Claims (10)
- 1.付着性動物細胞を培養するための方法であって、該方法において付着性動物 細胞が培養容器に入れた栄養培地で浮遊状態で培養される、付着性動物細胞の培 養方法において、該栄養培地中の総無機イオンおよび総アミノ酸のモル比が、殆 どまたは全く細胞凝集を起こさない水準に維持されていることを特徴とする付着 性動物細胞を培養するための方法。
- 2.栄養培地中の総無機イオンおよび総アミノ酸のモル比が、約10:1から約 1:1の範囲内に維持される請求の範囲第1項に記載の方法。
- 3.栄養培地中の総無機イオンおよび総アミノ酸のモル比が、総ナトリウムイオ ン濃度を75から120mMの範囲内に、総塩化物イオン濃度を50から90m Mの範囲内に、さらに総アミノ酸含量を20から50mMの範囲内に調節するこ とによって達成される請求の範囲第1項または第2項に記載の方法。
- 4.該付着性動物細胞が哺乳類細胞である先行するいずれかの請求項に記載の方 法。
- 5.該哺乳類細胞がヒト、ラット、マウスまたはハムスター起源である請求の範 囲第4項に記載の方法。
- 6.該哺乳類細胞がチャイニーズハムスター卵巣細胞である請求の範囲第4項に 記載の方法。
- 7.付着性動物細胞の浮遊培養に使用するための栄養培地であって、資化性炭素 源、窒素、アミノ酸、無機イオン、微量元素並びに任意に脂質および成長促進物 質または調整物質を混合状態で含む栄養培地において、該培地中に存在する総無 機イオンおよび総アミノ酸のモル比が、約10:1から約1:1の範囲にあるこ とを特徴とする付着性動物細胞の浮遊培養に使用するための栄養培地。
- 8.総無機イオンおよび総アミノ酸のモル比が、約5:1から約1:1の範囲に ある請求の範囲第7項に記載の栄養培地。
- 9.総ナトリウムイオン濃度が75から120mMの範囲内にあり、総塩化物イ オン濃度が50から90mMの範囲内にあり、さらに総アミノ酸濃度が20から 50mMの範囲内にある請求の範囲第7項または第8項に記載の栄養培地。
- 10.細胞培養によって動物細胞生成物を得るための方法であって、(1)培養 容器に入れた栄養培地中で浮遊状態で前記生成物を産生する付着性動物細胞を培 養し、(2)前記生成物が蓄積するまで培養を続け、さらに(3)前記生成物を 回収する工程を含む動物細胞生成物を得るための方法において、栄養培地中の総 無機イオンおよび総アミノ酸のモル比が、殆どまたは全く細胞凝集を起こさない 水準に維持されていることを特徴とする、細胞培養によって動物細胞の生成物を 得るための方法。
