本特許出願は、2006年10月23日に出願の米国仮特許出願第60/862540号の優先権を主張する。
本発明は、CB1化合物に関するものである。
(a)鏡像体純度が80%の鏡像体過剰率を超える化合物、
(b)鏡像体純度が90%の鏡像体過剰率を超える化合物、

標題の化合物(1.81kg、8.82モル)をメチルt−ブチルエーテル(3L、2.2Kg)、水(500mL)及び飽和炭酸水素ナトリウム水溶液(500mL)の入った22Lの分液漏斗に満たし、10分間撹拌して明るい黄色の水層を分離し、有機層を22Lのフラスコに移した。硫酸マグネシウム(200g、1.66モル)をフラスコに添加し、10分間撹拌し更に濾過した。濾液を濃縮して油状物とし、標題の化合物を1.64kg(90.6%)の重量の油状物として提供するためアセトニトリル(2×3L)で二回共蒸発させた。1H NMR(DMSO−d6,500MHz):δ8.85(d,1H,J=2.5Hz),8.10(dd,1H,J=2.8Hz),7.81(d,1H,J=8Hz),5.42(s,1H),1.47(s,6H)。

塩の形成:トシレート − 1当量のp−トルエンスルホン酸一水和物を添加し、イソプロパノールから結晶化させた。収率85%、MS(m/z):592(M+1)。
塩の形成:塩酸塩 − 1当量の塩酸を含むジエチルエーテルを添加して塩酸塩を形成させ、イソプロパノールから再結晶化させた。収率63%、MS(m/z):592(M+1)。
塩の形成:トシレート−1当量のp−トルエンスルホン酸一水和物を添加し、イソプロパノール−ヘプタンから結晶化させた。80%、MS、m/zを得る:592(M+1)。
抗体捕捉 SPA GTP−γ− 35 S結合:
ヒト又はラットのCB1又はCB2受容体を発現する細胞膜において、(3R,5R)−3−[1−メチル−1−(6−トリフルオロメチル−ピリジン−3−イル)−エチルアミノ]−1−(4−トリフルオロメチル−フェニル)−5−(3−トリフルオロメトキシ−フェニル)−ピロリジン−2−オン 4−メチルベンゼンスルホネート(「実施例2」と称する)の、アンタゴニスト/逆アゴニストとしての機能的GTP−結合を試験した。改良された抗体捕捉技術(DeLappら、1999)を使用した96穴フォーマットでGTP−γ35S結合を測定した。GTP−結合アッセイ緩衝液(20mMのHepes、100mMのNaCl、5mMのMgCl2、pH7.4)、CB1又はCB2を発現するCHO又はSf9細胞膜(Applied Cell Sciences社、Gaithersburg、MD;PerkinElmer Life Sciences社、ボストン、MA、前述(DeLappら、1999)したとおり膜を調製)実施例2及び500pMのGTP−γ−35S(PerkinElmer Life Sciences社、ボストン、MA)を、室温で30分間簡単に培養した。飽和量の完全アゴニスト(メタナンダミド(methanandamide))の存在下、アンタゴニスト用量反応を行った。0.27% Nonidet P40界面活性剤(Roche社、インディアナ州インディアナポリス)、抗Gi抗体(1:300の最終希釈;Covance社、ニュージャージー州プリンストン)及び1.25mgの抗ウサギ抗体シンチレーション近接アッセイビーズ(GE Healthcare社、ニュージャージー州ピスカタウェイ)を含有する混合物を添加し、プレートを封止し、更に3時間培養した。Beckman GS−6R遠心分離機を使用してプレートを700×gで10分間遠心分離し、Wallac MicroBeta TriLuxシンチレーションカウンタ(PerkinElmer社、マサチューセッツ州ボストン)を使用して1ウェルあたり1分間カウントした。
Kb=IC50/(1+[アゴニスト]/EC50)を使用して決定した

Cheng YC及びPrusoff WH.1973、Relationship between the inhibition constant(Ki) and the concentration of inhibitor which causes 50 per cent inhibition(I50) of an enzymatic reaction(阻害定数(Ki)と酵素反応の50%阻害(I50)を引き起こす阻害剤濃度との関係)、Biochem Pharmacol 22:3099−3108。
NIH雄Swissマウス(Harlan Sprague−Dawley、体重20〜25g)を試験の7〜10日前に入手した。1ケージあたり12匹のマウスを収容した。25〜30gの体重の動物だけを試験した。試験の日、投薬の少なくとも1時間前に、動物を試験室に入れた。投薬の際、各投薬の間に6〜8分間の間隔を置き、マウスにビヒクル(1%CMC、0.5%SLS、0.08%ポビドン、0.05%消泡剤)又は実施例2を経口投与した。次に、マウスを無菌のケージに入れた(1ケージあたり4匹のマウス)。90分後、実施例2又はビヒクルによる90分の前処理に続き、試験を開始した。
NIH−Swissマウスを、6cmまで22〜25℃の水を入れた無菌のプラスチック製円筒容器(直径:10cm、高さ:25cm)に6分間入れた。6分間の試験のうちの最後の4分間における不動時間を記録した。マウスが身動きせず浮いているか、又はその頭を水上に保つために必要な動作のみを行っているとき、不動とみなした。post−hoc Dunnett試験(α=0.05;JMP)を用いて、ANOVAによりデータを解析した。ビヒクルのコントロールに対して統計的に有意な不動時間の減少が観察されるときの、化合物の最小用量として、最少有効用量(MED)を記録した。
バイオアベイラビリティを評価する方法は当該技術分野で高く評価されている。1つのかかる参考文献としては、Medicinal Research Reviews 第21巻 第5号 382−396(2001)が挙げられる。

実験前1週間の間、雄ラット(155〜175g)を収容した。1%カルボキシメチルセルロースナトリウム、0.5%ラウリル硫酸ナトリウム、0.05%消泡剤、0.085%ポビドンを含むビヒクル中懸濁液中の実施例2の製剤を調製し、ラット(例えば雄SDラット、155〜175g)に経口投与(投与量:1又は10mg/kg)した。コントロールのラットには、ビヒクルを投与した。1時間後に、0.4%乳酸のビヒクル溶液中8mg/mLの濃度のクロザピン(Sigma社)を、8mg/kgの投与量で、s.c.投与した。クロザピン又はクロザピン用ビヒクル投与の2時間後に、断頭によって動物を殺した(1群あたりn=7〜8)。迅速に全脳を摘出し、その直後にドライアイス上でイソペンタン(2−メチルブタン)中に浸漬した。前頭前皮質(PFC)、側座核(NAc)及び背側線条体(DL−Str)を通る冠状断面を14μmで裁断し、Fos免疫組織化学検査を実施した。以下の式を使用して、非定型インデックス(Robertsonら、1994)を評価した:
非定型インデックス=(NAcD−NAcV)−(DL−StrD−DL−StrV)
式中、Fos−li神経単位の平均数、NAc=側座核、D=薬剤、V=ビヒクル、DL−Str=背側線条体。更にNewman−Keul’s事後検定(Graph Pad Prism 4.03)により、一方向の分散分析(ANOVA)を使用してデータを分析した。P<0.05を有意水準とした。
クロザピン及び実施例2の製剤はともに、個々に皮質及び皮質下のFos反応性を強化する傾向、又は顕著な強化を示した。実施例2の製剤は、1及び10mg/kg(経口)ともに、クロザピン単独での作用を強化することが観察された。
結論:
実施例2により、ラットの脳の前頭前皮質及び皮質下領域のcFos発現が強化された。クロザピンによっても、神経活性化に関する同様のスペクトラムが示された。クロザピンへの実施例2の製剤の添加により、認識機能及び陰性症状の制御にきわめて重要となる、脳域における、クロザピンにより誘発されるcFos発現の強化がもたらされた。クロザピン対する実施例2の製剤の全体的な効果により、腹側対背側の線条体のインパクトに関する、大きな非定型性の神経サインが生じることが観察された。
方法:
実験の5〜7日前に、カニューレ(BAS社、ウェストラファイエット、IN)を用いて、雄SDラット(260〜300g、Taconic Farms社、Germantown、NY)の前頭前皮質(PFC)に移植した。実験開始の約16時間前に、カニューレにより、カニューレの下に延びる4mmの膜チップを用いて、同心タイプのプローブを挿入し、前頭前皮質から微小透析液を回収し、モノアミン及びその代謝産物に関して分析した。全ての微小透析データを、3回の最終的な薬剤の事前注入における数値の平均(各群5〜6匹のラット)を100%と定義した場合の、透析液のベース濃度からの増減率として算出した。ANOVA、更にpost−hocボンフェローニ試験によりデータを分析した。
実施例2の製剤の投与量がわずか1mg/kg(経口)でも、ラット皮層のモノアミン及びその代謝産物のレベルが増加した。
アルコール嗜好ラットにおける、12時間のエタノール消費(Pラット):
高レベルのエタノール経口摂取量で選抜されたラット(Pラット)におけるエタノール消費を減少させる能力に関して、実施例2を評価した。更に、モチベーション又は挙動のコントロールに対する化合物の効果がエタノールによって評価できる条件下で、実施例2の効果を検討した。
継続して自由にアクセスできるパラダイム下で、雌のアルコール嗜好(P)ラットにおけるアルコール消費に対する実施例2の効果を検討した。比較のため、標準的なオピオイドアンタゴニストであるナルトレキソンを用いて試験した。15%(v/v)エタノールの自発的な消費をモニターした。ビヒクル(1%CMC、0.5%SLS、0.08%ポビドン、0.05%ダウコーニング消泡剤1510 US)中に実施例2の製剤を懸濁し、暗サイクル開始の3時間前にp.o.投与した。更に、赤外線センサにより自発運動を測定した。
水ではなくエタノールの消費が、実施例2の製剤の経口投与によって用量依存的な態様で減少した。ナルトレキソンは、高用量において、エタノールの摂取量を減少させることができた。実施例2の製剤は、10mg/kgまでの投与量で、これらのラットの歩行活動を顕著に変化させなかった。
エタノール摂取量を制御する動機的動因を減少させる、実施例2の能力を評価した。
方法:
応答により15%エタノール(v/v)を生じさせるPRスケジュール下で、雌のアルコール嗜好(P)ラットを検討した。PRスケジュール下では、エタノール配送に必要となる反応を、3回のエタノール供給の後、1〜2回から2回分増加させた。
実施例2の製剤の投与により、エタノール要求行動及び消費エタノール量が用量依存的に減少した。実施例2の製剤は、用量依存的[F(4,20)=4.52、p=0.009]に、区切点(ラットが一定量のエタノールを得るための作業量)を減少させた。
方法:
食餌誘発性肥満(DIO)の雄ロングエバンスラットに、実施例2の製剤を投与した。離乳児の段階から、少なくとも12週間、40%の脂肪、39%の炭水化物及び21%のタンパク質のカロリー含量の食餌を自由摂取させることによって、DIOを作り上げた。T17(17gのビヒクル群との相違を創出するのに必要となる投与量)によって、化合物の効果を定義した。これはすなわち、ビヒクル処理と比較し、2週後の3〜4%の最小生物学的体重減少に相当する。
14日間の検討期間にわたり1日1回実施例2の製剤を経口投与することにより、累積的に摂食が減少した。摂食の減少と一致して、実施例2の製剤による14日間の経口治療後の累積的体重減少から、T17が0.13mg/kgであると推定された。身体組成分析を定量的核磁気共鳴(QNMR)により測定し、最も高い投与量においても除脂肪体重の変化が最小でありながら、0.1〜10mg/kgの投与量範囲で、体脂肪量の顕著な減少を示した。
方法:
痩せたメスの成体SDラットを、通常の齧歯類用の餌であるPurina LabDiet 5001(脂肪12.3%)及び水を自由摂取させる条件で飼育した。1つの群(n=7)を、1〜14日目にビヒクル(1%乳酸)で処理し、一方、残りをオランザピン(2mg/kg、po)を含有するビヒクルで処理した。摂食続いて、2週間の処理期間にわたり、体重及び体脂肪量の変化をモニターした。14日間の薬物送達の後、オランザピン処理された動物(群あたりn=8)を分け、15〜28日目に、第1の群を0.3mg/kgの実施例2の製剤+オランザピンで処理し、第2の群を1mg/kgの実施例2の製剤+オランザピンで処理し、最後の群をビヒクル+オランザピンで処理した。
ビヒクル処理のコントロールと比較し、オランザピンを14日間にわたり1日1回処理した場合において、処理後の累積的な摂食、体重及び体脂肪量の増加を観察した。実施例2の製剤とオランザピンとによる処理では体除脂肪量を変化させることなく、実施例2の製剤を投与した2つの場合において、体脂肪量が顕著に減少した。
ビヒクル処理のコントロールと比較し、実施例2の製剤を14日間1日1回経口投与することにより、高エネルギー餌料を維持した食餌誘発性肥満(DIO)の雄ロングエバンスラットにおいて体重の顕著な減少が見られた。コントロールに比べて体重変化を創出するのに要した実施例2の有効投与量(T17)は、0.13mg/kg/日であると推定された。身体組成の変化を分析した結果、観察された体重減少は、10mg/kg(試験の中で最も高い投与量)までの投与量において、体脂肪量の顕著な減少によるものであることがわかった。2週間にわたるオランザピン処理と並行した、14日間にわたる実施例2の製剤の1日1回経口投与を受けた雌SDラットでは、オランザピン処理コントロールと比較し、体脂肪量の顕著な減少が助長されると評価できる。
