Description
マンナン含有量36%、水分8.2%のパームカーネルミール(不二製油株式会社製)100部に、酵素ヘミセルラーゼGM「アマノ」(天野製薬株式会社製)0.25部を溶解した酵素液150部を、60℃で12時間作用させた後、流動層乾燥装置(大河原製作所製)にて水分9.8%まで乾燥させ、乾燥粉体102部を得た。この乾燥粉体のマンノース含有量及びβ−1,4−マンノビオース含有量をイオン交換クロマトグラフィ法で測定したところ、マンノースが1.44部、β−1,4−マンノビオースが10.58部(マンナンに対し29.4%;乾物換算で11.5%)生成していた。
コプラミール(マンナン含有量30%、水分4.2%)100部に、酵素ヘミセルラーゼGM「アマノ」(天野製薬株式会社製)0.25部を溶解した酵素液150部を、60℃で12時間作用させた後、流動層乾燥装置にて水分9.3%まで乾燥させ、乾燥粉体106部を得た。この乾燥粉体のマンノース含有量及びβ−1,4−マンノビオース含有量を測定したところ、マンノースが1.36部、β−1,4−マンノビオースが12.35部(マンナンに対し41.2%;乾物換算で12.9%)生成していた。
パームカーネルミール(マンナン含有量36%、水分8.2%)100部に、酵素セルロシンGM5(エイチビィアイ株式会社製)0.3部を溶解した酵素液150部を、60℃で72時間作用させた後、流動層乾燥装置にて水分7.8%まで乾燥させ、乾燥粉体100部を得た。この乾燥粉体のマンノース含有量及びβ−1,4−マンノビオース含有量を測定したところ、マンノースが11.52部、β−1,4−マンノビオースが2.57部(マンナンに対し7.14%;乾物換算で2.79%)生成していた。
各種糖類のサルモネラ菌凝集抑制試験を以下のように行った。サルモネラ菌サルモネラ・エンテリティディス(伊藤忠飼料株式会社にて分離したSalmonella enteritidis KTE−61株 菌数1×108cfu/ml)の懸濁液0.1ml及び蒸留水0.1mlを十分混合させ、これに酵母キャンディダ・アルビカンス(社団法人動物用生物学的製剤協会より購入したCandida albicans KI−102001)懸濁液0.1mlを加え混合した。この混合液に凝集が生じることを実体顕微鏡(オリンパス社製)で確認する予備試験を行った。
実施例1、実施例2及び比較例1で得られた乾燥粉体に水を加え10倍量の懸濁液とした。また、マンノース試薬の1%水溶液を準備した。以上4点の懸濁液又は水溶液に鶏盲腸内容構成菌培養物であるインテクリーン(伊藤忠飼料株式会社製)を接種し、37℃にてマンノース、β−1,4−マンノビオースの代謝を調査した。培養開始時及び20時間培養後の懸濁液又は溶液をサンプリングし、10000rpmで10分間の遠心分離後、その上澄をイオン交換クロマトグラフィにてそれぞれの成分の含有量(mg/g)を調べた。その結果を表2に示す。表2から明らかなように、比較例1のマンノースは66%減少し、マンノース試薬においても69%が減少しており、マンノースは腸内細菌により非常に代謝されやすいことが判明した。これに対し、実施例1のβ−1,4−マンノビオースの減少量は16%であり、実施例2においてもβ−1,4−マンノビオースの減少量は17%であり、マンノースに比して明らかに代謝率が低いことが判明した。
市販コマーシャル採卵鶏初生オスヒナ150羽を3分し、市販コマーシャルブロイラー用配合飼料(商品名:アマタケS、薬剤無添加/伊藤忠飼料株式会社製)を給与した50羽の対照区、同配合飼料に実施例2で得られた乾燥粉体を0.1%添加した飼料を給与した50羽の区、及び比較例1で得られた乾燥粉体を0.1%添加した飼料を給与した50羽の区の合計3区を設定し、不断給餌、自由飲水にて飼育した。3日令でサルモネラ菌(Salmonella enteritidis KTE−61株)、3.9×108cfu/羽を経口接種させた。サルモネラ菌接種後5日、10日、17日において各区、盲腸内容物中のサルモネラ菌数(対数)を調べた。その結果を表3に示し、それをグラフで表したものが第1図である。
コプラミール100部に酵素スミチームACH(新日本化学工業株式会社製)0.25部を溶解した酵素液125部を50℃、22時間作用させた後、流動層乾燥装置にて水分7.1%まで乾燥、乾燥粉体103部を得た。この乾燥粉体にはマンノース1.17部、マンノビオース14.18部(マンナンに対し47.3%)が生成していた。
コプラミール100部に酵素セルロシンGM5(エイチビィアイ株式会社製)0.3部を溶解した酵素液150部を60℃、72時間作用させた後、流動層乾燥装置にて水分6.4%まで乾燥、乾燥粉体100部を得た。この乾燥粉体にはマンノース13.72部、マンノビオース0.64部(マンナンに対し2.13%)が生成していた。
市販コマーシャルブロイラー初生ヒナ45羽を3分し、市販コマーシャルブロイラー用配合飼料(商品名アマタケS、薬剤無添加/伊藤忠飼料株式会社製)を給与した15羽を対照区とし、同配合飼料に実施例6で得られた乾燥粉体を0.1%添加し給与した区15羽、比較例2で得られた乾燥粉体を0.1%添加した給与した区15羽の合計3区を設定し、不断給餌、自由飲水にて1週間予備飼育した。1週令でサルモネラ菌(Salmonella enteritidis HY−1株)、2.27×107cfu/羽を経口接種した。サルモネラ菌接種後1週間後、2週間後、3週間後において各区5羽ずつ解剖し、盲腸内容物中のサルモネラ菌数(対数)を調べた。
7週令採卵鶏中雛30羽を3分し、市販コマーシャルブロイラー用配合飼料(商品名アマタケS、薬剤無添加/伊藤忠飼料株式会社製)を給与した10羽を対照区とし、同配合飼料に実施例6で得られた乾燥粉体を0.1%添加し給与した区10羽、比較例2で得られた乾燥粉体を0.1%添加した給与した区10羽の合計3区を設定し、不断給餌、自由飲水にて1週間予備飼育した。その後、サルモネラ菌(Salmonella enteritidis HY−1株)、1.13×108cfu/羽を経口接種した。サルモネラ菌接種後1週間後、2週間後、3週間後、4週間後において盲腸便を採取し、盲腸便におけるサルモネラ菌数(対数)を調べた。
Claims (3)
- マンナン含有天然物にマンナン分解酵素を作用させて得られ、乾物換算で少なくとも10重量%以上のβ−1,4−マンノビオースを含むβ−1,4−マンノビオース含有組成物からなることを特徴とする飼料添加用サルモネラ菌定着抑制剤。
- マンナン含有天然物が、パーム核ミール及び/又はコプラミールであることを特徴とする請求項1記載の飼料添加用サルモネラ菌定着抑制剤。
- 請求項1又は2記載の飼料添加用サルモネラ菌定着抑制剤を飼料に配合し、β−1,4−マンノビオースが0.001〜1重量%含有され、家畜又は家禽の腸内でのサルモネラ菌の定着を抑制しうることを特徴とする飼料。
