頭部血管の血管周囲腔中に存在する神経ペプチドは、片頭痛発作中の侵害受容性入力の重要な媒介物質として関与するとされている。下垂体アデニル酸シクラーゼ活性化ポリペプチド(PACAP)は、感覚三叉神経ニューロン中に存在し、神経系の異なるレベルにおいて侵害受容を調節することができる。ヒトの実験研究は、PACAP38が頭痛及び片頭痛様発作の両方を誘発する(Schytz,et al.,2009,“PACAP38 induces migraine−like attacks in patients with migraine without aura”,Brain:132 pp16−25)ことを示しており、PAC1受容体拮抗薬が片頭痛の予防的及び/または急性治療に使用され得るという考えを支持している。
ヒトPAC1(hPAC1)タンパク質を含むPAC1タンパク質に結合する抗体が、本明細書に提供される。提供される抗体は、本明細書に説明される通り、その中へ1つ以上の相補性決定領域(CDR)が組み込まれる及び/または連結されるポリペプチドである。幾つかの抗体では、CDRは、CDR(単数または複数)の適切な抗原結合特性が達成されるようにCDR(単数または複数)を配向する「フレームワーク」領域内へ組み込まれる。概して、提供される抗体は、PAC1とそのリガンド(単数または複数)、例えば、PACAP−38などのPACAPとの間の相互作用に干渉する、それを遮断する、低減する、または調節することができる。

提供される抗体は、PAC1に結合するモノクローナル抗体を含む。モノクローナル抗体は、当該技術分野において既知である任意の技術を使用して、例えば、免疫付与計画の完了後に遺伝子導入動物から採取した脾臓細胞を不死化することによって、産生することができる。脾臓細胞は、当該技術分野において既知である任意の技術を使用して、例えば、それらを骨髄腫細胞と融合させて、ハイブリドーマを産生することによって、不死化することができる。ハイブリドーマ産生融合手順における使用のための骨髄腫細胞は、好ましくは、非抗体産生であり、高い融合効率と、それらが所望の融合細胞(ハイブリドーマ)のみの増殖を支持する特定の選択的培地内で増殖することを不可能にする酵素欠損とを有する。マウス融合における使用に好適な細胞株の例としては、Sp−20、P3−X63/Ag8、P3−X63−Ag8.653、NS1/1.Ag4 1、Sp210−Ag14、FO、NSO/U、MPC−11、MPC11−X45−GTG1.7、及びS194/5XXO Bulが挙げられ、ラット融合において使用される細胞株の例としては、R210.RCY3、Y3−Ag1.2.3、IR983F、及び4B210が挙げられる。細胞融合に有用な他の細胞株は、U−266、GM1500−GRG2、LICR−LON−HMy2、及びUC729−6である。
前述の配列に基づくキメラ及びヒト化抗体も提供される。治療剤としての使用のためにモノクローナル抗体は、使用前に種々の方法で改変されることができる。1つの例は、共有結合で連結されて、機能性免疫グロブリン軽もしくは重鎖またはその免疫学的に機能的な部分を産生する異なる抗体に由来するタンパク質セグメントから構成される抗体である、キメラ抗体である。概して、重鎖及び/または軽鎖の一部分は、特定の種に由来するか、または特定の抗体のクラスまたはサブクラスに属する抗体内の対応する配列と同一または相同であり、一方で鎖(単数または複数)の残りの部分は、別の種に由来するか、または別の抗体のクラスまたはサブクラスに属する抗体内の対応する配列に同一または相同である。キメラ抗体に関連する方法に関しては、例えば、参照により本明細書に援用される米国特許第4,816,567号及びMorrison et al.,1985,Proc.Natl.Acad.Sci.USA81:6851−6855を参照されたい。CDR移植は、例えば、米国特許第6,180,370号、同第5,693,762号、同第5,693,761号、同第5,585,089号、及び同第5,530,101号に説明される。
完全ヒト抗体も提供される。人間を抗原に曝露することなく所与の抗原に特異的な完全ヒト抗体を作製するための方法が、利用可能である(「完全ヒト抗体」)。完全ヒト抗体の産生を実施するために提供される1つの具体的な手段は、マウス体液性免疫系の「ヒト化」である。内因性Ig遺伝子が不活性化されたマウスへのヒト免疫グロブリン(Ig)遺伝子座の導入は、任意の望ましい抗原で免疫付与され得る動物、マウスにおいて完全ヒトモノクローナル抗体(mAb)を産生する1つの手段である。完全ヒト抗体の使用は、マウスまたはマウス由来mAbを治療剤としてヒトに投与することによって引き起こされ得ることがある免疫原性及びアレルギー性応答を最小にすることができる。
提供される抗体はまた、上記に説明される1つ以上のCDRまたは1つ以上の可変領域を含む二重特異性及び二重機能性抗体も含む。二重特異性または二重機能性抗体は、場合により、2つの異なる重/軽鎖対と2つの異なる結合部位とを有する人工ハイブリッド抗体である。二重特異性抗体は、ハイブリドーマの融合またはFab’断片の連結が挙げられるが、これらに限定されない多様な方法によって産生することができる。例えば、Songsivilai and Lachmann,1990,Clin.Exp.Immunol.79:315−321、Kostelny et al.,1992,J.Immunol.148:1547−1553を参照されたい。
提供される抗体の幾つかは、上に開示される抗体の改変体形態である。例えば、抗体の幾つかは、上記に列挙される重鎖または軽鎖、可変領域またはCDRのうちの1つ以上の中に1つ以上の保存的アミノ酸置換を有する。

抗体は、天然の種に見出されるものとは異なるか、またはそれから変更されたグリコシル化パターンを有することができる。当該技術分野において既知である通り、グリコシル化パターンは、タンパク質の配列(例えば、下記に考察される、特定のグリコシル化アミノ酸残基の有無)、またはタンパク質が産生される宿主細胞もしくは有機体の両方に依存することができる。特定の発現系は、下に考察される。
幾つかの実施形態では、抗原結合は、1つ以上の標識を含む。「標識基」または「標識」という用語は、任意の検出可能な標識を意味する。好適な標識基の例としては、以下のものが挙げられるが、これらに限定されない:放射性同位体もしくは放射性核種(例えば、3H、14C、15N、35S、90Y、99Tc、111In、125I、131I)、蛍光基(例えば、FITC、ローダミン、ランタニドリン光体)、酵素基(例えば、セイヨウワサビペルオキシダーゼ、β−ガラクトシダーゼ、ルシフェラーゼ、アルカリフォスファターゼ)、化学発光基、ビオチニル基、または二次レポーターによって認識される所定のポリペプチドエピトープ(例えば、ロイシンジッパー対配列、二次抗体のための結合部位、金属結合ドメイン、エピトープタグ)。幾つかの実施形態では、標識基は、種々の長さのスペーサーアームを介して抗体に結合されて、潜在的な立体障害を低減する。タンパク質を標識するための種々の方法は、当該技術分野において既知であり、適切と思われるように使用することができる。
一態様は、特定のハイブリダイゼーション条件下で他の核酸(例えば、表1A及び1Bに列挙されるヌクレオチド配列を含む核酸)をハイブリダイズする核酸をさらに提供する。核酸をハイブリダイズするための方法は、当該技術分野において周知である。例えば、Current Protocols in Molecular Biology,John Wiley&Sons,N.Y.(1989),6.3.1−6.3.6を参照されたい。本明細書に定義される通り、適度に厳密なハイブリダイゼーション条件は、5x塩化ナトリウム/クエン酸ナトリウム(SSC)、0.5%のSDS、1.0mMのEDTA(pH8.0)を含有する予洗浄液、約50%のホルムアミド、6xSSCのハイブリダイゼーション緩衝剤、及び55℃のハイブリダイゼーション温度(または他の類似のハイブリダイゼーション溶液、例えば、約50%のホルムアミドを含有し、42℃のハイブリダイゼーション温度を有するものなど)、及び0.5xSSC、0.1%のSDS中での60℃の洗浄条件を使用する。厳密なハイブリダイゼーション条件は、45℃にて6xSSC中でハイブリダイズした後、68℃にて0.1xSSC、0.2%のSDS中で1回以上洗浄する。さらに、当業者は、典型的には互いに少なくとも65%、70%、75%、80%、85%、90%、95%、98%、または99%同一であるヌクレオチド配列を含む核酸が、互いにハイブリダイズされたままであるように、ハイブリダイゼーション及び/または洗浄条件を操作して、ハイブリダイゼーションの厳密さを増加または減少させることができる。
別の態様は、ポリペプチドをコードする核酸またはその一部(例えば、1つ以上のCDRまたは1つ以上の可変領域ドメインを含有する断片)を含むベクターを提供する。ベクターの例としては、プラスミド、ウイルスベクター、非エピソーム哺乳類ベクター、及び発現ベクター、例えば、組み換え発現ベクターなどが挙げられるが、これらに限定されない。組み換え発現ベクターは、宿主細胞内での核酸の発現に好適な形態の核酸を含むことができる。組み換え発現ベクターは、発現のために使用される宿主細胞に基づいて選択される1つ以上の調節配列を含み、それは、発現される核酸配列に作動可能に連結される。調節配列としては、多くの種類の宿主細胞においてヌクレオチド配列の構成的発現を方向付けるもの(例えば、SV40初期遺伝子エンハンサー、ラウス肉腫ウイルスプロモーター、及びサイトメガロウイルスプロモーター)、特定の宿主細胞においてのみヌクレオチド配列の発現を方向付けるもの(例えば、組織特異的な調節配列、その全体が参照により本明細書に援用されるVoss et al.,1986,Trends Biochem.Sci.11:287,Maniatis et al.,1987,Science236:1237を参照されたい)、及び特定の処理または条件に応答してヌクレオチド配列の誘導性発現を方向付けるもの(例えば、哺乳類細胞内におけるメタロチオネインプロモーター、ならびに原核及び真核細胞系の両方におけるtet応答性及び/またはストレプトマイシン応答性プロモーター(同上参照)が挙げられる。発現ベクターの設計は、形質転換される宿主細胞の選択、所望のタンパク質の発現のレベルなどの因子に依存し得ることが当業者によって理解されるであろう。発現ベクターは、宿主細胞内に導入されて、それによって、融合タンパク質またはペプチドを含む、本明細書に説明される核酸によってコードされるタンパク質またはペプチドを産生することができる。
提供される非ヒト抗体は、例えば、マウス、ラット、ウサギ、ヤギ、ロバ、または非ヒト霊長類(サル(例えば、カニクイザルまたはアカゲザル)または類人猿(例えば、チンパンジー)など)などの任意の抗体産生動物に由来することができる。非ヒト抗体は、例えば、インビトロの細胞培養及び細胞培養に基づいた用途、あるいは該抗体に対する免疫応答が発生しないかもしくはわずかである、防止され得る、懸案事項ではない、または所望される任意の他の用途に使用することができる。ある特定の実施形態では、抗体は、上記に説明される通り、当該技術分野において既知である方法を使用して動物を免疫付与することによって産生することができる。抗体は、ポリクローナル、モノクローナルであってもよく、または組み換えDNAを発現することによって宿主細胞内で合成されてもよい。完全ヒト抗体は、上に説明される通り、ヒト免疫グロブリン遺伝子座を含有する遺伝子導入動物を免疫付与することによって、またはヒト抗体のレパートリーを発現しているファージディスプレイライブラリを選択することによって調製することができる。
少なくとも1つの上記に説明されるポリヌクレオチドを含むプラスミド、発現ベクター、転写または発現カセットの形態の発現系及び構築物、ならびにかかる発現系または構築物を含む宿主細胞もまた、本明細書に提供される。
抗体は、生物学的試料中のPAC1の検出及びPAC1を産生する細胞または組織の特定に有用である。例えば、PAC1抗体は、診断的分析、例えば、組織または細胞内で発現されるPAC1を検出及び/または定量化するための結合分析に使用することができる。PAC1に特異的に結合する抗体は、PAC1に関連する疾患の治療を必要とする患者における、PAC1に関連する疾患の治療に使用することもできる。それに加えて、PAC1抗体は、PAC1がそのリガンドPACAP(例えば、PACAP−38)と複合体を形成することを阻害し、それによって細胞または組織中のPAC1の生物学的活性を調節するために使用することができる。調節することができる活性の例としては、血管拡張の阻害及び/または神経性炎症の減少が挙げられるが、これらに限定されない。したがって、PAC1に結合する抗体は、他の結合化合物との相互作用を調節及び/または遮断することができ、したがって、PAC1に関連する疾患の改善における治療的使用を有することができる。
ヒトPAC1に関連付けられる疾患または状態としては、患者におけるその発症が、少なくとも部分的に、PAC1のそのリガンドPACAP(例えば、PACAP−38)との相互作用によって引き起こされる任意の疾患または状態が挙げられる。疾患または状態の重篤度もまた、PAC1のPACAP(例えば、PACAP−38)との相互作用によって増加または減少され得る。本明細書に説明される抗体で治療することができる疾患及び状態の例としては、頭痛、例えば、群発頭痛、片頭痛(migraine headaches)を含む片頭痛(migraine)など、慢性疼痛、糖尿病(II型)、炎症、例えば、神経性炎症など、循環器疾患、ならびに内毒素血症及び敗血症に伴う血行動態異常が挙げられる。
本明細書に説明される抗体は、PAC1に関連付けられる疾患及び/または状態を検出、診断、または観察するために、診断的目的のために使用することができる。当業者に既知である伝統的な免疫組織学的方法(例えば、Tijssen,1993,Practice and Theory of Enzyme Immunoassays,Vol15(Eds R.H.Burdon and P.H.van Knippenberg,Elsevier,Amsterdam)、Zola,1987,Monoclonal Antibodies:A Manual of Techniques,pp.147−158(CRC Press,Inc.)、Jalkanen et al.,1985,J.Cell.Biol.101:976−985、Jalkanen et al.,1987,J.Cell Biol.105:3087−3096)を使用した、試料中のPAC1の存在の検出のための方法も提供される。PAC1の検出は、インビボまたはインビトロで実施することができる。

PAC1抗体
A.抗PAC1抗体の生成
完全ヒト抗体を、標準的な方法を使用して、例えば、実質的にUS特許公開第US2010−0172895A1号に詳述される通りに、PAC1細胞外ドメインタンパク質、T細胞エピトープタグでタグ付けされたDNA、全長ヒトPAC1を発現するL1.2細胞、及び他のhPAC1抗原によるXENOMOUSE動物の免疫付与を通じて生成した。
ハイブリドーマ上清を、PAC1への結合に関して、また機能性拮抗薬活性に関しても、PACAP(例えば、PACAP−27またはPACAP−38)かまたは選択的な外因性ペプチドリガンド(マキサディラン)かのいずれかによるPAC1の活性化によるcAMPの生成を遮断するそれらの能力を検出する分析においてスクリーニングし、次に関連する受容体VPAC1及びVPAC2に対してカウンタースクリーニングした。望ましい機能及び選択性を有するその上清を、標準的な方法を使用して、例えば、実質的に参照により本明細書に援用されるUS特許公開第US2010−0172895A1号に詳述される通り、配列決定及びクローン化し、組み換え的に発現させ、精製し、機能及び選択性に関して再び試験した。
実験を実施して、関連する受容体PAC1、VPAC1、及びVPAC2に対する3つのPAC1作動薬(PACAP、VIP、及びマキサディラン)の活性及び選択性を評価した。これらの実験のデータは下の表7に示され、PACAPは、3つの全ての受容体において類似の作動薬活性を有し、VIPは、PAC1において幾らかの活性を有するが、VPAC1及びVPAC2において(約10〜100倍)高い活性を有し、マキサディランは、VPAC1及びVPAC2と対比してPAC1に高度に選択的であることを実証している。
cAMP機能分析におけるPAC1特異性遮断モノクローナル抗体の活性
A.抗PAC1抗体の活性
選択された本明細書に説明されるhPAC1抗体を、インビトロのPAC1媒介cAMP分析においてスクリーニングして、固有の有効性を決定した。分析は、SH−SY−5Y、hPAC1を内因的に発現するヒト神経芽細胞腫細胞株(Biedler,J.L.,et al.,1978,“Multiple neurotransmitter synthesis by human neuroblastoma cell lines and clones”.Cancer Res.38(11Pt1):3751−7、Lutz,E.M.,et al.,“Characterization of novel splice variants of the PAC1 receptor in human neuroblastoma cells:consequences for signaling by VIP and PACAP.”Mol Cell Neurosci2006;31:193−209)、及び以下の受託番号を有する、組み換えPAC1、VPAC1&VPAC2構築物を発現する複数の細胞株を用いた:ヒトPAC1(hPAC1):NM_001118.4、hPAC1短(アミノ酸残基89〜109が欠失されたhPAC1(使用範囲:1〜88、110〜468)):NM_001199637.1、カニクイザルPAC1:XM_005549858.1、ラットPAC1:NM_133511.2、マウスPAC1:NM_001025372.2、ヒトVPAC1(hVPAC1):NM_004624.3、ヒトVPAC2(hVPAC2):NM_003382.4、ラットVPAC2:NM_017238.1、マウスVPAC2:NM_009511.2。
受容体遮断抗体のKi決定のための放射性リガンド結合分析
どちらもPerkin Elmer(Billerica,MA)から得た125Iで標識されたPAC−1抗体(PAC1結合分析のため)または125Iで標識されたPEG−PACAP38(PACAP38結合分析のため)、及びhPAC−1 AequoScreen CHO(PerkinElmer Life and Analytical Sciences,Boston,Massachusetts)から調製した細胞膜を、種々の濃度の試験抗体の存在下で放射性リガンド結合実験に使用して、対応するKi値を決定した。
細胞を、Ham’sF12、10%のFBS、100μg/mlのストレプトマイシン、400μ/mlのG418(受容体発現選択)及びZeocin(エクオリン発現選択)中で、40xT225フラスコ内で集密になるまで増殖させた。フラスコを、Ca/Mgを含まないPBSで1回洗浄した。PBSを完全に除去した。5mlのVERSENEを各フラスコに添加し、約5分間待機した。フラスコの側部を叩き、全ての細胞を払い落とした。全てのフラスコからのVERSENE/細胞懸濁液を、50mlの円錐形管に移し、氷の上に定置した。全ての残留細胞を収集するために、フラスコを、Ca/Mgを有さず、プロテアーゼ阻害薬を有する氷冷PBSで濯ぎ、溶液を、管内の細胞に添加した。管を、1000rpmで10分間遠沈させた。ペレットを、−80℃で保管した。
PAC1結合分析を、120μl結合緩衝剤(20mMのトリスHCl、pH7.5、5.0mMのMgSO4、150mMのNaCl、0.1%のBSA(Sigma)、CompleteTM/50ml緩衝剤の錠剤1個(プロテアーゼ阻害薬))、10μlの試験化合物(20X)、50μlのヒトPAC1CHO膜懸濁液(1ウエルあたり0.2μg)、及び20μlの125I−PAC−1抗体(10X、PAC1結合分析のため)かまたは125I−PEG−PACAP38(10X、PACAP38結合分析のため)かのいずれかを含有する96ウエルプレート内で、室温で設定した。
KINEXAによって測定される結合KD
A.hPAC1を発現する全細胞を使用した実験。
抗Pac1抗体のCHOK1/huPac1細胞との結合を、KinExA上で試験した。手短に述べると、UltraLink Biosupport(Pierceカタログ番号53110)を、ヤギ抗huFc(Jackson Immuno Researchカタログ番号109−005−098)で予備コーティングし、BSAで遮断した。10pM、30pM、100pM、及び300pMの抗Pac1抗体を、1%のFBS及び0.05%アジ化ナトリウムを含有する培地F12中で、huPac1を発現する種々の密度(1.7x101〜9x106細胞/mL)のCHOK1細胞と共に定温放置した。抗Pac1抗体及び全細胞を含有する試料を、室温で4時間、回転させながら定温放置した後、4℃で5分間、220gで遠心分離して、全細胞及び抗Pac1抗体細胞の複合体を未結合抗Pac1抗体から分離した。遊離抗Pac1抗体を含有する上清を、0.22μmフィルタを通して濾過した後、ヤギ抗huFcコーティングビーズをKinExA上に充填した。ビーズに結合した抗Pac1抗体の量を、蛍光(Alexa Fluor647)標識された抗huIgG(H+L)抗体(Jackson Immuno Researchカタログ番号109−605−088)によって定量化した。結合シグナルは、各細胞密度における溶液中の遊離抗Pac1抗体の濃度に比例する。平衡解離定数(KD)を、KinExATM Proソフトウエア(Sapidyne Instruments,Inc.,Boise,ID)におけるn曲線分析によって概算した。
抗Pac1AbのhuPac1(1−135)::6xHisとの結合を、KinExA上で試験した。手短に述べると、UltraLink Biosupport(Thermo Fisher Scientificカタログ番号53110)を、huPac1(1−135)::6xHisで予備コーティングし、上記に説明される通りBSAで遮断した。30、100、及び300pMのAbを、種々の濃度のhuPac1(1−135)::6xHisと共に室温で少なくとも8時間定温放置した後、huPac1(1−135)::6xHisでコーティングしたビーズを通して走らせた。ビーズに結合したAbの量を、蛍光(DyLight649)標識されたヤギ抗huIgG(H+L)抗体(Jackson Immuno Researchカタログ番号109−495−088)によって定量化した。結合シグナルは、結合平衡における遊離Abの濃度に比例する。平衡解離定数(KD)を、1サイト均質結合モデル(KinExATM Pro software)を使用して競合曲線の非線形回帰から取得した。
血小板食作用分析
任意の潜在的なFcγ受容体誘発血小板減少に対処するために、本明細書に説明される抗体のサブセットを、食作用分析において試験した。
ヒト及びカニクイザルの両方の血小板を、以前に説明されている通りに調製した(Semple,J.W.,et al.,Blood(2007)109:4803−4805)。手短に述べると、健康なカニクイザルドナーからの全血液試料を、15分間制動せずに170xgで遠心分離した。血小板富化血漿(PRP)を、上層から収集した。試料の下層を、2000xgで10分間遠心分離して、血小板欠乏血漿(PPP)層及びバフィーコートを得た。PRP画分中の血小板を、20μMのCell Tracker Green CMFDAで、室温で30分間標識した。標識された血小板を、PBSで洗浄し、10%のFBSを伴うRPMI−1640中で再懸濁し、1x108細胞/mLの濃度に調節した。PBLを、同一のドナーから得たバフィーコートから調製した。赤血球を、BD Pharm−lyseを用いて、販売者によって提供されるプロトコルに従って溶解した。溶解後、PBLを、10%のFBSを伴うRPMI−1640で2回洗浄し、最終濃度を5x106細胞/mlに調節した。
食作用反応を、各100μLのCMFDAで標識された血小板及びPBMCを、試験物品と共に暗闇で定温放置することによって開始した。6時間の定温放置の後、プレートを氷の上に定置することによって反応を停止した。細胞外蛍光を、0.1%トリパンブルーと共に2分間定温放置することによって消光した。氷冷PBSで2回洗浄した後、細胞を、ヨウ化プロピジウムおよび抗CD14と共に4℃で30分間定温放置した。洗浄後、細胞を、BD LSRII Flow Cytometerによって分析した(図2)。先行経験に基づき、単球貪食血小板は、負の対照(a−SAかまたはa−DNPかのいずれか)の少なくとも2倍以上のCMFDA中央蛍光強度値を有する(2)。
Claims (30)
- ヒトPAC1に特異的に結合する単離された抗体またはその抗原結合断片であって、
(A)(i)表4Aに記載される軽鎖CDR1配列から成る群から選択されるアミノ酸配列、(ii)表4Aに記載される軽鎖CDR1配列から成る群から選択されるアミノ酸配列に少なくとも90%同一であるアミノ酸配列、または(iii)表4Aに記載される軽鎖CDR1配列から成る群から選択されるアミノ酸配列に少なくとも95%同一であるアミノ酸配列を含む、軽鎖CDR1と、
(B)(i)表4Aに記載される軽鎖CDR2配列から成る群から選択されるアミノ酸配列、(ii)表4Aに記載される軽鎖CDR2配列から成る群から選択されるアミノ酸配列に少なくとも90%同一であるアミノ酸配列、または(iii)表4Aに記載される軽鎖CDR2配列から成る群から選択されるアミノ酸配列に少なくとも95%同一であるアミノ酸配列を含む、軽鎖CDR2と、
(C)(i)表4Aに記載される軽鎖CDR3配列から成る群から選択されるアミノ酸配列、(ii)表4Aに記載される軽鎖CDR3配列から成る群から選択されるアミノ酸配列に少なくとも90%同一であるアミノ酸配列、または(iii)表4Aに記載される軽鎖CDR3配列から成る群から選択されるアミノ酸配列に少なくとも95%同一であるアミノ酸配列を含む、軽鎖CDR3と、
