Abstract

Description
本願は、仮特許出願第60/605,097、60/604,941および60/605,074号(それぞれ、2004年8月27日に出願し、これらは出典明示により本明細書に組み入れる)の優先権を主張する。
タンパク質およびポリペプチドは、治療剤としてますます重要になっている。大抵の場合、治療タンパク質およびポリペプチドは、目的の特定のタンパク質またはポリペプチドを非常に高レベルで産生するように設計および/または選択された細胞から、細胞培養にて生産される。細胞培養条件の制御および最適化は、タンパク質およびポリペプチドの商業生産の成功にとって非常に重要である。
本発明は、細胞培養におけるタンパク質および/またはポリペプチドの大規模生産のための改善されたシステムを提供する。例えば、本発明は、1つまたはそれ以上の:i)約70mMを超える単位体積あたりの累積アミノ酸量;ii)約2未満の累積アスパラギンに対するモル累積グルタミン比;iii)約0.2未満の総累積アミノ酸に対するモル累積グルタミン比;iv)約0.4〜1の総累積アミノ酸に対するモル累積無機イオン比;またはv)約16mMを超える単位体積あたりのグルタミンおよびアスパラギン濃度を合わせた累積量により特徴付けられる培地を利用する、商業規模の(例えば、500L以上)培養法を提供する。当業者は、上で用いられる「累積」が、培養開始時に加えられた成分およびその後加えられた成分を含む、細胞培養の過程において加えられた特定の成分(複数でも可)の総量を意味することは理解できるだろう。本発明のある種の好ましい具体例において、培養物の「フィード」を最小限にすることが望ましく、したがって、初期に存在する量を最大限にすることが望ましい。もちろん、培地成分は培養中に代謝され、同じ累積量の所定の成分の培養物は、それらの成分が異なる時間に加えられた場合には(例えば、すべて初期に存在することに対して、いくつかはフィードにより加える)、異なる絶対値を有するだろう。

本発明は、細胞培養によるタンパク質および/またはポリペプチドの製造に関する改善されたシステムを提供する。特に、本発明は、細胞成長、生存率および/またはタンパク質産生または質に有害である1つまたはそれ以上の代謝産物の産生を最小限にするシステムを提供する。本発明の好ましい具体例において、細胞培養は、バッチまたはフェドバッチ培養である。本発明の他のある種の好ましい具体例は、以下に詳細に記載する。しかしながら、当業者は、これらの好ましい具体例の種々の改良されたものが本発明の範囲に含まれることは理解できるだろう。本発明の請求の範囲は、具体例の記載に限定されるものではない。

本発明に従って望ましく発現されうるポリペプチドは、目的の生物学的または化学的活性に基づいて選択されるだろう。例えば、本発明は、医薬的または商業的関連がある酵素、受容体、抗体、ホルモン、調節因子、抗原、結合剤等を発現するのに用いることができる。
現在、医薬品または他の市販の薬剤、抗体の製品として使用され、研究されている多くの所定の抗体が、本発明における特定の目的の抗体である。抗体は、特定の抗原に特異的に結合する能力を有するタンパク質である。ホスト細胞において発現することができるいずれもの抗体を、本発明に従って用いることができる。好ましい具体例において、発現される抗体は、モノクローナル抗体である。
医薬および/または市販の薬剤として有効であることが示されているポリペプチドの他の群は、受容体を含む。受容体は、典型的には、細胞外シグナリングリガンドを認識することにより機能する膜貫通糖タンパク質である。典型的には、受容体は、リガンド認識ドメインに加え、細胞内での発育変動または代謝性変化を誘導する、結合リガンドでの標的細胞内分子のリン酸化によりシグナリング経路を開始する、タンパク質キナーゼドメインを有する。一の具体例において、目的の受容体は、膜貫通および/または細胞内ドメイン(複数でも可)を除去し、その代わりにIg−ドメインに結合できるように修飾されている。好ましい具体例において、本発明により製造される受容体は、受容体チロシンキナーゼ(RTK)である。RTKファミリーは、種々の機能の多数の細胞型に欠かせない受容体を含む(例えば、Yarden and Ullrich, Ann. Rev. Biochem. 57:433-478, 1988;Ullrich and Schlessinger, Cell 61:243-254, 1990(出典明示により本明細書に組み入れる)を参照)。RTKsの非限定的な例としては、繊維芽細胞成長因子(FGF)受容体ファミリーの一員、表皮成長因子受容体(EGF)ファミリーの一員、血小板由来成長因子(PDGF)受容体、免疫グロブリンを有するチロシンキナーゼおよびEGF相同ドメイン−1(TIE−1)およびTIE−2受容体(Sato et al., Nature 376(6535):70-74 (1995)(出典明示により本明細書に組み入れる)を参照)およびc−Met受容体が挙げられ、これらのいくつかは、直接的または間接的に血管新生を促進することが示唆されている(Mustonen and Alitalo, J. 医薬および/または市販の薬剤として有効であることが示されているポリペプチドの他の群は、成長因子および他のシグナリング分子を含む。成長因子は、典型的には、細胞により分泌され、他の細胞に結合し、受容体を活性化し、受容体細胞の代謝性変化または発育変動を生じさせる、糖タンパク質である。一の具体例において、目的のタンパク質は、ActRIIB受容体の細胞外ドメインおよび抗体のFc部を含有する、ActRIIB融合ポリペプチドである(例えば、Wolfman, et al., ActRIIB fusion polypeptides and uses therefor、US2004/0223966A1を参照)。他の具体例において、成長因子は、修飾GDF−8プロペプチドであってもよい(例えば、Wolfman, et al., Modifed and stabilized GDF propeptides and uses thereof、US2003/0104406A1を参照)。別として、目的のタンパク質は、ホリスタチン−ドメイン含有タンパク質であり得る(例えば、Hill, et al., GASP1: a follistatin domain containing protein, US 2003/0162714 A1, Hill, et al., GASP1: a follistatin domain containing protein, US 2005/0106154 A1, Hill, et al., Follistatin domain containing protein、US2003/0180306A1を参照)。
医薬および/または市販の薬剤として有効であることが示されているポリペプチドの他の群は、成長因子および他のシグナリング分子を含む。G−タンパク質共役受容体(GPCRs)は、7回膜貫通ドメインを有するタンパク質である。リガンドのGPCRへの結合において、シグナルは、細胞の生物学的または生理学的特性の変化が生じる細胞内で伝達される。
当業者には明らかであるように、遺伝子制御因子は、ポリペプチドまたはタンパク質の遺伝子発現をするために用いることができる。かかる遺伝子制御因子は、関連するホスト細胞で活性化するように選択されるべきである。制御因子は、構造的に活性であってもよく、または所定の条件下で誘導性であってもよい。誘導制御因子は、発現したタンパク質が毒性であるか、または細胞成長および/または生存率に悪影響を及ぼすものである場合に、特に有用である。このような場合、誘導制御因子が、細胞生存率、細胞密度および/または発現ポリペプチドまたはタンパク質の総収率を改善することにより、ポリペプチドまたはタンパク質の発現を調節する。本発明の実施に有用な多くの制御因子は、当該分野で公知であり、入手可能である。
目的のポリペプチドまたはタンパク質の発現を行うの十分な哺乳動物ホスト細胞核酸の導入に適した方法は、当該分野でよく知られている。例えば、Gething et al., Nature, 293:620-625 (1981); Mantei et al., Nature, 281:40-46 (1979); Levinson et al.;EP117,060;およびEP117,058(出典明示により本明細書に組み入れる)を参照のこと。
細胞培養およびポリペプチドの発現に感受性であるいずれの哺乳動物細胞または細胞型を、本発明の方法に従って利用することができる。本発明の方法により用いることができる哺乳動物細胞の非限定的な例としては、BALB/cマウス骨髄腫細胞株(NSO/l、ECACCNo:85110503);ヒト網膜芽細胞(PER.C6(CruCell, Leiden, The Netherlands));SV40によりトランスフェクトされたサル腎臓CV1株(COS−7、ATCCCRL1651);ヒト胚腎臓株(懸濁培養物の成長に関する、サブクローン化した293または293細胞、Graham et al., J. Acad. Sci., 383:44-68 (1982));MRC5細胞;FS4細胞;およびヒト肝臓癌株(HepG2)が挙げられる。特に好ましい具体例において、本発明は、CHO細胞株からのポリペプチドおよびタンパク質の培養および発現に用いることができる。
目的のポリペプチドを製造する典型的な方法は、バッチおよびフェドバッチ培養の両方を含む。バッチ培養法は、慣用的には、大規模生産培養物を特定の細胞密度の種培養で接種し、細胞成長および生存を促す条件下で細胞を培養し、細胞が特定の細胞密度に達した場合に回収し、発現したポリペプチドを精製することを含む。フェドバッチ培養法は、細胞の成長の間に消費される栄養分および他の成分をバッチ培養物に供給する付加的な工程を含む。慣用的なバッチおよびフェドバッチ培養に付随する難解な問題は、代謝廃棄物の産生であり、これは、細胞成長、生存率および発現ポリペプチドの産生に悪影響を及ぼす。特に悪影響を及ぼす2つの代謝廃棄物は、乳酸塩およびアンモニウムであり、これらは、それぞれ、グルコースおよびグルタミン代謝の結果として産生される。グルタミン代謝の結果としてのアンモニウムの酵素産生に加えて、また、アンモニウムは、時間とともに非代謝性分解の結果としても細胞培養物中に蓄積される。本発明は、細胞培養物におけるこれらの廃棄物の蓄積を遅らせ、逆進さえさせることにより、アンモニウムおよび乳酸塩の悪影響を最小限にした、ポリペプチドの改善された大規模生産法を提供する。限定するものではないが、バッチ、フェドバッチおよび潅流システムを含む、細胞を培養するいずれのシステムにおいても用いることができることは、当業者には明らかだろう。本発明のある種の好ましい具体例において、細胞を、バッチまたはフェドバッチシステムにおいて成長させる。
市販されている培地、例えばハムF10(Sigma)、Minimal Essential培地([MEM]、Sigma)、RPMI−1640(Sigma)およびダルベッコ修飾イーグル培地([DMEM]、Sigma)を含む、慣用的な培地組成は、他のアミノ酸と比較して、相対的に高レベルのグルコースおよびグルタミンを含有していた。これらの成分は、細胞の一次代謝エネルギー源であるので、大量に必要とされることが考えられる。しかしながら、これらの栄養分の急激な消費により、上記したような乳酸塩およびアンモニウムの蓄積が誘発される。加えて、高い初期濃度のグルコースおよびグルタミンならびにその後の乳酸塩およびアンモニウムの蓄積の結果として、高オスモル濃度となり、この環境が、細胞成長、細胞生存率およびポリペプチドの産生に悪影響を与える。
バッチおよびフェドバッチ培養によるタンパク質またはポリペプチドの生産用の哺乳動物細胞の種々の製造方法は、当該分野でよく知られている。上記したように、発現するのに十分な核酸(典型的には、目的のポリペプチドまたはタンパク質をコードする遺伝子およびいずれもの作用的に結合した遺伝子制御因子を含有するベクター)を、ホスト細胞株に多くのよく知られた技術のいずれかにより導入することができる。典型的には、細胞は、ベクターを取り込み、目的のポリペプチドまたはタンパク質を発現するホスト細胞を決定するために、細胞をスクリーニングする。哺乳動物細胞により発現された特定の目的のポリペプチドまたはタンパク質を検出する慣用的な方法は、限定するものではないが、免疫組織化学、免疫沈降、フローサイトメトリー、免疫蛍光顕微鏡検査法、SDS−PAGE、ウエスタンブロット、酵素免疫測定吸着法(ELISA)、高速液体クロマトグラフィー(HPLC)法、生物活性アッセイおよびアフィニティクロマトグラフィーを含む。発現ポリペプチドまたはタンパク質を検出する他の適当な方法は、当業者には明らかだろう。複数のホスト細胞が目的のポリペプチドまたはタンパク質を発現する場合、記載した方法のいくつかまたはすべてを、ポリペプチドまたはタンパク質を最も高レベルに発現する細胞を決定するのに用いることができる。
一旦、生産バイオリアクターに上記のように播種すると、細胞培養物を、細胞培養物の成長および生存に適した条件下で、初期成長フェーズにおいて保持する。正確な条件は、細胞型、細胞をデリバリーする生物および発現ポリペプチドまたはタンパク質の性質および特性に応じて変化するだろう。
本発明の教示に従って、初期成長フェーズの終わりに、第2の培養条件を適用し、培養物に代謝シフトが生じるように、少なくとも1つの培養条件をシフトしてもよい。阻害代謝物、最も顕著には乳酸塩およびアンモニアの蓄積は、成長を阻害する。細胞培養物の温度、pH、重量オスモル濃度または化学誘導剤レベルを変更することにより為される代謝シフトは、特定のグルコース消費速度に対する特定の乳酸塩産生速度の割合を減少させることにより特徴付けられる。一の非限定的具体例において、培養温度をシフトさせることにより培養条件をシフトする。しかしながら、当該分野で知られているように、温度のシフトは、適当な代謝シフトを達成することができるただ一つのメカニズムというわけではない。例えば、かかる代謝シフトは、限定するものではないが、pH、重量オスモル濃度および酪酸ナトリウムレベルを含む他の培養条件をシフトすることによっても達成することができる。上記したように、培養シフトのタイミングは、本発明の実施者により、ポリペプチドまたはタンパク質生産要求量または細胞自体の要求に基づいて決定されるだろう。
本発明に従って、一旦、細胞培養条件を上記したようにシフトさせると、細胞培養物を、細胞培養物の生存および商業的に十分なレベルでの所望のポリペプチドまたはタンパク質の発現に適した、第2の培養条件下で、次生産フェーズの間保持する。
本発明のある種の具体例において、実施者は、成長細胞培養物の特定の条件を定期的にモニターすることが有益または必要であると感じるかもしれない。細胞培養条件をモニタリングすることにより、実施者は、細胞培養物が、組み換えポリペプチドまたはタンパク質を最適なレベルで産生しているがどうか、あるいは、培養物が、最適な産生フェーズに入っているかどうかを判断することができる。ある細胞培養条件をモニターするために、分析用の少量の培養物アリコートを取り出す必要があるだろう。当業者は、かかる採取により、細胞培養が汚染される可能性があることは理解でき、かかる汚染のリスクを最小限にするために適切に注意すべきだろう。
一般的に、典型的には、本発明により発現したタンパク質またはポリペプチドを単離および/または精製することが望ましいだろう。好ましい具体例において、発現ポリペプチドまたはタンパク質は培地中に産生され、かくして、細胞および他の固体を、例えば精製工程の最初に、遠心分離または濾過除去により除去してもよい。この具体例は、特に、本明細書に記載の方法および組成物は高い細胞生存率を与えるので、本発明において用いる場合に有用である。結果として、培養プロセスの間に死亡する細胞がより少なく、発現ポリペプチドまたはタンパク質の収率を減少させる可能性がある培地中へのタンパク質分解酵素の放出がより少なくなる。
本発明のある種の好ましい具体例において、産生ポリペプチドまたはタンパク質は、薬理活性を有し、医薬の製造において有用であるだろう。上記した本願の組成物は、対象に投与されるか、あるいは、最初に、限定するものではないが、非経口(例えば、静脈内)、皮内、皮下、経口、経鼻、気管支、眼、経皮(局所)、経粘膜、直腸および膣経路を含む可能な経路によるデリバリー用に処方することができる。本発明の医薬組成物は、典型的には、哺乳動物細胞株から発現した精製ポリペプチドまたはタンパク質、デリバリー剤(すなわち、上記したカチオン性ポリマー、ペプチド分子トランスポーター、界面活性剤等)を医薬上許容される担体と組み合わせて含みうる。本明細書で用いられる場合、「医薬上許容される担体」なる用語は、医薬投与に適当しうる、溶媒、分散媒、コーティング剤、抗菌剤および抗真菌剤、等張および吸収遅延剤等を含む。また、追加の活性化合物を、組成物中に組み入れることもできる。
実施例1:抗GDF−8フェドパッチ法に関する、強化培地1
細胞株を培養する慣用的なフェドバッチ法は、供給物を与えるのに必要な時間と労力および大規模バイオリアクターにおいて特別な装置を必要とすることを含む、いくつかの欠点を有する。本願の目的は、最低限の供給で、大規模バイオリアクターにおける目的のタンパク質の生産用のバッチ培地を構築することである。
株および培地:チャイニーズ・ハムスター・卵巣(「CHO」)細胞を、成長および分化因子8(「抗GDF−8細胞」)に対するモノクローナル抗体を発現するために設計した(Veldman et al., Neutralizing Antibodies Against GDF-8 and Uses Therefor、US20040142382A1を参照)。抗GDF−8細胞を、新規バッチ培地の試験に用いた。培地1および培地2を、高細胞密度および生存率を裏付けるために、その能力を比較した。これらの培地ならびに培地3の組成は、表1に記載する。培地は、FeSO4・7H2Oを除くすべての成分を添加することにより作成した。ついで、培地を、pH7.25に調節し、オスモル濃度を記録し、ついで、FeSO4・7H2Oを加えた。
図1は、フラスコ実験における、抗GDF−8細胞の成長速度は、培地1および培地2で同じであったことを示している。
図2は、バイオリアクターにおいて、培地1が、培地3よりも、有意に高い最終細胞密度および生存率を示すことを示す。また、最終力価は、基本的な方法の551mg/Lから培地1で976mg/Lまで有意に増加した(データは示していない)。温度を、5日目に37℃から31℃にシフトした。予期せぬ高い細胞成長のため、培養物を、2体積%の20X培地4または3体積%の16X培地4のいずれかを、5日目以降、毎日供給した。かくして、これは、当初予定したバッチ実験とは正確には異なる。フィード培地へのアスパラギンおよびチアミンの供給を10日目から開始した。
実施例1において、培地1に用いる抗GDF−8細胞を培養するバッチ法を構築した。そのプロセスにおいて得られた高細胞密度のため、グルコースおよびグルタミンに加えて栄養物を供給することが有益であることが分かった。しかしながら、8X培地4フィード培地を有するバッチを補足することにより、培養物が過剰に希釈される。より濃縮したフィード培地を、この問題を解決するために構築した。
表2は、表3〜4の処方に用いられる、培地4A−1、培地4B、トレースBおよびトレースDの組成を示す。
第1の濃縮培地を、20X培地4として構築した。20X培地4の培地の構成を表3に示す。
フィード培地の最終pHを7.0に調節し、オスモル濃度は約1064mOsmであった。2%フィードにより、培養物が2g/Lのグルコース、2mMのグルタミンおよび14mOsmのオスモル濃度、増加する。
オスモル濃度の増加を軽減するために、フィード培地を、20X培地4(毎日2体積%)から16X培地4(毎日3体積%)に変更した。16X培地4の培地構成を、表4に示す。
16X培地4の構成を変更した。鉄ストック溶液をフィードに加え、結果として、個々のフィードに0.45μMを添加した。さらに、グルコースを戻し、フィード毎に1.5g/Lを加えた。この修飾16X培地4の培地構成を表5に示す。
ここで、フィード培地(16X培地4)を、いくつかのバッチと同様に、3つの別個のフィードの代わりに複合培地で作成した。葉酸を必要な濃度で溶解できることを確実にし、鉄または葉酸がいずれも、4℃または室温のいずれかで6日間貯蔵した後も溶液から沈殿しないことを確実にするために試験を行った。複合16X培地4の培地構成を表6に示す。
ここで、フィード培地をいくらか変更する。培地4B粉末を、培地4Bにおける個々の成分を添加する代わりに用いた。培地4B粉末を、グルコースと混ぜ合わせ、溶液をpH10.25まで滴定することにより、塩基性条件下で別々に溶解した。アミノ酸およびビタミン分析結果が、フェドバッチプロセスの終わりまでに、これらの二つの成分が排出されることを示したので、付加的なアスパラギンおよびチアミンを加えた。12Xの培地4の使用は、さらに、培養物に供給する場合、オスモル濃度の増加を軽減した。12X培地4の培地構成を表7に示す。
CHO細胞は、生存するために初期の培地にグルタミンを必要とする。慣用的には、初期グルタミンレベルは高く、グルタミンは、5日目以降フェドパッチ工程の最後まで毎日供給される。慣用的なフェドバッチプロセスは、通常、細胞培養において、高い乳酸濃度およびアンモニウムレベルを生じ、このことが、細胞成長、細胞密度および組み換えタンパク質発現の効果を阻害することが知られている。グルコースを培養物にゆっくりと添加するフェドバッチ工程は、乳酸産物を低下させ、細胞成長、細胞密度および組み換えタンパク質発現を改善することが示された。しかしながら、グルコース添加の操作に関する従来の方法は、大規模製造で実用化されていない。そこで、低い初期グルタミンレベルおよびフィードからグルタミンを除いた培養培地を利用することにより、アンモニウムおよび乳酸の産生が低レベルであり、細胞生存率を増加させることが示された。加えて、グルタミン不足培地において、組み換えタンパク質発現は増加し、最終オスモル濃度は減少した。
株および培地:抗GDF−8細胞を、1Lバイオリアクター中、培地1においてフェドバッチモードで培養した。
培養条件:細胞を、1Lバイオリアクターにおいて12日間成長させた。温度を、細胞成長に応じて、4日目または5日目に37℃から31℃にシフトした。3つのフェドバッチプロセス:通常(対照)プロセス、非グルタミンフィードプロセスおよびグルタミン不足プロセスを試験した。これらのプロセスの関連細目を、表8および表9に示す。
図3は、非グルタミンフィードまたは対照フェドバッチ条件において成長した培養物の細胞密度を示す。両方の場合において、細胞密度は、実験期間中同様であった。
図4は、非グルタミンフィードまたは対照フェドバッチ条件において成長した培養物の細胞生存率を示す。非グルタミンフィード培養物は、6日目から実験の終わりにかけて非常に高い細胞生存率を示した。
図5は、非グルタミンフィードまたは対照フェドバッチ条件において成長した培養物中のアンモニウムレベルを示す。非グルタミンフィード培地は、4日目から実験の終わりにかけてアンモニウムレベルの顕著な減少を示した。
図6は、非グルタミンフィードまたは対照フェドバッチ条件において成長した培養物中の乳酸レベルを示す。乳酸レベルは、実験の期間中、非グルタミンフィード培養物においてわずかに低下した。
図7は、非グルタミンフィードまたは対照フェドバッチ条件において成長した培養物における抗GDF−8抗体力価を示す。最終抗GDF−8抗体力価は、非グルタミンフィード培養物においてより高かった。
図8は、グルタミン不足または対照フェドバッチ条件において成長した培養物の細胞密度を示す。両方の場合において、細胞密度は、実験期間中同様であった。
図9は、グルタミン不足または対照フェドバッチ条件において成長した培養物における細胞生存率を示す。両方の場合において、細胞生存率は、実験期間中同様であった。
図10は、グルタミン不足または対照フェドバッチ条件において成長した培養物におけるアンモニウムレベルを示す。グルタミン不足培養物は、実験期間中、アンモニウムレベルの顕著な減少をを示した。
図11は、グルタミン不足または対照フェドバッチ条件において成長した培養物における乳酸レベルを示す。グルタミン不足培養物は、4日目から実験の終わりにかけて、乳酸レベルの顕著な減少をを示した。
図12は、グルタミン不足または対照フェドバッチ条件における抗GDF−8抗体力価を示す。最終抗GDF−8抗体力価は、グルタミン不足培養物においてより高かった。
培地1を、培地2よりも栄養物でさらに濃縮した。培地1における細胞成長最適な鉄レベルを、細胞培養期間の鉄欠乏に関する問題を回避するために測定した。
抗GDF−8細胞を、培地1または培地2においてディッシュ中で、1継代培養した。これらの培地の鉄濃度を、異なる量の貯蔵鉄溶液を添加することにより操作した。最終細胞密度を、CEDEXにより測定した。
図13は、異なる鉄濃度を有する培地1および培地2における抗GDF−8細胞のFe投与応答を示す。培地2において、細胞密度は、3μM〜15μMの範囲の鉄濃度に対して相対的に一定であった。培地1において、細胞密度は、鉄濃度の増加と共に増加したが、約5μMで最大に達した。この違いは、培地1は高含有量で栄養分を含み、培地における鉄のキレート化の結果として、細胞が利用できる鉄が減少したためと考えられる。これらの結果は、培地1にける鉄欠乏に関する問題を回避するために、5μM以上を維持すべきであることを示している。
抗ルイスY細胞培養プロセスにおいて、グルタミンをグルタミン酸に置換することによる効果を、3つの実験で試験した。
実験を、10Lバイオリアクター中、pH7.1、30%酸素溶解および開始温度37℃、5日目に31℃にシフトの条件で行った。散布ガスおよびヘッドスペースガスは、88%の93%空気/7%CO2混合物および12%酸素であった。すべての実験において、出発培地は、グルタミンを含有する培地1であった。補足グルコースおよびグルタミンフィードを含む、フィード培地およびフィードスケジュールを、表10に示す。「グルタミン酸塩」のカラムは、グルタミンを含有しないが、標準培地5におけるモルグルタミン濃度と等しいモル濃度のグルタミン酸塩を含有する、修飾培地5で与える。
各々の実験において、細胞密度は、図14に示すように同様であった。グルタミン2およびグルタミン酸塩2の実験において、pHが3日後に約6.7になったために細胞密度が低い。グルタミン3およびグルタミン酸塩3の実験における、6および7日目の間の密度の低下は、6日目での29%培地フィードによるものである。
この実験の目的は抗ルイスY細胞の培養において、グルコースおよびグルタミンを下記表11に示すフィード培地で置き換えることによる効果を試験することであった(Bogheart et al., Antibody-targeted chemotherapy with the calicheamicin conjugate hu3S193-N-acetyl gamma calicheamicin dimethyl hydrazide targets Lewisy and eliminates Lewisy-positive human carcinoma cells and xenografts, Clin. Can. Res. 10:4538-49 (2004)を参照)。細胞密度、細胞生存率、抗ルイスY力価およびアンモニウムレベルを測定した。
実験を、250mlの振盪フラスコ開始容量75mlで行った。すべての振盪フラスコに、培地2で0.25×106細胞/mlで播種した。フラスコを、インキュベーターにおいて、37℃、7%のCO2で、14日間インキュベートした。3および4日目に、フラスコに、5体積%の培地6フィード培地をフィードした。培地6の組成を表11に示す。5〜13日目に、フラスコに、表12に示すフィード溶液の1つをフィードした。各々の条件で、2回行った。試料を毎日採取し、細胞をCEDEXにより計数し、アンモニウム、グルコースおよび乳酸塩に関してアッセイした。
フィード培地中グルコースまたはガラクトースの存在下、グルタミン酸塩またはグリシルグルタミンをグルタミンの代わりに用いた場合に、最も高い細胞密度が観察された。細胞密度は、グルコース/グルタミン、ガラクトース/グルタミンまたはグルコース単独をフィードする培養物において、一般的に低い(図20)。最終生存率は、グルコース単独をフィードする培養物で最も高く、ついで、グルコース/グルタミン酸塩をフィードする培養物で高かった。グルコースまたはガラクトースと組み合わせたグルタミンまたはアスパラギンをフィードする培養物において最も低い生存率が観察された(図21)。
典型的なフェドバッチ生成法は、培養期間の間複数回のフィードを必要とする。これらのフィードは、細胞により消費されるか、あるいはバッチ期間中分解されうる培地中の栄養物を元に戻すように設計される。これらのフィードにより、大きなリアクターにスケールアップした場合に、インペラー・ジャンプ(impeller jump)(図24を参照)の必要性のような複雑な問題が生じる。さらに、フィードは、すでに培養物に分泌された抗GDF−8の量を希釈し、したがって、採集力価に影響を与える。バッチプロセスの使用により、フィードに適合するように部分容量で植菌する代わりに最大容量でバイオリアクターの植菌をすることが可能になり、インペラー・ジャンプの必要性が取り除かれ、生産性への希釈影響を大きく減少させるだろう。
抗GDF−8細胞を、以下の4つの成長条件に従って、12日間、lLのバイオリアクター中で成長させた。すべての条件に関するバイオリアクターパラメータを、同一に維持した。空気を散布することにより、溶解した酸素を維持し、空気飽和度の23%より高く維持し、pHを、0.58Mの二酸化炭素および0.71Mの炭酸ナトリウムを含有する溶液を添加することにより、7.00で維持した。すべての培養物の温度を、バッチの最初の4日間37度に維持した。バッチの4日目に、すべてのバイオリアクターの温度を31度に下げ、バッチ期間中この温度を維持した。対照およびフェドバッチ培養物に、8%、12%および8%の総リアクター容量のフィード培地を、それぞれ、5、7および10日目にフィードした。

最初の4日間の細胞成長は、対照とバッチプロセスにおいて同様であるが、グルタミン欠乏フェドバッチプロセスは、細胞密度が有意に低く、バッチ後には少し低めであった。両方のバッチプロセスは、恐らく、有意な希釈が無かったために、バッチ期間中、高い細胞密度を維持した(図25を参照)。すべての培養物の生存率は、8日目まで同じであった。しかしながら、11日目に、補給がないバッチプロセスの生存率は、他の3つのバイオリアクターよりも低く、最終日までに有意に減少することに注目することは興味深い。このことは、補給バッチプロセスがフェドバッチバイオリアクターと同様の生存率を示しているので、バッチ培地はさらに最適化されうることを示唆している(図26を参照)。
実施例2、5および6において、グルタミン欠乏が、2つの細胞において、フェドバッチ培養に、細胞成長、細胞生存率および力価の増加ならびに乳酸塩およびアンモニウムの産生の減少を含む有益性をもたらすことが実証された。また、アスパラギンもバッチ培地において同様の役割を果たすと考えられる。
抗GDF−8細胞を、異なる濃度のグルタミンおよびアスパラギンを有する修飾培地9において、1Lのバイオリアクター中で12日間培養した。基本培地9の組成を表14に示す。この基本組成の実験バリエーションを、表15に示す。培養物を、温度コントロールの問題のために最初の日に30℃であってリアクター4を除き、最初の5日間37℃でインキュベートした。培養物を、6日目に31℃にした。7日目に、培養物に、1回、グルタミンを欠く5体積%の培地5をフィードした。培養物を、毎日、細胞密度、抗GDF−8力価、乳酸塩およびアンモニウムレベルに関して測定した。
図30、31、32および33は、それぞれ、種々の実験条件下での実験における、抗GDF−8細胞の細胞成長、抗GDF−8力価、乳酸レベルおよびアンモニウムレベルを示す。
実施例8において、アスパラギンレベルに拘わらず、初期濃度4mMのグルタミンを含有する培地9が、1mMのグルタミンを含有する培地よりも良好であることが実証された。この実施例は、13mMグルタミンレベルおよび種々のアスパラギンレベルを含有する培地の影響を実証する。
抗GDF−8細胞を、表16に示す異なる濃度のグルタミンおよびアスパラギンを有する修飾培地9において、1Lバイオリアクター中で12日間培養した。この培養物を、最初の3日間37℃でインキュベートした。ついで、培養物を、4日目に31℃にシフトした。7日目に、培養物に、1回、グルタミンを欠く5体積%の培地5をフィードした。培養物を、毎日、細胞密度、細胞生存率、乳酸塩、アンモニウムレベルおよびグルタミンレベル、抗GDF−8力価およびオスモル濃度に関して測定した。
図34、35、36、37、38、39および40は、それぞれ、それぞれ、種々の実験条件下での実験における、抗GDF−8細胞の細胞成長、抗GDF−8細胞の生存率パーセント、乳酸レベル、アンモニウムレベル、グルタミンレベル、抗GDF−8力価およびオスモル濃度を示す。
実施例2、5および6において、グルタミン欠乏条件下で成長した培養物が、培養プロセスの最後に、低い乳酸塩およびアンモニウムレベルを示すことが見出された。しかしながら、非グルタミン欠乏条件下で培地9にて成長した培養物が、培養プロセスの最後に、さらに低い乳酸塩およびアンモニウムレベルを示した。この効果は、グルタミン欠乏が乳酸塩およびアンモニウムのレベルの低下に必要であると考えられる培地1のような他の培地では観察されなかった。培地9と培地1は、アスパラギン(培地9において20mMに対して、培地1+フィードで合計11mM)および酸性システイン(培地9において1.5mMに対して、培地1において0.95mM)のレベルの点で異なる。この実施例は、これら2つの成分が培養の終わりで観察される乳酸塩およびアンモニウムレベルの減少に関与するかどうかを試験する。
抗GDF−8細胞を、1Lのバイオリアクターで12日間培養した。細胞を、最初に37℃で培養し、4日目または5日目に、8〜10×106/mlで、31℃にシフトした。表17に、試験した種々の実験条件を示す。試料を毎日採取し、力価分析をタンパク質A HPLCで行った。
培地9において成長した抗GDF−8細胞は、培養を4mMまたは13mMのグルタミンのいずれかで開始したかに拘わらず、培養工程の最後で、低い乳酸塩およびアンモニウムレベルを示した(図42および43を参照)。対照的に、培地1は、培養が4mMのグルタミンで開始された場合にのみ培養工程の最後で、低い乳酸塩およびアンモニウムレベルを示した(図42および43を参照)。追加のアスパラギンおよびシステインを13mMのグルタミンを含有する培地1に添加すると、培養工程の最後で、乳酸塩およびアンモニウムレベルは低くなかった(図42および43を参照)。
実施例10は、培地1および培地9間のアスパラギンおよびシステインレベルの違いが、グルタミンが欠乏していない培地9における培養工程の最後での乳酸塩およびアンモニウムレベルの減少に関係しているかどうかを試験する。これらの因子が観察された減少に関与していないことが示された。培地1および培地9は、また、これらのアミノ酸およびビタミン濃度が異なる。この実施例は、2つの培地間のアミノ酸およびビタミン濃度の違いが、観察された減少に関係しているかどうかを試験する。
抗GDF−8細胞を、1Lのバイオリアクター中で12日間試験した。細胞を、最初に37℃で培養し、4日目に8〜10×106/mlで31℃にシフトした。表18に、種々の試験した実験条件を示す。アミノ酸、ビタミン、ヒドロコルチゾンおよびプトレシン、微量元素E(組成を表19に示す)および鉄を、種々の実験培地1条件に加え、これらの成分のレベルを培地9のレベルと等しくした。試料を毎日採取し、タンパク質A HPLCにより力価分析を行った。
試験したすべての条件は、付加的なアミノ酸を含有する培地1を除いて培養工程の最後に、低い乳酸塩およびアンモニウムレベルを示し、培地1と比較して高い培地9のアミノ酸レベルは、恐らく、乳酸塩およびアンモニウムレベルの減少に関与していないことを示している(図49および50を参照)。しかしながら、付加的なビタミン、ヒドロコルチゾンおよびプトレシン、微量元素Eおよび鉄を含有する培地1は、付加的なアミノ酸を含有する培地1を比較して、培養工程の最後に、より低い乳酸塩およびアンモニウムレベルを示した(図49および50を参照)。これは、これらの成分が、培地9で観察された減少に関与しうることを示している。
実施例11において、培地1に対して高いレベルの培地9におけるビタミン、ヒドロコルチゾンおよびプトレシン、微量元素Eおよび鉄は、培養工程の最後に観察される乳酸塩およびアンモニウムレベルの減少に関与しうることが決定された。ここでは、これらの成分を、個々におよび組み合わせて、減少に影響を与えるかを決定するために試験する。
抗GDF−8細胞を、1Lのバイオリアクター中で12日間培養した。細胞を、最初37℃で培養し、4日目に約6×106細胞/mlでシフトする、微量元素Eを含有する培地1を除いて、4日目に8〜10×106細胞/mlで31℃にシフトした。表20に、試験した種々の実験条件を示す。ヒドロコルチゾンおよびプトレシンを、すべての培地1条件に加え、これらの成分のレベルを培地9のレベルと等しくした。ビタミン、微量元素E(組成は表19に示す)および鉄を種々の実験培地1条件に加え、これらの成分のレベルを培地9のレベルと等しくした。試料を毎日採取し、タンパク質A HPLCにより力価分析を行った。
試験したすべての条件の内、13mMのグルタミンを含有する培地9および微量元素Eを含有する培地1だけが、培養工程の最後で、乳酸塩およびアンモニウムのレベルの減少を示した(図54および55を参照)。微量元素Eを含有する培地1で観察されたレベルの減少は、この培養物が、細胞が約6×106細胞/mLである時に温度シフトされた事実によるものでありうることを注意すべきである。
13mMのグルタミンを含有する培地9は、培地1構成物より高い抗GDF−8力価を示した。
この実験は、培地1、3および9を用いる細胞成長および抗GDF−8力価の違いを測定するために行った。
抗GDF−8細胞は、種々の培地、表21に記載のフィード条件下で培養した。関連する培地の情報を表22に示す。細胞を1Lのバイオリアクター中で12日間成長させ、4日目に37℃から31℃にシフトした。
培地9で培養した抗GDF−8細胞は、最も高い細胞密度および抗GDF−8力価を示し、一方、培地3で培養した抗GDF−8細胞は、最も低い細胞密度および抗GDF−8力価を示した(図57および58を参照)。培地9が培地1よりも結果が優れているという事実から、複数回フィードにより培地成分を供給するより、むしろ出発培地に培地成分を提供することがより好ましいことが示されている。加えて、培地1および培地9の両方が、培地3よりも良好に行われるという事実から、70mM未満の濃度でアミノ酸を提供するよりも、約70mMより高い濃度でアミノ酸を提供することがより優れた結果をもたらすことが示されている。最後に、約70mMより高い濃度で出発物質にアミノ酸を提供することが、最も高い細胞密度および力価を与えた(培地1と培地9を比較した)。
物質および方法
抗GDF−8細胞を、1Lのバイオリアクター中で培養し、表23に示す日に37℃から31℃にシフトした。最終力価を、グルタミン単独の最適レベルおよびグルタミンおよびアスパラギンを合わせた最適レベルを決定するために、T−テストに付した。表23に、培地9で成長した抗GDF−8細胞の関連実験条件および最終結果を要約した。
図59は、種々のレベルのグルタミン単独およびグルタミンおよびアスパラギンの組合せの推定抗GDF−8力価を示す。表24は、2〜15mMのグルタミンレベルおよびこの範囲外のグルタミンレベルの標準化力価を比較する、T−試験の結果を示す。表25は、16〜36mMのグルタミンおよびアスパラギンレベルの組合せおよびこの範囲外のレベルのグルタミンおよびアスパラギンの組合せを比較するT−試験の結果を示す。
この実施例は、高密度播種培養物を用いる中規模での3つの細胞培養培地バリエーションの能力を評価する。試験したすべての培地は、小規模バイオリアクターデータに基づいて、フェーズ1培地(培地11フィード培地をフィードした培地10)において改善が見られることが期待された。
ヒト化抗AベータペプチドIgG1モノクローナル抗体(「抗Aベータ細胞」)を発現するCHO細胞を、表26に示すように、種々の培地で試験した(Basi et al., Humanized Antibodies that Recognize Beta Amyloid Peptide、WO02/46237を参照)。pHの下点を、フェーズ1を除いて0.95MのNa2CO3+0.05MのK2CO3で制御して7.0に設定し、フェーズ1は、0.58Mの重炭酸ナトリウムおよび0.71Mの炭酸ナトリウムを含有する溶液で制御した。溶存酸素を、必要により空気を散布することにより30%に制御し、60rpmで撹拌し、フィード培地は培地5(上記のように、グルタミンを含む、または含まない)であった。03P49B501を除くすべての培養物を130L規模で成長させ、03P49B501は500L規模で成長させた。簡単には、培地1は、相対的な取り込み速度の考慮なしにすべての栄養物が豊富であり、一方、培地12は、栄養分が無差別に豊富なバージョンから明らかに不必要な栄養分を除去することによりバランスをとった。培地10、11および12の比較を表27に示す。
これらの実験を通じて、培地の変更により一様に改善された。細胞成長、生存率、低乳酸レベル、低アンモニウムレベルおよび力価の点で、低グルタミンレベルは、高いもの(図60〜64を参照)よりもより優れており、バランス化(バッチ)培地は、リッチ培地(培地1、図60〜64を参照)よりも優れていた。高密度接種で開始した培養物は、低密度接種で開始した培養物よりも高い最終力価を示した(図64を参照)。
物質および方法
IgG1のFc部位に結合したヒト75キロダルトン(p75)腫瘍壊死因子受容体(TNFR)の細胞外リガンド−結合部からなる二量体融合タンパク質を発現するCHO細胞(「TNFR−Ig細胞」)を、潅流バイオリアクターから高密度で播種し、生産バイオリアクター工程に関して培地9で3×106生存細胞/mlに希釈した。
図66、67、68、69、70、71および72は、それぞれ、細胞成長、細胞生存率、残存グルコース、グルタミンレベル、乳酸塩濃度、アンモニウム濃度および相対生産力価を示す。プロセスの軽い修飾の範囲では、すべての条件で、良好な細胞成長、高い細胞生存率および高い総最終力価が得られた。
物質および方法
培養のサイズが、関連する培養特性に影響を与えるかどうかを決定するために、抗GDF−8細胞を、小規模1リットルバイオリアクターまたは大規模6000リットルバイオリアクターで成長させた。細胞を37℃で成長させ、4日目に31℃にシフトした。
図73、74、75および76(それぞれ、細胞密度、力価、乳酸レベルおよびアンモニウムレベルを示す)に見ることができるように、6000リットルの大規模および1リットルの小規模培養間にこれらの特定に関して違いはなかった。乳酸塩およびアンモニウムレベルの両方とも、4日目に温度をシフトした後減少が始まった。これらの実施例は、培養物が同様の成長条件に付される場合、培養の規模が、細胞密度、細胞生存率、乳酸レベルおよびアンモニウムレベルに影響を与えないことを実証している。

細胞培養条件下で発現し、抗−Aベータをコードする遺伝子を含有する哺乳動物細胞;および
グルタミンを含有し、(i)約70mMを超える単位体積あたりの累積アミノ酸量、(ii)約2未満の累積アスパラギンに対するモル累積グルタミン比、(iii)約0.2未満の総累積アミノ酸に対するモル累積グルタミン比、(iv)約0.4〜1の総累積アミノ酸に対するモル累積無機イオン比、(v)約16mMを超える単位体積あたりのグルタミンおよびアスパラギンを合わせた累積量、およびその組合せからなる群から選択される培地特性を有する培地;

を含む細胞培養物を供給すること;
初期成長フェーズにおいて、該培養物が第1の培養条件下に維持された場合に、可能な最大生存細胞密度の約20%〜80%の範囲内まで該細胞に再生産させるのに十分な第1の期間、第1の培養条件下で該培養物を維持すること;

グルタミンを含有し、i)約70mMを超える出発アミノ酸濃度、ii)約2未満のアスパラギンに対するモルグルタミン比、iii)約0.2未満の総アミノ酸に対するモルグルタミン比、iv)約0.4〜1の総アミノ酸に対するモル無機イオン比およびv)約16mMを超えるグルタミンとアスパラギンの組合せ濃度からなる群から選択される少なくとも3つの培地特性を有する合成培地;
を含む細胞培養物を供給すること;
初期成長フェーズにおいて、該培養物が第1の培養条件下に維持された場合に、可能な最大生存細胞密度の約20%〜80%内で該細胞に再生産させるのに十分な第1の期間、第1の培養条件下、該培養物を維持すること;
少なくとも1つの培養条件を変更して、第2の培養条件を適用すること;
第2の条件下、第2の期間、該培養物を維持して、抗−Aベータを細胞培養物中に堆積させること;
を含む方法。 - 大規模生産細胞培養における抗−Aベータの製造方法であって、下記工程:
細胞培養条件下で発現し、抗−Aベータをコードする遺伝子を含有する哺乳動物細胞;および
グルタミンを含有し、i)約70mMを超える出発アミノ酸濃度、ii)約2未満のアスパラギンに対するモルグルタミン比、iii)約0.2未満の総アミノ酸に対するモルグルタミン比、iv)約0.4〜1の総アミノ酸に対するモル無機イオン比およびv)約16mMを超えるグルタミンとアスパラギンの組合せ濃度からなる群から選択される少なくとも4つの培地特性を有する合成培地;
を含む細胞培養物を供給すること;
初期成長フェーズにおいて、該培養物が第1の培養条件下に維持された場合に、可能な最大生存細胞密度の約20%〜80%内で該細胞に再生産させるのに十分な第1の期間、第1の培養条件下、該培養物を維持すること;
少なくとも1つの培養条件を変更して、第2の培養条件を適用すること;
第2の条件下、第2の期間、該培養物を維持して、抗−Aベータを細胞培養物中に堆積させること;
を含む方法。 - 大規模生産細胞培養における抗−Aベータの製造方法であって、下記工程:
細胞培養条件下で発現し、抗−Aベータをコードする遺伝子を含有する哺乳動物細胞;および
グルタミンを含有し、i)約70mMを超える出発アミノ酸濃度、ii)約2未満のアスパラギンに対するモルグルタミン比、iii)約0.2未満の総アミノ酸に対するモルグルタミン比、iv)約0.4〜1の総アミノ酸に対するモル無機イオン比、およびv)約16mMを超えるグルタミンとアスパラギンの組合せ濃度により特徴付けられる合成培地;
を含む細胞培養物を供給すること;
初期成長フェーズにおいて、該培養物が第1の培養条件下に維持された場合に、可能な最大生存細胞密度の約20%〜80%の範囲内まで該細胞に再生産させるのに十分な第1の期間、第1の培養条件下で該培養物を維持すること;
少なくとも1つの培養条件を変更して、第2の培養条件を適用すること;
第2の条件下、第2の期間、該培養物を維持して、抗−Aベータを細胞培養物中に堆積させること;
を含む方法。 - 大規模生産細胞培養における抗−Aベータの製造方法であって、下記工程:
細胞培養条件下で発現し、抗−Aベータをコードする遺伝子を含有する哺乳動物細胞;および
グルタミンを含有し、約16mMを超える単位体積あたりのグルタミンおよびアスパラギンを合わせた累積量を有する培地;
を含む細胞培養物を供給すること;
初期成長フェーズにおいて、該培養物が第1の培養条件下に維持された場合に、可能な最大生存細胞密度の約20%〜80%内で該細胞に再生産させるのに十分な第1の期間、第1の培養条件下、該培養物を維持すること;
少なくとも1つの培養条件を変更して、第2の培養条件を適用すること;
第2の条件下、第2の期間、該培養物を維持して、抗−Aベータを細胞培養物中に堆積させること;
を含む方法。 - 該培地が、グルタミンを含有し、(i)約70mMを超える出発アミノ酸濃度、(ii)約2未満の出発アスパラギンに対する出発グルタミン比、(iii)約0.2未満の出発総アミノ酸に対するモル出発グルタミン比、(iv)約0.4〜1の出発総アミノ酸に対するモル出発無機イオン比、(v)約16mMを超える出発グルタミンと出発アスパラギンの組合せ濃度、およびその組合せからなる群から選択される培地特性を有する培地を含む、請求項1記載の方法。
- 該乳酸レベルが、該培地特性を欠くが他は同一の培地における他は同一の条件下で観察される乳酸レベルよりも低く、
該アンモニウムレベルが、該培地特性を欠くが他は同一の培地における他は同一の条件下で観察されるアンモニウムレベルよりも低く、
生産された抗−Aベータの総量が、該培地特性を欠くが他は同一の培地における他は同一の条件下で観察される量より少なくとも多い、
請求項1〜6または70〜75いずれか1項記載の方法。 - 該培養物に、該抗−Aベータの生産の間さらなる成分を補足しない、請求項1記載の方法。
- 該培養物に、該抗−Aベータの生産の間さらなるグルタミンを補足しない、請求項1記載の方法。
- 該培養物におけるグルタミン濃度が、第2の培養条件に変更する段階の前に実質的に無くなる、請求項1記載の方法。
- 該培養物におけるグルタミン濃度が、第2の培養条件に変更する段階とほぼ同時に実質的に無くなる、請求項1記載の方法。
- 該培養物において、グルタミンの代わりにグリシルグルタミンを用いる、請求項1記載の方法。
- 該培地が、(i)約70mMを超える単位体積あたりの累積アミノ酸量、(ii)累積アスパラギンに対して約2未満の比であるモル累積グルタミン、(iii)総累積アミノ酸に対して約0.2未満の比であるモル累積グルタミン、(iv)総累積アミノ酸に対して約0.4〜1の比であるモル累積無機イオンおよび(v)約16mMを超えるグルタミンおよびアスパラギンを合わせた累積量を含有する、請求項1記載の方法。
- 該培地が、(i)約70mMを超える単位体積あたりの累積アミノ酸量、(ii)総累積アミノ酸に対して約0.2未満の比であるモル累積グルタミン、(iii)総累積アミノ酸に対して約0.4〜1の比であるモル累積無機イオンおよび(iv)約16amMを超える単位体積あたりのグルタミンおよびアスパラギンを合わせた累積量を含有する、請求項1記載の方法。
- 該培地における、単位体積あたりのヒスチジン、イソロイシン、ロイシン、メチオニン、フェニルアラニン、プロリン、トリプトファン、チロシンおよびプロリンの総累積量が約25mMを超える、請求項1記載の方法。
- 該培地における、単位体積あたりのヒスチジン、イソロイシン、ロイシン、メチオニン、フェニルアラニン、プロリン、トリプトファン、チロシンおよびプロリンの総累積量が約35mMを超える、請求項1記載の方法。
- 該培地における、単位体積あたりのヒスチジン、イソロイシン、ロイシン、メチオニン、フェニルアラニン、プロリン、トリプトファン、チロシンおよびプロリンの初期総量が約25mMを超える、請求項1記載の方法。
- 該培地における、単位体積あたりのヒスチジン、イソロイシン、ロイシン、メチオニン、フェニルアラニン、プロリン、トリプトファン、チロシンおよびプロリンの初期総量が約35mMを超える、請求項1記載の方法。
- 該培地が:
(i)約1.7mMを超える、単位体積あたりのヒスチジンの総累積量;
(ii)約3.5mMを超える、単位体積あたりのイソロイシンの総累積量;
(iii)約5.5mMを超える、単位体積あたりのロイシンの総累積量;
(iv)約2.0mMを超える、単位体積あたりのメチオニンの総累積量;
(v)約2.5mMを超える、単位体積あたりのフェニルアラニンの総累積量;
(vi)約2.5mMを超える、単位体積あたりのプロリンの総累積量;
(vii)約1.0mMを超える、単位体積あたりのトリプトファンの総累積量;
(viii)約2.0mMを超える、単位体積あたりのチロシンの総累積量;および
(ix)約2.5mMを超える、単位体積あたりのプロリンの総累積量、
からなる群から選択される培地特性を有する、請求項1記載の方法。 - 該培地が:
(i)約1.7mMを超える、単位体積あたりのヒスチジンの初期容量;
(ii)約3.5mMを超える、単位体積あたりのイソロイシンの初期容量;
(iii)約5.5mMを超える、単位体積あたりのロイシンの初期容量;
(iv)約2.0mMを超える、単位体積あたりのメチオニンの初期容量;
(v)約2.5mMを超える、単位体積あたりのフェニルアラニンの初期容量;
(vi)約2.5mMを超える、単位体積あたりのプロリンの初期容量;
(vii)約1.0mMを超える、単位体積あたりのトリプトファンの初期容量;
(viii)約2.0mMを超える、単位体積あたりのチロシンの初期容量;および
(ix)約2.5mMを超える、単位体積あたりのプロリンの初期容量、
からなる群から選択される培地特性を有する、請求項1記載の方法。 - 該培地における単位体積あたりのセリンの総累積量が約7mMを超える、請求項1記載の方法。
- 該培地における単位体積あたりのセリンの総累積量が約10mMを超える、請求項1記載の方法。
- 該培地における単位体積あたりのアスパラギンの総累積量が約8mMを超える、請求項1記載の方法。
- 該培地における単位体積あたりのアスパラギンの総累積量が約12mMを超える、請求項1記載の方法。
- 該培地における単位体積あたりのアスパラギンの総累積量が約8mMを超える、請求項1記載の方法。
- 該培地における単位体積あたりのアスパラギンの総累積量が約12mMを超える、請求項1記載の方法。
- 該培地における単位体積あたりのリンの総累積量が約2.5mMを超える、請求項1記載の方法。
