Abstract
【解決手段】2−(4−ヒドロキシフェニル)−エタノールを有効成分とする美白剤。好ましくは穀類由来成分を更に含有する、医薬品、医薬部外品又は飲食品である美白剤含有内用又は外用組成物。

マウスB16細胞を用いたメラニン生成抑制試験
<実験方法>
マウスメラノーマ由来B16細胞(理化学研究所)を、マイクロタイタープレートに播種した。詳細には、培地としてDulbecco’s Modified Eagle’s Medium(SIGMA)+10%FBSを用い、12ウエルマイクロタイタープレートに1wellあたり1×104のマウスメラノーマ由来B16細胞を播種した。翌日、細胞が底面に付着していることを確認した後、0.0002〜0.02%の2−(4−ヒドロキシフェニル)−エタノールを培地に添加し、5日間培養を行った(5%CO2存在下37℃)。そして、細胞を剥離させ、細胞懸濁液をマイクロチューブに移して培地を生理食塩水に交換した。その後、遠心により細胞を沈殿させ、細胞ペレットの色によりメラニンの生成量を評価した。
<結果>
図1は、2−(4−ヒドロキシフェニル)−エタノールを添加しなかった場合(control)及び各種濃度(0.0002%、0.002%、0.02%)で添加した場合における、実験後の細胞ペレット写真である。図1から明らかなように、2−(4−ヒドロキシフェニル)−エタノールを添加すると、control(0%)と比較し、顕著なメラニン生成抑制効果を奏することが確認された。また、メラニン生成量は、2−(4−ヒドロキシフェニル)−エタノールの添加量(濃度)に依存することも確認された。尚、2−(4−ヒドロキシフェニル)−エタノール添加による細胞数の変化は認められず、安全性も確認された。
白米を粉砕機にかけ、白米の粉砕物1kgを得た。この粉砕物に液化酵素5gと水3Lを添加し、60℃で4時間放置した。その後、加熱昇温させ、冷却した後、濾過器を用いて固液分離を行い、ろ液2.5Lを得た。これを85℃で30分加熱し、米エキスを得た。その後、当該米エキス中の2−(4−ヒドロキシフェニル)−エタノールの濃度が0.04%となるように前記化合物を添加し、本実施例品を得た。そして、(1)実施例品、(2)同濃度(0.04%)の2−(4−ヒドロキシフェニル)−エタノールのみ、及び、(3)10倍濃度(0.4%)の2−(4−ヒドロキシフェニル)−エタノールのみ、を夫々培地量の5%添加し、実施例1と同様の試験を行った。試験後、電子写真を撮影すると共に、当該写真を画像解析に付して黒化度を算出した。その結果を図2に示す。ここで、図2中、「エキス+0.002%」、「水+0.002%」及び「水+0.02%」が、夫々(1)、(2)及び(3)の黒化度である(尚、括弧中の数値は培地中の有効成分濃度を示す)。図2から明らかなように、「エキス+0.002%」と「水+0.002%」は、有効成分{2−(4−ヒドロキシフェニル)−エタノール}の含有量が同量であるにも係らず、遥かに強い美白効果が確認できた。更に、「エキス+0.002%」と「水+0.02%」は、有効成分{2−(4−ヒドロキシフェニル)−エタノール}の含有量が10倍も相違するにも係らず、同等の美白効果を奏することが確認できた。このように、本有効成分を米エキスと組み合わせた場合には、美白に関する相乗効果を奏することが判明した。
