本出願は、参照によりここに援用されるところの、2005年4月11日出願の米国特許仮出願番号第60/670,520号の優先権並びに利益を請求するものである。
本発明は、界面活性剤を用いたタンパク質の精製に関する。
生体高分子、特にタンパク質の生産工程には、物理的ならびに物理化学的性質を利用した高純度化工程を含むことが多い。このような加工工程にあっては、可溶性分子と不溶性分子を分離する条件の決定、比較的低率な各処理過程後の目的分子の収率、処理過程の中での生理活性の喪失、pHなどの加工工程の条件へのタンパク質の感受性を含むが、これに限定されない問題が生じる。
本発明は標的タンパク質および混入タンパク質を含む混合物からの、標的タンパク質の精製法を提供するものであって、混合物を、混入タンパク質を選択的に沈降させるのに効果的な量の陽イオン界面活性剤に曝露させるステップおよび標的タンパク質を回収するステップを含む。

0.1mg/mlのインターフェロンベータ溶液から200μlをカラムに分注した。
タンパク質は陽電荷、負電荷両方を持つため、両性電解質である。溶液のpHおよび、タンパク質と相互作用する荷電分子は、そのタンパク質の正味荷電に影響する。タンパク質の正味電荷が中性(等電点)であるとき、タンパク質間で強い相互作用が起こりえる。溶液のpHがタンパク質の等電点より下である場合、そのタンパク質は正味の正電荷を帯びており、他のタンパク質を含むカチオン性分子との間に静電反発力を生じる場合がある。
以下の実施例は本発明の理解を助けるために提供するものであり、決して本発明を限定することを意図するものではなく、またそう解釈されるものではない。
実施例 1.CPCを用いた、組換え哺乳類ウリカーゼの精製
医薬品グレードのウリカーゼは、基本的に非ウリカーゼタンパク質を全く含まないものでなければならない。E.coliで産生された哺乳類ウリカーゼ(等電点8.67)は、さらなる精製処理を施すために容易に単離できる、封入体(IB)と称される細胞内小器官に似た沈殿物に、細胞内で蓄積した。IBはスクランブル/ミスフォールドされて発現されたタンパク質を含むものと従来考えられてきたが、対照的に、これらのIB様エレメントは正しい折りたたみ構造を有するウリカーゼを沈殿物の形態で含んでいる。ウリカーゼIB様エレメントのアルカリ性pH、例えば約pH9〜11への曝露によって、沈殿しているタンパク質は再溶解した。可溶化されたIB様エレメントのウリカーゼ含有率は約40〜60%であり、均質なウリカーゼ調製物を得るためには、高度な精製を要した。ここに、多様な方法によって評価可能である、CPCを用いた、ウリカーゼならびにその他のタンパク質の精製を示す。例えば、哺乳類ウリカーゼの純度は、比活性度の測定、SDS−PAGEゲルでの電気泳動および染色処理後に現れるバンドの数、HPLCサイズ排除クロマトグラフィー分析で現れるピークのサイズと数によって評価することができる。
1.2.1.50 mM NaHCO3 バッファー (pH 10.3)
このバッファーは、NaHCO3を最終濃度が50mMになるように溶解して調製した。pHは10.2〜10.4になるように調整した。pHの初期値によって、0.1M HClまたは1N NaOHを用いることができる。
1.2.2.10% CPC 溶液
組換え哺乳類ウリカーゼ(尿酸オキシダーゼ)は、参照することによって全文を本明細書に含む、デューク大学の国際公開第00/08196号パンフレットおよび米国特許番号第60/095,489号に記載のとおり、E.coli K−12株W3110 F−で発現させた。
バクテリアは、カゼイン加水分解産物、酵母抽出物、塩、グルコース、アンモニアを含有する成長培地中で、37℃で培養した。
1.2.5.細胞破壊および回収
回収した細胞ペレットをpH8.0の50mMトリスバッファーと10mMのEDTA溶液に懸濁し、最終的に乾燥菌体重量(DCW)のほぼ20倍の体積となるように調製した。撹拌中のペレット懸濁液に、2000−3000 units/mlの濃度でライソザイムを加え、4℃〜8℃で、16〜20時間インキュベートした。
封入体(IB)ペレットをpH10.3〜0.1±0.1の50 mM NaHCO3バッファーに懸濁した。懸濁液を25±2℃で約0.5〜2時間インキュベーションし、IB由来のウリカーゼが溶解できるようにした。
ホモジェナイズしたIB(pH10.3)のアリコットに、目的のCPC濃度が得られるよう、活発に混和しながら10%CPC溶液を加えた。示したとおりに、サンプルを1時間から24時間インキュベートし、この間に沈殿物のフレークが生じた。サンプルを、12,000 x gで15分間遠心した。ペレットと上精を分離し、ペレットは50mM NaHCO3 バッファー(pH 10.3)に、元の体積となるよう懸濁した。それぞれの分画の酵素活性を測定し、分画は濃縮し透析して、残ったCPCを除いた。
処理したIBサンプルと非処理のIBサンプルのアリコットの、タンパク質含有量を、改変ブラッドフォード法によって測定した(Macart and Gerbaut (1982) Clin Chim Acta 122:93−101)。
1.2.9.1.酵素活性
ウリカーゼの活性を、UV法(Fridovich,I.(1965)The competitive inhibition of uricase by oxonate and by related derivatives of s−triazines.J Biol Chem,240,2491−2494を、1mg/ml BSAを含めることで改変)用いて測定した。酵素反応速度は、2つのサンプルで、尿酸のアラントインへの酸化による、292nm波長分光の吸光度の減少を測定することによって求めた。1活性単位は、定められた条件下で、25℃で1分間に1μモルの尿酸を分解するのに要するウリカーゼの量と定義する。ウリカーゼの力価は、1mgタンパク質あたりの活性単位(U/mg)として表す。

VS=反応液に分注した希釈したサンプル(単位はμl)
未変性のウリカーゼ酵素ならびに混入物の、量およびその相対的な割合は、Superdex 200カラムを用いたHPLCによって得た溶出プロファイルより定量された。2つのウリカーゼ溶液のサンプルをカラムに分注した。それぞれのピークならびに総エリアに占める割合は自動的に計算され、これを図の次に示した表にまとめた。
1レーンあたり〜20□gのタンパク質を含むサンプルを、15%SDS−PAGEゲル上で分離した。得られたゲルは、クーマシーブリリアントブルーで染色した。
上精に回収されたウリカーゼ活性および純度への、CPC処理(0.005%〜0.075%、1〜24時間)の影響は、表1および図1に示した。(pH10.3の下での)CPC処理前にあっては、タンパク質濃度は1.95mg/mlであり、比酵素活性度は3.4−4.67U/mgであった。図1Bに示した結果は、それぞれのインキュベーション時間で、CPC濃度が増えるにつれ、上精のタンパク質濃度が減少していることを示している。0.04%以下の濃度では、CPCのタンパク質濃度に与える影響は比較的小さかった。0.04%〜0.075%の濃度では、CPCはタンパク質濃度を元の濃度の約50%にする場合がある。

ウリカーゼを含有するIBを、1.3に記載したとおりの方法で単離し、可溶化した。CPC処理前、およびCPCにより沈殿したタンパク質を濾過で除いた後の可溶物のサンプルを分析した。
可溶化したIBのHPLC分析は、ウリカーゼのピーク(保持時間(RT)〜25.5分)が、粗IBサンプルのタンパク質の約46%を占めることを示した(図2A)。CPC処理後は、ウリカーゼのピークがタンパク質の約92%にまで増加し(図2B)、RT15〜22分の間で溶出する混入物が顕著に減少している(図2A)。ウリカーゼピークのエリアは、図2Aの約70%である。従って、これらの結果は、CPC処理による非ウリカーゼタンパク質の除去によって、ウリカーゼの純度が2倍になったことを示している。
(表2に示した)結果は、ウリカーゼ活性の物質収支が、処理過程を通じて保持されていたことを示している。CPCへの曝露は、溶液中の総タンパク質の60%を沈殿させることが示された。溶液の酵素活性の85%以上が残存したので、外来タンパク質の除去は、生成された上精の比酵素活性度を110%以上増加させたことになる。ほとんどの精製処理でそうであるように、目的の活性の一部は、ペレットに残存した。この例では、元の活性の17.6%のみがペレットに残存し(これを、分析のため50mM重炭酸ナトリウム(7m Si、pH 10.3)で抽出した)、総量に占める割合は比較的低かった。

粗ウリカーゼのCPC処理前および、その後の可溶性物質と不溶性物質の分離、CPC処理、遠心処理による分画、遠心によって得たペレットの再構成、の各処理過程から得た、等量のタンパク質を含むサンプルを、SDS−PAGEによって分析した。結果(図3参照)は、CPC処理前では、混入タンパク質が存在していることを示している。CPC処理後では、ペレットにほとんどの混入タンパク質が含まれており、上精ではウリカーゼが主要な単バンドを形成していた。
2.1.材料と方法
2.1.1.緩衝液
2.1.1.1.封入体溶解液
溶解バッファーは、6M尿素、50mMトリス、1mM EDTAおよび0.1Mシステインを含有する。バッファーのpHは8.5に滴定した。
フォールディングバッファーは、1M尿素、0.25mM NaCl、1mM EDTAおよび0.1Mシステインを含有する。バッファーのpHは10.0に滴定した。
scFv抗体(pI8.9)は、参照することによって全文を本明細書に含むPCT公報WO02/059264に記載の、カルボキシ末端にシステイン−リシン−アラニン−リシンを有するscFvをコードするベクターで形質転換したE.coliで発現させた。
scFvを含有するバクテリア細胞は、誘導を行う前培養として、5時間、最終濃度0.5%となるようにL−アルギニンを加え、pHを7.2に調整した最小培地で培養した。scFv発現の誘導は、培地のグルコース量を限定することで行った。scFvを含有するバクテリア細胞の培地からの回収は、限外濾過によって行った。
回収した細胞ペレットをpH8.0の50mMトリスバッファーと10mMのEDTA溶液に懸濁し、最終的に乾燥菌体重量(DCW)の20倍の体積となるように調製した。撹拌中のペレット懸濁液に、2000−3000 units/mlの濃度でライソザイムを加え、4℃で、16〜20時間インキュベートした。
IBを多く含んだペレットを、封入体溶解バッファー(上記参照)に懸濁し、室温で5時間インキュベーションし、in vitroでアルギニン/酸化型グルタチオンをベースとする溶液でリフォールドさせた。リフォールディング後、タンパク質に透析処理を施し、尿素/リン酸を含有するバッファーに対して、タンジェンシャルフロー濾過によって濃縮した。
scFvを含有するリフォールディング混合物に、最終濃度が0.02%となるよう10%CPC溶液を加えた。室温で1〜2時間インキュベーションした後、沈殿物は濾過によって除去した。上精が、scFv抗体を含有する。
2.2.1.回収可能なscFv抗体への、CPC濃度の影響
(pH7.5または10における)CPC処理の、scFv抗体の純度および回収に与える影響は、表3に示してある。CPC処理前の、IBタンパク質の初期量は73mgであり、Superdex 75によるHPLC分析で15.87mgのscFv抗体を含んでいることが示された。scFv抗体を含有するピークの保持時間(RT)は約20.6分であった。結果は、CPC濃度の増加に伴って総タンパク質の回収量が減少し、CPC濃度が0.03%未満であるときは、scFv抗体の回収量は80%より高い範囲に留まったことを示していた。pH10の条件下に比べて、pH7.5の条件下では、より効率的な混入タンパク質の除去が達成された。従って、scFv抗体の精製は、0.01から0.03% CPC処理によって達成された。

2.3.1.CPC処理後の、 scFv 回収物のHPLC分析
リフォールドされたタンパク質のHPLC分析は、scFv抗体のピーク(保持時間(RT)〜20.6分)が総タンパク質量の22.7%を占めることを示した(図4B)。図4Cのクロマトグラムは、0.02%CPC処理の後に、上精のscFv抗体のピークが、注入した総タンパク質の約75.9%を占めたことを示しており、これは3.3倍の精製となる。従って、CPC処理は、scFv抗体溶液のタンパク質混入物を除去した。
結果(図5参照)は、CPC処理前には、サンプルには多くの種類のタンパク質が、相当量含まれていたことを示している。同様に、CPC処理後では、ペレットに多くの種類のタンパク質が含まれていた。これに比して、CPC処理後の上精は、scFv抗体の、1本の大きなバンドを示した。
インターフェロンベータ(IFN−beta、pI8.5〜8.9)は、既知の方法によって、E.coliで発現させた。Nagola,S.et al.,Nature,284:316(1980)、Goeddel,D.V.et al.,Nature,287:411(1980)、Yelverton,E.et al.,Nuc.Acid Res.,9:731(1981)、Streuli,M.et al.,Proc.Nat’l Acad.Sci.(U.S.),78:2848(1981)、1981年5月6日公開の欧州特許出願番号第28033号、1981年6月15日公開の第321134号、1981年8月26日公開の、第34307号、および1981年7月1日発行のベルギー特許第837379号は、組換えDNA技術を用いたベータインターフェロンの産生法を記載している。バクテリアによって生産したIFNの回収および精製の方法は、米国特許番号第4,450,103号、第4,315,852号、第4,343,735、第4,343,736号、およびDerynckら,Nature(1980)287:193−197および Scandella and Kornberg,Biochemistry,10:4447(1971)に記載されている。IFN−betaを含む封入体を単離し、可溶化した。
a Vydac C4カラムによって定量
b SECプロファイルはいくつかのピークを含んでいた。13分(保持時間13分)で溶出したピークは、CPC処理によって減少し、この領域は高分子量のタンパク質ならびにその変異型が溶出するところにあたる。
CPCを、ヒル凝固Xa因子阻害物質の精製に用いた。ヒル凝固因子阻害剤(FXaI、pI 8.4〜9.1)は、米国特許番号第6,211,341号および国際公開第94/23735号パンフレットに記載されている方法によって生産されうる。FXaIを含有する封入体(IB)の単離の後、実施例1に記したとおりに、FXaIはこの封入体から実質的に精製された。IBペレットの溶解の後、調整物を10%CPC溶液と共にインキュベーションした。この後、混合物を12,000xgで15分間遠心処理した。ペレットと、上精が分離された。ペレットを50 mM NaHCO3バッファーに、元の体積に等しくなるように懸濁した。ペレットと上精は、それぞれ別に濃縮および透析し、残存したCPCを除いた。含有タンパク質ならびに活性を測定し、FXaIが、上精で主要なコンポーネントを占めており、ペレットにはほとんど存在していないことが示された。これらの結果は、CPC処理が、回収したFxaIの収率ならびに純度を向上させたことを示している。
CPBを発現しているクローンから得た等量の封入体を、8M尿素pH 9.5で可溶化した(対照及び試験)。CPBの生成法については、国際公開第96/23064号パンフレットならびに米国特許番号第5,948,668号に記載されている。試験サンプルは、0.11%CPCで処理し、リフォールディング前に濾過によって不溶物を除いた。コントロールならびにテストサンプルのリフォールディングは、溶液をリフォールディングバッファーに1:8で希釈することによって行った。室温で一晩、エンドプロテイナーゼで処理した後、等量のコントロール溶液ならびに試験溶液を、DEAEセファロースカラムにロードした。カラムをウォッシュし、引き続いて活性酵素を、60mM塩化ナトリウムによって、20mMトリスバッファーpH8に溶出した。
(*) タンパク質含有量はブラッドフォード法によって測定した。
(**) リフォールディング前には、タンパク質は不活性であった。
Claims (43)
- 標的タンパク質の精製法であって、
a) 可溶化された標的タンパク質および可溶化された1つまたはそれ以上の混入タンパク質の混合物を含む溶液であり、標的タンパク質はアルカリ条件下で正電荷を帯び、1つまたはそれ以上の混入タンパク質は多価陰電荷を帯びている溶液を得る工程、
b) 前記の可溶化された標的タンパク質および可溶化された1つまたはそれ以上の混入タンパク質の混合物を含む溶液を、1つまたはそれ以上の混入タンパク質を選択的に沈殿させるのに効果的な量の、1つまたはそれ以上の陽イオン界面活性剤で処理し、溶液中に残存する標的タンパク質の比率を大きくする工程、および
c) 前記の可溶化された標的タンパク質を回収する工程。 - 少なくとも1つまたはそれ以上の陽イオン界面活性剤が両親媒性アンモニウム化合物である、請求項1に記載の方法。
- 両親媒性アンモニウム化合物が、一般式QN+で表される第4アンモニウム化合物と、一般式RNH3 +で表されるパラフィン鎖第1アンモニウム化合物、およびそれらの塩から成る群から選択される、請求項2に記載の方法。
- 両親媒性アンモニウム化合物を、セチルピリジニウム塩、ステアリン酸アミド−メチルピリジニウム塩、ラウリルピリジニウム塩、セチルキノリニウム塩、ラウリルアミノプロピオン酸メチルエステル塩、ラウリルアミノプロピオン酸金属塩、ラウリルジメチルベタイン、ステアリルジメチルベタイン、ラウリルジヒドロキシエチルベタインおよびベンゼトニウム塩からなる群から選択する、請求項3に記載の方法。
- 両親媒性アンモニウム化合物が、塩化ヘキサデシルピリジニウム、デカリニウムアセテート、塩化ヒキサデシルピリジニウム、塩化セチルトリメチルアンモニウム、混合塩化n−アルキルジメチルベンジルアンモニウム、塩化セチルピリジニウム、塩化N,N−ジメチル−N−[2−[2−[4−(1,1,3,3,−テトラメチルブチル)−フェノキシ]エトキシ]エチル]ベンゼンメタンアンモニウム、塩化アルキル−ジメチルベンジル−アンモニウム、塩化ジクロロ−ベンジルジメチル−アルキルアンモニウム、臭化テトラデシルトリメチルアンモニウム、臭化ドデシルトリメチルアンモニウム、臭化セチルトリメチルアンモニウム、ラウリルジメチルベタイン、ステアリルジメチルベタイン、ラウリルジヒドロキシエチルベタインから選択される、請求項4に記載の方法。
- 両親媒性アンモニウム化合物がセチルピリジニウム塩である、請求項4に記載の方法。
- セチルピリジニウム塩がハロゲン化物塩である、請求項6に記載の方法。
- セチルピリジニウムハロゲン化物塩が塩化セチルピリジニウムである、請求項7に記載の方法。
- 両親媒性アンモニウム化合物が、6〜20炭素原子を有する脂肪族鎖を少なくとも1本有する、請求項8に記載の方法。
- 脂肪族鎖が炭素原子8〜18個を有する、請求項9に記載の方法。
- 溶液が、さらに1つまたはそれ以上の細胞成分を含む、請求項1に記載の方法。
- 1つまたはそれ以上の細胞成分が微生物由来である、請求項11に記載の方法。

- 標的タンパク質の等電点が7以上である、請求項1に記載の方法。

a) 可溶化された標的タンパク質および可溶化された1つまたはそれ以上の混入タンパク質を含む溶液であって、標的タンパク質はアルカリ条件下で正電荷を帯び、1つまたはそれ以上の混入タンパク質は多価陰電荷を帯びている溶液を得る工程、
b) 前記の可溶化された標的タンパク質および可溶化された1つまたはそれ以上の混入タンパク質を含む溶液を、1つまたはそれ以上の混入タンパク質を選択的に沈殿させるのに効果的な量の、1つまたはそれ以上の両親媒性アンモニウム化合物である陽イオン界面活性剤で処理する工程、および
c) 前記の可溶化された標的タンパク質を回収する工程。 - 下記の工程を含む、複数のタンパク質の溶液において標的タンパク質の割合を増加させる方法;
a.標的タンパク質と混入タンパク質を含んでおり、標的タンパク質が溶液の総タンパク質重量に対して第1の百分率を為している、複数のタンパク質が溶解した溶液であって、標的タンパク質はアルカリ条件下で正電荷を帯び、1つまたはそれ以上の混入タンパク質は多価陰電荷を帯びている溶液を得る工程、および
b.前記の溶液を、混入タンパク質を選択的に沈殿させるのに効果的な量の、1つまたはそれ以上の陽イオン界面活性剤で処理する工程;
ここで、標的タンパク質が工程bの溶液において総タンパク質重量に対して第2の百分率を為しており、この第2の百分率は、第1の百分率よりも大きい。
