Description
即ち本発明は、
(1) 少なくとも炭酸塩を含有する培地成分を収容し、非通気性材料で密封された第1の容器と、該炭酸塩以外の培地成分を含有し、密封された第2の容器からなる培養用培地セット、
(2) 非通気性材料の二酸化炭素透過係数が約1000cm3/m2・24hr・atm未満である(1)に記載の培養用培地セット、
(3) 非通気性材料の水蒸気透過係数が約1000g/m2・24hr・atm未満である(1)に記載の培養用培地セット、
(4) 第2の容器が通気性材料からなる(1)に記載の培養用培地セット、
(5) 通気性材料の酸素透過係数が約100〜5000cm3/m2・24hr・atmである(4)に記載の培養用培地セット、
(6) 通気性材料の二酸化炭素透過係数が約1000〜20000cm3/m2・24hr・atmである(4)に記載の培養用培地セット、
(7) 第1の容器および第2の容器に連通可能な閉鎖手段を設けた(1)に記載の培養用培地セット、
(8) 炭酸塩が炭酸水素ナトリウムである(1)に記載の培養用培地セット、および、
(9) (1)〜(8)に記載の培養用培地セットを用いて、培地成分として少なくとも炭酸塩を含有する培地を非通気性雰囲気下で保存することを特徴とする培地保存方法に関する。
第1の容器は、内層が低密度ポリエチレン、中間層がポリ(エチレン−ビニルアルコール)コポリマー、外層に低密度ポリエチレンの2枚のシートを用意し、2枚のシートの間にポリエチレン−ポリプロピレン高分子ブレンド(混合比3:7)製の弱シール形成用シートを所定長分埋没するように配置する。次いで、2枚のシートの側縁および弱シール用シートを含んだ3枚のシートが重なる部分を通る帯状部分を熱溶着して、口部を有する袋状の容器を作製する。この袋状の容器に、炭酸水素ナトリウム水溶液30mlを収容し、袋状の容器の口部を熱溶着して作製する。
内層が低密度ポリエチレン、中間層がポリ(エチレン−ビニルアルコール)コポリマー、外層に低密度ポリエチレンの2枚のシートを用意し、2枚のシートを重ね合わせ、培地が収容でき、かつチューブが接続できるポートを設置したうえで、シートの側縁を熱溶着して、第1の容器1を作成した。さらに、上記ポートから炭酸水素ナトリウム水溶液30mlを収容した。
次に第1の容器と第2の容器を接続する。第1の容器及び第2の容器に設けたそれぞれのポートに、連通ピース(折れ棒)を設けたポリ塩化ビニル製チューブを接続して、図2に示す培養用培地セットを作製した。

Claims (7)
- 少なくとも炭酸塩を含有する培地成分を収容し、非通気性材料で作成された第1の容器と、該炭酸塩以外の培地成分を収容した第2の容器、及び第1の容器と第2の容器との間に設けた連通可能な閉鎖手段を備えた培養用培地セット。
- 非通気性材料の二酸化炭素透過係数が1000cm3/m2・24hr・atm未満である請求項1に記載の培養用培地セット。
