Description

本出願は、2007年5月17日に出願された米国仮特許出願第60/930,585号の優先権の利益を主張し、この内容全体は参照により本明細書に組み込まれる。
本発明は、PVDF(ポリフッ化ビニリデン)基材に熱結合しているポリスルホンまたはポリエーテルスルホン膜を含む膜構造体に関する。
現在のところ、濾過装置を製造する現場において予備滅菌され、濾過装置を使用する現場において無菌化される、生物学的流体からウイルスを除去する濾過装置を提供することが望まれている。一般に、製造現場における滅菌はガンマ線照射により実施され、無菌化は使用の現場において苛性アルカリにより実施される。苛性アルカリによる無菌化は、最初に苛性アルカリ水溶液(典型的には0.1規定の水酸化ナトリウム)により、次いで滅菌水により、次いで滅菌緩衝液により実施される。従って、濾過装置がガンマ線照射および苛性アルカリの両方に起因する分解に対して耐性であることが必要である。
濾過装置は、一般にポリスルホン(PS)膜またはポリエーテルスルホン(PES)膜を含む濾過膜が結合している支持プレートを含むエレメントを利用する。なぜなら、このエレメントにより高性能限外濾過(UF)を実施することができるからである。
現在のところ、PSまたはPES膜はアクリル樹脂基材に熱結合させることができることが公知である。アクリル樹脂は耐苛性アルカリ性でないので、苛性アルカリによる滅菌を要求する方法におけるアクリル樹脂の使用は望ましくない。
PSまたはPES膜は、ポリスルホン基材(プレート)に加熱により容易に結合させることができる。なぜなら、材料が類似しているからである。しかしながら、プレートを一緒に結合させることは困難である。プレートを一緒に結合させるために典型的に使用される方法は、接触溶接と称される。この方法は、2つのプラスチックプレートをプラスチックが溶融し始めるまで加熱器に接触させることを含む。次いで、プレートを加熱器から取り出し、加熱器を移して離し、プレートを一緒に加圧する。この方法は、材料が冷却したときに2つのプレートが完全に溶接されるように、2つのプレートを一緒に加圧するまで溶融したままにするためのプラスチック材料を要求する。この方法を使用してポリスルホンまたはポリエーテルスルホンプレートを同一または類似の材料のプレートに溶接することは困難である。溶融したPSまたはPESは、加熱エレメントが溶融したPSまたはPESから除去されたとき、プレートとプレートとの良好な結合の形成を困難にする薄膜を急速に形成することが見出されている。
PSまたはPESプレートを一緒に溶接する他の方法が試みられており、困難であること、または完成品に対して何らかの悪影響を生じさせることが判明している。他の方法の幾つかは、レーザ溶接、無線周波数(RF)溶接、振動溶接、超音波溶接および溶媒溶接を含む。レーザ溶接およびRF溶接は、十分に強い結合を生じさせない。振動溶接および超音波溶接は、不所望なPSまたはPES粒子の生成をもたらす。塩化メチレンのような溶媒による溶媒溶接は、膜が類似の材料から構成されており、溶媒により変化または溶解し得るので困難である。
従って、接触溶接することができ、上述の悪影響を有しない、基材に熱結合している膜を含む濾過カートリッジのエレメントを提供することが望まれる。さらに、ガンマ線照射または苛性アルカリへの曝露による分解に対して耐性でもあるこのようなエレメントを提供することが望まれる。
本発明は、ポリフッ化ビニリデン(PVDF)基材に熱結合しているPSまたはPES膜を有する濾過カートリッジエレメントを提供する。さらに、2つ以上のPVDF基材を容易に単純に一緒に結合させる。熱結合は、加熱エレメントをPSもしくはPESまたはこれらのコポリマーまたはこれらのブレンドを含む膜に圧力下で当て、次いでこの膜をPVDF基材と接触させてPVDF基材を溶融したPVDFがPSまたはPES膜ポアに浸入する程度に部分溶融させることにより実施する。加熱エレメントを除去し、溶融したPVDFを冷却して凝固させる。得られた膜の基材への結合は、膜を基材から取り出そうとすると基材に結合したままの膜残留物をもたらすほど十分に強い。さらに、2つ以上のPVDF基材を互いに熱結合させることができる。

本発明によれば、膜は、任意の構成、例えば対称または非対称(薄膜化(skinned)または非薄膜化(unskinned))などの構成を有することができる。さらに、膜は、任意の多孔度、例えば逆浸透(RO)膜、限外濾過(UF)膜、精密濾過(MF)膜などの多孔度を有することができる。PSまたはPES膜は、一般に約428°F(140℃)から約446°F(230℃)の融点を有する。このような膜は公知であり、Millipore Corporation of Billerica、Massachusettsを含む多くの製造業者により、Millipore Express(登録商標)膜のような商標のもと作製されている。好ましい膜は、実質的に同時に互いに流延させる2種以上のポリマー溶液から複合膜を形成する共流延法により作製される単位膜である。共流延法により、特有の膜、例えば非対称または対称の2つの区域を有する膜、対称の区域を有する非対称の区域を有する膜などを製作することができる。これらの区域のそれぞれのポアサイズおよび厚さを変えることもできる。US7,208,200は、このような膜を作製する1つの方法を示している。
PSおよび/またはPES膜が結合しているPVDF基材は、供給物、透過物および/または保持物のための内部ポート、ならびに所望の流体流を実施するための流体流路を含むように設計することができる。PVDFは、一般に約284°F(140℃)から約356°F(180℃)の融点を有する。1つまたは複数のPSおよび/またはPES膜のPVDF基材への結合を実施するため、加熱エレメントを提供する。膜の基材への結合が悪影響を受けないように、膜と接触する加熱エレメントの部分に抗粘着性表面、例えばポリテトラフルオロエチレン(PTFE)を提供することが好ましい。加熱エレメントは約490°F(254℃)から約530°F(277℃)の温度に加熱し、加熱エレメントは膜に、PVDF基材の少なくとも一部と重なる所望の結合領域において膜と基材との密着を確保するために圧力下で当てる。圧力下での加熱は、PVDFを所望の結合領域において溶融させ、溶融したPVDFを膜ポア中に浸入させるのに十分な時間実施する。次いで、加熱エレメントを膜との接触から除去し、得られた積層体を冷却させて溶融したPVDFを凝固させる。
次いで、膜を所望により含有することができ、または含有することができない第2のPVDF基材を、PSまたはPES膜が上述の通り既に封止されている第1のPVDF基材近傍に移動させる。第2の基材は、供給物、透過物および/または保持物、ガス排気などのための1つまたは複数のポートを所望により含有してよい。
第2の基材は加熱エレメントにより加熱し、次いで膜を含有する第1のPVDF基材の表面に対して加圧して第1のPVDF基材および第2のPVDF基材を一緒に封止する。
以下の実施例は本発明を例示するものであり、本発明を限定するものではない。
図1に示す通り、ポリエーテルスルホンUF膜10を、流体流出口14を有する円形のPVDF支持プレート12に結合させた。使用した加熱エレメントの温度は、510°F(265℃)から540°F(282℃)であった。封止時間は、20psigから30psigの圧力において4秒から10秒であった。次いで、流体流入口18を有する円形のPVDF上部プレート16をPVDF支持プレート12に周縁部20において熱結合させ、全ての流体供給物が流出口14を導通する前に膜10を導通することを要求されるPES膜10、流体流入口18および流体流出口14を有する濾過カートリッジを形成した。好ましくは、上部プレート16を支持プレート12に、膜10が支持プレート12に結合している領域の外側において結合させる。
本実施例によるデバイスは、支持プレートに一体的に封止されている膜および下部プレートに液密的に一体的に封止されている上部プレートを提供する。製造は単純、迅速および容易であり、過去の問題を回避する一方、ガンマ線照射および苛性アルカリの両方に安定であるデバイスを提供する。
必要に応じて図示の通り、別のポート19を一方(図示)または両方のプレート12、16上で使用することもできる。ポート19は、ポート19に結合している適切なガスフィルタ(図示せず)、例えばMillipore Corporation of Billerica、Massachusettsから入手可能なMILLEX(登録商標)ガス排気フィルタによるガス排気に使用することができる。または、ポート19は、タンジェンシャルフロー濾過において見られるような再循環のための供給に後に戻すことができる保持物のための導管(図示せず)に連結させることができる。
図1は丸型のデバイスを示しているが、他の型を使用することもできる。例えば、図2は、カセット式フィルタデバイスに使用されることが多い典型的なプレートおよびフレーム設計を示している。図2において、PSまたはPES膜30は、第1のカセットフレーム34の表面32に封止ライン36により図示の通り結合させる。次いで、第2のカセットフレーム38を第1のカセットフレーム34に、膜が第1のカセットフレーム34に結合している領域36の外側において結合させる。種々のポート40は、カセットフレーム34および38中に示している。2つ以上のフレームを使用してよい。複数のカセットを互いに積層して好適に寸法化されたデバイスを形成することができる。カセットは、少なくとも最も外側のカセット上に各端部上で封止されている外表面を有してよく、またはカセットは、当該技術分野において公知である、カセットの外表面に類似の様式で結合している同様にPVDFから構成されている別個の追加の一体型端部プレート(図示せず)を有してよい。

Claims (18)

  1. ポリフッ化ビニリデンを含む基材に熱結合しているポリスルホン、ポリエーテルスルホン、これらのブレンドおよびこれらのコポリマーからなる群から選択される少なくとも1つの多孔性膜を含む積層体。
  2. 前記膜がポリスルホンを含む、請求項1の積層体。
  3. 前記膜がポリエーテルスルホンを含む、請求項1の積層体。
  4. ポリフッ化ビニリデンを含む基材に熱結合しているポリスルホンおよびポリエーテルスルホンのブレンドを含む少なくとも1つの多孔性膜を含む積層体。
  5. ポリフッ化ビニリデンを含む基材に熱結合しているポリスルホンおよびポリエーテルスルホンのコポリマーを含む少なくとも1つの多孔性膜を含む積層体。
  6. 複数の膜を有する、請求項1の積層体。
  7. 複数の膜を有する、請求項5の積層体。
  8. 前記基材が少なくとも1つの流体流路を含む、請求項1の積層体。
  9. 前記基材が少なくとも1つの流体流路を含む、請求項3の積層体。
  10. 前記基材が少なくとも1つの流体流路を含む、請求項5の積層体。
  11. ポリスルホン、ポリエーテルスルホン、これらのブレンドおよびこれらのコポリマーからなる群から選択される少なくとも1つの多孔性膜、ポリフッ化ビニリデンを含む第1の基材(膜が第1の基材の表面上に熱結合している。)、およびポリフッ化ビニリデンを含む第2の基材(膜を含む第1の基材の表面に結合している。)を含むデバイス。
  12. 第1の基材が、該基材の面上に膜が結合している表面の反対側において形成されたポートを有し、ポートは、第1の基材を通り膜に隣接する領域に達する開口部を有し、ならびに第2の基材が、該基材の面上に第2の基材が第1の基材に結合している表面の反対側において形成されたポートを有する、請求項11のデバイス。
  13. 濾過デバイスを形成する方法であって、ポリスルホン、ポリエーテルスルホン、これらのブレンドおよびこれらのコポリマーからなる群から選択される少なくとも1つの多孔性膜を提供すること;第1の支持プレートおよび第2の支持プレートを提供すること(第1の支持プレートおよび第2の支持プレートはPVDFから形成されており、支持プレートは互いに面する第1の表面および各プレートの第1の表面の反対側の第2の表面を有し、ならびにプレートは各プレートの第1の表面から第2の表面に形成された少なくとも1つのポートをそれぞれ有する。);少なくとも1つの膜を第1の支持プレートの第1の表面に熱結合させること;および第1のプレートの第1の表面を第2のプレートの第1の表面に、膜が結合している第1のプレートの第1の表面の領域の外側の領域内で熱結合させることを含む、方法。
  14. 第1のプレートの第1の表面を第2のプレートの第1の表面にそれぞれの周縁部において結合させる、請求項13の方法。
  15. 約490°F(254℃)から約530°F(277℃)の温度に加熱した加熱エレメントを提供すること、および加熱エレメントを膜に、第1の支持プレートの第1の表面の少なくとも1部と重なる所望の結合領域において当てること、および加熱エレメントを除去すること、および結合を冷却させることをさらに含む、請求項13の方法。
