1a)末端のシアル酸の非還元末端でタンパク質反応性アルデヒドを形成するために過ヨウ素酸ナトリウムでのCA(大腸菌由来のα-2,8結合PSA)の酸化を示す、そして
2)タンパク質のアミノ基と安定的で不可逆的な共有結合を形成するためにシアノ水素化ホウ素ナトリウム(NaCNBH3)とシッフ塩基の選択的還元を示す。
炭酸アンモニウム、エチレングリコール、PEG(8KDa)、シアノ水素化ホウ素ナトリウム(> 純度98% )、メタ過ヨウ素酸ナトリウム、トリエタノールアミン、塩化ナトリウム、硝酸ナトリウム、アジ化ナトリウム、PBSタブレット及び分子量マーカーはSigma Chemical Laboratory, UKから購入された。使用されたCAである直鎖のα-(2→8)-結合大腸菌K1 PSA(22.7 kDa 平均, 高い多分散度 1.34; 39 kDa, p.d. 1.4; 11kDa, p.d. 1.27)はCamida, Irelandから購入された。他の材料は2,4ジニトロフェニルヒドラジン(2,4 DNPH)(Aldrich Chemical Company, UK)、透析チューブ(3.5kDa及び10kDaカットオフ限度; Medicell International Limited, UK), Sepharose SP HiTrap, PD-10カラム(Pharmacia, UK)、トリス-グリシンポリアクリルアミドゲル(4-20%)、トリス-グリシンドデシル硫酸ナトリウムのランニングバッファー及びローディングバッファー(Novex, UK), Sepharose Q FF及びDEAE(Amersham Biosciences, UK)、トリス-ボラート-EDTA(TBE) ポリアクリルアミド ゲル(4-20%及び20%)、TBEバッファーとローディングバッファー(Invitrogen, UK)を含んでいた。脱イオン水はElgastat Option 4 water purification unit(Elga Limited, UK)から得られた。使用したすべての試薬は分析グレードであった。プレートリーダー(Dynex Technologies, UK)はタンパク質の分光光度測定およびCA分析に使用された。
タンパク質とコロミン酸測定
(シアル酸のような)PSAの定量的推定は他でも述べられているように[Gregoriadis et. al.,1993; Fernandes and Gregoriadis, 1996, 1997]レゾルシノール法[Svennerholm 1957]によって行われた。タンパク質はビシンコニン酸(BCA)比色法によって測定された。
1a CAの活性化
新しく調製された0.02 Mメタ過ヨウ素酸ナトリウム(NaIO4; CAより6倍モル過剰)溶液が20℃でCAと混合され、反応混合物は暗所で15分間磁気撹拌された。酸化されたCAは70%(最終濃度)エタノールを用い、20分間3000gの遠心分離によって沈澱された。上清は取り除かれペレットは最小量の脱イオン水に溶解された。CAは再び70%エタノール沈澱させられ、それから12,000 gで遠心分離された。ペレットは最小量の水で溶解され、凍結乾燥しさらなる使用まで-20℃で保存された(図1; 段階1)。
酸化されたCA(CAO; 22.7kDa)は水素化ホウ素ナトリウムの存在下で還元された。新しく調製された15mMの水素化ホウ素ナトリウム(NaBH4; 希硫酸溶液で希釈することによってpH8-8.5まで希釈されたNaOH中)は20℃でCAO(100mg CA/ml)と混合され、反応混合物は暗所で2時間までの間撹拌された。pHは反応の完了まで7に下げられた。酸化/還元されたCA(CAOR)は4℃でpH 7の0.01%炭酸アンモニウムバッファーに対して透析された(透析チューブに対して3.5 kDa分子量カットオフ)。限外濾過は透析チューブからのCAOR溶液の濃縮のために使用された。濾液は凍結乾燥され、さらに必要となるまで4℃で保存された。どのようなアルデヒド量の測定も’CA酸化の測定’下で述べられているように測定された(図1; 段階2)。
アルデヒド含量がないことを確認した後、CAORが(1時間までの)より長い時間の間過ヨウ素酸溶液とともにインキュベートされた点を除いてはCAの活性化の下で報告されているようにCAORは再び酸化された。CAORO生産物の酸化度もまたこの段階から得られる凍結乾燥粉末で測定された(図1:段階3)。
コロミン酸酸化度の定量的推定は2,4 DNPHで行われ、それはカルボニル化合物との相互作用でわずかに溶解性の2,4 ジニトロフェニルヒドラゾンを生じる。非酸化(CA)、酸化(CAO)、還元(CAOR)及び再酸化(CAORO)(各々5mg)が2,4-DNPH試薬(1.0ml)に加えられ、溶液は振られてから結晶性沈澱が観察されるまで37℃で放置された[Shriner et. al., 1980]。CA(定量的)酸化度はアルカリ溶液中でのフェリシアン化物イオンのフェロシアン化第二鉄(ペルシアブルー)への還元に基づいた方法[Park and Johnson, 1949]で測定され、それは630nmで測定された。この例では、グルコースがスタンダードとして使用された。
(10-100 mg/ml)でCAOは300倍モル過剰のNH4Clを含む脱イオン水の2mlに50 mlチューブ中で溶解され、それからNaCNBH4(1 N NaOH(aq)中の5 Mストック)を最終濃度5 mg/mlで加えられた。混合物は3日間室温でインキュベートされた。コントロール反応はまたCAOの代わりにコロミン酸で行なわれた。生産物のコロミン酸アミン誘導体は5 mlの氷冷エタノールの添加によって沈澱された。沈澱はベンチトップ型遠心分離機で室温下、30分、4000 rpmで遠心分離することによって回収された。ペレットは確保し2 mlの脱イオン水で再度懸濁し、それから10 mlの超遠心分離チューブで5 mlの氷冷エタノールで再度沈澱された。沈澱は室温で30分間30,000 rpmで遠心分離によって集められた。ペレットは2 mlの脱イオン水で再度懸濁され、凍結乾燥された。
TNBS(ピクリルスルホン酸, すなわち2,4,6-トリ-ニトロ-ベンゼン スルホン酸)分析は生産物に存在するアミノ基の量を測定するために使用された[Satake et. al., 1960]。マイクロタイタープレートのウェル中でTNBS(15 mM TNBSの0.5 μl)が0.1 Mホウ酸バッファー pH 9.5の90μlに加えられた。これにCAアミドの50 mg/ml溶液10 μlが加えられた。405nmの吸光度を測る前にプレートは20分間室温で放置された。グリシンはスタンダードとして0.1から1mMの濃度範囲で使用された。TNBSは第一級アミン基をトリニトロフェニル化する。アミンのTNP付加物が検出される。2回の冷却エタノール沈澱で精製した生産物のTNBS分析を使用した検査は90%近い変換を示した。
上の実施例1cで合成されたCAOROは20℃、2時間、0.1M酢酸ナトリウム中でN-[β-マレイミドプロピオン酸] ヒドラジドの5モル当量と反応された。生産物のヒドラゾンはエタノール中で沈澱され、酢酸ナトリウム中で再懸濁し、エタノール中で再び沈澱され、水に再度溶解し凍結乾燥した。生産物はタンパク質とペプチドのシステイン部分のチオール基への位置特異的結合にとって有用である。
CAサンプル(CA, CAO, CAOR及びCAORO)はNaNO3(0.2M), CH3CN(10%; 5mg/ml) に溶解され、 屈折率による検出(GPC system: VE1121 GPC 溶媒ポンプ、VE3580 RI検出器及びTrisec 3 ソフトウェアでの照合(Viscotek Europe Ltd)とともに2x GMPWXLカラムでクロマトグラフされた。サンプル(5mg/ml)は0.45μmナイロン膜で濾過され、移動相として0.2M NaNO3とCH3CN(10%)で7 cm/minで実行された(図3)。
過ヨウ素酸塩と水素化ホウ素での処理後に内部のα-2,8結合Neu5Ac残基の完全性はGPCよって分析され、酸化されたもの(CAO)、酸化還元されたもの(CAOR)、2度酸化されたもの(CAORO)、アミノCA(CA-NH2)材料で得られたクロマトグラフがネイティブのCAのものと比較された。酸化されたもの(15分)(CAO)、還元されたもの(CAOR)、2度酸化されたもの(1時間)(CAORO)及びネイティブのCAは、連続の酸化、還元段階がポリマー鎖の重大なフラグメント化を生じさせるという証拠はなく、ほとんど一致した溶出プロフィールを示すことがわかった(図3)。小さなピークはバッファーの塩を示す。
2.1 大規模の分別
XK50カラム(Amersham Biosciences, UK)は900 mlのSepharose Q FF(Amersham Biosciences)を詰められ、フローレート50ml/minで洗浄バッファー(20mM トリエタノールアミン; pH 7.4)の3カラム容量で平衡化された。CA(200 ml洗浄バッファー中に25グラム)がシリンジ口を経由して毎分50 mlでカラムにかけられた。この後に洗浄バッファーの1.5カラム容量(1350ml)でカラムを洗浄した。
次のサンプルはより小さな規模(1-75ml マトリックス;コロミン酸0.2-3グラム)で同一の洗浄と勾配系を使用して分別された。
CAとその誘導体(22.7 kDa)は集団の異なる%を有する46 kDaまでの分子量平均をもち多分散度が1.1未満の様々な狭いスピーシーズに首尾よく分別された(表1-2と図4-8)。表1は75mlの規模での22.7kDaの材料を分別した結果を示す。図7はGPC結果であり、図4-6はCA分画のnative PAGEである。
CAの分別に影響を与える様々な因子(例えば洗浄容量など)は研究された[図7-14]。
CAの分別に影響を与える様々な因子が研究された。結合研究はカラム(5mlマトリックス)上へのCAの50、100、150及び200mgのローディングによってなされた。CAの200mgを使用して、CAの99%超がカラムに結合した(図9)。段階的勾配によって、溶出バッファーの1カラム容量でカラムが洗浄された時、ポリマーの多分散度は1.1を超えることが分かった(図11)。1.5カラム容量でのカラムの洗浄は1.1未満の多分散度を有するCA分画を生じた(図10)。
参考例2で調製されたCAO(22.7 KDa)と狭分散-CAO(27.7kDa pd= 1.09; 40.9 kDa. pd = 1.02)はGH複合体の調製のために使用された。
成長ホルモンは0.15 M PBS(pH 7.4)に溶解され、異なるCA(CAO及びNCAO)に共有結合された。異なるCA(22kDa, CAO; 27.7kDaと40.9kDa, NCA)は個々にCA:GHモル比(12.5:1)でGH(2mg)に加えられ、シアノ水素化ホウ素ナトリウムが4 mg/mlの最終濃度で加えられた。反応混合物は密閉され35±2℃で24時間磁気撹拌された。その混合物はそれから70% w/v飽和度に達するまで連続的に撹拌しながらゆっくり塩を加えることによる硫酸アンモニウム((NH4)2SO4)沈澱にかけられて、4℃で1時間撹拌され、それから15分間回転(5000xg)させ、ペレットは(NH4)2SO4の飽和溶液に再懸濁され、15分間再度回転させた(5000xg)。回収された沈澱は1 ml PBS pH7.4に再度溶解され、同じバッファーに対して4℃で長く(24 h)透析された。コントロールは非酸化CAの存在下またはCAの非存在下でネイティブのタンパク質を複合体化手順にかけることを含んでいた。タンパク質の付随した変性を避けるために振盪は最小限にしておいた。ポリシアリル化GHはSDS-PAGEによって特徴付けされた。ポリシアリル化GHはアニオン交換クロマトグラフィーを通過し、生産物の分画はSDS PAGEにかけられた(図15)。
GH-CA複合体は首尾よく合成された。SDS-PAGEの結果(図15)は(GHと)コントロールでのサンプルの移動は新しいGHのそれとよく似ていることを示す。複合体レーンではポリシアリル化-GHを示す質量の増加を典型的に示すバンドの移動がある。バンド幅は広分散ポリマーとの複合体に比較して狭分散ポリマーとの複合体の場合には顕著に狭くなる。さらに、(広分散ポリマーとの)GH複合体はアニオン交換クロマトグラフィーによって異なるスピーシーズに首尾よく分離された(図19)。
ディファレンシャルエタノール沈澱はコロミン酸の異なる鎖長を沈澱させるのに使用された[図16, 17]。
CA(22.7; pd 1.34)は異なるエタノールの濃度を使用して狭分散の分画に沈澱することはできなかった(図17)。しかしながら、ディファレンシャルエタノール沈澱はより小さく幅が狭いCAはより多くのエタノール(EtOH)を必要とすることを示した。幅が広い22.7 kDaのポリマーは生産物ポリマーの収量>80%を与えて70% EtOHで沈澱した。80%EtOHの濃度はより低いMW6.5KDa(pd< 1.1)の> 80%を沈澱させるために必要とされた。この方法はまた生産物に混入した塩の一部を取り除く。
22.7kDaのサンプルは異なる分子量カットオフ膜(5, 10, 30, 50及び100 kDa)上での限外濾過により精製された。すべての場合で濃縮液はGPCとnative PAGEによって検査された[図18]。
異なる分子量カットオフ膜上での限外濾過によって精製された22.7 kDaのサンプルは膜のカットオフの増加に伴いポリマーの多分散度の減少とより高分子量へのシフトがあったことを示した(図18)。図16はアニオン交換クロマトグラフィー(左)と濾過(右)によるCAの分別を示す。CAのIECは濾過による分別に比べてずっと狭い分散のCAを有する分画を生じた。
分別されたCAポリマーはD2Oを使用して(もしあるなら)不純物に対して1H(400 MHz)と13C(100 MHz)NMR分光法によって特徴付けされた(図20)。
狭分散ポリマーの分別された物質の1Hと13C NMRには不純物がない。加えて、1H NMRスペクトルでのH-3プロトンと13C NMRスペクトルのC-2炭素の化学シフトはポリマーが実際に期待されるα-2,8結合シアル酸物質であることを確認する。
8.1 大規模での分別
XK50カラム(Amersham Biosciences, UK)は900 ml Sepharose Q FFで詰められて、50ml/minのフローレートで洗浄バッファー(20mM トリエタノールアミン; pH 7.4)3カラム容量で平衡化された。CA(200 ml洗浄バッファー中に12.5g)がシリンジ口を経由して毎分50 mlでカラムにかけられた。この後に洗浄バッファーの1.5カラム容量(1350ml)でカラムを洗浄した。
CAはまた異なる塩濃度(0, 200, 250, 300, 350, 375, 400, 425, 450, 475, 500, 525, 550及び575mM NaCl)を含む同一のバッファー系(20mMトリエタノールアミン; pH 7.4)を使用するSepharose Q FF(5ml マトリックス、あらかじめ詰められている; Amersham Biosciences, UK)を使用して小規模(CAの200mg)で分別された。結合したCAは通常のトリエタノールアミンバッファー中の1000mM NaClを使用したカラムの最終洗浄とともに1ml/minのフローレートで1.5カラム容量(7.5ml)でカラムを洗浄することによって溶出された。
図21と23はそれぞれ大規模(12.5 g CA; 900 mlマトリックス)と小規模(200 mg CA; 5 ml マトリックス)での39 kDa CAポリマーを分別した結果として様々な分画のCAの%集団を示す。図22と24は大及び小規模でのCAの分別の結果として得られたnative PAGEである。表4は様々な塩濃度でのIECによって得られた分画のGPC結果である。GPCデータは97 kDaまでのスピーシーズが分別方法によって生じていることを示す(さらに詳細は実施例9を参照)。これらのより大きな分子量のポリマーはホスフェート分析によってより低い分子量のものに比べてより大きな割合でCAの還元末端に存在するホスフェート部分を有することが示され、それは無機リンの存在の検査である(Rouser et. al. 1970)。
例えばCA、CAO、CAOR又はCAORO(4-5 mg/ml)のどれかを0.2 M NaNO3/0.1% NaN3/10% アセトニトリル(1 ml)(または、他には10mM PBS, pH 7.4)中に溶解し、それから0.2μm ナイロン膜(Whattman, UK)上で結果溶液を濾過することによって新しく調製されたCAサンプルは、、GPCによって分析された。

10.1 native PAGEによるCAの分析の最適化
分別された及び分別されていないCAはゲルでのこれらのポリマーの分離を最適化するためにnative PAGEでTBEとトリス-グリシンゲルによってさらに分析された。一般的に、狭又は広分散のCAの40μgが、ゲルのウェル毎にローディングバッファーの10μl を含む20μl としてロードされた。ゲルは3種の異なる速度(150, 25, 15 mV/cm)で作動され、それからアリカンブルーで染色され、その後に2%酢酸で脱色された。
CA(22.7 kDa, pd 1.34)のIEC分別から得られた高圧限外濾過の濾液はVivaflow(mwco 3 kDa)により濃縮された。これらのCAサンプルはポリマーのどんな分解に対しても4-20% TBEゲルを使用したnative-PAGE(図27)によって高圧限外濾過の対応するフィルターとともに分析された。
多分散CAはまたNaClのようなどの塩も不存在下でpH 7.4でのトリエタノールアミン濃度範囲を使用してSepharose Q FF(1ml マトリックス、あらかじめ詰められている)によって分別された。従って、CA(40mg; 1ml)(22.7kDa; pd 1.34)はQ FFカラム(1ml マトリックス; あらかじめ詰められている; Amersham Biosciences)にロードされた。結合したCAは1ml/minのフローレートで各々のトリエタノールアミンバッファー(50, 100, 200, 300, 400, 500, 600, 700, 800, 900, 1000, 1100及び1200mMトリエタノールアミン)の1mlをカラムに通すことにより2000mMトリエタノールアミンを使用したカラムの最終洗浄を伴って溶出された。サンプルは直接凍結乾燥され、それからnative PAGE(アルシアンブルーによって染色されたTBEゲル)によって分析された。

HEPESとエタノールアミンバッファー系はpH段階的勾配を作るために使用された。pH 7.6, 7.8及び8.0でのバッファーはHEPESの濃度を10から50mMまで増加し、pHをNaOHでおおよその値に設定することを用いて調整された。ナトリウムイオンの濃度は36mMを超えなかった。pH 8.2, 8.3, 8.5, 8.7, 8.9, 9.1, 9.3, 9.5及び9.7は1M エタノールアミン(20から70mM最終濃度)のおおよその量をHEPES 50mM(10から50mM最終濃度)と混合し、ナトリウムイオン濃度が30mMを超えないことを確認しながら、pHをNaOHで調整することにより作られた。最終のバッファーは70mM エタノールアミン pH 11であった。

Claims (21)
- 多分散多糖の水溶液がカラム中のイオン交換樹脂に接触し、該多糖は次に水性溶出バッファーによって選択的溶出に供されて、多糖が溶出分画から回収される、多分散系のイオンにより電荷を有する多糖を異なる平均分子量の分画に分離する方法であって、
選択的溶出がカラムの樹脂を各々が連続的に異なる一定のイオン強度及び/又はpHを有する少なくとも3種類の溶出バッファーで洗浄することを含み、2番目とそれに続くバッファーは直前の段階のバッファーより高いイオン強度及び/又はpHを有することを特徴とし、
前記連続するバッファーのイオン強度間の相違が実質的に同じであり、
前記の相違が0.005から0.1Mであり、
前記分画が1.1以下の多分散度を有する多糖を含み、該多糖が(i)シアル酸ポリマー、(ii)シアル酸コポリマー、(iii)(i)又は(ii)の加水分解物、又は(iv)(i)、(ii)、又は(iii)の官能化誘導体であり、
前記多分散多糖の重量平均分子量が少なくとも5kDaである、方法。 - 溶出バッファーがすべて同じpHを有する請求項1に記載の方法。
- イオン強度が強無機酸と強無機塩基の塩の濃度を変化させることによって変えられる請求項1又は2に記載の方法。
- 溶出バッファーが連続的により高いpHの値を有する請求項1に記載の方法。
- 連続する段階のpH間の相違が実質的に同じである請求項4に記載の方法。
- 前記の相違が0.2pH単位である請求項5に記載の方法。
- 各々のバッファーのpHが7.4−13の範囲にある請求項1〜6のいずれかに記載の方法。
- 2番目とそれに続くバッファーは直前の段階のバッファーより高いイオン強度及びpHを有する請求項4〜7のいずれかに記載の方法。
- 各々の溶出バッファーが少なくとも1.0カラム容量の量で使用される請求項1〜8のいずれかに記載の方法。
- 溶出バッファーが揮発性の酸又は塩基を含みかつ回収が溶出分画からの揮発性の酸又は塩基の揮発を含む請求項1〜9のいずれかに記載の方法。
- バッファーがアミンである塩基を含む請求項10に記載の方法。
- イオン交換樹脂が強い又は弱いアニオン又はカチオン交換媒体である請求項1〜9のいずれかに記載の方法。
- 選択的溶出が少なくとも5段階を含む請求項1〜12のいずれかに記載の方法。
- 最初の溶出バッファーのイオン強度が0.001から1Mの範囲にある請求項1〜13のいずれかに記載の方法。
- 多糖が溶出分画で実行される最初の限外濾過の段階を含む段階によって回収される請求項1〜14のいずれかに記載の方法。
- 多糖が乾燥により溶液から最終的に分離される請求項1〜15のいずれかに記載の方法。
- イオン交換樹脂と接触する前に非溶媒沈澱の予備的な段階を含む請求項1〜1619のいずれかに記載の方法。
- 多糖がポリ(2,8−結合シアル酸)、ポリ(2,9−結合シアル酸)又は交互の2,8−2,9−結合PSAである請求項1〜17のいずれかに記載の方法。
- 多糖がCA、又はそれの酸化、還元、アミノ化及び/又はヒドラジド誘導体である請求項18に記載の方法。
- 分画された多糖をタンパク質に結合させることを含む請求項1〜19のいずれかに記載の方法。
- 分散系のイオンにより電荷を有するタンパク質−多糖複合体を分離する請求項1〜20のいずれかに記載の方法。
