る。スペーサーXの好ましい例は、下記の(8)〜(11)で表されるものである。また、スペーサーXは、化合物が二つのリン酸基を距離選択的に認識できるように二つのDpaが夫々有するピリジン環の間を連結している。
本発明の化合物は、様々な機能分子を修飾することが可能であり、修飾に用いることができる分子として例えば以下のものを挙げることが出来る。水溶性向上のためにエチレングリコール鎖、あるいは発光・蛍光センシングのための、例えば、ルミノール、イソルミノール、ルシフェリン、ジオキセタン、フルオレセイン、ローダミン等の発色性物質・発光性物質。また、核磁気共鳴法による検出のための常磁性体、磁性粒子や核磁気共鳴活性核種、γカウンター検出のための123I、201Tl、67Ga、99mTc、111Inなどの放射性核種、15O、13N、11C、18Fなどの陽電子放出種。さらに、治療のための薬剤など。特に、化合物の水溶性向上のためのエチレングリコール鎖が好ましい。また、常磁性体としてはガドリニウム、磁性粒子として酸化鉄微粒子をそれぞれ好適に用いることができる。また、核磁気共鳴活性核種として1H、13C、15N、19F、23Na、29Si、31Pなどを好適に用いることができる。
本発明に従う発光性化合物として、既述の式(16)で表されるZn(Dpa) Stilbazole型錯体1(図3)を以下のスキームで合成した。また比較のため、Dpaを一つ有する化合物2(図4)も合成した。図5に合成スキームを示す。
100ml三口ナスフラスコに、2-ピリジンカルボン酸塩酸塩 5.0g (31.7mmol)、塩化チオニル 15mlを入れ氷浴上で攪拌した。その溶液に、蒸留水 0.5ml (31.7mmol / 1eq)をゆっくり滴下し、滴下後に加熱還流を開始した。反応追跡はTLC (Hexane / EtOAc = 1 / 1) により行い、3日後に反応の終了を確認した。溶媒を減圧留去し、さらにトルエンを加えて減圧留去を行った。残渣にトルエンを30ml加え、氷浴で冷却し、MeOH (1.3eq)をゆっくり滴下すると固体が析出した。析出した固体をろ別し、トルエンで洗浄した。得られた固体をクロロホルムに溶解し、飽和炭酸水素ナトリウム水溶液で洗浄した。有機層を硫酸マグネシウムにより乾燥した後、溶媒を減圧留去した。精製はカラムクロマトグラフィー (シリカゲル, Hexane / EtOAc = 3 / 1)により行い、白色固体 2.52g (46.3%)を得た。同定は1H-NMR、FAB-MSにより行った。(1H-NMR (400MHz、CDCl3、25℃、TMS) δ/ ppm ; 4.02 (3H, s), 7.49 (1H, dd, J=2.0, 4.8 Hz), 8.14 (1H, ds J=2.0 Hz), 8.65 (1H, d, J=4.8 Hz). (実施例1(12)ターゲットとする多点リン酸化ペプチドの設計)
タウタンパク質のリン酸化部位の中でも特に(i, i+2)の位置でリン酸化を受けている部分配列を採用し、それらを設計・合成した。また、(i, i+4)や(i, i+6)の位置に二つのリン酸基を有しているタウ配列ペプチドもコントロールペプチドとして合成した。さらに、本発明の化合物分子が、特定のリン酸化部位のみを認識可能であるかを評価するため、(i, i+4, i+6)の位置がリン酸化されている三リン酸化ペプチドも同時に合成した(Tau204-216 3P)。図6に合成したリン酸化ペプチドを示す。
全てのペプチドは、ペプチド自動合成機 (ABI 433A, Applied Biosystems)で行った。ソフトウェアは、Standard Fmoc-based FastMoc coupling chemistryを用いた。ペプチド合成に用いたFmoc保護アミノ酸及び試薬は渡辺化学工業から購入した。Amide resin樹脂(導入率0.64mmol / g, 0.1mmolスケール)に対して4等量のFmocアミノ酸(0.4mmol)を合成用バイアルに入れ、自動合成した (N末端アミノ酸の脱保護まで行った)。樹脂からの切り出し及び脱保護は以下の操作に従って行った。得られた樹脂を50mlのナスフラスコに入れ、切り出し・脱保護用試薬(m-クレゾール, チオアニソール, トリフルオロ酢酸)をそれぞれ必要量加え、室温で1時間撹拌した。各試薬の必要量は、樹脂300mgに対し、m-クレゾール 0.06ml, チオアニソール 0.36ml, トリフルオロ酢酸2.58mlである。樹脂をろ別し、ろ液を減圧留去した。TBMEを加え、生じた粗ペプチドの沈澱物をろ別し、デシケーター中で減圧乾燥した。得られた粗ペプチドを蒸留水に溶解させ、不溶物をメンブレンフィルターによりろ過したものをHPLCでの精製に用いた。HPLCの条件は次のとおりである。カラム:ODS-A (YMC社 , 10mm×250mm , 30mm)、移動相: A / B= 5 / 95〜40 / 60 , 40分かけてグラジエント。A:アセトニトリル(0.1% TFA)、B:蒸留水(0.1% TFA)、流速:3ml / min、検出波長:220nm。同定はMALDI-TOF MSにより行い、その結果は次のとおりである。Tau210-220-2P;計算値 ([M-H]-) 1512.61、測定値1506.83。Tau231-238-2P;計算値 ([M-H]-) 1117.39、測定値1111.81。Tau227-238-2P;計算値 ([M-H]-) 1542.47、測定値1552.0。Tau204-216-3P;計算値 ([M-H]-) 1808.47、測定値1813.8。Tau204-216-2P (i,i+4);計算値 ([M-H]-) 1730.72、測定値1722.75。Tau204-216-2P (i,i+6);計算値 ([M-H]-) 1730.72、測定値1733.9。分取したペプチド溶液は凍結乾燥した。
(Tau210-2202Pを用いた検討)
蛍光スペクトル測定により、化合物1及び化合物2のペプチド認識能の評価を行った。
測定は、[化合物1あるいは2] = 10mM、 光路長1cm、 pH7.2、 50mM HEPES buffer、25℃で行った。励起波長は次のとおりである。化合物1はλex = 323nm, Ex / Em = 15nm / 10nm,、Tau210-220-2Pのみλex = 323nm, Ex / Em = 5nm / 10nmとした。化合物2 はλex = 305nm, Ex / Em = 10nm / 12nmとした。図7に示すように、化合物1にTau210-2202Pを添加すると、350nmの蛍光強度が減少し、430nm付近の長波長側の蛍光強度が増加した。この変化はペプチドを過剰に加えていくと飽和をむかえた。それぞれの蛍光変化から会合定数を算出したところほぼ105M-1オーダーであった。一方、図8Aに示すように、化合物2にTau210-2202Pを滴下した結果、蛍光が全体的に緩やかに減少するだけであった。以上の結果から、化合物1における蛍光の二波長変化は、化合物1の二つの亜鉛錯体がペプチド上の二つのリン酸基を認識したためであると示唆された。化合物1及び化合物2自体の蛍光スペクトルを比較すると、化合物1より化合物2が短波長側の蛍光が極端に小さく、長波長側の蛍光が大きいことが明らかとなった(図8B)。測定は、[化合物1あるいは2] = 10mM, λex = 305nm, Ex / Em = 10nm / 12nm、50mM HEPES buffer, 1cm セル, 25℃で行った。この結果から、化合物1のペプチド添加に伴った蛍光の二波長変化は、ペプチド上のリン酸基への結合に伴ったスペーサー(ビニル基)に連結するピリジンへの亜鉛イオンの配位状態の変化が要因であることが明らかとなった。これはスチルバゾール骨格の回転抑制を要因とする当初の予想とは異なる結果であった。
化合物1に、各ペプチドを滴下した際の各波長における結合乗数を図9に示す。二つのリン酸基がi, i+2の距離に位置しているペプチドを滴下した場合には、いずれも蛍光の二波長変化が観測された。また、蛍光変化から算出された各ペプチドとの結合定数も全てほぼ105オーダーの値を示した。一方、二つのリン酸基がi, i+4やi, i+6の距離に位置しているペプチドでは、顕著な蛍光変化は観測されなかった。また、小分子リン酸種であるフェニルリン酸においても、顕著な蛍光変化は観測されなかった。これらの結果の中でも特筆すべきはTau204-216 3Pでは蛍光の二波長変化が生じたが、Tau204-216 2P (i, i+4)やTau204-216 2P (i, i+6)では顕著な蛍光変化が生じなかったことである(図9、図10)。以上の結果から、化合物1は、ペプチド上のi, i+2のリン酸化状態を選択的に認識することが明らかとなった。また、同一配列上に複数のリン酸基が存在する中でも、特定の位置に存在するリン酸基に特異的に結合していることも明らかとなった。
以上の検討により、化合物1にi, i+2の位置にリン酸基を有するタウ配列ペプチドを添加すると、蛍光の二波長変化が生じた。この蛍光変化が結合に伴った変化であるかを評価するため、特にTau210-220 2P、Tau204-216 3P、Tau204-216 2P (i, i+6)の三種類のペプチドを用いてITC測定による親和性の評価を行った。ITC測定の条件は次のとおりである。Tau210-2202P : [化合物1] = 100μM, [peptide] = 2mM。Tau204-216 3P : [化合物1] = 50μM, [peptide] = 1mM。Tau204-216 2P (i, i+6) : [化合物1] = 100μM, [peptide] = 3.24mM。この濃度で滴定回数 : 24回, 測定温度 : 25℃、pH7.2、50mM HEPES buffer中で行った。化合物1にTau210-220 2Pを添加した際の熱量変化を図11に、三種類のペプチドそれぞれにおける熱量変化から得られた各種熱力学的パラメータを図12に示す。ペプチド滴下に伴った熱量変化は、エントロピー駆動的な吸熱過程であった(ΔS > 0、ΔH > 0)。この変化は、化合物分子やリン酸化ペプチドに水和している水分子が結合に伴って脱水和したことに由来すると考えられる。Tau210-220 2PやTau204-216 3Pにおいて得られた会合定数(K = 3.33 x 105 M-1、K = 2.88 x 105 M-1)は、蛍光変化により算出した値とほぼ同等の値であった。この結果は、化合物1にTau210-220 2PやTau204-216 3Pを添加した際の蛍光変化が、結合を反映した変化であることを示している。一方、Tau204-216 2P (i, i+6)では、N = 0.51となり、ペプチド上の二つのリン酸基それぞれに化合物二分子がそれぞれ個別に相互作用していると考えられる。また、算出されたKaもモノZn / Dpa錯体と同等の値となったことも架橋構造ではなく、1 : 2の相互作用をしていることを示唆している。以上の結果から、化合物1がTau210-220 2PやTau204-216 3P上のi, i+2の位置に存在する二つのリン酸基を選択的に架橋認識していることが明らかとなった。
化合物1のTau210-2202Pへの相互作用モードを評価するため、化合物分子へのペプチド添加前後におけるCDスペクトル変化を測定した。CDスペクトル測定は次の条件で行った。[化合物1] = 20uM, [Tau210-2202P] = 0 or 20uM。 pH8.0 Borate buffer, 2mmセル, 室温、スキャンスピード : 200nm / min, 積算 : 10回, レスポンス : 2sec, バンド幅 : 1.0nm。化合物1にTau210-2202Pを添加したところ、化合物分子の吸収極大波長をゼロとし、正と負のコットンピークが観測された(図13A)。この結果は、化合物分子がTau210-2202P上の二つのリン酸基に架橋的に結合することで、ペプチドの不斉が化合物に反映されていることを示す。次に、化合物1へのTau210-2202P添加に伴ったCDスペクトル変化(λ = 291nm)を利用して、Job's Plotを作成し化学量論比の決定を行った。
Job's plotの作成には次の条件で測定を行った。[化合物1] + [Tau210-2202P] = 20uM。 pH7.2 HEPES buffer, 1cmセル, 室温、スキャンスピード : 200nm / min, 積算 : 10回, レスポンス : 2sec, バンド幅 : 1.0nm。[Tau210-2202P] : [化合物] = 1 : 1の場合に最大値をとるようなプロットが得られ(図13B)、Tau210-2202Pと化合物1が1 : 1で結合していることが明らかとなった。以上の検討から、化合物1がTau210-2202P上の二つのリン酸基に架橋認識して1 : 1で結合していることが明らかとなった。
以上の結果をまとめると、化合物分子自身の骨格を剛直にするため、スペーサー(ビニル基)にZn / Dpa錯体を直結させ、スペーサーをDpaのピリジン環の4位に直接導入した新規Stilbazole型化合物を合成した(化合物1及び化合物2)。化合物1は、タウタンパク質の多点リン酸化配列上のi, i+2の位置に離れて存在するリン酸基を架橋認識して1 : 1で結合し、蛍光の二波長変化によって読み出すことに成功した(Ka = 〜105 M-1)。さらに化合物1は、複数のリン酸基が存在する中でi,i+4やi,i+6に位置する二つのリン酸基には結合せず、i, i+2に位置する二つのリン酸基のみを選択的に認識することが明らかとなった。既に報告されている、スペーサーが亜鉛錯体部分の窒素原子に結合している多点リン酸化ペプチド認識化合物では、リン酸基の距離認識性は3倍程度の親和性の差で示されている。詳細はJ. Soc., (2003) 125, 10184-10185, Akio Ojida et al.を参照のこと。すなわち、弱くではあるがリン酸基距離の異なる複数のジリン酸化ペプチドと相互作用する。用いられている基質ペプチド配列が本実施例の配列と異なるため直接的な比較は出来ないが、蛍光強度変化を指標にした場合には、本発明の化合物1は、ジリン酸間における距離認識性が向上していると言える。
本発明に従う別の発光性化合物として、既述の式(12)で表されるBODIPY-Zn(Dpa)(化合物9とする)を以下のように合成した。図14に合成スキームを示した。
100ml三口ナスフラスコに、以下の物質を加え、100℃で攪拌した。3,5-ジヒドロキシ-ベンズアルデヒド 1.0g (7.24mmol)、炭酸カリウム 2.5g (18.1mmol, 2.5eq)、リチウムブロマイド 628.7mg (7.24mmol, 1.0eq)、乾燥DMF 35ml。その後、反応溶液にトリエチレングリコールモノクロロヒドリンの乾燥DMF溶液 (2.31ml (15.9mmol, 2.2eq) / 10ml)を滴下し、100℃で引き続き攪拌した。反応追跡は、TLC (CHCl3 / MeOH = 10 / 1) により行い、五日後に反応の終了を確認した。不溶物をろ別後、ろ液を減圧留去した。酢酸エチル及び10% w/v 炭酸カリウム水溶液を加え、酢酸エチルにより抽出を行った。その後、有機層を10% w/v 炭酸カリウム水溶液により洗浄し、硫酸ナトリウムで乾燥し、溶媒を減圧留去した。精製は、カラムクロマトグラフィー (シリカゲル, CHCl3 / MeOH = 15 / 1)により行い、薄茶色オイル状化合物を得た(収量 (収率):2.75g (98.0%))。同定は1H-NMRにより行った。
二口50mlナスフラスコに化合物11 1.0g (2.57mmol)、2,4-ジメチルピロール514.3mg (0.56ml / 2.1eq)、乾燥CH2Cl2、TFA一滴を入れ、室温で攪拌した。反応追跡は、TLC (CHCl3 / MeOH = 10 / 1) により行い、24時間後に反応の終了を確認した。反応溶液をCH2Cl2により希釈し、飽和炭酸水素ナトリウム水溶液で洗浄した。有機層を硫酸ナトリウムにより乾燥し、溶媒を減圧留去した。精製は、カラムクロマトグラフィー (シリカゲル, CHCl3 / MeOH = 10 / 1)により行い、茶色オイル状化合物を得た(収量 (収率) : 824.2mg (55.7%))。同定は1H-NMRにより行った。
二口50mlナスフラスコに次の物質を入れ、0℃で攪拌した。化合物12 824mg (1.43mmol)、ヨウ素 798.5mg (3.15mmol / 2.2eq)、炭酸カリウム 593mg (4.29mmol / 3.0eq)、メタノール 10ml。反応追跡は、TLC (CHCl3 / MeOH = 10 / 1) により行い、12時間後に反応の終了を確認した。クロロホルムにより抽出を行い、飽和チオ硫酸ナトリウム水溶液により洗浄した。有機層を硫酸マグネシウムにより乾燥し、溶媒を減圧留去した。褐色オイル状化合物を得た(収量 (収率) : 886.3mg (75.1%))。同定は1H-NMRにより行った。
二口50mlナスフラスコに、化合物13 880.0mg (1.07mmol)、DIEA 6.52ml (35eq / 37.5mmol)、乾燥CH2Cl2 15mlを入れ、0℃で10分攪拌した。その後、BF3-OEt2 5.95ml(48eq / 51.4mmol)を加え、0℃でさらに攪拌を行った。反応追跡は、TLC (CHCl3 / MeOH = 10 / 1) により行い、2時間後に反応の終了を確認した。反応溶液を蒸留水及び2N NaOH水溶液により洗浄し、有機層を硫酸ナトリウムにより乾燥し、溶媒を減圧留去した。黒赤色オイル状化合物を得た(収量 (収率) : 866.3mg (92.8%))。同定は1H-NMRにより行った。
脱気アルゴン置換を行った二口50mlナスフラスコに次の物質を入れ、室温で10分攪拌した。化合物14 465.4mg (0.53mmol)、ピリジンボロン酸 739.7mg (1.33mmol / 2.5eq)、dry DMF 10ml、Pd(OAc)2 (S. M.に対して5%mol)、PPh3 (S. M.に対して10%mol)。その後、2M Na2CO3水溶液 2ml加え、70℃で攪拌した。反応追跡は、TLC (CHCl3 / MeOH = 10 / 1, シリカゲル) により行い、2時間後に反応の終了を確認した。溶媒を減圧留去し、残渣にCH2Cl2を加え、不溶成分をろ別し、ろ液を減圧留去した。精製は、カラムクロマトグラフィー (シリカゲル, CHCl3 / MeOH = 10 / 1)により行い、赤色オイル状化合物を得た(収量 (収率) : 283.5mg (58.0%))。同定は1H-NMRにより行った。
二口50mlナスフラスコに、化合物15 280mg (0.30mmol)、AcOH-H2O (3 / 2) 15mlを入れ、加熱還流を行った。反応追跡は、TLC (CHCl3 / MeOH = 10 / 1, シリカゲル) により行い、3時間後に反応の終了を確認した。反応溶液を氷に注ぎ、炭酸カリウムにより中和を行った。その後、クロロホルムにより抽出し、有機層を硫酸ナトリウムで乾燥し、溶媒を減圧留去した。赤色オイル状化合物を得た(収量 (収率) : 56.0mg)。同定は1H-NMRにより行った。
二口50mlナスフラスコに、化合物16 56.0mg (0.067mmol)、乾燥CH2Cl2 10ml、酢酸2滴、アミノメチルピコリン21.4mg (2.5eq)を入れ、室温で10分間攪拌した。その後、NaBH(OAc)3 61.3mg (3.0eq)を加え、室温で攪拌した。反応追跡は、TLC (CHCl3 / MeOH = 10 / 1, NH3aq入り, シリカゲル) により行い、24時間後に反応の終了を確認した。反応溶液に飽和炭酸水素ナトリウム水溶液を加え、クロロホルムにより抽出を行った。有機層を硫酸ナトリウムにより乾燥し、溶媒を減圧留去した。精製は、カラムクロマトグラフィー (シリカゲル, CHCl3 / MeOH = 30 / 1, NH3aq入り)により行い、赤色オイル状化合物を得た(収量 (収率) : 44.7mg (63.6%))。同定は1H-NMRにより行った。
一口50mlナスフラスコに、化合物17 26.5mg (0.025mmol)、単蒸留アセトニトリル / メタノール 10mlを加え、均一溶液になるまで攪拌を行った。その後、305.99mM Zn(NO3)2水溶液 161.91μl (1.95eq)を加え、室温で一時間攪拌した。反応溶液を減圧留去し、少量の蒸留水を加え、凍結乾燥を行った。得られた固体を酢酸エチル及びヘキサンにより洗浄し、減圧乾燥した。橙色固体を得た(収量 (収率) : 30.5mg (84.5%))。同定は1H-NMRにより行った。
タウタンパク質のリン酸化部位の中で(i, i+4)の位置でリン酸化を受けている部分配列ペプチド(Tau 2P)を合成した。231番目のトレオニンと235番目のセリンがリン酸化されている。また、コントロールペプチドとしてリン酸化なしペプチド(Tau 0P)、およびiの位置にリン酸基を有しているモノリン酸化タウ配列ペプチド(Tau 1P)も合成した。ペプチド合成は(実施例1(13))に従って行った。図15に合成したリン酸化ペプチドを示す。
蛍光スペクトル測定により、BODIPY-Zn(Dpa)化合物9(以後、BODIPY-Zn(Dpa)と略す)のリン酸化ペプチド認識能の評価を行った。測定は、[BODIPY-Zn(Dpa)] = 5 mM、pH7.2、50mM HEPES buffer、25℃で行った。励起波長は、λex = 530 nmとした。図16Aに示すように、BODIPY-Zn(Dpa)にTau 2Pを添加すると、540 nmの蛍光強度が増加した。この変化はTau 2Pを過剰に加えていくと飽和をむかえた。蛍光の変化から会合乗数を算出したところ2.75x105 M-1であった。一方、BODIPY-Zn(Dpa)にTau 0PもしくはTau 1Pを滴下した結果、ペプチドの添加に伴って、蛍光強度は緩やかな減少を示したが大きな変化は認められなかった。(図16B)。これらの結果を表9に示す。この結果より、Tau P2添加系における蛍光強度の増加はBODIPY-Zn(Dpa)の二つの亜鉛錯体がペプチド上の二つのリン酸基を認識したためであると考えられる。以上の結果から、BODIPY-Zn(Dpa)はTau 2P、ここでは「i」と「i+4」番目のアミノ酸がリン酸化されたタウペプチドの検出が可能であることがわかった。
タウタンパク質のリン酸化部分配列ペプチドTau 1P、BODIPY-Zn(Dpa)、ならびに231番目トレオニンをリン酸化するGSK−3βを用いたキナーゼアッセイを試みた。実験条件は次のとおりである。Tau 1Pの濃度は20μM、BODIPY-Zn(Dpa)濃度は20μM、 ATPを100μM, BSAを2μg, 50 mM HEPES, 10mM MgCl2, 10mM DTT, 2μM EDTA, pH7.2、30℃とした。励起波長は540 nmとした。結果を図18に示した。図18から明らかなように、時間と共に蛍光強度の増加が確認された。これは、GSK−3βによるTau 1Pの231番目のトレオニンのリン酸化をBODIPY-Zn(Dpa)が認識し相互作用した結果である。また図19から明らかなように、GSK−3βの濃度依存性が認められた。以上の結果より、BODIPY-Zn(Dpa)は2つのリン酸化部位を認識し、キナーゼによるリン酸化をリアルタイムに検出できることが明らかとなった。
脳組織内の過剰リン酸化タウタンパク質の検出が可能かどうかを調査するため、BODIPY-Zn(Dpa) を用いたヒトアルツハイマー(AD)脳ならびに正常脳の海馬組織切片の蛍光染色実験を行った。具体的には、ヒトADもしくは正常脳海馬組織切片を脱パラフィン処理(キシレン浸漬5 minを3回繰り返した後、100%、90%、80%、70% エタノール水溶液に段階的に浸漬(各2 min)し、最後に水洗5 minを2回した。次に、リポフスチン除去処理を以下のように行なった。PBS 洗浄2 min×2 、0.25 wt%過マンガン酸カリウム/PBS浸漬30 min、PBS 洗浄2 min2回、1 wt% Oxalic acid、1 wt% Potassium pyrosulfate/PBS 浸漬6 min、最後にPBS 洗浄2 minを2回。続けて、トリプシン処理(室温でPBS-Tween 洗浄2 min2回、37℃で0.05 % Trypsin/PBSを15 min)、最後に室温でPBS-Tween 洗浄5minを2回した。その後、抗体ならびにBODIPY−Zn(Dpa)で共染色した。まず10%ヤギ血清(37℃, 30 min)でブロッキングを行い、10%ヤギ血清を除去後、1次抗体を添加し、4℃で17-19 時間反応させた。その後、PBS-Tween 洗浄を2 分間を5回(アイスバス上)行い、2次抗体(AlexaFluor 633 ラベルヤギIgG 抗体(抗マウスIgG ))を添加し、37℃、1 h反応させた。PBS-Tweenで2分間、3回洗浄(アイスバス上)した。その後、10 μM BODIPY-Zn(Dpa), 0.0001% DAPI/HBS溶液100 μLを室温, 10 min反応させ、0.5 mM Zn(NO3)2 水溶液で2分間洗浄を2回(アイスバス上)した。その後、PermaFluorTM(Beckman Coulter製)で封入した。本実験では、一次抗体として、以下の抗体を用いた。抗アミロイドベータタンパク抗体(Aβ42;Aβ染色kit(Wako)付属の溶液)。抗リン酸化タウ抗体(AT8;エピトープpSer202/pThr205、1: 5000希釈)。抗タウ抗体(Tau-2;リン酸化タウならびに非リン酸化タウいずれも認識する抗体、1: 5000希釈)。染色切片の観察は、共焦点レーザー顕微鏡観察を用いた。DAPIは励起波長351 nm、蛍光波長は400-500 nmで観察した。BODIPY-Zn(Dpa)は励起波長488 nm、蛍光波長500-555 nm、ならびにAlexaFluor 633は励起波長633nm Laser、蛍光波長645-745 nmで観察した。染色結果を図20に示す。
BODIPY-Zn(Dpa) の蛍光が、AT8(抗リン酸化タウ抗体)とBODIPY-Zn(Dpa) の蛍光が
共存していることがわかった。このことは、BODIPY-Zn(Dpa)がリン酸化タウタンパクに
結合することが可能であることを示している。一方、ヒトAD脳海馬組織において、Aβ42
(抗Aβ抗体)とBODIPY-Zn(Dpa) の蛍光は共存しなかった。このことは、
BODIPY-Zn(Dpa)はアミロイドベータタンパクには結合しないことを示している。さらに興味深いことに、ヒト正常脳海馬組織においては、BODIPY-Zn(Dpa) の蛍光が見られないことから、BODIPY-Zn(Dpa)はアルツハイマー脳特異的な化合物であることが示された。以上の結果より、本発明に従うBODIPY-Zn(Dpa)は、リン酸化タウタンパクを高い選択性を持って捕捉し、染色することが可能であり、一方でアミロイドβ凝集体を認識しない化合物であることが理解される。
BODIPY-Zn(Dpa)がリン酸化アミノ酸を認識して、組織切片上のタウタンパクを染めているのかを明らかにするために、ヒトAD脳海馬組織切片に対し、脱リン酸化酵素(PP2A)処理を行い、BODIPY-Zn(Dpa)染色像の変化を調べた。染色前に、脱リン酸化酵素処理(PP2A; 0.5 units / 50 μL, 37℃, 24 h)を行い、実施例2(12)の手順に従って、海馬組織切片の蛍光染色実験を行った。図21に染色結果を示した。PP2Aで組織上のタウタンパクを脱リン酸化することでBODIPY-Zn(Dpa)染色およびAT8染色で見られた蛍光のドットが消失した。一方でTau-2染色像でははっきりとした蛍光のドットが確認され、PP2A
処理後も組織切片中にタウタンパク凝集体が存在することが確認された。
類似の構造を持つZn/Dpa2核錯体化合物(図23a-d)についてヒトAD脳海馬組織を用いた蛍光染色実験を行った。それらを用いたヒトAD脳海馬組織の染色結果を図24に示した。全てのZn/Dpa2核錯体化合物で抗リン酸化タウ抗体AT8と重なる蛍光のドット(図中矢印)が観測され、これらのZn/Dpa2核錯体化合物がリン酸化タウタンパクプローブとして有用である可能性が示された。
Zn/Dpa2核錯体化合物の血液脳関門(BBB; Blood Brain Barrier)透過性を見積もるために水−オクタノール系での分配係数(Pow)を評価した。Pow > 0.1でBBBを単純拡散で透過する可能性がある。10 μM のZn/Dpa2核錯体化合物水溶液 150 μLに1−オクタノール 150 μLを加え、激しく混合した。遠心分離(1500 rpm)を2分間行った後、水相中の化合物濃度を測定することで分配係数を決定した。図25には実施例2(14)で用いられたZn/Dpa2核錯体化合物a-dの最大吸収波長と分配係数(Pow)を示した。全ての化合物でPow は0.1以上となり、単純拡散によるBBB透過の可能性が示された。
BODIPY-Zn(Dpa)の19F−NMR測定を行った結果、−120 ppmにシングルピークでFのNMR信号を検出することが可能であった。この結果より、本発明の化合物BODIPY-Zn(Dpa)は、リン酸化タウタンパク質あるいはリン酸化タウペプチドを19F−NMRあるいは19F−MRIで検出可能であることが示された。
ICRマウス(male、7週齢)にBODIPY-Zn(Dpa)を静脈投与し、インビボにおける脳移行性を測定した。具体的には、投与量1 mg/kgとなるように、BODIPY-Zn(Dpa) (50 μM HBS溶液,400 μL)を尾静脈より注入し、投与後2分または30分に脳を採取した。脳(424 〜488 mg)は採取後、3 mL のHBS溶液中でスパーテルを用いてホモジナイズし、超音波照射によって均一化した。逆相HPLCにより、脳内に含まれていたBODIPY-Zn(Dpa) の定量を行い、投与量に対する脳内のBODIPY-Zn(Dpa)含有量(% ID/g: % injected dose par gram of brain)を求めた。HPLCでの検出は、化合物由来の蛍光(励起520nm/蛍光545nm)を利用した。
大腸菌により発現させたタウタンパク質(8μM)とヘパリン(1.6μM)を37℃で20日間インキュベートすることでタウ凝集体を調製した。GSK-3βによりin vitroでリン酸化したタウタンパク質を用いて、リン酸化タウ凝集体を同様にして調製した。in vitroのリン酸化は、SDS-PAGE後のPro-Q diamond染色結果から、リン酸化度 = 6 mol P/mol Tauであることを確認した。
に10μMのチオフラビンT(ThTと略す事がある)を添加した。添加後、0.5 mM Zn(NO3)2で2回洗浄した後、染色されたリン酸化タウ凝集体の懸濁液(0.5 μL)をカバーガラス上で風乾させ、共焦点レーザー走査型顕微鏡で観察した(OLYMPUS FV-1000, Obj. lens100x)。チオフラビンTの観察には励起波長458nm、蛍光波長470-490 nmで、BODIPY-Zn(Dpa)の観察には、励起波長488 nm、蛍光波長560-580 nmで行った。
合していることがわかった。図27(n-Tau)からは、ThT蛍光では凝集体が観察されたが、BODIPY-Zn(Dpa)蛍光では凝集体が観察されないことがわかった。この結果は、リン酸基が凝集体上になければBODIPY-Zn(Dpa)では染まらないことを意味しており、BODIPY-Zn(Dpa)はリン酸基を認識してp-Tau凝集体と結合していることが確認された。
図27(Aβ)では、ThT蛍光では凝集体が観察されたが、BODIPY-Zn(Dpa)蛍光では凝集
体が観察されなかった。この結果は、BODIPY-Zn(Dpa)はAβ凝集体を染色せず、pTau凝
集体を染色することを意味しており、p-Tau凝集体への結合選択性を示すものである。以上の結果より、本発明にかかるBODIPY-Zn(Dpa)のリン酸化タウ凝集体への高い結合選択性
が確認された。
BODIPY-Zn(Dpa)に対してリン酸化タウ凝集体、タウ凝集体、Aβ凝集体を滴定し、凝集体が相互作用する濃度範囲を調べた。100 nMのBODIPY-Zn(Dpa)に対し、タウ濃度0 − 320 nM (0 −14 μg/mL)の範囲で滴定を行った。溶媒はHBS (10 mM HEPES (pH7.4), 150 mM NaCl) に10% DMSO、10 μM Zn(NO3)2を含むものを用いた。37°C, 60 分のインキュベート後に蛍光を測定した(励起波長:490 nm、蛍光波長:545 nm)。
リン酸化タウ凝集体、タウ凝集体、Aβ凝集体に対してBODIPY-Zn(Dpa)を滴定することで、EC50(蛍光強度変化の最大値(ΔFmax)の半分の値を示すプローブ濃度)を求めた。一定濃度の凝集体(1 μg/mL)にBODIPY-Zn(Dpa)を滴定し、各濃度における蛍光強度の変化量(ΔF)をプロットした。溶媒はHBS (10 mM HEPES (pH7.4), 150 mM NaCl) に10% DMSO、100 μM Zn(NO3)2を含むものを用いた。37°C, 30 分のインキュベート後に蛍光を測定した(励起波長:490 nm、蛍光波長:545 nm)。

- 二つの2,2’−ジピコリルアミン(Dpa)とスペーサーXとからなる式(1)で表される構造
を有し、該スペーサーXが式(2)、式(3)、式(4)および式(5)のいずれかから選択される化合物。
