実施例1で同定したうちの4菌株を液体培地に植菌してから、37℃で培養した。培養後、1.5mlの培養液を遠心分離し、沈殿を回収。沈殿物にTAE(0.04M:Tris・acetate、0.001M:EDTA)、アルカリ溶液を添加し60℃で70分処理した。熱処理後、フェノールクロロホルム溶液を加えてから、再度60℃で15分処理し、遠心分離してから、上清を回収、プロテイナーゼKを加えて60℃で60分処理した。その後、水、酢酸ナトリウム、エタノールを加え、遠心分離後、沈殿を回収し、水を加えてLPS簡易精製サンプルを調製した。抽出・簡易精製したLPSを電気泳動で確認した。図1は、SDS電気泳動後のゲルを銀染色した後の写真である。レーンMは分子量マーカー、レーン○1、○8は精製した小麦から単離したグラム陰性細菌LPS、レーン○2から○7は、小麦、林檎、柚子から単離したグラム陰性細菌から簡易精製した6種のLPSを電気泳動している。この結果、レーン○5の林檎から単離したグラム陰性細菌3−4株(パントエア・バガンスと同定)由来のLPSが、その他のLPSに比較して、高分子量側LPSのサイズが小さいことがわかった。
LPSはマクロファージ細胞に働きかける事で、体の免疫力を活性化する。そこで、実施例2と同様の4菌株のLPSについて、NO産生量を指標として、マクロファージ細胞の活性化能を評価した。
実施例2において、分子量分布が、従来のLPSと異なっていたパントエア・バガンス3−4株を選び、20Lジャーファンメンターで培養した。培地(Glycerol、酵母エキス、NH4Cl、K2HPO4、KH2PO4、FeSO4・7H2O、CaCl2・7H2O、MgSO4・7H2O)10Lに、パントエア・バガンスの前培養液を5%添加し、30℃、pH6.5〜7.0、200〜300rpm、通気量20L/minで、48時間培養した。培養液を加熱処理し、遠心分離した上清から徐々にLPSを精製した。具体的には、上清に活性炭を混和し、遠心分離で活性炭を除去した後に、さらにセライトろ過、膜ろ過を行って食品用LPS素材(FL)を調整した。さらにFLからエタノール沈殿を行い、沈殿物を回収、凍結乾燥を経て、化粧品用LPS素材(CL)を得た。
実施例4のように調整したパントエア・バガンスLPS化粧品用原料を1%配合した化粧水を製造した。全成分は下記のとおりである。
水、BG、グリセリン、ジグリセリン、パントエア/コメヌカ発酵エキス液、ヒアルロン酸Na、シロキクラゲ多糖体、ソルビトール、セリン、グリシン、グルタミン酸、アラニン、リシン、アルギニン、トレオニン、プロリン、ベタイン、PCA-Na、ポリオクタニウム-51、ポリソルベート-20、セルロースガム、PEG-150、1,2-ヘキサンジオール、クエン酸、クエン酸Na、乳酸Na、メチルパラベン、プロピルパラベン
この化粧水について100人の女性にモニタ試験を行ったところ、半数以上の女性が肌の保湿感が高まり、肌が柔らかくなったと回答し、スキンケア製品に対する有用性が示された。

- パントエア・バガンスからリポ多糖を得ることを特徴とするリポ多糖製造方法。
- 請求項1記載のリポ多糖が配合されていることを特徴とするリポ多糖配合物。
- 前記リポ多糖配合物が医薬品、動物用医薬品、医薬部外品、化粧品、食品、機能性食品、飼料、又は浴用剤であることを特徴とする請求項3記載のリポ多糖配合物。
