Claims (15)

NAD(H)依存フマル酸還元酵素をコードするヌクレオチド配列の発現に際して、NAD(H)依存フマル酸還元酵素が細胞質ゾル中で活性である、細胞。 - 細胞がコハク酸の形成を触媒する酵素をコードするヌクレオチド配列を発現し、ヌクレオチド配列がNAD(H)依存フマル酸還元酵素をコードして、配列番号3、および/または配列番号6のアミノ酸配列と少なくとも70%の配列同一性を有するアミノ酸配列を含む、請求項1に記載の細胞。
- NAD(H)依存フマル酸還元酵素がトリパノソーマ(Trypanosoma)種に由来する、請求項1または2に記載の細胞。
- ピルビン酸カルボキシラーゼをコードするヌクレオチド配列を過剰発現する、請求項1〜3のいずれか一項に記載の細胞。
- 異種のホスホエノールピルビン酸カルボキシキナーゼをコードするヌクレオチド配列をさらに含む、請求項1〜4のいずれか一項に記載の細胞。
- リンゴ酸デヒドロゲナーゼをコードするヌクレオチド配列の発現に際して、細胞質ゾル中で活性であるリンゴ酸デヒドロゲナーゼをコードするヌクレオチド配列をさらに含む、請求項1〜5のいずれか一項に記載の細胞。
- リンゴ酸のフマル酸への変換を触媒する酵素をコードするヌクレオチド配列の発現に際して、細胞質ゾル中でリンゴ酸のフマル酸への変換を触媒する酵素をコードするヌクレオチド配列をさらに含む、請求項1〜6のいずれか一項に記載の細胞。
- ジカルボン酸輸送体をコードするヌクレオチド配列をさらに含む、請求項1〜7のいずれか一項に記載の細胞。
- アルコールデヒドロゲナーゼをコードする少なくとも1つの遺伝子が機能性でない、請求項1〜8のいずれか一項に記載の細胞。
- グリセロール−3−リン酸デヒドロゲナーゼをコードする少なくとも1つの遺伝子が機能性でない、請求項1〜9のいずれか一項に記載の細胞。
- コハク酸デヒドロゲナーゼをコードする少なくとも1つの遺伝子が機能性でない、請求項1〜10のいずれか一項に記載の細胞。
- アスペルギルス(Aspergillus)、好ましくはアスペルギルス・ニガー(Aspergillus niger)である、請求項1〜11のいずれか一項に記載の細胞。
- サッカロミセス・セレヴィシエ(Saccharomyces cerevisiae)である、請求項1〜11のいずれか一項に記載の細胞。
- 請求項1〜13のいずれか一項に記載の真核細胞を、コハク酸が調製される適切な発酵培地中で発酵させるステップを含む、コハク酸を調製する方法。
- 請求項14に記載の方法によって得られる、コハク酸を含む発酵ブロス。
