Description
本願は、2002年2月13日出願の米国特許出願番号10/076,802の一部継続出願であり、該出願の開示内容すべては、参考として本明細書で援用される。
この出願は、ポリペプチドを効率的に発現および分泌させる特定の真核生物シグナル配列を、特に原核宿主細胞で用いる方法に関する。また、本発明は真核生物シグナル配列を用いてポリペプチド提示ライブラリーを産生およびスクリーニングする方法に関する。
細胞膜を介して細胞から分泌されるタンパク質は、通常、タンパク質の細胞膜横断の手助けに必要とされる付加的なペプチド配列をアミノ末端に有する「プレタンパク質(preprotein)」と呼ばれる前駆体の形態で細胞内に産生される。この付加的なペプチド酸配列は、「シグナル配列(signal sequence)」または「リーダー配列(leader sequence)」と呼ばれている。大腸菌(E.coli)等の原核生物では、シグナルペプチドは、タンパク質分泌をペリプラズムおよび外膜へ指向させる。真核生物細胞では、シグナル配列を含むプレタンパク質が粗面小胞体(RER)膜を貫くようにして挿入されており、それによってプレタンパク質が分泌経路に導かれる。このプロセスの過程で、シグナル配列はシグナル認識粒子(SRP)と呼ばれる粒子と相互作用し、続いて該粒子がSRP受容体またはドッキングタンパク質(docking protein)と呼ばれるRER膜タンパク質によって認識される。RERへのプレタンパク質の挿入後または挿入と同時に、シグナルペプチダーゼと呼ばれる酵素によって、プレタンパク質からシグナル配列が切り出され、それによって成熟タンパク質がRERに放出される。いったんタンパク質が分離してERの内腔に入ると、該タンパク質はゴルジ体に移動し、さらに分泌小胞に移る。分泌小胞が原形質膜と融合すると、小胞の含有物が細胞外環境に放出される。原形質膜および細胞壁の両方を持つ生物(例えば、酵母)では、概して小胞の内容物が上記膜と上記細胞壁との間のペリプラズム空間に放出される。
一態様では、本発明は、Fabフラグメントを発現するための方法を提供する。この方法は、(i)真核生物のシグナル配列にそれぞれが作動可能に結合した抗体重鎖および抗体軽鎖をコードするジシストロニック(dicistronic)な転写ユニットに作動可能に結合したラムノースプロモーターを含むベクターを有する細菌細胞の培養物を提供する工程;(ii)培養物にラムノースを添加してラムノースプロモーターを誘導することで、抗体重鎖および抗体軽鎖ならびにそれらに結合したシグナル配列を発現させ、ペリプラズムに分泌させ、シグナルペチド配列が抗体重鎖および抗体軽鎖からプロセスされ、抗体重鎖と抗体軽鎖とが結合して標的分子に特異的に結合するFabフラグメントを形成する工程;ならびに(iii)前記細菌細胞の培養物から前記Fabフラグメントを回収する工程、を含む。それらの方法では、真核生物シグナル配列の各々がMRTLAILAAILLVALQAQA(配列番号2)またはMKILILGIFLFLCSTPAWA(配列番号1)の配列を有するシグナルペプチドをコードする。
(I.概観)
1つの種に由来するシグナル配列が他の種で機能的および/効率的であるかどうかは、一般に予測不可能である。このことは、特に原核生物と真核生物との間のシグナル配列の機能的置換に関して当てはまる(Humphreysら、前出)。そのような予測不可能なこともまた、本発明者によって示されている。試験した合計19種類の真核生物シグナルペプチドのうち、たったの3種類のみが適切にプロセッシングされ原核生物発現系に作動可能に結合したポリペプチド配列の効率的な発現および分泌を導いた(実施例2〜3)。
特に明記しない限り、本明細書で使われる技術用語および科学用語のすべてが本発明の技術分野で当業者により通常理解されるものと同様の意味を有する。本明細書に記載したものと類似または同等の方法および材料のいずれも、本発明の実施または試験に用いることができるが、好ましい方法および材料が記載される。以下の定義は、本発明を実施する上で読者の手助けとなるものである。
本発明は、宿主細胞、特に原核宿主細胞(例えば、大腸菌(E.coli))で目的のタンパク質またはポリペプチドを真核生物シグナル配列を用いて発現させるための方法およびベクターを提供する。好ましくは、真核生物シグナル配列は、MKILILGIFLFLCSTPAWA(配列番号1)またはMRTLAILAAILLVALQAQA(配列番号2)の配列を有するシグナルペプチドをコードする。配列番号1はヒトセルロプラスミンシグナル配列(受託番号CERU_HUMAN)に対応し、配列番号2はヒト好中球ディフェンシン1,2,3前駆体シグナル配列(受託番号DEFN_HUMAN)に対応する。本発明に用いられ得る別の好ましい真核生物シグナルペプチド配列は、MGALAVFAVACLAAVASVAHA (配列番号3)である。それはクラミドモナス(Chlamydomonas reinhardtii)アリールスルファターゼ前駆体のシグナルペプチドである(受託番号ARS_CHLRE)。目的のタンパク質が免疫グロブリンまたはFabフラグメントである場合、Ig重鎖および軽鎖をコードするポリヌクレオチドは各々が真核生物シグナル配列に対して作動可能に結合している。そのようなベクターによって宿主細胞およびアセンブリから機能性免疫グロブリン分子またはFabフラグメントへの発現および分泌が可能となる。
目的の様々なポリペプチドを本発明の発現ベクターによって発現させることができる。いつくかの方法では、目的のポリペプチドが抗体またはそのフラグメントである。発現されるポリペプチドは、マウス抗体、ヒト抗体、またはキメラ抗体であり得る。いくつかの好ましい実施形態では、目的のポリペプチドはFabフラグメントである。発現される抗体は、重鎖配列および軽鎖配列がすでに知られている特定の抗体であり得る。別の用途では、抗体は目的の特定の標的についての親和性に対するポリペプチド掲示ライブラリースクリーニングを介して同定される(例えば、実施例4〜7を参照せよ)。セクションIVで、より詳細に開示されるように、セクションIVでは、スクリーニングと濃縮化の後、抗体の重鎖および軽鎖をコードするポリヌクレオチド配列が提示ベクターから発現ベクターに移される。原核細胞系で機能的抗体の発現に真核生物シグナル配列を用いることの有効性は、実施例6〜7に例示されている。
本発明は、少なくとも1つの転写ユニットを含む発現ベクターを提供する。この転写ユニットは、目的のポリペプチドをコードするポリヌクレオチドに作動可能に結合した真核生物シグナル配列を含む。ベクターのいくつかも転写ユニットに作動可能に結合した細菌プロモーターを含む。いくつかのベクターにおいて、細菌プロモーターはラムノース誘導プロモーターである。さらに、発現ベクターは、宿主細胞内のポリペプチドを適当に転写およびそれに続く翻訳にとって必要な、または好ましい他の要素(例えば宿主細胞によって認識される複製起点)、終止コドン、および任意の他のポリヌクレオチド配列も含み得る。ベクターもまた、少なくとも1つの選択可能なマーカーを含む。
本発明の発現ベクターにとって好ましい宿主細胞は、原核細胞(例えば、大腸菌(E.coli))である。発現ベクターからの目的のポリペプチドの発現は、当該技術分野で日常的に実施されている方法(例えば、Sambrookら、上掲、およびAusubel、上掲)を用いて、原核宿主細胞内で実行することができる。原核宿主細胞系において、作動可能に結合した真核生物シグナル配列によって、目的のポリヌクレオチドを発現させる方法もまた、セクションVで詳細に説明する。ラムノース誘導プロモーターを用いた場合、宿主細胞は大腸菌(E.coli)rhaB+株または大腸菌(E.coli)rhaB−株のいずれかであり得る。これらの宿主細胞でのラムノースによる発現の誘導は、以下の実施例に記載したようにして実施することができる。ラムノース誘導プロモーターからのポリペプチドの発現もまた、当該技術分野では記載されている(例えば、EganおよびSchleif, 上掲;Haldimannら、J. Bacteriol.177:4121−30, 1998;ならびにWilmsら、Biotech. and Bioengin. 73:95−103,2001)。
本発明は、ポリペプチド提示ライブラリーの産生およびスクリーニングの改善された方法を提供する。該方法で用いられる提示ベクターは、宿主系(例えば、大腸菌(E.coli)等の原核宿主細胞)での効率的な分泌を可能とする真核生物シグナル配列(例えば、表3および4に示す配列)を含む。続いて、掲示ライブラリーをスクリーニングすることによって同定される目的のポリペプチドは、発現ベクターにクローニングされ得る(例えば、原核生物発現ベクター)また、同定されたポリペプチドを産生する宿主系に、発現ベクターを導入し得る。抗体掲示ライブラリーのスクリーニングの際、重鎖および/または軽鎖可変ドメインをコードするポリヌクレオチドをファージ提示ベクターから発現ベクターにサブクローニングする。発現ベクターを宿主に導入して発現させ、シグナルペプチドを含む軽鎖および重鎖を産生させる。次に、重鎖および軽鎖によって成熟抗体が形成されて宿主から放出される。
複製可能な遺伝子パッケージとは、細胞、胞子、またはウイルスを意味する。複製可能な遺伝子パッケージは、真核細胞または原核細胞のものであり得る。掲示ライブラリーの作製は、掲示される外来性ポリペプチドをコードする核酸を複製可能な遺伝子パッケージのゲノムに導入して、内在性タンパク質との融合タンパク質を作ることによっておこなわれる。この融合タンパク質は、複製可能な遺伝子パッケージの外面から通常発現される。融合タンパク質の発現、外表面およびアセンブリへの移動は、遺伝子パッケージの外表面由来の外来性ポリペプチドの掲示に帰する。
抗体ライブラリーの多価ファージ提示を、米国特許第6,057,098号の記載にしたがって作ることができる。すなわち、出発材料は、ファージのライブラリーであり、ライブラリー構成要素は、ファージ外殻タンパク質、抗体軽鎖可変ドメインまたは抗体重鎖可変ドメイン、およびタグから構成される融合タンパク質を外表面から提示することが可能なファージを有する。少なくともいくつかの構成要素では、抗体重鎖または抗体軽鎖がパートナー抗体重鎖可変ドメインまたはパートナー軽鎖可変ドメイン鎖と複合体を形成し、スクーリングすべきFabフラグメントが該錯体によって形成される。融合タンパク質および/またはパートナー抗体重鎖もしくは軽鎖は、ファージゲノムのセグメントによってコードされる。さらに、掲示ベクターは、抗体重鎖または軽鎖(例えば、表3および4に示すように)に作動可能に結合した少なくとも1つの真核生物シグナル配列を含む。Fabフラグメントの掲示が意図される場合、掲示ベクターは、重鎖配列に作動可能に結合した第1の真核生物シグナル配列および軽鎖配列に作動可能に結合した第2の真核生物シグナル配列を含む。第1および第2のシグナル配列は、同一または異なった配列であり得る。いくつかの方法では、シグナル配列は配列番号1、2、または3に示すシグナルペプチド配列をコードする。例えば、軽鎖に結合したシグナル配列は、配列番号1に示すシグナルペプチド配列をコードする配列番号43であり得、重鎖に結合するシグナル配列は配列番号2に示す(実施例8に例示したような)シグナルペプチドをコードする配列番号45であり得る。あるいは、両方の鎖を、配列番号1に示す同一のシグナルペプチド配列をコードするシグナル配列(例えば配列番号43および44)に作動可能に結合させることができる。
抗体ライブラリーは、一本鎖または二本鎖である。一本鎖抗体ライブラリーは、1個の抗体のみの重鎖または軽鎖、あるいはその可変ドメインを有し得る。しかし、より典型的には、一本鎖抗体ライブラリーの構成要素は、連続する単一のタンパク質内で、ペプチドスペイサーによって分離された重鎖可変ドメインまたは軽鎖可変ドメインの融合から形成される。(例えば、Ladnerら、WO 88/06630;McCaffertyら、WO 92/01047 を参照のこと)二本鎖抗体は、重鎖および軽鎖、またはその結合フラグメントの、非共有結合または共有結合によって形成される。二本鎖抗体はまた、2個の抗体結合部位を有する安定した二量体を形成する2個の一本鎖抗体から形成され得る。(Schierら、J. Mol. Biol. 255, 28−43 (1996); Arndtら、Biochemistry 37, 12918−12926 (1998)を参照のこと)抗体ライブラリーの多様性は、免疫化B細胞または非免疫化B細胞の非クローン個体群のような、天然由来の抗体コード配列を獲得することによって起こり得る。それとは別に、またはそれに加えて、多様性は、他のタンパク質について論じられるような、人工的変異誘発によってもたらされることもある。
本発明の多価提示ライブラリーを作成およびスクリーニングするための改良された方法は、キメラまたはヒト抗体ライブラリーを包含する。キメラ抗体は、例えば、抗体が第一の種由来の可変領域および第二の種由来の定常領域を有するものを含む、少なくとも1のキメラ抗体鎖を有する。Fabフラグメントのようないくつかのキメラ抗体は、キメラ重鎖およびキメラ軽鎖を含む。いくつかのキメラ軽鎖は、第一の種由来の軽鎖可変領域、および第二の種由来の定常領域を含む。同様に、いくつかのキメラ重鎖は、第一の種由来の重鎖可変領域、および第二の種由来の重鎖定常領域を含む。インタクトな抗体は、2コピーのキメラ軽鎖および2コピーのキメラ重鎖を含み得る。キメラFabフラグメントは、キメラ軽鎖およびキメラ重鎖を含み得る。第一の種由来の可変領域、および第二の種由来の定常領域を有するキメラ抗体について、その可変領域は、典型的には、マウス、ラット、ウサギ、モルモット、ウシ、ウマ、ヒツジ、ハゲワシ(vulcher)、サル、またはチンパンジーのような、非ヒト種から得られる。その定常領域は、典型的には、ヒトでの利用を意図したキメラについてヒト由来であるか、または獣医学的適用が意図される動物種由来である。Fabフラグメントにおいて、重鎖定常領域は通常CH1領域を含み、軽鎖定常領域はC−κまたはC−λのようなインタクトな軽鎖定常領域である。インタクトなIgG抗体において、重鎖定常領域は、典型的にCH1、ヒンジ、CH2およびCH3領域を含み、軽鎖は、インタクトなC−κ軽鎖またはC−λ軽鎖である。
ポリペプチドの複数のコピーを提示するライブラリーのメンバーは、提示ライブラリーにおいては比較的稀少である。そのライブラリーがスクリーニング標的と接触する前に、1つより多いポリペプチドを提示するライブラリーメンバーを富化するには、対になったタグとレセプターとを使用することができる。タグは、レセプターと結合するものなら何でもよいが、典型的には、短いペプチド配列である。レセプターは、そのタグに対して、特異的ではあるが、可逆的な結合を示し、支持体に固定化することができるものなら何でもよい。本発明に適した様々なタグ−レセプターの組み合わせが、米国特許第6,057,098号に開示されている。例えば、エピトープと抗体との対は、抗ダイノルフィン(dynorphin)mAb 32.39に対して高い親和性を有することが知られているヘキサペプチドリガンド(Barrettら、Neuropeptides 6,113−120(1985)およびCullら、PNAS 89,1865−1869(1992)を参照せよ)と、MAb 3E7を結合することが知られている種々の短いペプチド(Schatz,Biotechnology 11,1138−43(1993))を含む。タグと抗体とのもう別の組み合わせは、BlanarおよびRutter,Science 256,1014−1018(1992)が記述している。さらに別のタグ−レセプター対の例は、FLAGTMシステム(Kodak)である。
多価ライブラリーメンバーの選択は、ライブラリーメンバーのタグ成分に対するレセプターにライブラリーを接触させることにより実行する。通常、固相に固定化したレセプターにライブラリーを接触させ、ライブラリーメンバーをそれらのタグを介してレセプターに結合することにより、平衡に達することが可能となる。その後、複合体となったレセプターおよびライブラリーメンバーを、レセプターが親和性を有する固相の添加(例えば、アビジンで標識した固相を用いて、ビオチンで標識したレセプターを固定化することができる)によって、溶液から取り出す。あるいは、溶液中でライブラリーをレセプターに接触させることができ、この結果、レセプターが固定化される。大部分の提示されたタグがレセプターに平衡で結合するように、レセプターの濃度は、通常、液相結合の間、タグ/レセプターのKd以上にすべきである。レセプター−ライブラリーメンバーを固相に接触させる際、溶液中でライブラリーメンバーから固相が分離された後、少なくとも2つの提示されたタグを介してレセプターに結合するライブラリーメンバーのみが、固相に結合したままである。単一のタグを介してレセプターに結合されるライブラリーメンバーは、固相の分離および洗浄の間、固相より剪断されると推定される。未結合のライブラリーメンバーを除去した後、イオン強度もしくはpHを変化させるか、またはレセプターへの結合についてタグと競合する物質を添加することによって、結合したライブラリーメンバーをレセプターおよび固相から解離できる。例えば、アガロースに固定化されるとともにNi2+を含む金属キレートリガンドのヘキサヒスチジン配列への結合は、イミダゾールを溶液に添加して金属キレートリガンドの結合について競合させることにより、簡単に逆転できる。抗体−ペプチド結合は、多くの場合、pHを10.5以上に上げることによって解離できる。
Fabまたは他のポリペプチドの多価提示について富化されたライブラリーメンバーを、目的の標的への結合についてスクリーニングした。目的の標的は、結合パートナーを同定することが望まれるためいずれの目的の分子でもよい。この工程では、スクリーニングされるのは、提示されたポリペプチドであって、標的に結合するタグではないので、標的はタグに対する特異的結合親和性を欠くべきである。この工程でのスクリーニング手順は、親和性試薬がタグに対するレセプターではなく目的の標的である以外は、先の工程に非常に類似している。富化されたライブラリーメンバーを、その固定化を可能にする方法で通常標識した(例えば、ビオチンを用いて)標的に接触させる。結合により、平衡へと進むことが可能となり、その後、パンニングとして公知のプロセスで固相と接触させることにより、標的を溶液から取り出す(ParmleyおよびSmith、Gene 73、305−318(1988))。選択プロセス全体を通して、固相と結合したままのライブラリーメンバーには、それらと固定化した標的分子との間の多価結合よって同様のことを行う。未結合のライブラリーメンバーを洗浄して、固相から取り除く。
(A.サブクローニング)
提示ライブラリーメンバーのスクリーニングでは、代表的に、標的に対する特異的親和性を有するライブラリーメンバーの小集団が生じる。いくつかの方法では、ライブラリーメンバーのクローン性単離物が得られ、これらの単離物を様々な他の分析に直接用いる。他の方法では、ライブラリーメンバーのクローン性単離物を得て、抗体鎖をコードするポリヌクレオチド配列をそれぞれの単離物から増幅する。代表的に、発現ベクターに移す前に、同一のポリヌクレオチド分子の成分として重鎖および軽鎖を増幅するので、提示ベクター中に存在する重鎖および軽鎖の組み合わせは、発現ベクター中でも保存される。さらに、各シグナル配列もまた、発現ベクターに移され、その中で、抗体鎖に対するその作動可能な結合が保存される。発現ベクターは、上記に開示したような原核生物ベクターであり得る。発現ベクターは、また、真核生物ベクターであり得る。さらに、いくつかの方法では、1つの発現ベクター(例えば、セクションIIIで論じる原核生物発現ベクター)中の目的のポリヌクレオチド(例えば、Fabコード配列)を、別の発現ベクター(他の原核生物ベクターまたは真核生物ベクター)に移す。
セクションIIIおよびIVで論ずるような対象となるポリヌクレオチドを発現させるために様々な宿主系を用いることができる。セクションIIIで論ずるように、大腸菌(E.coli)は、本発明のポリヌクレオチドをクローニングするために特に有用な1つの原核宿主細胞である。使用に好適な他の細菌宿主としては、バチラススブチリス(Bacillus subtilis)等の棹菌(bacilli)と、サルモネラ菌(Salmonella)、セラチア属(Serratia)、および多種の シュードモナス(Pseudomonas)種等の他の腸内細菌(enterobacteriaceae) とが挙げられる。これらの原核細胞宿主では、発現ベクターを作製することもでき、宿主細胞に適合した発現制御配列を通常含む(例えば、複製起点)。さらに、ラクトースプロモーター系、トリプトファン(trp)プロモーター系、ベータラクタマーゼプロモーター系、またはファージラムダもしくはファージT7から得たプロモーター系等の様々な周知のプロモーターの任意の数を存在させる。プロモーターは、転写および翻訳を開始終了するために、通常、発現を制御し、任意でオペレーター配列を有し、リボソーム結合部位配列等を有する。
(発現ベクターの構築)
本明細書に記載したように、ポリヌクレオチドに作動可能に連結するlacおよびラムノースプロモーター等のプロモーターにしたがって発現するポリペプチドをコードするポリヌクレオチドを有する発現ベクターを構築した。
米国特許第6,057,098号の実施例17記載のものと同様の発現ベクターを、κ鎖上に異なるシグナル配列を有するFabをクローニングするために開発した。このベクターは修飾pBR322プラスミドであり、このベクターをAL1.3と命名した。これは、lacプロモーター(Plac)、アンピシリン耐性遺伝子、NsiI制限酵素切断部位、HindIII制限酵素切断部位、および部分的なテトラサイクリン耐性遺伝子を含む。
シグナル配列ありで、またはシグナル配列無しで、タンパク質をクローニングするため、米国特許第6,057,098号の実施例17に記載のものと類似した別の発現ベクターを開発した。ベクターは、AL2.2と命名された修飾pBR322プラスミドであり、ラムノースプロモーター(PrhaSB)を含んだrhaRおよびrhaS遺伝子と、アンピシリン耐性遺伝子と、NsiIおよびHindIII制限部位と、部分的なテトラサイクリン耐性遺伝子とを有する。
pBRSacH3ベクターを、AL2.2から誘導した。該ベクターは、NsiI部位がヒトセルロプラシミンシグナル配列と置き換えられる点を除き、あらゆる点でAL2.2と同じである。セルロプラシミンシグナル配列の3’末端最後の2塩基を修飾してSacI制限部位を生成した。SacIおよびHindIII部位の存在により、インサート導入の準備としてベクターを線状化することが可能となる。このコンストラクトは、その5’末端にヒトセルロプラシミンシグナル配列を使う抗体および抗原をクローニングするために作られた。シグナル配列をベクターに置くことにより、サブクローニング用のPCRプライマーをかなり小さくできる。したがって、PCR 反応がより効率的になる。必要とされる反応数がより少なくなるので、必要とされる時間および資源の両方が削減される。
(真核生物シグナル配列の選択)
真核生物シグナル配列を選択し、ポリペプチドをコードするポリヌクレオチドと共に発現ベクターに組み込んだ。ポリペプチドには、本明細書に記載されているような、抗体重鎖ポリペプチドおよび抗体軽鎖ポリペプチドが含まれる。

(真核生物のシグナル配列の評価と選択)
本明細書に記載する通り、宿主細胞中での抗体重鎖または軽鎖ポリペプチドの発現を含めた、宿主細胞中のポリペプチド発現のための多種類のシグナル配列を評価し、選択した。誘導ラムノースプロモーター経由のバクテリア宿主細胞中を含め、培養宿主細胞中でポリペプチドを発現させた。試験には、ペプチド発現の評価(例、収率、量)とペプチドからのシグナル配列のプロセシング(例、切断)を含めた。例えば、検出可能なSDS−PAGE分析によるものを含む検出可能な発現、および例えばSDS−PAGE分析で証拠付けられるようなものを含むポリペプチドからのシグナル配列のプロセシングの証拠は、発現のための真核生物のシグナル配列を選択することを可能にした。好ましい真核生物のシグナル配列は、効率的な発現(例えば、検出可能な、ごく弱いものでなく好ましくは強い、SDS−PAGEによるポリペプチドバンド)および効果的なおよび/または完全なプロセシング(例えば、シグナル配列および/または発現されたポリペプチド鎖それぞれについてのプロセシングを示すポリペプチド鎖バンドの早い泳動、シグナル標識の完全なプロセシングを示す2つ以下の単一バンドの存在)をもたらすものである。発現および/またはプロセシングを検出するための多種類の方法が、本明細書に記述したSDS−PAGE法を含め、当業者に周知であり、また利用されている。
実施例2に記述した通り、異なるプラスミドすなわちWC2A〜WC22Aを含むクローンを20セット構築し、誘導性ラムノースプロモーターにより抗C.difficileトキシンA抗体(CDTXA.22.2)を発現させるのに用いた。全てのケースで、重鎖は原核生物のアルカリホスファターゼシグナル配列を用い人工的に作った。コントロールセットであるWC2Aでは、κ鎖は原核生物のpelBシグナル配列を用いて作り、一方残りの19においては、それを19の異なる原核生物シグナル配列を用いて作った。全てのケースで、抗体重鎖に対してまた、そのC末端にアフィニティ精製を可能にするためにヘキサヒスチジンタグを設けた。これらのプラスミドに対して、培養中の選択を可能にするためテトラサイクリンおよびアンピシリン耐性遺伝子を持たせた。これらのプラスミドをE.coli株DH10B rha+株(Invitrogen,Carlsbad,CA)に電気穿孔した。それぞれのセットからの3つのクローンについて、CDTXA.22.2抗体の発現に関し評価した。

これら3つの真核生物シグナル配列の完全なプロセシングを、Perkin Elmer Applied Biosystems Model 494 Prociseタンパク質/ペプチドシーケンサー(PE Applied Biosystems,Foster City,CA))を使い、κ鎖上のN末端アミノ酸の配列を解読することで確かめた。これら3つのシグナル配列のポリヌクレオチド配列は表3に示す通りである。

(実施例4)
(モノクローナル抗体の調製)
本明細書に記載したrMET1およびrMET.10抗体を含め、モノクローナル抗体を選択し、調製した。
(P−(3−メルカプトプロピル)アミノ−d−メタンフェタミンの合成)
P−(3−メルカプトプロピル)アミノ−d−メタンフェタミン(還元メタンフェタミン誘導体)は米国特許第5,470,997号の実施例11に記載された通りに合成することができる。
還元メタンフェタミン誘導体とキーホールリンペットヘモシアン、ウシ血清アルブミン、およびアルカリホスファターゼとの結合体を次のように調製した。
ウシ血清アルブミン(BSA,20mg/ml)とSMCCとの反応を、SMCCのアセトニトリル溶液をBSA対SMCCのモル比率1:50でBSAに加え、さらにその溶液のpHを1N水酸化カリウムで7から7.5の間に保ちつつ1時間室温でかき混ぜることで、おこなった。反応しなかった物質から、0.1Mリン酸カリウム、0.02Mカリウムホウ酸塩、および0.15M塩化ナトリウム、pH7.0でのゲル濾過クロマトグラフィーにより、タンパク質を分離した。β−メルカプトエタノール(Fisher Scientific,Pittsuburgh,PA)をBSA−SMCCに、推定されるマレイミドの存在量を越える十分なモル濃度で加え、溶液を室温で最低1時間攪拌した。遊離メルカプトエタノールは、大規模な透析により、0.1Mリン酸カリウム、0.02Mカリウムホウ酸塩、0.15M塩化ナトリウム、pH7.0の中に取り除いた。
米国特許第6,057,098号の実施例1〜4および7に記載の通り、BS45ウラシル鋳型を用い、還元メタンフェタミンKLHにより免疫されたマウスの脾臓から精製されたRNAを用い、ファージ提示ライブラリーを作製した。還元メタンフェタミンAPビオチン結合体に結合した抗体ファージを、米国特許第6,057,098号の実施例15に記載の通り、選択した。還元メタンフェタミンAPビオチンよりはるかに大量のBSA−SMCCを抗体ファージに加え、何ラウンドかのパンニング法により、SMCCリンカーに特異的な抗体を取り除いた。パンニング法の5回目のラウンドの後、抗体ファージを、還元メタンフェタミンAPビオチン、およびそれらの薬剤と交差反応するメタンフェタミン抗体を取り除くための何種類かの過剰な薬剤と共に、インキュベートした。加えられた交差反応薬剤は、+エフェドリン、−エフェドリン、d−擬エフェドリン、ラニチジン、プロカインアミド、フェンメトラジン、およびトリメトベンズアミドであった。これらの交差反応物質の存在下で、2ラウンドの選択を行った。アビジン乳濁液と結合したファージのアリコートを、これらの交差反応物質の存在下で、100mmLB平板に播種し、米国特許第6,057,098号の実施例15に記載の通り、プラークをニトロセルロースフィルターでオーバーレイした。このフィルターは、米国特許第6,057,098号の実施例15に記載の通り、AP−還元メタンフェタミン結合体を用いて開発した。ポジティブなプラークは、米国特許第6,057,098号の実施例18に記載の通り、pBRncoH3発現プラスミド中にサブクローニングした。これらのクローンのうち1つをrMET1と名付けた。
(発現ベクターへのクローニング)
本明細書中に記載されているrMET10モノクローナル抗体をコードするポリヌクレオチドを含む、ポリペプチドをコードするポリヌクレオチドを発現ベクターにクローニングした。
モノクローナル抗体rMET10は、実施例4に記載の選択プロセスとクローニングプロセスによって得た。元の構成では、κ鎖および重鎖の分泌を指示するために、pelBシグナル配列およびアルカリホスファターゼシグナル配列をそれぞれ用いた。これらのシグナル配列をヒトセルロプラスミンシグナル配列に変換するため、κ鎖および重鎖を別々に増幅し、その後、サブクローニングプロセスの際にκ鎖−重鎖カセットに再構成した。κ鎖は、プライマーSとプライマーG(表2)とを用いて増幅した。プライマーSは、rMET10に特異的な配列を有することに加えて、PrhaSBの3’末端に対応する約20ヌクレオチドのAL2.2ベクター配列をその5’末端に含み、それに続き、κ鎖と同じフレームにあるヒトセルロプラスミンシグナル配列をコードする配列を含む。プライマーGは、ファージ提示ベクター内のκ鎖の終止コドンの下流にあり、かつ重鎖のリボソーム結合部位の上流にある領域に結合する。重鎖は、プライマーTとプライマーR(表2)とを用いて増幅した。プライマーTは、rMET10に特異的な配列を有することに加えて、プライマーGに相補的な約20ヌクレオチドをその5’末端に含み、それに続き、重鎖と同じフレームにあるヒトセルロプラスミンシグナル配列をコードする配列を含む。
(発現の用量試験)
本明細書中に記載されているrMET10抗体を含む、ポリペプチド発現の用量試験を行った。
rMET10抗体の発現用プラスミドは、実施例5の記載と同様に構築した。このコンストラクト中のκ鎖および重鎖は、ともに、ヒトセルロプラスミンシグナル配列をもつように加工した。このプラスミドをE.coli DH10B rha+株に電気穿孔することで、クローンrMET10.LS.1を生成し、10μg/mlのテトラサイクリン選択下で培養した。このプラスミドをまた、別のE.coli ECL339 rha−株(ATCC)に電気穿孔することで、クローンrMET10.ECL.20を生成した。宿主株がゲノムにテトラサイクリン耐性遺伝子をもつため、100μg/mlのアンピシリン選択下でこのクローンを培養した。
(発現の追加用量試験)
本明細書中に記載されているrMET1抗体を含む,ポリペプチド発現の追加用量試験を行った。
プラスミドpBRsacH3(実施例1)を、rMET1抗体(実施例9)のクローニングに用いた。この実験で、κ鎖および重鎖を加工して、それぞれヒトセルロプラスミンシグナル配列およびヒト好中球ディフェンシンシグナル配列を、持つようにした(実施例9)。このプラスミドをE.coli DH10B rha+株に電気穿孔することで、クローンrMET1.LS.1を生成し、10μg/mlのテトラサイクリン選択下で培養した。このプラスミドを、E.coli ECL339 rha−株に電気穿孔することで、クローンrMET1.ECL.1を生成し、100μg/mlのアンピシリン選択下で培養した。
(真核生物シグナル配列をもった提示ベクターの構築)
本明細書に記載されている抗体ファージ提示ベクターを含む、真核生物シグナル配列をもった提示ベクターを構築した。
(ポリペプチドの提示と選択)
抗体ファージ提示ベクターを含む、真核生物シグナル配列をもった提示ベクターをポリペプチドの提示と選択に利用した。
米国特許第6,057,098号、実施例3および実施例4の記載とほぼ同様に、カッパ鎖および重鎖可変領域の一本鎖DNA配列をrMet1用に調製した。オリゴヌクレオチド41と971(表7)はカッパ鎖可変領域を増幅するのに用いられ、オリゴヌクレオチド204と1170(表8)は、重鎖可変領域を増幅するのに用いられた。一本鎖DNA反応(それぞれ4X100μl)は、オリゴヌクレオチド1のみを重鎖用に用い、オリゴヌクレオチド2のみをカッパ鎖用に用いて行った(表7および表8)。一本鎖DNAを精製し、ウラシル鋳型BS60に結合させた。これは、250μgスケールのみのウラシル鋳型が用いられたことを除き、米国特許第6,057,098号、実施例7の記載と同様に行われた。アニーリングは、試料を70℃で2分間インキュベートし、37℃より低い温度にゆっくり冷却することによって行われた。エクステンションおよびライゲーションの後、90μlの突然変異誘発停止緩衝液を混合液に加え、1μlの希釈DNAを、米国特許第6,057,098号実施例8の記載と同様に、大腸菌(E.coli)DH12Sの電気穿孔用コンピテントセル40μl(Invitrogen,Carlsbad,CA)に電気穿孔した。電気穿孔した細胞のアリコートを、100mmプレートの上にプレーティングした。プレート上のプラークにニトロセルロースフィルターをかぶせ、米国特許第6,057,098号、実施例13の記載と同様に、AP−メタンフェタミンでフィルターを展開した。機能的陽性プラークを選択し、50μlの2倍YTにとった。
(ポリクローナルファージ富化)
本明細書に記載されているパンニング法を含めたポリペプチド発現およびポリペプチド提示に用いる、真核生物シグナル配列をもったポリクローナルファージの富化を行った。
BS60ウラシル鋳型(実施例8)を用い、米国特許第6,057,098号、実施例7とほぼ同様にして、第1ラウンド抗体ファージを調製したが、ここでは以下のような相違を伴っていた。表7および表8に示すオリゴヌクレオチドを用い、米国特許第6,057,098号、実施例3の記載と同様にして、二本鎖PCR産物を調製した。相補的cDNAの独立した調製(米国特許第6,057,098号、実施例2)は行わなかった。代わりに、プラチナTaq(Platinum Taq)によるスーパースクリプトワンステップRT−PCR(Superscript One Step RT−PCR)(Invitrogen, Carlsbad, CA)を用いて、RNA鋳型から始まるDNA増幅を、製造元の推奨にしたがって行った。一本鎖DNAを二本鎖DNAから分離するHPLCの勾配(米国特許第6,057,098号の実施例4)を短くした。以下の熱プロフィールを用い、ウラシル鋳型とss−DNAインサートのアニーリングを行った。すなわち、70℃を2分間30秒、70℃から42℃までの傾斜(ramp)を10分以上、42℃で10秒間維持(hold)、4℃で保存(store)である。突然変異誘発DNAは、キアクイック(QIAquick)PCR精製カラム(Qiagen, Valencia, CA)を用い、製造元の説明にしたがって精製した。30μLの溶出緩衝液で突然変異誘発DNAをカラムから溶出し、1μLをDH12Sの電気穿孔用コンピテントセル40μLに電気穿孔した。電気穿孔された細胞を、一晩培養したXL1細胞(0.6mL)と2YT(0.4mL)との混合液1mLに再懸濁した。1mLの試料すべてを、150mmのLBアガープレート上にプレーティングした。プレートを37℃で4時間、さらにその後、20℃で一晩インキュベートした。第1ラウンドの抗体ファージ試料は、米国特許第6,057,098号、実施例15の記載と同様にして、プレートから溶出した。
(ポリペプチドのクローニング、発現、精製、および修飾)
本明細書に記載したように、抗体産生の免疫原として有用なポリペプチド等のポリペプチドのクローニング、発現、精製、および修飾をおこなった。
抗体産生に用いた免疫原は、バイオサイト社(Biosite Incorporated)によって調製した。PCRプライマーは、ヒトproMMP9の5’末端の配列およびヒトproMM9の3’末端のコード配列とに一致させた(Genbank受託番号J05070)。上記プライマーは、6つのヒスチジンコドン、プライマーFF、およびGGを含み、それぞれがコード配列の一端と停止コドンとのあいだあり、かつ金属キレートアフィニティクロマトグラフィによる組み換えタンパク質の精製を補助する(表2)。5’プライマーも同様に、EcoRI部位およびすぐ上流の配列に対応する5’末端に21塩基対のpEAK12ベクター配列(Edge BioSystems, Gaithersburg,MD)を含む。3’プライマーは、約20ヌクレオチドのベクター配列を有し、その5’末端に6塩基のNotI部位およびすぐ下流の配列が含まれる。これらのプライマーの5’末端にあるベクター配列は、T4DNAポリメラーゼ処理によって、pEAK12ベクターのものに対して特異的かつ相補的である単一鎖のオーバーハングを形成する。proMMP9遺伝子インサートのPCR増幅を、5’プライマー(FF)100pmol、3’プライマー(GG)100pmol、エクスパンドポリメラーゼ2.5単位、2mM dNTP 10μl、10xエクスパンド反応緩衝液10μl、鋳型としてクロンテッククイック(Clontech Quick)−クローンヒト脾臓cDNA(Clontech Laboratories, Palo Alto, CA) 1μl、および水100μlを含む2x100μl反応スケールでおこなった。反応は、実施例18(米国特許第6,057,098号)の記載にしたがって、パーキンエルマーサーマルサイクラー(Perkin−Elmer thermal cycler)でおこなった。PCR産物を沈殿させ、アガロースゲル電気泳動で画分化して、全長の産物をゲルから切り出し、精製し、さらに水に再懸濁した(実施例17、米国特許第6,057,098号)。インサートの挿入に備え、pEAK12ベクターをNotIおよびEcoRI((New England BioLabs,Beverly,MA)による消化によって調製した。T4エクソヌクレアーゼ消化のために、DNA1.0μgに10×緩衝液A1.0μlを添加し、さらに水によって最終容量を9μlにすることで、インサートおよびEcoRI/NotI消化pEAK12ベクターを調製した。試料を1μl(1U/μl)のT4DNAポリメラーゼで30℃、4分間にわたり消化した。T4DNAポリメラーゼを70℃、10分間インキュベートすることで、加熱による不活性化をおこなった。試料を冷やし、軽く遠心し、さらに新しい微小遠心管で消化インサート45ngを消化pEAK12ベクター100ngに添加した。1.0μlの10倍アニーリング緩衝液を加えた後、水で容量を10μlにした。混合物を70℃、2分間加熱し、20分間以上冷やして室温にし、インサートおよびベクターのアニーリングを可能にした。アニールDNAを希釈水によって1対4に希釈し、30μlのエレクトロコンピテント大腸菌(E.coli)DH10B株(Invitrogen,Carlsbad,CA)を電気穿孔(実施例8、米国特許第6,057,098号)にかけた。形質転換細胞を2倍YTブロスで1.0mlに希釈し、アンピシリン(75μg/ml)添加LB寒天プレート上に10μl、100μl、300μlプレーティングし、さらに37℃で一晩増殖させた。コロニーを取り、2倍YT(37℃で75μg/mlアンピシリン)で一晩増殖させた。翌日、長期間−80℃保存のためにグリセロールストックを作製した。これらのクローン(proMMP9ピーク12)の配列を、実施例1に記載したように、pEAK12ベクター内のインサートの5’および3’側に結合したオリゴヌクレオチドプライマーHHおよびII(表2)をそれぞれ用いて、マックコーネルリサーチ(MacConnell Research)で確認した。ヒトproMMP9のトランスフェクションおよびそれに続く発現および精製に好適なプラスミドを、エンドフリープラスミドメガキット(EndoFree Plasmid Mega Kit)を用い、かつ製造元の推奨(Qiagen,Valencia,CA)に従って、クローンproMMP9ピーク12.2から調製した。
(真核生物シグナル配列を用いたポリペプチド抗原のクローニング)
ポリペプチド抗原をクローン化し、真核生物シグナル配列(本明細書に記載したようにヒトセルロプラスミンシグナル配列を含む)を用いて、発現させた。
成熟マカクインターロイキン8抗原の5’末端のコード配列およびマカクインターロイキン8抗原の3’末端のコード配列、ならびにプライマーYおよびZにそれぞれ対応したPCRプライマーを作製した(表2)。特に、5’プライマーは、pBRsacH3ベクターの3’末端に対応する22塩基対のベクター配列をその5’末端に有し、また7つのヒスチジンコドン、および該コード配列の上流にあるエンテロキナーゼ切断部位を有する。上記3’プライマーは、5’末端にあるHindIII部位に対して3’側の24個のヌクレオチドのベクター配列に加えて、HindIII消化によって取り除かれるtetプロモーターの19ヌクレオチドを含む(実施例17、米国特許第6,057,098)。
(ポリペプチドの発現と精製)
本明細書に記載されているポリペプチド抗原を含むポリペプチドを発現させ、精製した。
このクローンは、実施例12の記載と同様に、マカクIL−8抗原を発現するプラスミドで電気穿孔された宿主大腸菌(E.coli)DH10B rha+株からなった。この構築物は、N末端の加工により、セプタヒスチジンタグ、エンテロキナーゼ切断部位、およびヒトセルロプラスミンシグナル配列をこの順序でもった。細胞は、10μg/mLのテトラサイクリン選択下で培養した。
Claims (12)
- Fabフラグメントを発現させる方法であって、以下:
MKILILGIFLFLCSTPAWA(配列番号1)、MRTLAILAAILLVALQAQA(配列番号2)またはMGALAVFAVACLAAVASVAHA(配列番号3)に示されたシグナルペプチド配列をコードする真核生物のシグナル配列にそれぞれが作動可能に結合した抗体重鎖および抗体軽鎖をコードするポリヌクレオチドを含むベクターを有する細菌細胞の培養物を提供し、それによって、該抗体重鎖および該抗体軽鎖ならびにそれらに結合したシグナル配列を発現させ、ペリプラズムに分泌させ、該シグナルペプチド配列が該抗体重鎖および該抗体軽鎖からプロセッシングされ、該抗体重鎖と該抗体軽鎖とが組み立てられて標的分子に特異的に結合するFabフラグメントを形成する工程と、
該細菌細胞の培養物から該Fabフラグメントを回収する工程と、
を包含する、方法。 - 請求項1に記載の方法であって、
(a)前記重鎖に作動可能に結合した前記シグナル配列がMRTLAILAAILLVALQAQA(配列番号2)をコードし、前記軽鎖に作動可能に結合した前記シグナル配列がMKILILGIFLFLCSTPAWA(配列番号1)をコードするか、または
(b)該重鎖に作動可能に結合した該シグナル配列がMKILILGIFLFLCSTPAWA(配列番号1)をコードし、該軽鎖に作動可能に結合した該シグナル配列がMRTLAILAAILLVALQAQA(配列番号2)をコードするか、または
(c)該重鎖に作動可能に結合した該シグナル配列がMGALAVFAVACLAAVASVAHA(配列番号3)をコードし、該軽鎖に作動可能に結合した該シグナル配列がMRTLAILAAILLVALQAQA(配列番号2)をコードするか、または
(d)該重鎖に作動可能に結合した該シグナル配列がMRTLAILAAILLVALQAQA(配列番号2)をコードし、該軽鎖に作動可能に結合した該シグナル配列がMGALAVFAVACLAAVASVAHA(配列番号3)をコードするか、または
(e)該重鎖に作動可能に結合した該シグナル配列がMGALAVFAVACLAAVASVAHA(配列番号3)をコードし、該軽鎖に作動可能に結合した該シグナル配列がMKILILGIFLFLCSTPAWA(配列番号1)をコードするか、または
(f)該重鎖に作動可能に結合した該シグナル配列がMKILILGIFLFLCSTP
AWA(配列番号1)をコードし、該軽鎖に作動可能に結合した該シグナル配列がMGALAVFAVACLAAVASVAHA(配列番号3)をコードする、方法。 - 請求項1に記載の方法であって、
(a)前記重鎖に作動可能に結合した前記シグナル配列と、前記軽鎖に作動可能に結合した前記シグナル配列とが共にMKILILGIFLFLCSTPAWA(配列番号1)をコードするか、または
(b)該重鎖に作動可能に結合した該シグナル配列と該軽鎖に作動可能に結合した該シグナル配列とが共にMRTLAILAAILLVALQAQA(配列番号2)をコードするか、または
(c)該重鎖に作動可能に結合した該シグナル配列と該軽鎖に作動可能に結合した該シグナル配列とが共にMGALAVFAVACLAAVASVAHA(配列番号3)をコードする、方法。 - 前記ポリヌクレオチドが、さらに作動可能にプロモーターに結合される、請求項1に記載の方法。
- 前記ポリヌクレオチドが、さらに作動可能にラムノースプロモーターに結合される、請求項1に記載の方法。
- 前記培養物中の培地から前記Fabフラグメントを回収する、請求項1に記載の方法。
- 前記回収工程に先立って、前記細胞を溶解して前記Fabフラグメントを放出させる工程をさらに含む、請求項1に記載の方法。
- ファージからFabフラグメントを提示する方法であって、該方法は、抗体重鎖および抗体軽鎖をコードするファージ提示ベクターを細菌細胞内で発現させる工程を包含し、該軽鎖が第1のシグナルペプチド配列に作動可能に結合しており、そして該重鎖がファージ外表面タンパク質および第2のシグナルペプチド配列に作動可能に結合しており、それによって該抗体の鎖が発現されて該細胞のペリプラズムに分泌され、そしてプロセッシングされることで、該シグナルペプチド配列から該抗体の鎖が分離され、標的分子に対する特異的結合親和性を持ち、ファージ粒子の外表面から掲示されるFabフラグメントとして該抗体の鎖が組み立てられ、
該第1のシグナルペプチド配列および第2のシグナルペプチド配列の各々が配列MKILILGIFLFLCSTPAWA(配列番号1)、MRTLAILAAILLVALQAQA(配列番号2)またはMGALAVFAVACLAAVASVAHA(配列番号3)を有する真核生物のシグナル配列である、方法。 - 請求項8に記載の方法であって、
(a)前記第1のシグナルペプチド配列がMKILILGIFLFLCSTPAWA(配列番号1)であり、前記第2のシグナルペプチド配列がMRTLAILAAILLVALQAQA(配列番号2)であるか、または
(b)該第1のシグナルペプチド配列がMRTLAILAAILLVALQAQA(配列番号2)であり、該第2のシグナルペプチド配列がMKILILGIFLFLCSTPAWA(配列番号1)であるか、または
(c)該第1のシグナルペプチド配列がMRTLAILAAILLVALQAQA(配列番号2)であり、該第2のシグナルペプチド配列がMGALAVFAVACLAAVASVAHA(配列番号3)であるか、または
(d)該第1のシグナルペプチド配列がMGALAVFAVACLAAVASVAHA(配列番号3)であり、該第2のシグナルペプチド配列がMRTLAILAAILLVALQAQA(配列番号2)であるか、または
(e)該第1のシグナルペプチド配列がMGALAVFAVACLAAVASVAHA(配列番号3)であり、該第2のシグナルペプチド配列がMKILILGIFLFLCSTPAWA(配列番号1)であるか、または
(f)該第1のシグナルペプチド配列がMKILILGIFLFLCSTPAWA(配列番号1)であり、該第2のシグナルペプチド配列がMGALAVFAVACLAAVASVAHA(配列番号3)である、方法。 - 請求項8に記載の方法であって、
(a)前記第1のシグナルペプチド配列および前記第2のシグナルペプチド配列が、共にMKILILGIFLFLCSTPAWA(配列番号1)であるか、または
(b)該第1のシグナルペプチド配列および該第2のシグナルペプチド配列が、共にMRTLAILAAILLVALQAQA(配列番号2)であるか、または
(c)該第1のシグナルペプチド配列および該第2のシグナルペプチド配列が、共にMGALAVFAVACLAAVASVAHA(配列番号3)である、方法。 - 前記ベクターがプロモーターを含む、請求項8に記載の方法。
