Abstract
Description
SH3YL1は細胞質タンパク質で存在し、細胞の移動に関係するが、細胞の炎症誘発刺激であるLPS(100ng/ml)処理後、HAEC(human aortic endothelial cell,Lonza,cc−2535)、HPMEC(human pulmonary microvascular endothelial cell,Promo Cell,c−12281)でSH3YL1の発現レベルを確認した。
炎症刺激因子であるLPSによって発現が増加したSH3YL1が細胞外部へ分泌されるのかを確認した。
糖尿病性腎症患者の血漿で実質的にSH3YL1タンパク質の濃度が増加するかを確認した。
Claims (10)
- SH3YL1遺伝子の発現レベルまたはSH3YL1タンパク質のレベルを測定することができる物質を含有する腎症診断用の組成物。
- 上記SH3YL1遺伝子の発現レベルは発現の可否、発現量及び発現パターンで構成された群から選択されることを特徴とする請求項1に記載の腎症診断用の組成物。
- 上記SH3YL1タンパク質のレベルは上記タンパク質の存在の可否、発現量及び発現パターンで構成された群から選択されることを特徴とする請求項1に記載の腎症診断用の組成物。
- 上記SH3YL1遺伝子の発現レベルを測定することができる物質はSH3YL1遺伝子を増幅することができるプライマー対または上記SH3YL1遺伝子とハイブリダイズすることができるプローブであることを特徴とする請求項1に記載の腎症診断用の組成物。
- 上記SH3YL1タンパク質のレベルを測定することができる物質はSH3YL1遺伝子がコードするタンパク質に特異的に結合する抗体またはアプタマーであることを特徴とする請求項1に記載の腎症診断用の組成物。
- 腎症診断に必要な情報を提供するために、請求項1に記載の腎症診断用の組成物を生物学的試料に処理して、試料中のSH3YL1遺伝子の発現レベルまたはSH3YL1タンパク質のレベルを検出する方法。
- 生物学的試料は、組織、細胞、血液、血清、血漿、唾液及び尿で構成された群から選択されることを特徴とする請求項6に記載の方法。
- SH3YL1遺伝子またはその断片とハイブリダイズすることができるプローブが固定されている腎症診断用のマイクロアレイ。
- SH3YL1遺伝子またはその断片を増幅することができるプライマー対、上記SH3YL1遺伝子とハイブリダイズすることができるプローブまたはSH3YL1遺伝子がコードするタンパク質に特異的な抗体またはアプタマーを含有する腎症診断用のキット。
- 次の段階を含む腎症の予防または治療用の物質をスクリーニングする方法:
(a)SH3YL1遺伝子を発現する細胞に候補物質を接触させる段階;
(b)SH3YL1遺伝子の発現レベル及び/またはSH3YL1タンパク質のレベルを測定する段階;及び
(c)SH3YL1遺伝子の発現レベル及び/またはSH3YL1タンパク質のレベルが減少する場合の候補物質を腎症の予防または治療用の物質として選別する段階。
