定常ドメインとの間のジスルフィド結合を形成していてもよい。TRBC1又はTRBC2は、追加的に、定常ドメインの75位でのシステイン→アラニン変異及び定常ドメイン89位でのアスパラギン→アスパラギン酸変異を含んでいてもよい。αβヘテロ二量体に存在する細胞外定常ドメインの一方又は両方は、一方又は両方のC末端で、例えば15まで又は10まで又は8まで又は7以下のアミノ酸が欠失されていてもよい(短縮化)。α鎖細胞外定常ドメインのC末端は、8アミノ酸が欠失されていてもよい(短縮化)。

本明細書に記載の組成物及び方法における使用に適切な抗体フラグメント及びバリアント/アナログとしては、数例挙げるとすれば、ミニボディ、Fabフラグメント、F(ab')2フラグメント、dsFv及びscFvフラグメント、ナノボディTM(Ablynx(ベルギー)から市販され、ラクダ科動物(例えば、ラクダ又はラマ)抗体に由来する合成の単鎖免疫グロブリン可変重鎖ドメインを含む構築物)及びドメイン抗体(Domantis(ベルギー);これは、親和性成熟単鎖免疫グロブリン可変重鎖ドメイン又は免疫グロブリン可変軽鎖ドメインを含む)又は抗体様結合特性を示す代替のタンパク質足場、例えばアフィボディ(Affibody(スウェーデン);工学的に操作されたプロテインA足場を含む)若しくはアンチカリン(Anticalins)(Pieris(ドイツ));工学的に操作されたアンチカリンを含む)が挙げられる。免疫エフェクターは、複数ドメイン抗原結合性ポリペプチドのTCR部分に連結される。ここで、好ましくは、免疫エフェクターは抗CD3抗体である。
1つの更なる観点において、本発明は、本発明の複数ドメイン結合性分子をコードする核酸を提供する。幾つかの実施形態において、核酸はcDNAである。幾つかの実施形態におて、核酸はmRNAであり得る。核酸は、天然に存在しない及び/又は精製された及び/又は操作された核酸であり得る。核酸配列は、利用する発現系に従って最適化されたコドンであり得る。当業者に公知のように、発現系は、細菌細胞、例えばE. coli、若しくは酵母細胞、若しくは哺乳動物細胞、若しくは昆虫細胞を含んでいてもよく、又は無細胞発現系であり得る。
種々の宿主細胞におけるポリペプチドのクローニング及び発現のための系は周知である。適切な宿主細胞としては、細菌、哺乳動物細胞、酵母及びバキュロウイルス系が挙げられる。異種ポリペプチドの発現のために当該分野において利用可能な哺乳動物細胞株としては、チャイニーズハムスター卵巣細胞、HeLa細胞、ベイビーハムスター肝臓細胞、NSOマウスメラノーマ細胞などが挙げられる。一般的な好適な細菌宿主はE. coliである。原核細胞(例えばE. coli)における抗体及び抗体フラグメントの発現は、当該分野において十分に確立されている。概要については、例えばPluckthun,Bio/Technology 9:545-551 (1991)を参照。培養中の真核細胞における発現もまた、特異的結合性分子の製造のための1つの選択肢として、当業者に利用可能である。最近のレビューとして、例えばReff,Curr. Opinion Biotech. 4:573-576(1993);Trillら,Curr. Opinion Biotech. 6:553-560(1995)を参照。
よって、本発明の1つの更なる観点は、本明細書に開示される核酸を含有する宿主細胞を提供する。1つの尚更なる観点は、宿主細胞に該核酸を導入することを含んでなる方法を提供する。導入には任意の利用可能な技法を用い得る。真核細胞については、適切な技法としては、リン酸カルシウムトランスフェクション、DEAE-デキストラン、エレクトロポレーション、リポソーム-媒介トランスフェクション及びレトロウイルス又は他のウイルス(例えばワクシニア又は昆虫細胞についてはバキュロウイルス)を用いる形質導入が挙げられ得る。細菌細胞については、適切な技法としては、塩化カルシウム形質転換、エレクトロポレーション及びバクテリオファージを用いるトランスフェクションが挙げられ得る。導入に続いて、例えば宿主細胞を遺伝子発現条件下に培養することにより、核酸からの発現を引き起こすか又は可能にし得る。
当該分野において周知であるように、分子は翻訳後修飾に付されていてもよい。グリコシル化はそのような修飾の1つであり、TCR鎖中の規定のアミノ酸へのオリゴ糖部分の共有結合的付着を含む。例えば、アスパラギン残基又はセリン/スレオニン残基は、周知のオリゴ糖付着位置である。特定のタンパク質のグリコシル化状況は、タンパク質配列、タンパク質コンホメーション及び特定の酵素の利用可能性を含む幾つかの因子に依存する。更に、グリコシル化状況(すなわち、オリゴ糖タイプ、共有結合及び付着の総数)は、タンパク質の機能に影響することがある。したがって、組換えタンパク質を製造するときには、グリコシル化の制御が望ましい場合が多い。制御されたグリコシル化は、抗体ベースの治療薬を改善するために用いられる(Jefferisら(2009) Nat Rev Drug Discov Mar;8(3):226-34.)。本発明の分子のTCR部分について、グリコシル化は、例えば特定の細胞株(哺乳動物細胞株、例えばチャイニーズハムスター卵巣(CHO)細胞又はヒト胚性肝臓(HEK)細胞を含むがこれらに限定されない)を用いることにより、又は化学的修飾により制御されてもよい。このような修飾は、グリコシル化が薬物動態を改善し、免疫原性を低減させ及び天然型ヒトタンパク質をより厳密に模擬することができるので、望ましくあり得る(Sinclair and Elliott(2005) Pharm Sci.Aug;94(8):1626-35)。
医薬組成物は、任意の適切な経路、例えば非経口経路(皮下、筋内、髄腔内又は静脈内経路を含む)、経腸(経口又は直腸経路を含む)、吸入又は鼻内経路による投与に適合していてもよい。このような組成物は、薬学分野において公知の任意の方法により、例えば活性成分をキャリア又は賦形剤と滅菌条件下で混合することにより、製造されてもよい。
本発明の分子の投薬量は、治療すべき疾患又は異常、治療すべき個体の年齢及び状態などに依存して広範に変化し得る。本発明の分子に適切な用量範囲は、25ng/kg~50μg/kg又は1μg~1gであり得る。医師が、使用すべき適切な投薬量を最終的には決定する。
本発明の複数ドメイン抗原結合性ポリペプチド、医薬組成物、ベクター、核酸及び細胞は、実質的に純粋な形態、例えば、少なくとも80%、少なくとも85%、少なくとも90%、少なくとも91%、少なくとも92%、少なくとも93%、少なくとも94%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%又は100%純粋で提供されてもよい。
・医薬における使用のため、好ましくはガン又は腫瘍の治療法における使用のための、本発明の複数ドメイン抗原結合性ポリペプチド、核酸、医薬組成物又は細胞;
・ガン又は腫瘍の治療用医薬の製造における、本発明の複数ドメイン抗原結合性ポリペプチド、核酸、医薬組成物又は細胞の使用;
・本発明の複数ドメイン抗原結合性ポリペプチド、核酸、医薬組成物又は細胞を患者に投与することを含んでなる、患者におけるガン又は腫瘍の治療法;
・本発明の複数ドメイン抗原結合性ポリペプチド、核酸、医薬組成物又は細胞を含む、ヒト対象に投与するための注射可能製剤。
本発明の各々の観点の好適な特徴は、他の観点の各々についても同様である(ただし、必要な変更は加える)。本明細書において言及した先行技術文献は、法が許容する最大範囲まで組み込まれる。

図2は、Fcドメインを組み込まれたTCR-抗CD3融合体が抗原陽性標的細胞の存在下でT細胞活性化を媒介することができることを示す。3つのIgG1-Fc融合体のデータを示す。
図3A)はアルブミン結合性ペプチドを組み込まれたTCR-抗CD3融合体が抗原陽性標的細胞の存在下でT細胞活性化を媒介することができることを示す。アルブミン結合性ペプチドがC-α(F1)、N-α(F2)又はC-β(F3)のいずれかに付着する3つの形式が示されている。B)は、アルブミン結合性ナノボディを組み込まれているTCR-抗CD3融合体が抗原陽性標的細胞の存在下でT細胞活性化を媒介できることを示している。アルブミン結合性ペプチドがC-α(R)又はC-β(Y)のいずれかに付着する2つの形式が示されている。C)は、アルブミン結合性ドメイン抗体(AlbudabTM)を組み込まれているTCR-抗CD3融合体が抗原陽性標的細胞の存在下でT細胞活性化を媒介することができることを示している。AlbudabTMがTCR-抗CD3融合体のC-αに付着する1つの形式が示されている。
図4A)は、マウス血清中でのTCR-抗CD3-AlbudabTM融合体のPK特性を示す。B)は、マウスにおける評価に基づく、TCR-抗CD3-AlbudabTM融合体のヒトにおける理論的PKを示す。
以下の実施例は、本発明の複数ドメイン結合性分子を説明する。当該分子は、TCR-抗CD3-Fc融合タンパク質又はTCR-抗CD3-アルブミン結合性融合タンパク質と呼ぶこともある。
実施例1(Fc融合体)
a)TCR-抗CD3-Fc融合タンパク質の設計
本実施例において、PRAMEのHLA-A*02制限ペプチドに結合する高親和性TCRを含んでなるTCR-抗CD3融合タンパク質を用いた。このような分子の例はWO2018234319に提供される。
ヒトIgG1 Fcドメインを、リンカーを介して、TCR-抗CD3のC末端に融合した(概略については図1Aを参照)。当該分野において公知であるヒトIgG1 Fcの機能的バリアントを含んでなる更なる2つの構築物を作製した。バリアント1は、Fcγレセプター(FcγRs)にも補体タンパク質C1qにも結合せず、したがって機能上サイレントである。バリアント2はFcRnに対して増大した結合性(延長したインビボ半減期をもたらし得る)を示す。各場合で、Fcドメインは、ヘテロ二量体化を容易にするための既知のノブ・イン・ホール変異Y86T及びT22Yを含んでなる。
図1Bは、機能上サイレントなヒトIgG1 Fcを含んでなるTCR-抗CD3-Fc融合体(バリアント1)の配列を示す。
浮遊培養適応チャイニーズハムスター卵巣(CHO)細胞(ExpiCHO Expression system, Thermo Fisher)に基づく一過性発現系を用いて、Fc融合体の発現を行った。細胞は、種々のIg Fcドメインに融合した該当するTCR鎖を含有する哺乳動物発現プラスミドを用いて、製造業者の指示に従ってトランスフェクトした。収集後、細胞培養上清を、保冷遠心分離機において4000~5000×gにて30分間遠心分離することにより明澄化した。上清を0.22μmフィルターに通してろ過し、更なる精製のために集めた。
精製のため、Fc融合体を、先ず緩衝剤で調整後、mAbselect Sureプレパックカラム(GE Healthcare又は同等な樹脂)を製造業者のガイドラインに従って用いて精製した。特定のタンパク質含有画分をプールし、生理学的に適切な緩衝剤中で適切なカラム(GE Healthcare)を用いるサイズ排除クロマトグラフィーにより更に精製した。特定のタンパク質含有画分をプールして、下流の試験及び貯蔵のために濃縮した。
TCR-抗CD3-Fc融合タンパク質を、抗原提示T2細胞に対するCD3+ T細胞の強力な再指向化を媒介する能力について評価した。インターフェロン-γ(IFN-γ)放出をT細胞活性化についての読取値として用いた。
アッセイは、ヒトIFN-γ ELISPOTキット(BD Biosciences)を製造業者の指示に従って用いて行った。簡潔には、T2細胞を標的細胞として用いて、5nM PRAMEペプチドでパルスした。標的細胞は、アッセイ培地(10%熱不活化FBS及び1%ペニシリン-ストレプトマイシン-L-グルタミンを含有するRPMI 1640)中に1×106/mlの密度にて調製し、体積50μl中50,000細胞/ウェルにてプレートした。新鮮なドナー血液から単離した末梢血単核細胞(PBMC)をエフェクター細胞として用いて、体積50μl中35,000細胞/ウェルにてプレートした。TCR-抗CD3-Fc融合タンパク質を10nM~0.0001nMの最終濃度に滴定して、ウェルに体積50μlで添加した。
図2に示す結果は、TCR-抗CD3-Fc融合タンパク質が抗原陽性標的細胞の存在下で有効なT細胞活性化を媒介することを証明している。Ec50値はpM範囲であった(IgG1-Fc、IgG1-Fc-バリアント1及びIgG1-Fc-バリアント2との融合体に関してそれぞれ238pM、257pM及び25pM)。抗原陰性標的細胞を用いたコントロール実験は、機能上サイレントなIgG1-バリアント2が無視し得るバックグランド活性をもたらしたことを証明している。したがって、機能上サイレントなFcドメインが治療用途に最も好適であると考えられた。
a)TCR-抗CD3-アルブミン結合性融合タンパク質の設計
第1の設計において、TCR-抗CD3融合タンパク質は、gp100のHLA-A*02制限ペプチドに結合する高親和性TCRを含んでいた。この分子のアミノ酸配列はWO2011001152に開示されている。具体的には、TCR-抗CD3融合体は、WO2011001152の配列番号45のα鎖(ここで、配列番号45の番号付けに基づいて、アミノ酸1~109はWO2011001152の配列番号8で置換され、1位のアミノ酸はAである);及びWO2011001152の配列番号36のβ鎖(ここで、残基259~370はWO2011001152の配列番号27に相当し、1位及び2位のアミノ酸はそれぞれA及びIである)を含んでなっていた。アミノ酸配列QRLMEDICLPRWGCLWEDDFを有するアルブミン結合性ペプチド(Dennisら,J Biol Chem. 2002 Sep 20;277(38):35035-43に記載される通り)を、TCR-抗CD3融合体にリンカーを介して付着させた。適切なリンカーはGGGGSである。アルブミン結合性ペプチドを3つの異なる付着部位に融合した3つのバリアントを調製した:C-α(F1)、N-α(F2)又はC-β(F3)。
第3の設計において、アルブミン結合性ドメイン抗体をTCR-抗CD3融合体α鎖のC末端に付着させた。抗体はAlbudabTMプラットフォームに属する。このドメイン抗体の2つのバリアントを用いた;DOM 7h-10-14 dAb及びDOM 7h-11-15 dAb(WO201010893中でそれぞれ配列番号26及び27で提供される)。抗体はリンカーを介して、直接的に(すなわち、リンカーを有さずに)付着された。適切なリンカーはGGGGSである。TCR-抗CD3融合タンパク質は、MAGEA3のHLA-A*01制限ペプチドに結合する高親和性TCRを含んでいた。この分子のアミノ酸配列はWO2013041865に提供される。
TCR-抗CD3融合タンパク質に関して当該分野において公知のもの(例えば、WO2011001152、実施例2)と同じ方法を用いて、TCR-抗CD3-アルブミン結合性融合タンパク質をE. coli中で封入体として発現させ、その後、リフォールディングさせて精製した。
TCR-抗CD3-アルブミン結合性融合タンパク質を、抗原陽性ガン細胞に対するCD3+ T細胞の強力な再指向化を媒介する能力について評価した。インターフェロン-γ(IFN-γ)放出をT細胞活性化についての読取値として用いた。
アッセイは、ヒトIFN-γ ELISPOTキット(BD Biosciences)を製造業者の指示に従って用いて行った。簡潔には、アルブミン結合性ペプチドを含んでなる融合体については、メラノーマMel526細胞を標的細胞として用いた。標的細胞は、アッセイ培地(RPMI 1640+150μMヒト血清アルブミン(HSA)及び1%ペニシリン-ストレプトマイシン-L-グルタミン)中に1×106/mlの密度にて調製し、体積50μl中50,000細胞/ウェルにてプレートした。新鮮なドナー血液から単離した末梢血単核細胞(PBMC)をエフェクター細胞として用いて、体積50μl中30,000細胞/ウェルにてプレートした。TCR-抗CD3-Fc融合タンパク質を10nM~0.0001nMの最終濃度に滴定して、ウェルに体積50μlで添加した。
実施例1cに記載の通りにプレートを調製して発色させた。
図3A-Cは、アルブミン結合性部分とTCR-抗CD3融合タンパク質との融合体が、抗原陽性標的細胞の存在下で有効なT細胞活性化を媒介することを示している。Ec50値はpM範囲であった(137.4/178.0/137.1)。
a)TCR-抗CD3-アルブミン結合性融合タンパク質のPK評価
TCR-抗CD3-AlbudAb融合体のPK特性をマウス血清中で調べた。
マウスに0.1mg/kgの融合タンパク質を静脈内ボーラス注射により投与し、血清サンプルを120時間にわたって一定間隔で採取した。ELISAベースアッセイを用いてPK評価を行った。簡潔には、ビオチン化pHLA複合体をストレプトアビジン-被覆プレートに付着させた後、血清サンプルを添加した。抗CD3 scFvに対する一次ヤギ抗体と、450nmでのTMBを用いる比色検出に関して活性化されたHRP-接合抗ヤギIgGとを用いて検出工程を行った。生成した結果を用いて、系列希釈及び標準曲線に対する分析によってTCR-抗CD3-Albudab融合体の存在及び結合活性を確証した。結果を%活性として表した。この結果を用いて、Cmax対時間のプロットを作成した。
図4AのPKデータを用いて、TCR-抗CD3-AlbudAb融合体のヒトにおける理論的なPKプロフィールを算出した(図4B)。Albudabとの融合体は、インビボ半減期が7時間から264時間へ延長されたと予測される。
Claims (17)
- i)T細胞結合性免疫エフェクターに連結された、ペプチド-主要組織適合性複合体(pMHC)結合性部分と、
ii)免疫グロブリンFc又はアルブミン結合性ドメインを含んでなる半減期延長ドメインと
を含んでなる複数ドメイン結合性分子。 - pMHC結合性部分がT細胞レセプター(TCR)又はTCR及び/若しくは抗体可変ドメイン及び少なくとも1つの定常ドメインを含んでなるTCR様抗体である、請求項1に記載の複数ドメイン結合性分子。
- TCRがヘテロ二量体α/βTCRポリペプチド対である、請求項2に記載の複数ドメイン結合性分子。
- TCRが単鎖α/βTCRポリペプチドである、請求項2に記載の複数ドメイン結合性分子。
- T細胞結合性免疫エフェクタードメインが、CD3及び/又はTCR/CD3複合体との相互作用によりT細胞を活性化するCD3エフェクタードメインである、請求項1~4のいずれか1項に記載の複数ドメイン結合性分子。
- CD3エフェクタードメインが抗体scFv又は抗体様足場を含んでなる、請求項5に記載の複数ドメイン結合性分子。
- 半減期延長ドメインが免疫グロブリンFcドメインである、請求項1~6のいずれか1項に記載の複数ドメイン結合性分子。
- 半減期延長ドメインがアルブミン結合性ドメインを含んでなる、請求項1~6のいずれか1項に記載の複数ドメイン結合性分子。
- 半減期延長ドメインがpMHC結合性部分のC若しくはN末端又はT細胞結合性免疫エフェクターのC若しくはN末端に連結されている、請求項1~8のいずれか1項に記載の複数ドメイン結合性分子。
- 半減期延長ドメインがpMHC結合性部分又はT細胞結合性免疫エフェクターにリンカーを介して連結されている、請求項1~8のいずれか1項に記載の複数ドメイン結合性分子。
- 医薬として使用するための請求項1~10のいずれか1項に記載の複数ドメイン結合性分子。
- 請求項1~10のいずれか1項に記載の複数ドメイン結合性分子を含んでなる医薬組成物。
- 請求項1~10のいずれか1項に記載の複数ドメイン結合性分子をコードする核酸。
- 請求項13に記載の核酸を含んでなる発現ベクター。
- 請求項13に記載の核酸又は請求項14に記載のベクターを含んでなり、複数ドメイン結合性分子をコードする核酸が単一オープンリーディングフレーム又はそれぞれα鎖及びβ鎖をコードする2つの別個のオープンリーディングフレームとして存在する、宿主細胞。
- 請求項15に記載の宿主細胞を核酸の発現に適切な条件下で維持し、複数ドメイン抗原結合性ポリペプチドを単離することを含んでなる請求項1に記載の複数ドメイン結合性分子の製造方法。
- 請求項1~10のいずれか1項に記載の複数ドメイン結合性分子又は請求項12に記載の医薬組成物を必要とする患者に投与することを含んでなる治療方法。
