Abstract

本発明の分野は、概略的には、抗CD26抗体および他のポリペプチド、特には、ヒト化抗CD26抗体ならびにCD26の発現に関連する疾患の治療のための抗体および他のポリペプチドを使用するための方法に関する。
CD26は、広範に分布する110kDaの細胞表面糖タンパク質である。CD26は、当初、T細胞活性化抗原として定義された(Foxら(1984) J. Immunol. 133, 1250-1256、 Fleischer (1987) J. Immunol. 138, 1346-1350、およびMorimotoら(1989) J. Immunol. 143, 3430-3439)。この分子は、その細胞外ドメインにおいてジペプチジルペプチダーゼIV(DPPIV;EC3.4.14.5)活性および広範な組織分布を有することが示された(Hegenら(1990) J. Immunol. 144, 2908-2914およびUlmerら(1990) J. 本発明は、本明細書中に記載される抗体等のポリペプチドをコードするポリヌクレオチドをさらに提供する。いくつかの実施形態では、ポリヌクレオチドは、本明細書中に記載される重鎖可変領域または軽鎖可変領域を含むポリペプチドをコードする。ポリヌクレオチドを含む、発現ベクター等のベクターもまた提供される。本発明のベクターおよび/またはポリヌクレオチドを含む宿主細胞がさらに提供される。いくつかの実施形態では、ポリヌクレオチドは単離される。
本発明は、本明細書中に記載されるポリペプチド(たとえば抗体)および/またはポリヌクレオチドを含む組成物をさらに提供する。たとえば、本発明は、本明細書中に記載されるポリペプチドを含む医薬組成物を提供する。ポリペプチドを含むキットもまた提供される。
本発明のポリペプチド(抗体等)は、診断方法および治療処置方法を含むがこれらに限定されない様々な用途に有用である。CD26を発現する細胞の増殖を阻害するための方法もまた提供される。
CM03 Fabの結合親和性およびVHおよびVLの配列
CM03(本明細書中において14D10とも言われる)は、ヒトCD26に対して産生されたマウスモノクローナル抗体である。CM03のVHおよびVLの配列は、後述の実施例における変異体を設計するためのリード物として使用した。マウスモノクローナル抗体CM03のVLおよびVHの配列にそれぞれ対応する軽鎖可変領域(VL)および重鎖可変領域(VH)を含むFabもまた生産した。このCM03 Fabは、本明細書中でしばしば「X369」または「CM03 X369」等と呼ばれる。
ヒト化CM03変異体のFab
A.配列
CM03抗体の重鎖可変領域および軽鎖可変領域をヒト化のために使用し、様々な変異体重鎖可変領域および変異体軽鎖可変領域を生成した。
大腸菌細胞株は、図7〜10に示される重鎖軽鎖Fab変異体を発現させるために使用した。
以下の表3は、Fab X369(CM03 VHおよびVL)、X391、X392、X396、ならびにX399の様々な理論的な物理的特性の概要を示す。たとえば、分子量、荷電した残基の数、疎水性残基、等電点、および全体電荷を示す。
CM03 Fab X369、およびCM03 VHに連結されたX377 VL変異体を含むFabの特異性分析を行った。データを図12A、12B、および12Cに示す。データは、Biacore(登録商標)技術を使用して作成した。CD26結合パートナーは、以下に記載されるとおりであった。
以下の表4は、CM03 X369およびCM03 VL変異体X376、X377、X378、X379、X380、またはX381のそれぞれと組み合わせられたCM03 VHを含むFabの、KDを含む親和性分析の概要を示す。このデータは、Biacore(登録商標)技術を使用して作成した。CD26結合パートナーは、以下に示されるとおりであった。
変異体VHおよび変異体VLの配列の対を含む選択されたFabの特異性分析を行った。図14は、CD26に対する結合について、未精製腹水マウスMAb CM03(14D10)と競合したCM03 X369 Fabならびにヒト化CM03 Fab X389、X390、X391、X392、X393、X395、およびX396を示す。CD26に対する14D10の結合を、CM03変異体により予め占有されたC26に対する14D10の結合と比較した。クローン化CM03の組換えFabは陽性対照として使用した。分析は、Biacore(登録商標)を使用して、実施例1に記載されるように行った。Fabはすべて、CD26に対する14D10の結合において少なくとも80%の減少をもたらし、これは、そのFab変異体対が、14D10と類似するか少なくとも重複する結合部位を有することを示す。
ヒト化重鎖可変領域およびヒト化軽鎖可変領域の様々な対を含むFabを、細胞増殖を阻害するそれらの性能について分析した。
ヒト化CM03 IgG1抗体
A.配列
完全長ヒト化抗体は、選択されたVHおよびVLの対、ヒトIgG1重鎖定常領域、およびヒトκ軽鎖定常領域を使用して作製した。図16は、IgG1鎖間相互作用およびヒンジ領域を示す。図17Aは、重鎖定常領域アミノ酸配列を示す。図17Aはまた、重鎖におけるVH変異体およびリーダー配列の位置を示す。図17Bは、軽鎖定常領域アミノ酸配列を示す。図17Bはまた、軽鎖におけるVL変異体およびリーダー配列の位置を示す。
図18は、HEK293細胞において一過性に発現させ、プロテインA親和性カラム(Procep-A、Millipore社、カタログ#113111827)を通してメーカーのマニュアルに従って精製した完全長組換えヒト化モノクローナル抗体(rhuMAb)409、411、410、および412を示す。抗体は、100mM酢酸ナトリウムpH3.0により溶離し、次に、2M Tris pH9.0による即時中和を行った。次いで、試料を、不溶性材料を除去するためにJS4.2ローターで4,000rpmで30分間回転させ、保管前の滅菌のために0.2μmフィルターを通して濾過した。図18はまた、チャイニーズハムスター卵巣(CHO-DG44)細胞において安定して発現したrhuMAb 411を示す。各場合において、形質移入は、リポフェクトミン(lipofectomin)を介して実行した。
CD26に対するrhuMAb409、410、411、および412のCD26結合親和性を後述の表9に示す。それらの分析は、Biacore(登録商標)を使用して、結合パートナーが上記に述べられたrhuMAbであることを除き、実施例1に記載されるように実行した。rhuMAb 411は、rhuMAb409のKDの3.7倍を有していた。rhuMAb410および412は、rhuMAb409のKDの0.5倍および0.4倍をそれぞれ有していた。
特定の組換えヒトMAbの抗増殖性効果を、抗体により誘導された凝集を評価した形態学的アッセイでJurkat CD26+細胞において分析した。図21は、対照処理(上の像)と比べた、rhuMAb409(左下の像)およびrhuMAb410(右下の像)で処理した細胞の像を示す。両rhuMAbは、対照処理と比べて、細胞増殖の阻害の証拠を示した。
エピトープマッピング実験は、CD26に対するヒト化モノクローナル抗体の有望な結合部位を決定するために実行した。エピトープマッピングは、ペプチドマイクロアレイを使用して実行した。

例示的なMTTアッセイプロトコールは以下のとおりである:
Jurkat細胞を、完全長のヒトCD26をコードするプラスミドで形質移入し、200μg/ml G418下で選択する。細胞は、生存可能性を>95%に維持するために、3日毎に1:5で継代する。アッセイ前の約48時間、回転培地は、1100rpm、5分間回転させることにより新鮮な増殖培地(37℃)と完全に置換する。(アッセイ培地は以下の増殖培地と同じである: 87mL RPMI-1640液体培地(Hyclone社:SH30027-02);10mL FCS(56℃、30分間処理済);0.5mLペニシリン/ストレプトマイシン(100μg/mL);2mL G418(Calbiochem社、PBS中100mg/ml);および2mL L-グルタミン(200mM、Hyclone社、#SH30034.01)) このアプローチにより、アッセイを行うときに、細胞生存率を97〜99%とすることができる。細胞生存率は少なくとも95%でなければならない。

rhuMAb 411の結合親和性
rhuMAb 411は、グルタミン合成酵素発現系(GS系;Lonza Biologics社、スラウ、UK)を用いた哺乳動物細胞(チャイニーズハムスター卵巣)懸濁培養における発酵により生産した。ヒトCD26(hu-CD26)に対するRhuMAb 411の結合親和性を標準的なBiacore(登録商標)プロトコールを使用して決定した。ヒトCD26に対するマウスモノクローナル抗体14D10の結合親和性もまた決定した。これらの結合研究の結果を後述の表12に提示する。

Lonza Biologics社(スラウ、UK)のCHOK1SV細胞株であるGS-CHOを抗体rhuMAb 411を生産するために使用した。IgG1抗体rhuMAb 411をコードする重鎖遺伝子および軽鎖遺伝子を含む二重遺伝子ベクターで形質移入したGS-CHO細胞株を構築した。
