本発明は、概して、結核菌(Mycobacterium tuberculosis)抗原の抗原および/または免疫原の組合せを含む組成物、ならびに、結核の診断、治療、および予防におけるそれらの使用に関する。

ID120は、Rv2389、Rv3810、Rv1813、Rv3620およびRv2608に由来する融合パートナーを含む融合構築物である。ID83-pET28.aベクターをKpnI/HindIIIで消化し、挿入断片を除去した。事前に作製しておいた36f.1-pET28.aベクターにID83挿入断片を連結した(また、KpnI/HindIIIで消化した)。融合構築物は、配列番号98に記載のポリヌクレオチド配列を有し、この配列は、配列番号99に記載の融合タンパク質をコードする。ID120を宿主BL-21plysSに発現させた(1L、2XYT増殖培地、37℃)。OD 0.50にて、1 mM IPTGで発現を誘導し、OD 1.41で細胞を回収した。細胞ペレットを溶解バッファー(20mM Tris pH8、100mM NaCl、2mM PMSF)中に懸濁し、凍結させた。ID120は封入体を形成し、ID83と同様に処理した。ID120は、一晩では十分に結合しなかったため、容量を8M尿素で2倍にし、BMEを10 mMまで添加した。室温で2時間、次に4℃で一晩、より低濃度の溶液をNi-NTA樹脂に結合させた。樹脂を洗浄し、前述のように溶出した。この精製物からの画分を20 mM Tris pH 8において透析した。

この実施例により、本発明のMtb抗原が、PPD+健康体ドナーからのヒトPBMC、および結核菌に感染したマウスから単離した脾細胞において記憶再生応答(memory recall response)を誘導することを実証する。
ヒトPBMCのin vitroでの刺激およびサイトカインELISA
PBMCは、アフェレーシスにより取得するか、7PPD-、および15PPD+健康体ドナー由来のヘパリン添加血液から精製した。PBMCを2〜2.5 x 105細胞/ウェルで96ウェル組織培養プレートに3重に塗布し、培地、PHA(10μg/ml)、結核菌(Mtb)溶解物(10μg/ml)、または各組換えタンパク質(50μg/ml)と一緒に72時間培養した。上清を回収し、特異的mAb(eBioscience)を製造者のプロトコルに従って用いた二重サンドイッチELISAによりIFN-γについて分析した。
結核菌に感染したマウスからの脾臓を、感染後の様々な時点で採取し、従来の方法により単一の脾細胞懸濁液を取得した。ELISPOTアッセイを用いて、IFN-γまたはTNF発現脾細胞の相対数を決定した。MultiScreen 96ウェルろ過プレート(Millopore、マサチューセッツ州ベッドフォード)に10μg/mlのラット抗マウスIFN-γ、またはTNF、捕獲Ab(eBioscience)をコーティングし、4℃で一晩インキュベートした。プレートをPBSで洗浄し、RPMI 1640および10%FBSを用いて室温で少なくとも1時間遮断してから、再度洗浄した。2 x 105細胞/ウェルで脾細胞を2重に塗布し、特異的rAg(10μg/ml)を用いて37℃で48時間刺激した。次いで、プレートをPBSおよび0.1%Tweenで洗浄し、PBS、0.5%BSA、および0.1% Tween中5μg/mlのビオチンコンジュゲート型ラット抗マウスIFN-γ、またはTNF、二次Ab(eBioscience)と一緒に、4℃で一晩インキュベートした。製造者のプロトコルに従って、Vectastain ABCアビジンペルオキシダーゼコンジュゲートおよびVectastain AEC基質キット(Vector Laboratories、カリフォルニア州バーリンゲーム)を用いて、フィルターを展開した。脱イオン水でプレートを洗浄することにより反応を停止し、プレートを暗所で乾燥させ、スポットを計数した。

結核菌H37Rv株での低用量エアロゾル暴露によりマウスを感染させ、感染後の様々な時点で脾臓を採取した。ELISPOTアッセイを用いて、48時間のin vitro培養の間の、Mtb組換えRv0164、Rv0455、Rv0496、Rv2608、Rv3020、Rv3478、Rv3619、Rv3620、Rv1738、Rv1813、Rv3810、Rv2389、Rv2866、Rv0054、Rv0655、Rv0831、Rv1009、Rv1240、Rv1288、Rv1410、Rv1569、Rv1789、Rv1818、Rv1860、Rv1886、Rv1908、Rv2220、Rv2032、Rv2875、Rv3044、Rv3310、Rv3881、Rv0577、Rv1626、Rv0733、Rv1253、Rv1980、Rv3628、Rv1884、Rv3875、Rv1511およびID83タンパク質に応答する、TNF発現脾細胞の相対数を決定した。
C57BL/6マウスにおけるMtb抗原に対する免疫応答、ならびにMtbのエアロゾルチャレンジに対する防御
この実施例では、本発明のMtb抗原によるマウスの免疫化が、免疫原性であり、エアロゾル結核菌チャレンジに対する防御を提供できることを実証する。
組換え抗原およびアジュバント製剤
上記のように組換えタンパク質を生産した。CpG 1826は、Coley Pharmaceuticals(マサチューセッツ州ウェルズリー)から取得した。
雌のC57BL/6マウスをCharles Riverから取得し、各実験において生齢を合わせた(5〜7週)。25μgのアジュバントCpGと一緒に製剤化した8μgの以下:組換えRv0164(配列番号1)、Rv0496(配列番号6)、Rv2608(配列番号26)、Rv3020(配列番号36)、Rv3478(配列番号41)、Rv3619(配列番号46)、Rv3620(配列番号51)、Rv1738(配列番号11)、Rv1813(配列番号16)、Rv3810(配列番号56)、Rv2389(配列番号21)、Rv2866(配列番号31)、Rv0831(配列番号115)、Rv1288(配列番号127)、Rv1569(配列番号133)、Rv1789(配列番号136)、Rv1818(配列番号139)、Rv1860(配列番号142)、Rv1886(配列番号145)、Rv2220(配列番号154)、Rv2032(配列番号151)、Rv2623(配列番号160)、Rv2875(配列番号163)、Rv3044(配列番号166)、Rv0577(配列番号184)、Rv1626(配列番号187)、Rv0733(配列番号190)、Rv3628(配列番号202)、およびRv1884(配列番号205)タンパク質で3回(3週間置きに)免疫した。生理食塩水、アジュバントのみ、およびBCG対照群のマウスには、それぞれ、3用量のPBS、3用量のアジュバントのみ、または1用量の5 x 104 BCG CFUを受けた。1匹当たり総用量100μlをマウスに皮下注射した。

組換え抗原およびアジュバント製剤
前記のように組換えタンパク質を作製した。CpG1826はColey Pharmaceuticals(マサチューセッツ州ウェルズリー)から入手した。

この実施例は、本発明のID83融合タンパク質を発現すべく操作されたアデノウイルスベクターによるマウスの免疫が、C57BL/6マウスにおいて免疫原性であることを実証するものである。
ウイルス構築および精製
AdEasy(商標)XL AdenoviralVector System(Stratagene #240010)を用いて、Ad5-ID83を構築した。手短には、PCRを用いて、プラスミドDNAからID83を増幅し、HinDIIIおよびEcoRVで消化し、pShuttle-CMVに連結して、ID83-pShuttleCMVを作製した。PmeIで消化することにより、ID83-pShuttleCMVを線状にし、大腸菌BJ5183-AD-1エレクトロコンピテントセル(Stratagene #200157)にエレクトロポレートした(2.4kV、186Ω、0.2cm ギャップキュベット)。PacIで消化することにより、組換えAd5-ID83プラスミドを同定した。Polyfect試薬(Invitrogen #301107)を用いて、60mmプレート中で、AD-239細胞にPacIで消化したAd5-ID83プラスミド(4μg)をトランスフェクトした。4日後、3mL媒地中に細胞を回収し、3サイクルの凍結/解凍により細胞を溶解させた。溶解物上清を、CsCl勾配遠心分離による精製用のウイルスを増幅するために用いた。

この実施例では、標準的抗生物質療法と併用した本発明の組換えタンパク質の混合物によるマウスの免疫化により、結核菌感染マウスの寿命を延長できることを実証する。
組換えタンパク質およびアジュバント製剤

結核菌によるエアロゾルチャレンジから2週間後、標準的抗生物質療法を開始した。飲用水に50 mg/lのリファンピンと85 mg/lのイソニアジドを添加して、これらをマウスに60日間与えた。いくつかのマウスにはさらに免疫療法を施し、感染から76日、97日、および118日後、20μgのアジュバントGLA-SEと一緒に製剤化したRv1813、Rv2608、およびRv3620組換えタンパク質各々6μgでこれらのマウスを免疫した。1匹当たり総量100μlをマウスに皮下経路で注射した。マウスの生存を225日間モニターした。

からなる群より選択される、請求項1に記載の組成物。 - Rv2608(配列番号26)、Rv1813(配列番号16)およびRv3620(配列番号51)を含む前記組合せが、以下:Rv1886(配列番号145)、Rv2389(配列番号21)、Rv3478(配列番号41)、Rv2875(配列番号163)、Rv2220(配列番号154)、Rv0733(配列番号190)、Rv0577(配列番号184)、Rv3044(配列番号166)、Rv1626(配列番号187)、Rv3619(配列番号46)およびRv3020(配列番号36)からなる群より選択される1種以上の抗原をさらに含む、請求項2(a)に記載の組成物。

- Rv3478(配列番号41)およびRv3619(配列番号46)を含む前記組合せが、以下:Rv1886(配列番号145)、Rv2389(配列番号21)、Rv1813(配列番号16)、Rv2875(配列番号163)、Rv2220(配列番号154)、Rv0733(配列番号190)、Rv0577(配列番号184)、Rv3044(配列番号166)、Rv1626(配列番号187)、Rv3620(配列番号51)、Rv2608(配列番号26)およびRv3020(配列番号36)からなる群より選択される1種以上の抗原をさらに含む、請求項2(c)に記載の組成物。
- 2種以上の抗原、またはその免疫原性断片は、融合ポリペプチドの形態で共有結合している、請求項1に記載の組成物。
- 前記融合ポリペプチドが、以下:ID83(配列番号91)、ID94(配列番号95)、ID93(配列番号226)、ID91(配列番号236)、ID71(配列番号245)、ID114(配列番号251)、ID125(配列番号257)、またはこれらと少なくとも90%の同一性を有する配列、からなる群より選択されるアミノ酸配列を含む、請求項8に記載の組成物。
- 前記免疫刺激剤が、以下:GLA、AS-2、ENHANZYN(商標)、MPL(商標)、3D-MPL(商標)、IFA、QS21、CWS、TDM、AGP、CpG含有オリゴヌクレオチド、トール様受容体アゴニスト、LeIF、サポニン、サポニン模擬物、ならびに生体および合成のリピドA、イミキモド、ガルジキモド(gardiquimod)、レシキモド、polyI:C、フラジェリン、またはこれらの組合せからなる群より選択される、請求項1に記載の組成物。

- 請求項11〜17のいずれか一項に記載の融合ポリペプチドをコードする、単離されたポリヌクレオチド。
- 請求項1〜10のいずれか一項に記載の有効量の組成物を、それを必要とする被験体に投与することを含む、防御免疫応答を刺激する方法。

(a)生体サンプルを以下:DID85(配列番号265)、DID92(配列番号273)、DID108(配列番号283)、およびDID93(配列番号291)からなる群より選択される融合ポリペプチドと接触させるステップと;
(b)生体サンプルにおいて、前記融合ポリペプチドに結合する抗体および/またはT細胞の存在を検出するステップ
を含む、上記方法。 - 生体サンプルにおける結核菌(Mycobacterium tuberculosis)感染を検出するための診断キットであって、
(a)以下:DID85(配列番号265)、DID92(配列番号273)、DID108(配列番号283)、およびDID93(配列番号291)からなる群より選択される融合ポリペプチドと;
(b)検出試薬
とを含む、上記診断キット。
