Abstract

Description
1.技術分野
本発明は、概して生体液を分析するためのチェンバに関し、特に生体液内の粒子状物質の計数を可能にするチェンバに関する。
完全血球算定(CBC)は、全血に対する最も頻繁に行われる試験の一式であり、とりわけ白血球数(WBC)、赤血球数(RBC)、および血小板数などの多数の個別分析を含む。使用される方法は、一式の検体の完全性、機器の複雑性および費用、および試験ごとの費用によって異なる。米国特許第4,156,570号に説明されるQBC(登録商標)方法などの、最も複雑でない方法は資本費用が最も安く、実施しやすいが、典型的に試験ごとの費用がより高い。QBC(登録商標)方法は、操作者の訓練が最小限であり、一日にわずかな試験しか行われない治療場面での状況に最も適している。反対に、病院または参照試験所で使用される大量血液分析器の資本費用は20倍高いが、大量方式で使用すると比較的に低い試験ごとの費用となり得て、そのような設定ではより費用効率が高くなる。

図1−5を参照して、生体液を分析するための本装置は、以下、チェンバ高さ16と呼ばれる距離によって互いから分離される、第1の平面部材および第2の平面部材によって画定される1つ以上のチェンバ2を含む。第1の平面部材および第2の平面部材のうちの少なくとも1つは、チェンバ2内に配置される生体液試料を画像化することができるように、十分に透明である。本発明の説明を容易にするために、平面部材は、以下、「最上」平面部材4および「底」平面部材3と呼ばれる。以下、最上平面部材4はまた、透明であるとしても説明される。ほかの実施例では、底平面部材3は、最上平面部材4よりもむしろ、または最上平面部材4に加えて、透明であってもよい。
Claims (25)
- 生体液試料内の1つ以上の特定要素を計数するための方法であって、
部材は略均一な高さによって互いから分離される、透明である第1の平面部材と第2の平面部材との間に形成されるチェンバを提供するステップと、
前記チェンバ高さは、前記試料が少なくとも前記チェンバの一部分に対して前記第1および第2部材間に延在するようなサイズであって、前記チェンバ高さは、前記チェンバ内に導入されると前記特定要素が前記試料内で不均一に分布するように、前記特定要素と相対的なサイズである、前記生体液試料を前記チェンバに導入するステップと、
前記チェンバ内の前記試料の実質的全体を検査して、前記特殊要素の少なくとも1つの全体を計数するステップと、
前記チェンバ内に含まれる試料の体積を決定するステップと、
単位体積ごとの前記特定要素の少なくとも1つの数を決定するステップと、
を含む方法。 - 前記生体液試料は、抗凝固処理全血液である、請求項1に記載の方法。
- 検査する、体積を決定する、および数を決定する前記ステップは、デジタル画像分析を利用する、請求項2に記載の方法。
- 前記特定要素の前記計数された少なくとも1つは白血球を含む、請求項3に記載の方法。
- 前記特定要素の前記少なくとも1つは、識別可能となり別々に計数されるように選択的に染色される表面エピトープを有する、白血球の小集団である、請求項4に記載の方法。
- 検査する、体積を決定する、および数を決定する前記ステップは、デジタル画像分析を利用する、請求項1に記載の方法。
- 前記試料内に不均一に分布される特定要素の全ては計数される、請求項1に記載の方法。
- 生体液試料内の1つ以上の特定要素を計数するための方法であって、
チェンバは既知の体積を有し、部材は略均一な高さによって互いから分離される、透明である第1の平面部材と第2の平面部材との間に形成されるチェンバを提供するステップと、
前記チェンバ高さは、前記試料が実質的に前記チェンバの全範囲にわたって前記第1および第2部材間に延在するようなサイズであって、前記チェンバ高さは、前記チェンバ内に導入されると前記特定要素が前記試料内で不均一に分布するように、前記特定要素と相対的なサイズである、前記生体液試料を前記チェンバに導入するステップと、
前記チェンバ内の前記試料の実質的全体を検査して、前記特殊要素の少なくとも1つの全体を計数するステップと、
単位体積ごとの前記特定要素の少なくとも1つの数を決定するステップと、
を含む方法。 - 前記生体液試料は抗凝固処理全血液である、請求項8に記載の方法。
