Description
本発明は、神経成長因子(NGF)に結合するヒトモノクローナル抗体に関する。疼痛および疼痛関連障害を処置するための組成物および方法も説明する。
米国では、毎日、2百万人を超える人々が、慢性疼痛によって能力が損なわれている(JessellおよびKelly、1991、「Pain and Analgesia」、PRINCIPLES OF NEURAL SCIENCE、第3版、(Kandel、Schwartz、およびJessell編)、Elsevier、New York)。残念なことに、現在の疼痛の処置は、ある程度しか有効ではなく、多くのこれらの処置自体が、衰弱または危険な副作用をもたらす。例えば、非ステロイド性抗炎症薬(「NSAID」)、例えば、アスピリン、イブプロフェン、およびインドメタシンは、炎症性疼痛に対して中程度に有効であるが、腎毒性でもあり、大量投与は、胃腸刺激、潰瘍、出血、および精神錯乱をもたらす傾向にある。オピオイドで処置された患者も、多くの場合、錯乱を経験し、長期のオピオイドの使用は、耐性および依存と関連する。局所麻酔薬、例えば、リドカインおよびメキシレチンは、疼痛を阻害するのと同時に、通常の感覚を喪失させる。

(1)抗体のエフェクター部分がヒトであるため、ヒト抗体は、ヒト免疫系の他の部分と良好に相互作用し得る(例えば、補体依存性細胞毒性(CDC)または抗体依存性細胞毒性(ADCC)によって標的細胞をより効率的に破壊する);

二重特異性抗体または二機能性抗体は、典型的には、2つの異なる重鎖/軽鎖対および2つの異なる結合部位を有する人工ハイブリッド抗体である。二重特異性抗体は、限定されるものではないが、ハイブリドーマの融合またはF(ab’)フラグメントの連結を含む様々な方法によって作製することができる。例えば、SongsivilaiおよびLachmann、1990、Clin.Exp.Immunol.79:315−321;Kostelnyら、1992、J.Immunol.148:1547−1553を参照されたい。
本発明は、ヒトNGFに結合する抗体を提供する。これらの抗体は、完全長NGFまたはそのフラグメントを用いた免疫化によって作製することができる。本発明の抗体は、ポリクローナル抗体またはモノクローナル抗体であっても良く、かつ/または組換え抗体であっても良い。好ましい実施形態では、本発明の抗体は、例えば、ヒト抗体を産生し得るトランスジェニック動物の免疫化によって調製されるヒト抗体である(例えば、国際特許出願公開第93/12227号を参照されたい)。

(大腸菌細胞からのヒトNGFタンパク質の生産)
(rHu−NGF(1〜120)のクローン化)
ヒトNGFをコードするヌクレオチド配列を、配列番号27および配列番号28に示されている配列を有するオリゴヌクレオチドプライマーならびに標準的なPCR技術を用いてcDNAから増幅した。5’プライマーは、成熟配列のコドン1(セリン)の直前にNdeI制限部位およびメチオニン開始コドンを形成する。3’プライマーは、終止コドンの直後にBamHI制限部位を形成する。得られたPCR産物を、ゲル精製し、制限エンドヌクレアーゼNdeIおよびBamHIで消化し、次いで、同様にNdeIおよびBamHIで消化されたベクターpCFM1656に連結した。連結されたDNAで、大腸菌株657のコンピテント宿主細胞を形質転換した。組換えタンパク質産物を産生する能力、および正確なヌクレオチド配列(すなわち、配列番号29)を有するプラスミドを保有する能力について、クローンをスクリーニングした。組換えヒトNGF1〜120のアミノ酸配列は、配列番号30に示されている。
NGF発現構築物(上記のような)を含む大腸菌細胞を、流加様式で富栄養培地で発酵させた。細胞を、600nmでのODが49になるまで30℃で増殖させ、次いで42℃に温度を変更することによって誘導した。細胞を、誘導の4時間後に遠心分離によって収集した。最終ODは、75であった。発現収率は、約0.15g/Lであることが決定された。
細胞ペーストを、Microfluidizerで溶解させ、10,000×gで30分間遠心分離し、ペレットを1%のデオキシコール酸で洗浄し、上記のように遠心分離し、次いで得られたペレットを冷水で洗浄して再び遠心分離した。得られたペレット(WIB−洗浄された封入体)を、変性剤(8MグアニジンHCl、50mM Tris pH8.5(10mM DTT含有))で再懸濁し、室温で1時間可溶化し、10,000×gで30分間遠心分離し、その上清を注意深くデカントし、次いで4℃のレドックス対を含む水性緩衝液で25倍に希釈して5日間置いた。次いで、得られたリフォールディング物を、pH3.0にまでpH調整し(titrate)、0.45μMのフィルターに通して濾過した。このリフォールディング物を、標準的なNaCl勾配を用いるSp−Sepharose高速カラムで精製した。続いて、陽イオン交換カラムからのプールを濃縮し、アリコートを−80℃で凍結させた。このタンパク質の純度を、SDS−ポリアクリルアミドゲル電気泳動(SDS−PAGE)によって評価し、クマーシーブルー染色によって分析した。精製されたタンパク質は、この方法によると、主バンドが90%を超えていた。
(神経成長因子(NGF)に対するヒトモノクローナル抗体の生産)
(トランスジェニックHuMabおよびKMマウス)
NGFに対する完全ヒトモノクローナル抗体を、それぞれがヒト抗体遺伝子を発現する、トランスジェニックマウスのHCo7株、HCo12株、HCo7+HCo12株、およびKM株を用いて調製した。これらのマウス株のそれぞれでは、内因性マウスκ軽鎖遺伝子は、Chenら(1993、EMBO J.12:811−820)に記載されているようにホモ接合性に破壊されており、内因性マウス重鎖遺伝子は、国際特許出願公開第01/09187号(参考として組み入れられる)の実施例1に記載されているようにホモ接合性に破壊されていた。これらの各マウス株は、Fishwildら(1996、Nature Biotechnology 14:845−851)に記載されているようにヒトκ軽鎖導入遺伝子KCo5を有する。HCo7株は、米国特許第5,545,806号、同第5,625,825号、および同第5,545,807号(参考として組み入れられる)に記載されているようにHCo7ヒト重鎖導入遺伝子を有する。HCo12株は、国際特許出願公開第01/09187号(参考として組み入れられる)の実施例2に記載されているようにHCo12ヒト重鎖導入遺伝子を有する。HCo7+HCo12株は、HCo7およびHCo12の両方の重鎖導入遺伝子を有し、それぞれの導入遺伝子に対して半接合性である。KMマウスは、Tomizukaら(1997、Nature Genet.16,133−143および2000、Proc.Natl.Acad.Sci、97,722−727)に記載されているようにSC20重鎖導入遺伝子を含む。この導入遺伝子は、マウス染色体には組み込まれないが、代わりに独立した染色体フラグメントとして増殖させられる。このフラグメントは、約15MBのヒト第14染色体を含む。これは、全てのVH、D、およびJH遺伝子セグメントならびに全ての重鎖定常領域アイソタイプを含むヒト重鎖遺伝子座全体を含む。全てのこれらの株は、本明細書ではHuMabマウスと呼ばれる。
NGFに対する完全ヒトモノクローナル抗体を作製するために、HuMabマウスを、抗原として大腸菌細胞由来の精製組換えNGFを用いて免疫化した(実施例1)。HuMabマウスの一般的な免疫化スキームは、Lonbergらに記載されている(それぞれの教示が参考として本明細書に組み入れられる、1994、Nature 368:856−859;Fishwildら、前出、および国際特許出願公開第98/24884号)。マウスは、抗原の最初の注入に際し、6〜16週齢であった。NGF抗原の精製組換え調製物(25〜100μg)を用いて、HuMabマウスを腹腔内(IP)または皮下(SC)で免疫化した。
モノクローナル抗体産生用のマウスを調製し、屠殺の2日前に抗原で静脈内に追加免疫を行い、その後に脾臓を取り出した。HuMabマウスからマウス脾細胞を単離して、標準的なプロトコルを用いてPEGでマウス骨髄腫細胞系に融合した。典型的には、各抗原に対して10〜20の融合を行った。
上記のようなELISAアッセイを用いて、NGF免疫原で陽性反応性を示すハイブリドーマをスクリーニングした。NGFに対して高い結合活性で結合するモノクローナル抗体を分泌するハイブリドーマをサブクローン化してさらに特徴付けた。液体窒素中に保存する5〜10のバイアルの細胞バンクを作るために、親細胞の反応性を維持している(ELISAによって決定)各ハイブリドーマ由来の1つのクローンを選択した。
(強力なNGF中和活性を有する抗NGF抗体の選択およびクローン化)
NGF活性のインヒビターとして(すなわち、NGF「中和」)、実施例2で最初に同定された抗体の有効性を、それぞれの改変されたペプチドが、バニロイドレセプター−1(VR1)発現のNGF誘導を阻止する能力を測定することによって評価した。
時期を見て妊娠させた、終末麻酔したSprague−Dawleyラット(Charles River、Wilmington,MA)の子宮から外科的に取り出した、胎齢19日(E19)のラットの全ての脊髄分節から、無菌条件下で後根神経節(DRG)を1つずつ切り離した。5%熱不活化ウマ血清(GibcoBRL、Grand Island,NY)を含む氷冷L−15培地(GibcoBRL)にDRGを集め、任意の疎性結合組織および血管を取り除いた。pH7.4の、Ca2+およびMg2+を含まないダルベッコリン酸緩衝生理食塩水(DPBS)(GibcoBRL)で、DRGを2回リンスした。次いで、DRGを、パパイン解離系(Worthington Biochemical Corp.、Freehold,NJ)を用いて単一細胞懸濁物へと解離させた。簡単に述べると、DRGを、アール平衡塩溶液(EBSS)において20U/mLのパパインを含む消化溶液中、37℃で50分間インキュベートした。1:1のMEM/Ham’s F12、1mg/mLオボムコイドインヒビター、および1mg/mlオボアルブミン、および0.005%デオキシリボヌクレアーゼI(DNase)からなる解離培地中で、先端熱加工(fire−polished)パスツールピペットを用いたすりつぶしによって細胞を解離させた。
プレーティングの2時間後に、細胞を、組換えヒトβ―NGF(Amgen)または組換えラットβ−NGF(R&D Systems、Minneapolis,MN)で、10ng/mLの濃度(0.38nM)で処理した。連続希釈した抗NGF抗体(R&D
Systems)を含む陽性コントロールを、それぞれの培養プレートに適用した。試験抗体を、3.16倍の連続希釈法を用いて10個の濃度で添加した。全てのサンプルを、培養物に添加する前に完全培地で希釈した。インキュベーション時間は、VR1発現の測定の前の40時間とした。
培養物を、4%パラホルムアルデヒドを含むハンクス平衡塩溶液で15分間固定し、Superblock(Pierce、Rockford,IL)でブロッキングし、0.25% Nonidet P−40(Sigma)を含むTris−HCl(Sigma)緩衝生理食塩水(TBS)を用いて室温で1時間、透過処理した。培養物を、0.1%Tween 20(Sigma)を含むTBSで1回リンスし、ウサギ抗VR1 IgGと共に室温で1時間半インキュベートし、続いてEu標識した抗ウサギ二次抗体(Wallac Oy、Turku,Finland)と共に室温で1時間インキュベーションした。それぞれの抗体のインキュベーション後に、TBSで洗浄した(ゆっくり振盪しながら3×5分)。増強溶液(150μL/ウェル、Wallac Oy)を培養物に添加した。次いで、蛍光シグナルを、時間分解蛍光光度計(Wallac Oy)で測定した。改変ペプチドで処理したサンプル中のVR1発現を、0〜1000ng/mLのNGF漸増標準曲線と比較することによって決定した。DRGニューロンにおけるVR1発現に対するNGF効果の阻害パーセント(最大可能阻害と比較)を、NGF処理されていないコントロールと比較することによって決定した。結果を表2および表5に示す。
精製した抗NGFハイブリドーマ抗体サンプルを、タンパク質の配列決定およびLC/MS分析のために調製した。容積が15mL未満になるまでAmicon centriprep−30を用いて培地を濃縮することにより、抗体を馴化培地から精製した。rProA(Pharmacia)レジンのバッチを、PBSで4回洗浄し、最後の洗浄の後にPBSで50%スラリーを形成した。適切な量のrProAレジン(レジン1μLあたり約5μgの抗体であるが、少なくとも50μLのレジンを使用する)を、抗体サンプルに添加し、4℃で一晩インキュベートした。Ab−レジン混合物を遠心分離し、未結合の画分を収集した。0.5mL PBSを添加して0.45μm Spin−X(CoStar)管に移した後、サンプルを10000rpmで3分間遠心分離した。次に、レジンを0.5mL PBSで少なくとも3回洗浄し、次いで0.1M グリシン(pH2.7)をレジンの1.5倍量で添加し、室温で10分間インキュベートし、続いて10000rpmで3分間さらに遠心分離し、上清を収集した。この溶出工程をさらに2回繰り返し、次いで合わせた上清を、1/25倍量の1.0M tris(pH9.2)で中和した。
最も強力なNGF結合モノクローナル抗体を発現するハイブリドーマ、4D4.D7を供給源として使用して、TRIzol(登録商標)試薬(Invitrogen)を用いて全RNAを単離した。第1鎖cDNAを、伸長アダプター(5’−GGC CGG ATA GGC CTC CAN NNN NNT−3’)(配列番号33)と共にランダムプライマーを用いて合成し、次いで5’RACE(cDNA末端の迅速増幅)調製アッセイ(preparative assay)を、GeneRacerTMKit(Invitrogen)を用いて製造業者の取扱説明書にしたがって行った。完全な軽鎖をコードするcDNAを調製するために、順方向プライマーを、GeneRacerTMネスト化プライマーとし、逆方向プライマーを、5’−GGG GTC AGG CTG GAA CTG AGG−3’(配列番号34)とした。重鎖可変領域をコードするcDNAを調製するために、順方向プライマーを、GeneRacerTMネスト化プライマーとし、逆方向プライマーを、5’−TGA GGA CGC TGA CCA CAC G−3’(配列番号35)とした。RACE産物を、pCR4−TOPO(Invitrogen)にクローン化して、その配列を決定した。コンセンサス配列を用いて、完全長の抗体鎖PCR増幅用のプライマーをデザインした。
TCC CCT GTT GAA GCT C−3’;配列番号37)をコードした。得られたPCR産物のフラグメントを精製し、XbaIおよびSalIを用いて消化し、次いでゲル単離し、哺乳動物発現ベクターpDSRα20に連結した(あらゆる目的に対して参考として本明細書に組み入れられる国際特許出願公開第90/14363号を参照されたい。pDSRα20は、部位特異的変異誘発によってpDSRa19のヌクレオチド2563を「グアノシン」から「アデノシン」に変更することによって作製した)。
(チャイニーズハムスター卵巣(CHO)細胞における抗NGF抗体の発現)
4D4抗NGF mAbの安定な発現を、リン酸カルシウム法を用いた、4D4−重鎖/pDSRα19 IgG2または4D4重鎖/pDSRα19 IgG1およびNGF−κ/pDSRα19プラスミドの、ジヒドロ葉酸レダクターゼ欠損(DHFR−)無血清適応チャイニーズハムスター卵巣(CHO)細胞への共トランスフェクションによって達成した。トランスフェクト細胞を、透析血清を含むがヒポキサンチン−チミジンを含まない培地で選択して、DHFR酵素を発現する細胞の増殖を確認した。トランスフェクトクローンを、アッセイ、例えば、ELISAでスクリーニングして、馴化培地中における4D4抗NGF mAbの発現を検出した。発現が最大のクローンに対して、DHFR増幅のためにメトトレキセート(MTX)の濃度を上昇させた。馴化培地中における4D4抗NGF mAbのさらに高い発現を検出するために、MTX増幅クローンを、アッセイ、例えば、ELISAでスクリーニングした。発現が最大のクローンを、サブクローン化に供して均一な集団を得て、細胞バンクを作製した。
(抗NGF 4D4抗体の活性の特徴付け)
スピナー(S)またはローラー(R)の条件下で増殖した細胞で作製された、一過性発現抗NGF 4D4抗体を、上記(実施例3)のように行われるDRGニューロンベースのNGF中和バイオアッセイにおいて、NGFを中和する能力を確認するために試験した。
(抗NGF抗体の生産)
抗NGF抗体を、CHO細胞のクローン系での発現によって生産する。それぞれの産生を行うために、1つのバイアル由来の細胞を無血清細胞培養培地で解凍する。この細胞を、まずTフラスコで増殖させ、続いてスピナーフラスコで増殖させ、次いで2000Lバイオリアクターまで規模が拡大するステンレススチールリアクターで増殖させる。生産は、流加培養法を用いて2000Lバイオリアクターで行い、ここでは細胞の増殖および培養物の生存度を維持するために濃縮された培地成分を含む栄養供給物を加える。生産は約2週間続き、この間、抗NGF抗体が上記細胞によって構成的に産生され、細胞培養培地に分泌される。
(他のニューロトロフィンとの交差反応性)
4D4抗体を、ヒトNT3についてのDRGニューロン生存アッセイおよびヒトBDNFについての培養DAニューロンにおけるDA取り込みのアッセイを含む様々なバイオアッセイにおいて、ヒトNT3またはヒトBDNFに対するその交差反応性について試験した。
プレーティングの2時間後、DRG細胞(実施例3で上記された単離手順)を、100ng/mL(3.8nM)の組み換えhNT−3で処理した。連続希釈した抗hNT3抗体(R&D)を陽性コントロールとして用いた。未知の物質(抗NGF Abサンプル)を、10ポイントの3.16倍連続希釈の様々な濃度で添加した。全てのサンプルを完全培地で希釈してから培養物に添加した。
培養物を、4%パラホルムアルデヒドを含むハンクス平衡塩溶液を用いて15分間固定し、Superblock(Pierce)で1時間ブロッキングし、0.25% Nonidet P−40(Sigma)を含むTris−HCl(Sigma)緩衝生理食塩水(TBS)を用いて室温(RT)で1時間、透過処理した。培養物を、0.1%Tween20(Sigma)を含むTBSで1回リンスし、マウス抗MAP2IgG(Chemicon、Temecula,CA)と共に室温で1.5時間インキュベートし、続いてEu標識した抗マウス二次抗体(Wallac Oy、Turku,Finland)と共に室温で1時間インキュベーションした。それぞれの抗体のインキュベーション後に、TBSで洗浄した(おだやかに振盪しながら3×5分)。増強溶液(150mL/ウェル、Wallac Oy)を培養物に添加し、次いで蛍光シグナルを、時間分解蛍光光度計(Wallac Oy)で測定した。
胎齢19日(E19)のSprague−Dawleyラット(Jackson Labs)を使用した。ドーパミン作動性ニューロンが豊富な腹側中脳組織を取り出して、Ca++およびMg++を含まない冷ダルベッコリン酸緩衝生理食塩水(DPBS)、pH7.4(Gibco)に移した。組織フラグメントは、パパイン解離系(Worthington Biochemical Corp.、Freehold,NJ)を用いて単一細胞懸濁物へと解離させた。簡単に述べると、組織フラグメントを、アール平衡塩溶液(EBSS)において20単位/mLのパパインを含む消化溶液中、37℃で50分間インキュベートした。1:1のMEM/Ham’s F12、1mg/mLオボムコイドインヒビター、および1mg/mLオボアルブミン、および0.005%デオキシリボヌクレアーゼI(DNase)からなる解離培地で、先端熱加工パスツールピペットを用いたすりつぶしによって細胞を解離させた。解離した細胞を、5分間、200×gでペレット化し、1mg/mLのオボムコイドインヒビター、1mg/mLのオボアルブミン、および0.005%DNaseを含むEBSSで再懸濁した。細胞懸濁物を、10mg/mLのオボムコイドインヒビター、10mg/mLのオボアルブミンを含む勾配溶液で、200×gで6分間遠心分離して細胞デブリを除去し;25μgNitexナイロンメッシュ(Tetko,Inc.)に通して濾過して凝集塊を除去した。解離した細胞を、100,000/cm2の密度で組織培養プレートにプレーティングした。このプレートは、既に記載したように、ポリ−オルニチン100μg/mL(Sigma)およびマウスラミニン1μg/mL(Gibco BRL)で事前にコーティングした(Louis JCら、J.Pharmacol.Exp.Ther.1992;262:1274−1283)。この培養培地は、1:1の最小必須培地(MEM)/Ham’s F12、12%ウマ血清(Gibco)、100μg/mLのトランスフェリン、および2.5μg/mLのインスリン(Sigma)から構成された。この培養物を、37℃、5%CO2、および100%の湿度で6日間維持した。
プレーティングの2時間後に、10ng/mLのBDNFを細胞に添加し、続いて連続濃度の抗NGF Abサンプルを添加した。抗BDNF抗体(Amgenで生成)を陽性コントロールとして用いた。
ドーパミン取り込みアッセイを、既に記載されているように行った(Friedman,L.およびMytilineou,C.、Neuroscience Letters 1987;79:65−72)。6日目に、培養物を、5.6mMグルコース、1.3mM EDTA、および0.5mMパルギリン、モノアミンオキシダーゼインヒビターを含む予め温めたクレブス−リンゲルリン酸緩衝液(pH7.4)で1回洗浄した。この培養物を、50nM[3H]DA(NEN)を含む取り込み緩衝液中、37℃で60分間インキュベートした。取り込み緩衝液を除去して取り込みを停止させ、培養物をクレブス−リンゲルリン酸緩衝液で3回洗浄した。細胞を液体シンチレーションカクテル、opticphase supermix(Wallac)を添加することによって溶解させ、[3H]DAを培養物に直接放出させた。次いで、この細胞溶解物を、microbeta−plus液体シンチレーションカウンター(Wallac,Inc.)で放射活性についてカウントした。0.5mM GBR12909、高親和性のDA取り込み部位の特異的なインヒビター(Heikkila REおよびMazino L、European Journal of Pharmacology 1984;103:241−8)を取り込み緩衝液に添加することによって低親和性のDA取り込みを評価して、この値を総取り込み量から減じて高親和性DA取り込み値を得た。
(抗NGF抗体のエピトープの同定)
(限定タンパク質分解によるエピトープマッピング)
5マイクログラム(μg)のNGFを、4D4(11μg)と共に0.1M Tris緩衝液、pH7.5中、4℃で30分間インキュベートした。次いで、この複合体をプロテアーゼ(サブチリシン)1μgを用いて37℃で1時間および2時間消化した。HPLCペプチドマップを、4D4抗体によって保護されたペプチドを探すために互いに比較した。NGFの限定タンパク質分解により、いくつかの大きなペプチドがNGFから最初に放出されたことが示された。特に興味深いペプチドS18.3、S18.5およびS34.4が産生され、タンパク質分解から抗体で保護された。他のピークは、有意に形成または保護されなかった。2つの実験での保護されたペプチド(1時間および2時間の消化)を表7に示す。
サブチリシンで消化された材料(それぞれ3μg)を、活性な4D4抗体および不活性なモノクローナル抗体(♯162)(8μg)と共に、0.1M Tris緩衝液、pH7.5中、4℃で30分間インキュベートした。結合/非結合のペプチドを、Microcon 10(Millipore Corp.、Bedford,Mass)によって分離し、抗体に結合したペプチドを探すために両方の画分(結合および非結合)をHPLCによって分析した。4D4抗体および♯162で処理し、Microcon分離した後、非結合画分のHPLC比較によって同定された2つの低下ピークを回収し、抗体結合ペプチドを示した。4D4結合ペプチドは:
S1(4.4)−−−−SRKAVRR(113〜119)(配列番号49)、C末端;および
S2(28.3)−−−−EVMVL(35〜39)(配列番号50)、ループ領域であった。

BSAを用いてさらに1時間ブロッキングした。次いで、プレートを、様々な濃度(0、3、10、30μg/mL)のヒト抗NGF抗体と共にインキュベートし、続いてヤギ抗hFc Ab−HRP(KPL)と共に2時間インキュベートした。シグナルをTMB基質で発色させて、停止溶液(KPL)の添加後に450nmで読み取った。
(KinExAによるモノクローナル抗体の親和性測定)
Ab 4D4(38859〜80)のhuNGF(29714〜91)に対する結合を、KinExAで試験した。簡単に述べると、Reacti−Gel 6×(Pierce)をhuNGFでプレコーティングして、BSAでブロッキングした。10pMおよび30pMのAb 4D4サンプルを、様々な濃度のhuNGF(Amgen)と共に室温で8時間インキュベートしてから、huNGFがコーティングされたビーズに通した。ビーズが結合した抗体の量を、蛍光(Cy5)標識ヤギ抗ヒトIgG抗体(Jackson Immuno Research)によって定量化した。結合シグナルは、平衡状態での遊離抗体の濃度に比例していた。解離平衡定数(KD)を、双対曲線一部位均一結合(dual−curve one−site homogeneous binding)モデル(KinExTMソフトウェア)を用いて競合曲線の非線形回帰から得た。KDは、Ab 4D4のhuNGFに対する結合について約4pMであった。
(さらなる抗NGF抗体の同定)
上記の実施例2および3に記載されたように作製され同定されたさらなる抗NGF抗体(14D10、6G9、7H2、14F11、および4G6と命名)をさらなる研究のために選択した。簡単に述べると、馴化培地を結合活性について試験した。培地から抗体を精製して配列決定した。予測質量を、馴化培地の抗体の質量分析データと比較した。抗体をクローン化した。クローンの内の2つが、CHO細胞で発現され、上記のように活性について試験した。この結果を表10に示す。
(ヒトにおける皮下NGF抗体の安全性および耐容性)
この試験の目的は、変形性関節症(OA)膝疼痛の被験体において、神経成長因子に対する抗体(NGF抗体)の複数回の皮下(SC)投与の安全性および耐容性を評価し、かつOA膝疼痛の被験体に対してNGF抗体の複数回SC投与を施した後、NGF抗体の血清薬物動態(PK)を調査することにあった。複数回SC投与の臨床上の利点を、Western Ontario and McMaster Universities Arthritis Index[WOMACTM3.1]によって評価した。
